Continuous spike and wave in slow-wave sleep (CSWS) is an electroencephalographic (EEG) pattern characterized by generalized spike-wave discharges occurring for at least 85% of non-rapid eye movement (non-REM) sleep, with marked attenuation during rapid eye movement (REM) sleep. It has been described in a large number of structural and nonstructural neurologic conditions and is associated with epilepsy, behavioral disturbances, and severe neuropsychiatric impairment. We describe continuous spike and wave in slow-wave sleep in 2 patients (one with Rett syndrome and the other with Lhermitte-Ducl

Landau-Kleffner syndrome (LKS) and the syndrome of continuous spikes and waves during slow wave sleep (CSWS) are two points on the spectrum of functional childhood epileptic encephalopathies. They are characterized by a severe paroxysmal EEG disturbance that may permanently alter the critical synaptogenesis by strengthening synaptic contacts that should have been naturally "pruned." The much more common benign epilepsy with centrotemporal spikes is also related to LKS and CSWS by a common pathophysiology. Although prognosis in LKS and CSWS for seizure control is good, cognitive function declin

BACKGROUND: The manipulation of specific brain oscillations by applying transcranial electrical stimulation techniques in order to enhance memory processes during sleep has become an intriguing field of research. A seminal study found a positive effect of slow-oscillatory transcranial direct current stimulation (so-tDCS) on sleep-dependent consolidation of declarative memories. Since then several studies have tried to replicate this result with inconsistent findings.
OBJECTIVE/HYPOTHESIS: This study aimed to reexamine effects of so-tDCS on declarative memory observed in young participants bas

Phase coherence (λ) between respiratory sinus arrhythmia (RSA) and respiration has emerged as a promising metric for assessing the role of autonomic nervous system (ANS) activity and slow wave sleep (SWS) activities in healthy subjects. This study aims to investigate how λ and SWS activity differ between Obstructive Sleep Apnea (OSA) patients with and without major depressive disorder (MDD) during overnight sleep and explore whether the correlation between λ and SWS activity exists among those OSA patients compared to healthy individuals. Overnight electroencephalogram (EEG), electrocardiogram

BACKGROUND AND OBJECTIVE: Rebound of slow-wave sleep (SWS) and rapid eye movement (REM) sleep is observed in patients who are on continuous positive airway pressure (CPAP) therapy for obstructive sleep apnoea (OSA); but, neither have been objectively defined. The pressure titration study often represents the first recovery sleep period for patients with OSA. Our aim was to objectively define and identify predictors of SWS and REM sleep rebound following CPAP titration.
METHODS: Paired diagnostic polysomnography and pressure titration studies from 335 patients were reviewed.
RESULTS: The mean

Benefits in long-term memory retention and generalization have been shown to be related to sleep-dependent processes, which correlate with neural oscillations as measured by changes in electric potential. The specificity and causal role of these oscillations, however, are still poorly understood. Here, we investigated the potential for augmenting endogenous slow-wave (SW) oscillations in humans with closed-loop transcranial alternating current stimulation (tACS) with an aim toward enhancing the consolidation of recent experiences into long-term memory. Sixteen (three female) participants were

STUDY OBJECTIVES: Emerging evidence suggests a role for sleep in contributing to the progression of Alzheimer disease (AD). Slow wave sleep (SWS) is the stage during which synaptic activity is minimal and clearance of neuronal metabolites is high, making it an ideal state to regulate levels of amyloid beta (Aβ). We thus aimed to examine relationships between concentrations of Aβ42 in the cerebrospinal fluid (CSF) and measures of SWS in cognitively normal elderly subjects.
METHODS: Thirty-six subjects underwent a clinical and cognitive assessment, a structural MRI, a morning to early afternoon

Using topographic EEG mapping, we studied the relationships between delta activity during slow-wave sleep (SWS) and the background EEG activity during wakefulness, in 11 normal subjects and 35 stroke patients with unilateral supra-tentorial lesions. Delta-1 power during SWS showed a significant positive correlation with alpha-1 power during wakefulness, in both hemispheres. Delta-1 and delta-2 power during SWS correlated positively not only with alpha-2 power, but also with delta-1 and delta-2 power during wakefulness in the affected hemisphere. these figures indicate that the amount of delta

BACKGROUND: Increases in slow-wave activity (SWA) during sleep are associated with improvements in overnight memory retention and cognitive performance in young adults. Links between SWA and overnight improvements in cognition among older populations, however, are poorly understood. This proof-of-concept pilot study used a single session of excitatory repetitive transcranial magnetic stimulation (rTMS) during wakefulness to increase SWA during subsequent sleep in older adults with subjective cognitive decline. We examined whether this single night increase in SWA led to improvements in overnig

BACKGROUND: Military personnel must maintain physical performance despite exposure to operational stressors such as sleep loss, caloric restriction and high cognitive load. Habitual sleep and specific sleep features are positively associated with fitness and may contribute to physical performance in operational settings. Further, by affecting muscle recovery, sleep may contribute to the ability to maintain performance across multiple days of exposure to operational stressors.
OBJECTIVES: We examined the role of individual differences in baseline sleep on baseline physical performance and on c

OBJECTIVE: Continuous Spike-Wave during slow Sleep (CSWS) syndrome associates a clinically important neurocognitive regression with strong activation of non-REM sleep spikes. Its mechanisms remain unknown, but a contribution of rare perinatal thalamic injuries has been highlighted. We determine the incidence of such lesions in a cohort of CSWS patients.
METHODS: N = 65 patients with CSWS and a control group (N = 51) were studied. Spikes were quantified in long-term ambulatory EEGs, brain Magnetic Ressonance Imaging (MRI) structural lesions were assessed and thalamic volumetry was performed. A

Sleep deprivation can acutely reverse depressive symptoms in some patients with major depression. Because abnormalities in slow wave sleep are one of the most consistent biological markers of depression, it is plausible that the antidepressant effects of sleep deprivation are due to the effects on slow wave homeostasis. This study tested the prediction that selectively reducing slow waves during sleep (slow wave deprivation; SWD), without disrupting total sleep time, will lead to an acute reduction in depressive symptomatology. As part of a multi-night, cross-over design study, participants wi

The pharyngeal constrictors have been hypothesized to play an important role in the regulation of upper airway (UAW) patency in patients with sleep apnea. However, little research has focused on the activation and control of muscles that determine the lateral and posterior wall of the retropalatal airway dimensions. Our aim was to investigate the effects of slow wave sleep (SWS) and rapid eye movement (REM) sleep on the activation of pharyngeal constrictor (thyropharyngeus; TP) and dilator (stylopharyngeus; SP) muscles during eupneic breathing and induced central apneas. In nine goats, we foun

The calculation of the correlation dimension (D2) was applied to the study of the profiles of EEG slow-wave activity in nine narcoleptic subjects and nine sex- and age-matched control subjects who, following a baseline night recording, were maintained on 16 h of diurnal sleep deprivation and, thereafter, submitted to a 32-h bed rest protocol. The reversibility test allowed us to reject the null hypothesis that the time series considered in our study were generated by a static transformation of a linear Gaussian random process. Similarly, all profiles showed a positive largest Lyapunov exponent

BACKGROUND: Slow-wave activity (0.5-4 Hz electroencephalographic activity) during non-rapid eye movement sleep is consistently associated with better cognitive performance in older adults. Slow-wave activity is known to regulate synaptic plasticity and may thereby mitigate excitotoxicity and accumulation of Alzheimer's pathology. Paradoxically, longer total sleep times in older adults are often associated with poorer cognition and general health. Conventional behavioral sleep treatments robustly increase sleep efficiency and sleep time, but do not consistently enhance slow-wave activity and ha

UNLABELLED: During non-rapid eye movement (NREM) sleep, cortical neurons alternate between ON periods of firing and OFF periods of silence. This bi-stability, which is largely synchronous across neurons, is reflected in the EEG as slow waves. Slow-wave activity (SWA) increases with wake duration and declines homeostatically during sleep, but the underlying mechanisms remain unclear. One possibility is neuronal "fatigue": high, sustained firing in wake would force neurons to recover with more frequent and longer OFF periods during sleep. Another possibility is net synaptic potentiation during w

Obstructive sleep apnoea (OSA) is associated with an increased risk for cognitive impairment and dementia, which likely involves Alzheimer's disease pathology. Non-rapid eye movement slow-wave activity (SWA) has been implicated in amyloid clearance, but it has not been studied in the context of longitudinal amyloid accumulation in OSA. This longitudinal retrospective study aims to investigate the relationship between polysomnographic and electrophysiological SWA features and amyloid accumulation. From the Mayo Clinic Study of Aging cohort, we identified 71 participants ≥60 years old with OSA (

The importance of sleep as a key component of cardiovascular health has been increasingly recognized. Our previous research demonstrated that auditory-enhanced slow waves increase cardiac function, but the underlying mechanisms behind these beneficial effects remain uncertain. In this study, we examined the influence of two types of slow waves on cardiovascular functions. 18 healthy middle-aged men participated in a three-night sleep lab experiment under three conditions: two auditory slow wave enhancement conditions and one SHAM control. We found that the strength of heart rate and blood pres
