Potential of Sorghum Polyphenols to Prevent and Treat Alzheimer's Disease: A Review Article.

Alzheimer's disease (AD) is characterized by the excessive deposition of extracellular amyloid-beta peptide (Aβ) and the build-up of intracellular neurofibrillary tangles containing hyperphosphorylated tau proteins. This leads to neuronal damage, cell death and consequently results in memory and learning impairments leading to dementia. Although the exact cause of AD is not yet clear, numerous studies indicate that oxidative stress, inflammation, and mitochondrial dysfunction significantly contribute to its onset and progression. There is no effective therapeutic approach to stop the progressi

Drying and processing protocols affect the quantification of cyanogenic glucosides in forage sorghum.

BACKGROUND: Cyanogenic glucosides are common bioactive products that break down to release toxic hydrogen cyanide (HCN) when combined with specific β-glucosidases. In forage sorghum, high concentrations of the cyanogenic glucoside dhurrin lead to reduced productivity and sometimes death of grazing animals, especially in times of drought, when the dhurrin content of stunted crops is often higher. The aim of this study was to develop harvesting protocols suitable for sampling in remote areas. RESULTS: Dhurrin concentration in air- and oven-dried leaves was the same as in fresh leaves, with no s

The Promising Prospective Technology for Biofuel Production with Mixed Cultures Using Sorghum Grain (Sorghum bicolor (L.) Moench) Grown in a Temperate Climate: Characteristics of Fermentation Factors and Volatile Compounds Analysis.

Sorghum grain (Sorghum bicolor (L.) Moench) grown in a temperate climate was used as a feedstock for bioethanol production. Mixed cultures of Zymomonas mobilis and Saccharomyces cerevisiae were used for fermentation under the conditions and parameters established through the design of experiments (DOE, 2(K-p) standard design). The most effective parameters for the simultaneous saccharification and fermentation of the liquefied and granular starch fermentation methods were selected. Analyses revealed that the granular starch fermentation method (with Stargen 002) combined with mixed cultures yi

Multi-Omics Integration Drives Precision Breeding in Sorghum: Molecular Dissection and Breeding Practice.

Sorghum (Sorghum bicolor L.), the fifth-most important cereal crop globally, serves as a cornerstone crop for food security, forage production, and bioenergy feedstock on arid and semi-arid marginal lands, owing to its high water use efficiency derived from C4 photosynthesis and exceptional tolerance to abiotic stresses. Conventional sorghum breeding has long been constrained by insufficient genetic dissection of complex traits, the "black box" of genotype-to-phenotype mapping, and poorly understood genotype-by-environment interactions, resulting in stagnant genetic gain. In recent years, rapi

Common genetic control for grain filling duration and kernel weight in grain sorghum.

The genetic control of GFD in sorghum closely overlaps with that of grain size, is influenced by both pre- and post-anthesis processes, offering new opportunities for yield improvement through targeted breeding and inter-racial trait introgression. The duration of the grain filling period has been associated with yield increases in cereals including maize and sorghum. The genetic control of grain filling duration (GFD) is however not known in sorghum. This study explored the genetic variation and extent of genetic control for GFD in a diverse panel of sorghum genotypes (n=904), in three enviro

Synergistic remediation of oil-contaminated soil using sorghum and oil sludge-derived biochar.

Oil sludge contamination poses a significant threat to agricultural soil health and productivity. While biochar is a promising soil amendment, the synergistic potential of combining oil sludge-derived biochar with plant systems for holistic remediation remains poorly quantified. In a 60-day greenhouse study using a full-factorial design, we investigated the interactive effects of oil sludge (0-0.5% w/w) and biochar produced from the same sludge (0-1% w/w) on soil biochemistry and forage sorghum (Sorghum bicolor) growth, providing the first quantitative evidence of true synergy through combined

Transcriptomic and targeted metabolomic insights into carotenoid-mediated color formation in sorghum grains.

INTRODUCTION: Sorghum is a major staple crop in semi-arid regions; however, its generally low grain carotenoid content limits its potential contribution to alleviating vitamin A deficiency. Elucidating the regulatory mechanisms underlying carotenoid accumulation is therefore essential for the nutritional improvement of sorghum. METHODS: Five sorghum varieties with distinct grain colors (white, gray, yellow, red, and black) were analyzed using integrated targeted carotenoid metabolomic and transcriptomic approaches to characterize carotenoid composition and its molecular regulation. RESULTS A

Utilizing genetic variation in perennial sorghum to improve host plant resistance to aphids.

With growing concerns over the sustainability of conventional farming systems, perennial crops offer an environmentally friendly and resilient alternative for long-term agricultural production. Perennial grain crops provide numerous benefits, such as low input investment, reduced tillage, soil conservation, better carbon sequestration, sustainable yields, and enhanced biodiversity support. Sorghum (Sorghum bicolor) is the fifth most-grown cereal crop grown for food, fuel, and food grain in the world. The development of perennial sorghum offers a substitute for traditional annual sorghum crops

Saline irrigation improves survival of forage sorghum but limits growth and increases toxicity.

Moderately saline water has been proposed as a potential irrigation resource for crops such as forage sorghum (Sorghum bicolor × Sorghum bicolor nothosubsp. drummondii) in drought-prone regions. However, it is not yet fully understood how salinity affects growth and potential toxicity of sorghum. Sorghum produces the cyanogenic glucoside dhurrin, which can cause hydrogen cyanide (prussic acid) poisoning in grazing animals. To address this, two glasshouse experiments were conducted. Experiment 1 assessed tolerance of sorghum to a range of salt treatments (0, 12.5, 25, 50, 100 and 150 mM NaCl).

Graphene oxide enhances aphid resistance in sorghum via the miR319-SbTCP7-SbLOX3 Pathway.

The aphid (Melanaphis sacchari) has emerged as a formidable pest, devastating sorghum plants and highlighting the need for sustainable management strategies. Graphene oxide (GO), as a novel material, has garnered attention for its use in crop cultivation and management, but its effects on biotic stresses remain elusive. Here, we used 10 mg/L GO to spray aphid-stressed sorghum seedlings four times in total. GO exposure reduced 50% H2O2 from the reactive oxygen species (ROS) burst induced by the aphid. Further analysis revealed that GO within the cells acts as a nanozyme, mimicking and enhancing

Valorisation of insect infested sweet sorghum reeds towards production of a fermented beverage.

Sweet sorghum variety (Sorghum bicolour (L)) commonly known as sweet reeds, Ntšhe, in Setswana, is a valuable cash crop mostly for small scale farmers in Botswana and other southern African countries. These reeds are widely consumed as a delicacy and contribute significantly to food security, employment, and rural incomes. However, infestations by the larval stages of Chilo partellus (stem borer moths) lead to substantial economic losses, as consumers reject worm-infested reeds. To mitigate these losses, valorisation of condemned sweet reeds is attractive. Here, we took advantage of our unders

Genome-wide identification and characterization of greenbug-inducible NAC transcription factors in sorghum.

BACKGROUND: Sorghum (Sorghum bicolor) is an important cereal crop grown worldwide because of its multipurpose uses such as food, forage, and bioenergy feedstock and its wide range of adaption even in marginal environments. Greenbug can cause severe damage to sorghum plants and yield loss. Plant NAC transcription factors (TFs) have been reported to have diverse functions in plant development and plant defense but has not been studied in sorghum yet. METHODS AND RESULTS: In this study, a comprehensive analysis of the sorghum NAC (SbNAC) gene family was conducted through genome-wide analysis. A

Comparative Transcriptome Analysis Reveals Genetic Mechanisms of Sugarcane Aphid Resistance in Grain Sorghum.

The sugarcane aphid, Melanaphis sacchari (Zehntner) (Hemiptera: Aphididae) (SCA), has become a major pest of grain sorghum since its appearance in the USA. Several grain sorghum parental lines are moderately resistant to the SCA. However, the molecular and genetic mechanisms underlying this resistance are poorly understood, which has constrained breeding for improved resistance. RNA-Seq was used to conduct transcriptomics analysis on a moderately resistant genotype (TAM428) and a susceptible genotype (Tx2737) to elucidate the molecular mechanisms underlying resistance. Differential expression

Matrix-Assisted Laser Desorption/Ionization-Mass Spectrometry Imaging of Metabolites during Sorghum Germination.

Dhurrin is the most abundant cyanogenic glucoside found in sorghum (Sorghum bicolor) where it plays a key role in chemical defense by releasing toxic hydrogen cyanide upon tissue disruption. Besides this well-established function, there is strong evidence that dhurrin plays additional roles, e.g. as a transport and storage form of nitrogen, released via endogenous recycling pathways. However, knowledge about how, when and why dhurrin is endogenously metabolized is limited. We combined targeted metabolite profiling with matrix-assisted laser desorption/ionization-mass spectrometry imaging to in

Artificial intelligence-based structural modeling of PSTOL1 receptor kinases in rice and sorghum.

Originally identified in rice (Oryza sativa OsPSTOL1), PHOSPHORUS-STARVATION TOLERANCE 1 (PSTOL1) proteins are essential determinants of phosphorus efficiency in several plant species, including Sorghum bicolor (SbPSTOL1). In contrast to OsPSTOL1, SbPSTOL1 homologs exhibit a transmembrane receptor-like kinase (RLK) architecture with polymorphic extracellular domains. We employed an AI-driven computational pipeline to investigate the structure and conformational dynamics of SbPSTOL1 proteins. Monomeric predictions using multiple deep-learning platforms yielded structures that were inconsistent

High-resolution microCT reveals relationships between stomata and interior leaf anatomy in sorghum.

Stomata are pores in the leaf epidermis that regulate the trade-off between CO2 uptake for photosynthesis and water vapor loss to the atmosphere. Stomatal patterning therefore influences water use efficiency and is a target for engineering to avoid drought stress. However, there is limited understanding of how internal leaf anatomy is coordinated with stomatal development, in part due to the technical challenges of assessing three-dimensional anatomy with sufficient resolution. C4 grasses are understudied, and this is a significant knowledge gap, given their file-like stomatal distribution and
