Social defeat stress in rats: escalation of cocaine and "speedball" binge self-administration, but not heroin.

RATIONALE: Exposure to intermittent episodes of social defeat stress can increase drug seeking and leads to intense drug taking in rats. OBJECTIVES: This study investigated the consequences of repeated, intermittent social defeat stress on patterns of drug self-administration in rats with access to heroin, cocaine, or a heroin-cocaine combination ("speedball"). METHODS: Male Long-Evans rats were either handled (controls) or subjected to 25-min social defeat stress episodes on days 1, 4, 7, and 10 during confrontations with an aggressive resident. Ten days following the last defeat, rats were

Glucocorticoid receptors in lateral septum are involved in the modulation of the emotional sequelae induced by social defeat.

The current research studied the behavior adopted in the elevated plus maze (EPM) of rats previously subjected to a social defeat using the resident-intruder paradigm. One day after defeat, intruder animals exhibited an anxiogenic-like behavior in the EPM. In addition, we also evaluated the role of the corticosteroid receptor system (minerlocorticoid - MR - and glucocorticoid - GR - receptors) from the lateral septum (LS) on the anxiety generated by social defeat. The LS is an area of the aversive circuitry that is preferentially activated in passive defensive postures, and participates - toge

Inflammation as a mediating pathway between social defeat and mental health in humans: A systematic review.

INTRODUCTION: Social Defeat (SD), an animal-derived concept characterised by experiences of subordination, may influence biological mechanisms affecting mental health. Despite evidence linking inflammation to mental disorders and animal studies suggesting SD's inflammatory effects, to date, no review has comprehensively examined these relationships in human populations. This systematic review synthesises evidence from human studies examining human SD, inflammation, and mental health, with particular attention to inflammation as a potential mediating pathway in these relationships, to better in

Comparison of R-ketamine and rapastinel antidepressant effects in the social defeat stress model of depression.

RATIONALE: The N-methyl-D-aspartate (NMDA) receptor antagonists, including R-ketamine and rapastinel (formerly GLYX-13), show rapid antidepressant effects in animal models of depression. OBJECTIVE: We compared the rapid and sustained antidepressant effects of R-ketamine and rapastinel in the social defeat stress model. RESULTS: In the tail suspension and forced swimming tests, R-ketamine (10 mg/kg, intraperitoneal (i.p.)) or rapastinel (10 mg/kg, i.p.) significantly attenuated the increased immobility time in the susceptible mice, compared with the vehicle-treated group. In the sucrose prefe

Cannabidiol prevents social avoidance, potentiation of cocaine reward and gene expression alterations induced by exposure to intermittent social defeat in mice.

Exposure to intermittent social defeat (ISD), a model of social stress, increased anxiety- and depression-like behaviors and enhanced sensitivity of mice to the rewarding effects of cocaine. In this study, we evaluated the role of cannabidiol on these behavioral effects of ISD and ISD-induced alterations in targets of the serotonin, endocannabinoid and hypothalamus-pituitary-adrenal (HPA) systems. Male mice were treated with cannabidiol (30 or 60 mg/kg) and exposed to four episodes of social defeat on PND 47, 50, 53 and 56. Control mice were not exposed to stress. In experiment 1, on PND 57-58

Ketamine attenuates the effects of intermittent social defeat on anxiety, social interaction and cocaine-induced conditioned place preference in male mice.

Mice exposed to intermittent social defeat (ISD) stress in late adolescence exhibit short-term anxiety- and depression-like behaviours and demonstrate greater sensitivity to the rewarding effects of cocaine in adulthood. Furthermore, the development of depression-like symptoms predicts subsequent enhanced vulnerability to cocaine reward. The aim of this study was to investigate whether ketamine, a non-competitive glutamate N-methyl-D-aspartate (NMDA) receptor antagonist with antidepressant properties, could prevent the short-term effects of ISD on anxiety- and depression-like behaviours and th

A Clinically Aligned Murine Model of Electroconvulsive Stimulation Reverses Social Aversion and Displays Fear Memory Impairment After Chronic Social Defeat Stress.

Electroconvulsive therapy (ECT) is the most effective treatment for patients with major depression, bipolar depression, mania, catatonia, and schizophrenia. Nonetheless, the mechanisms underlying its therapeutic effects largely remain unknown. While previous preclinical studies have noted a role for neurotropic signaling, neurogenesis, and alterations in monoamine neurotransmitter systems, these models were largely conducted using procedures that deviate from clinical practice. Therefore, we sought to develop a clinically relevant murine model of ECT, referred to as electroconvulsive stimulati

Male Wistar rats are more susceptible to lasting social anxiety than Wild-type Groningen rats following social defeat stress during adolescence.

Adolescence is an important period for the development of adult social competences. Social stress during adolescence may contribute not only to an inadequate social development but also to adult vulnerability to social anxiety. There seems to be a clear individual differentiation, however, in the vulnerability to the long-term negative consequences of social stress. The current study further explores this individual vulnerability and is aimed at the influence of social stress during adolescence on adult social anxiety and its context specificity. Rats from different strains (Wistar and Wild-ty

Adaptive reorganization of local sleep and prelimbic cortical circuits predict behavioral resilience to social defeat stress.

Understanding the biological mechanisms responsible for resilience to stress-the ability to overcome adverse conditions-remains a major challenge. While sleep is a known regulator of resilience, the localized cortical dynamics that facilitate this process remain unclear. We hypothesized that resilience to social stress is determined by local sleep changes within the prelimbic cortex (PrL), a region critical for top-down control of stress-responsive circuits. To test this, we conducted longitudinal single-unit and local field potential (LFP) recordings in the prelimbic (PrL) cortex of male mice

Housing conditions during self-administration determine motivation for cocaine in mice following chronic social defeat stress.

RATIONALE: Stress exposure has a lasting impact on motivated behavior and can exacerbate existing vulnerabilities for developing a substance use disorder. Several models have been developed to examine how stressful experiences shape drug reward. These range from locomotor sensitization and conditioned place preference to the propensity for drug self-administration or responding to drug-predictive cues. While self-administration studies are considered to have more translational relevance, most of the studies to date have been conducted in rats. Further, many self-administration studies are cond

Gene deficiency and pharmacological inhibition of soluble epoxide hydrolase confers resilience to repeated social defeat stress.

Depression is a severe and chronic psychiatric disease, affecting 350 million subjects worldwide. Although multiple antidepressants have been used in the treatment of depressive symptoms, their beneficial effects are limited. The soluble epoxide hydrolase (sEH) plays a key role in the inflammation that is involved in depression. Thus, we examined here the role of sEH in depression. In both inflammation and social defeat stress models of depression, a potent sEH inhibitor, TPPU, displayed rapid antidepressant effects. Expression of sEH protein in the brain from chronically stressed (susceptible

Decreased hepatic contents of coenzyme A molecular species in mice after subchronic mild social defeat stress.

Social stress may precipitate psychiatric disorders such as depression, which is related to the occurrence of the metabolic syndrome, including obesity and type 2 diabetes. We have evaluated the effects of social stress on central and peripheral metabolism using a model of depression in mice. In the present study, we focused on coenzyme A (CoA) molecular species [i.e. non-esterified CoA (CoASH), acetyl-CoA and malonyl-CoA] which play important roles in numerous metabolic pathways, and we analyzed changes in expression of these molecules in the hypothalamus and liver of adult male mice (C57BL/6

Early handling improves sociability and preference for social novelty in social-defeated male rats: Laterality in vermal Purkinje cells architecture.

Although the cerebellum is primarily known for motor functions, its involvement in cognitive and emotional functions has become more evident in recent years. In particular, the cerebellar vermis has been implicated in social behavior. Here, we examined the effects of social defeat stress (SDS) during late adolescence on sociability, preference for social novelty, and the morphological characteristics of cerebellar Purkinje cells (PCs). Additionally, we investigated the impact of early handling, an early-life mild stress, and its interaction with subsequent SDS, on social behavior and the struc

Dysfunction of Serotonergic and Dopaminergic Neuronal Systems in the Antidepressant-Resistant Impairment of Social Behaviors Induced by Social Defeat Stress Exposure as Juveniles.

BACKGROUND: Extensive studies have been performed on the role of monoaminergic neuronal systems in rodents exposed to social defeat stress as adults. In the present study, we investigated the role of monoaminergic neuronal systems in the impairment of social behaviors induced by social defeat stress exposure as juveniles. METHODS: Juvenile, male C57BL/6J mice were exposed to social defeat stress for 10 consecutive days. From 1 day after the last stress exposure, desipramine, sertraline, and aripiprazole were administered for 15 days. Social behaviors were assessed at 1 and 15 days after the l

STAT3 Signaling Mediates Agomelatine Restoration of Prefrontal Cortex Synaptic Plasticity in Chronic Social Defeat Stress Mice.

Major depressive disorder (MDD) is a major public health issue worldwide. It not only causes an increased socio-economic burden, but also negatively affects personal lives. It has been shown that depression leads to impairments in synaptic plasticity, such as impaired long-term potentiation (LTP) in the prefrontal cortex (PFC). A recent study reported that administration of agomelatine, a melatonin receptor agonist and 5-hydroxytryptamine (5-HT) 2C antagonist, which may be involved in synaptic functions, for 1 week has a rapid antidepressive effect. Moreover, the mechanism of Glycogen synthase

Intermittent voluntary wheel running promotes resilience to the negative consequences of repeated social defeat in mice.

A novel approach to reduce the incidence of substance use disorders is to promote resilience to stress using environmental resources such as physical exercise. In the present study we test the hypothesis that Voluntary Wheel Running (VWR) during adolescence blocks the negative consequences of stress induced by intermittent repeated social defeat (IRSD). Four groups of adolescent male C57BL/6 mice were employed in the experiment; two groups were exposed to VWR (1 h, 3 days/week) from postnatal day (PND) 21 until the first social defeat (PND 47), while the remaining two groups did not have acces

Taurine Alleviates Chronic Social Defeat Stress-Induced Depression by Protecting Cortical Neurons from Dendritic Spine Loss.

Abnormal amino acid metabolism in neural cells is involved in the occurrence and development of major depressive disorder. Taurine is an important amino acid required for brain development. Here, microdialysis combined with metabonomic analysis revealed that the level of taurine in the extracellular fluid of the cerebral medial prefrontal cortex (mPFC) was significantly reduced in mice with chronic social defeat stress (CSDS)-induced depression. Therefore, taurine supplementation may be usable an intervention for depression. We found that taurine supplementation effectively rescued immobility

Behavioral flexibility impacts on coping and emotional responses in male mice submitted to social defeat stress.

Behavioral flexibility permits the appropriate behavioral adjustments in response to changing environmental demands. The present study aimed to evaluate if variability in baseline flexibility can enable differences in coping strategies, changes in neuroplasticity, and behavioral outcomes in responses to chronic social defeat stress (CSDS). Male C57BL6 mice were submitted to the Morris Water Maze (MWM) using an extended protocol for reversal learning to assess. The animals were divided into low and high behavioral flexibility groups based on their performance on the last day of acquisition vers

Transcriptome-Wide Identification of G-to-A RNA Editing in Chronic Social Defeat Stress Mouse Models.

Emerging evidence suggests that RNA editing is associated with stress, neurological diseases, and psychiatric disorders. However, the role of G-to-A RNA editing in chronic social defeat stress (CSDS) remains unclear. We herein identified G-to-A RNA editing and its changes in the ventral tegmental area (VTA), a key region of the brain reward system, in CSDS mouse models under emotional stress (ES) and physiological stress (PS) conditions. Our results revealed 3812 high-confidence G-to-A editing events. Among them, 56 events were significantly downregulated while 23 significantly upregulated in

Subanesthetic ketamine reverses neuronal and astroglial metabolic activity deficits in a social defeat model of depression.

Depression is one of the most debilitating neuropsychiatric disorders. Most of the current antidepressants have long remission time and low recovery rate. This study explores the impact of ketamine on neuronal and astroglial metabolic activity in prefrontal cortex in a social defeat (SD) model of depression. C57BL/6 mice were subjected to a social defeat paradigm for 5 min a day for 10 consecutive days. Ketamine (10 mg/kg, intraperitoneal) was administered to mice for two consecutive days following the last defeat stress. Mice were infused with [1,6-13 C2 ]glucose or [2-13 C]acetate to assess
