Comparative effects of TGF-β2/Smad2 and TGF-β2/Smad3 signaling pathways on proliferation, migration, and extracellular matrix production in a human lens cell line.

The signaling pathway of transforming growth factor β2 (TGF-β2)/Smad plays an important role in the pathological process in posterior capsule opacification (PCO) after cataract surgery. Smad2 and Smad3 are both receptor-regulated Smads (R-Smads) of the TGF-β2 signaling pathway. We aim to find which among Smad2, Smad3, and Smad2&3 plays a key role in PCO pathology. The signal characteristics of TGF-β2 and Smad proteins in the human lens cell line HLE-B3 were investigated. Smad2, Smad3, or Smad2&3 were silenced using small interfering RNA. We then tested cell proliferation by 3-(4,5-dimethylthia

Ligustilide Alleviates Renal Fibrosis Through Fblim1-Dependent Inhibition of the TGF-β1/Smad3 Signaling Pathway.

Renal fibrosis constitutes a central pathological hallmark of chronic kidney disease (CKD), with its progression largely mediated through the transforming growth factor‑β1 (TGF‑β1)/Smad3 signaling pathway. Ligustilide (LIG) has been reported to possess anti‑fibrotic activity; however, its protective effects and underlying molecular mechanisms in renal fibrosis remain incompletely understood. This study examined the effects of LIG and explored its underlying mechanisms in a unilateral ureteral obstruction (UUO) mouse model and TGF‑β1-stimulated mouse renal tubular epithelial cell (TCMK1) cells.

Identification of SMAD3 as a Novel Mediator of Inflammation in Human Myometrium In Vitro.

Preterm birth remains the primary cause of early neonatal death and is a major determinant for long-term health consequences. Aberrant intrauterine inflammation and infection are known to augment the synthesis of proinflammatory cytokines and induce uterine contractions, which can subsequently lead to preterm birth. The transforming growth factor-β (TGF-β) superfamily members regulate numerous cellular processes through the activation of intracellular mediators known as mothers against decapentaplegic homolog (SMADs). Studies in nongestational tissues have shown that SMAD3 plays a role in immu

Role of Klotho, an antiaging protein, in pulmonary fibrosis.

Klotho is a recently discovered antiaging protein. Although many researchers are investigating the roles of Klotho in chronic kidney diseases and cancer, however, there are no studies on the roles of Klotho in chronic pulmonary diseases. The purpose of this study was to define the role of Klotho in pulmonary fibrosis using a murine model of ovalbumin (OVA)-induced chronic asthma and in BEAS-2B human bronchial epithelial cells. In an in vivo experiment, mice were sensitized by intraperitoneal injection of OVA (20 μg/mouse), followed 1 week later by an airway challenge with 1 % OVA solution deli

The role of Smad2 and Smad3 in regulating homeostatic functions of fibroblasts in vitro and in adult mice.

The heart contains an abundant fibroblast population that may play a role in homeostasis, by maintaining the extracellular matrix (ECM) network, by regulating electrical impulse conduction, and by supporting survival and function of cardiomyocytes and vascular cells. Despite an explosion in our understanding of the role of fibroblasts in cardiac injury, the homeostatic functions of resident fibroblasts in adult hearts remain understudied. TGF-β-mediated signaling through the receptor-activated Smads, Smad2 and Smad3 critically regulates fibroblast function. We hypothesized that baseline expres

NEDD9 targets COL3A1 to promote endothelial fibrosis and pulmonary arterial hypertension.

Germline mutations involving small mothers against decapentaplegic-transforming growth factor-β (SMAD-TGF-β) signaling are an important but rare cause of pulmonary arterial hypertension (PAH), which is a disease characterized, in part, by vascular fibrosis and hyperaldosteronism (ALDO). We developed and analyzed a fibrosis protein-protein network (fibrosome) in silico, which predicted that the SMAD3 target neural precursor cell expressed developmentally down-regulated 9 (NEDD9) is a critical ALDO-regulated node underpinning pathogenic vascular fibrosis. Ablation of the renal tubular gluconeogenic enzyme PCK1 drives AKI-to-CKD transition by negatively regulating the TGF-β/Smad3 signaling pathway.

The transition from acute kidney injury (AKI) to chronic kidney disease (CKD) is characterized by progressive renal fibrosis, yet the key tubular regulatory factors driving this maladaptive repair remain inadequately defined. In this study, we investigated the role of phosphoenolpyruvate carboxykinase 1 (PCK1), the rate-limiting enzyme of gluconeogenesis, in the AKI-to-CKD transition. Analysis of human single-cell RNA sequencing (scRNA-seq) datasets revealed a profound downregulation of PCK1 in injured proximal tubule cells, which correlated with the upregulation of pro-fibrotic markers. Analysis of transforming growth factor β signaling in chronic rhinosinusitis.

BACKGROUND: It has been reported that there is a significant difference in the local tissue concentration of transforming growth factor (TGF)-β1 between chronic rhinosinusitis without nasal polyps (CRSsNP) and chronic rhinosinusitis without nasal polyps (CRSwNP) patients. TGF-β has been reported to play an important role in regulating epithelial cell repair in lower airway remodeling and may be a critical factor involved in the remodeling process of chronic rhinosinusitis (CRS). METHODS: Ethmoidal mucosal samples collected from CRS and healthy control patients were analyzed for TGF-β1, TGF-β

FKBP10 Silencing Alleviates Gluteal Muscle Contracture by Inhibiting Fibrosis and Restoring Autophagy via HSP47/SMAD3 Pathway Inactivation.

FKBP prolyl isomerase 10 (FKBP10) plays a key role in fibrosis-driven progression of gluteal muscle contracture (GMC). This study investigated the molecular mechanism of FKBP10 in GMC progression. Expression levels of FKBP10, heat shock protein 47 (HSP47), SMAD3, autophagy, and fibrosis-related indicators were analyzed for correlations. The GMC rat model was constructed by injecting methanol penicillin. The interaction between FKBP10 and HSP47 was also assessed. FKBP10 expression was up-regulated in the gluteal muscle of patients with GMC and rats, accompanied by obvious damage and fibrosis. E

Canagliflozin alleviates high glucose-induced peritoneal fibrosis via HIF-1α inhibition.

The cardioprotective effects of sodium-glucose cotransporter type 2 (SGLT2) inhibitors have been demonstrated in many studies. However, their benefits for end-stage kidney disease patients, particularly those on peritoneal dialysis, remain unclear. SGLT2 inhibition has shown peritoneal protective effects in some studies, but the mechanisms are still unknown. Herein, we investigated the peritoneal protective mechanisms of Canagliflozin in vitro by simulating hypoxia with CoCl2 in human peritoneal mesothelial cells (HPMCs) and rats by intraperitoneal injection of 4.25% peritoneal dialysate simul

TGF-β1/Smad3 Pathway Targets PP2A-AMPK-FoxO1 Signaling to Regulate Hepatic Gluconeogenesis.

Maintenance of glucose homeostasis is essential for normal physiology. Deviation from normal glucose levels, in either direction, increases susceptibility to serious medical complications such as hypoglycemia and diabetes. Maintenance of glucose homeostasis is achieved via functional interactions among various organs: liver, skeletal muscle, adipose tissue, brain, and the endocrine pancreas. The liver is the primary site of endogenous glucose production, especially during states of prolonged fasting. However, enhanced gluconeogenesis is also a signature feature of type 2 diabetes (T2D). Thus,
