Environmentally-Induced (Extrinsic) Skin Aging: Exposomal Factors and Underlying Mechanisms.

As a barrier organ, the skin is an ideal model to study environmentally-induced (extrinsic) aging. In this review, we explain the development of extrinsic skin aging as a consequence of skin exposure to specific exposomal factors, their interaction with each other, and the modification of their effects on the skin by genetic factors. We also review the evidence that exposure to these exposomal factors causes extrinsic skin aging by mechanisms that critically involve the accumulation of macromolecular damage and the subsequent development of functionally altered and/or senescent fibroblasts in

Clinical Skin Aging Score and Risk of Degenerative Cardiovascular Diseases.

BACKGROUND: Skin aging can be a contributing factor in the prediction, follow-up, and early diagnosis of some disorders related to the aging process, including degenerative cardiovascular diseases. OBJECTIVE: We sought to explore the association between a clinical skin aging score and risk of degenerative heart diseases. METHODS: This study included two groups; a case group consisting of 44 patients older than 30 years who were admitted to the cardiology department with degenerative heart disease and 44 age- and sex-matched healthy individuals to act as a control group. The skin aging score wa

[Air pollution (particulate matter and nitrogen dioxide) and skin aging].

It has recently been discovered that air pollution can contribute to skin aging. This conclusion is based on both epidemiological and mechanistic evidence. Specifically, exposure to ambient relevant particulate matter and to nitrogen dioxide (NO2) is associated with an increased risk to develop facial pigment spots. In addition, genetic studies indicate the involvement of gene-environment interactions because women carrying certain genetic variants of the aryl hydrocarbon receptor (AHR) signaling pathway have a higher risk to develop facial pigment spots in response to exposure to particulate

Skin aging: molecular pathology, dermal remodelling and the imaging revolution.

Skin is a multifunctional organ but, alongside every other organ system, is subject to both intrinsic (chronological) and extrinsic (environmental) aging, resulting in a loss of functional capacity. Cutaneous aging manifests as an observable change in the external appearance of the skin, the major accelerator of the aging process being our interactions with our environment, such as chronic exposure to solar irradiation (UV, IR or visible wavelengths of light). The aim of this contribution, therefore, was to provide a review of the pathological mechanisms which may play roles in the development

Mapping epidermal and dermal cellular senescence in human skin aging.

Single-cell RNA sequencing and spatial transcriptomics enable unprecedented insight into cellular and molecular pathways implicated in human skin aging and regeneration. Senescent cells are individual cells that are irreversibly cell cycle arrested and can accumulate across the human lifespan due to cell-intrinsic and -extrinsic stressors. With an atlas of single-cell RNA-sequencing and spatial transcriptomics, epidermal and dermal senescence and its effects were investigated, with a focus on melanocytes and fibroblasts. Photoaging due to ultraviolet light exposure was associated with higher b

Targeting lymphoid-derived IL-17 signaling to delay skin aging.

Skin aging is characterized by structural and functional changes that contribute to age-associated frailty. This probably depends on synergy between alterations in the local niche and stem cell-intrinsic changes, underscored by proinflammatory microenvironments that drive pleotropic changes. The nature of these age-associated inflammatory cues, or how they affect tissue aging, is unknown. Based on single-cell RNA sequencing of the dermal compartment of mouse skin, we show a skew towards an IL-17-expressing phenotype of T helper cells, γδ T cells and innate lymphoid cells in aged skin. Protective Role of Melatonin and Its Metabolites in Skin Aging.

The skin, being the largest organ in the human body, is exposed to the environment and suffers from both intrinsic and extrinsic aging factors. The skin aging process is characterized by several clinical features such as wrinkling, loss of elasticity, and rough-textured appearance. This complex process is accompanied with phenotypic and functional changes in cutaneous and immune cells, as well as structural and functional disturbances in extracellular matrix components such as collagens and elastin. Because skin health is considered one of the principal factors representing overall "well-being

Polygoni Multiflori Radix Preparat Delays Skin Aging by Inducing Mitophagy.

BACKGROUND: As the skin is the largest organ of the human body, it is aging inevitably and produces cosmetic and psychological problems, and even disease. Therefore, the molecular mechanisms related to the prevention of skin aging need to be further explored. METHODS: Aging models were constructed by D-galactose. Mice were administrated with polygoni multiflori radix preparat (PMRP), PMRP and 3-methyladenine, or PMRP and rapamycin intragastrically. The apparent and viscera index of aged rats was measured. Then, the physicochemical property, antioxidant ability, histological structure, mitocho

Anti-Aging Potentials of Methylene Blue for Human Skin Longevity.

Oxidative stress is the major cause of skin aging that includes wrinkles, pigmentation, and weakened wound healing ability. Application of antioxidants in skin care is well accepted as an effective approach to delay the skin aging process. Methylene blue (MB), a traditional mitochondrial-targeting antioxidant, showed a potent ROS scavenging efficacy in cultured human skin fibroblasts derived from healthy donors and from patients with progeria, a genetic premature aging disease. In comparison with other widely used general and mitochondrial-targeting antioxidants, we found that MB was more effe

The skin barrier function: differences between intrinsic and extrinsic aging.

AIM: Transepidermal water loss (TEWL), stratum corneum hydration (SCH), and the skin surface pH are important parameters to characterize the skin barrier function. The aim of this exploratory study was to compare TEWL, SCH and skin surface-pH on sun-protected versus sun-exposed skin areas in relation to age. METHODS: Twenty four female subjects with healthy skin were recruited in three age groups. Lifetime sun exposure was assessed using a questionnaire. TEWL, SCH, and skin surface pH were measured on the right upper inner arm, right mid-volar forearm and right dorsal forearm under standardiz

Photoaging versus intrinsic aging: a morphologic assessment of facial skin.

Histologic studies have become increasingly important in recognizing morphologic differences in photoaged versus intrinsically aged skin. Earlier histologic studies have attempted to evaluate these changes by examining anatomical sites which are not comparable, such as face and buttocks. As part of a multicenter study, we have quantitatively examined a panel of 16 histologic features in baseline facial skin biopsies from 158 women with moderate to severe photodamage. When compared to the postauricular area (photo protected), biopsies of the crow's feet area (photo exposed) had a twofold increa

Human skin through the ages.

As the largest organ of the human body the skin offers a protective role, providing a tough but pliable covering that provides the major barrier between the internal organs and the environment. It actively regulates water loss and is both oxygen and carbon dioxide permeable, and influences temperature regulation and immunological functions through its sensory properties. Both intrinsic and enhanced environmental factors contribute to the progressive deterioration of the skin with increasing age. Cutaneous problems are therefore an unavoidable and inevitable consequence of aging skin, which can

Pigmentary changes of the ageing skin.

In subjects older than 25-30 years the number of enzymatically active melanocytes detectable by the dopa reaction decreases by about 10-20% per decade, with exposed skin having approximately twice as many pigment cells as unexposed skin. Chronic exposure to sunlight may stimulate the epidermal melanocyte system rather than accelerating chronological ageing. The number of melanocytic naevi declines with age. Despite the decreased melanocyte density, photoaged skin has irregular pigmentation and, frequently, there is hyperpigmentation. This may be due to greater positivity of dopa of chronically

Physiological background to skin ageing.

Synopsis Ageing and ultimate death, when survival offers no reproductive advantages to the species, are essential to the evolutionary process. Yet the prolongation of youth has always been a human dream. The prevention of ageing in visible areas of skin is desirable not for physiological but for social reasons; the skin proclaims our sexuality and asserts our social image. Has the aspiration any hope of success? Ageing of the skin involves changes in the dermis, epidermis, pigment cells, hair follicles, sebaceous and sweat glands, and sense organs. Some of these changes appear intrinsic to the

Microbiome-Derived Metabolic Signaling in the Gut-Skin Axis: Implications for Nutricosmetics and Healthy Skin Aging.

The gut-skin axis represents a bidirectional communication network linking the gut microbiota, microbial metabolism, immune regulation, and skin physiology. Natural bioactives derived from dietary plants, mushrooms, fermented foods, marine resources, and functional ingredients may influence skin health through both direct nutritional effects and host-microbiome metabolic interactions. This review provides a microbiome-centered, systems-level perspective on the relationships among dietary bioactives, gut microbial biotransformation, microbiome-derived metabolites, and skin aging-associated phys

Autophagy Declines with Premature Skin Aging resulting in Dynamic Alterations in Skin Pigmentation and Epidermal Differentiation.

Autophagy is a membrane traffic system that provides sustainable degradation of cellular components for homeostasis, and is thus considered to promote health and longevity, though its activity declines with aging. The present findings show deterioration of autophagy in association with premature skin aging. Autophagy flux was successfully determined in skin tissues, which demonstrated significantly decreased autophagy in hyperpigmented skin such as that seen in senile lentigo. Furthermore, an exacerbated decline in autophagy was confirmed in xerotic hyperpigmentation areas, accompanied by seve
