Computational analysis of missense mutations in squalene epoxidase associated with terbinafine resistance in clinically reported dermatophytes.

Dermatophyte infections, as a significant public health threats, are increasingly associated with antifungal drug resistance, particularly to terbinafine. Mutations in the squalene epoxidase (SQLE) gene have been linked to resistance by altering amino acid residues and interfering with drug-protein interactions. This study applied computational tools including I-Mutant, ConSurf, HOPE, DynaMut2, STRING, and molecular docking to assess the structural and functional impact of clinically reported SQLE missense mutations in terbinafine-resistant dermatophyte isolates. Twelve out of fourteen mutatio

Squalene is a potential endocrine modulator in rat: A proof-of-principle study with 3-methylcholanthrene-induced toxicity.

Oestrus urine was proved as a potential endocrine modulator in alleviating the toxicity induced by 3-methylcholanthrene (3-MC) in male rats. We, in this study, aimed to prove the attributing potential of toxicity alleviation to squalene, an oestrus-specific pheromone in rats. A single dose of 3-methylcholanthrene (25 mg/kg BW, i.p.) was administered to male Wistar rats with concurrent exposure to squalene sprayed in bedding material (Group III). Group II rats did receive 3-MC treatment but did not expose to squalene. Group I rats were intact control neither administered 3-MC nor sprayed with s

Gas chromatography-mass spectrometry and Raman imaging measurement of squalene content and distribution in human hair.

A sensitive and specific gas chromatography-mass spectrometry (GC-MS) method was developed and validated for the measurement of the squalene content from root to tip, in both Chinese black virgin and bleached hair. Deuterated squalene was used as the internal standard. For quantification, selective ion monitoring (SIM) at m/z 410.0 and 347.0 were monitored for squalene and deuterated squalene, respectively. Different methods for the extraction of squalene from ex vivo human hair were compared including organic solvent extraction and acid/alkali hydrolysis. The best extraction efficiency was ob

Peroxidated squalene induces the production of inflammatory mediators in HaCaT keratinocytes: a possible role in acne vulgaris.

In order to investigate whether products derived from the oxidation of sebum can be responsible for the induction of inflammatory processes, HaCaT keratinocytes were treated with peroxidated squalene. NF-kappaB activation, secretion, and expression of IL-6, as well as peroxisome proliferator-activated receptor alpha (PPARalpha) mRNA and protein levels, were measured at the end of the treatment and after 24 and 48 hours of recovery. Squalene peroxidation products were administered in amounts able to elicit significant hyperproliferation and to induce lipoxygenase (LOX) activity. The results sho

Hybrid nanogel system of quercetin-loaded squalene NLCs with zinc-HA network for dry eye syndrome.

Dry eye syndrome (DES) is a multifactorial ocular surface disorder characterized by tear film instability, inflammation, and ocular irritation. Conventional therapies are often limited by rapid precorneal elimination, low bioavailability, and frequent administration. To overcome these limitations, quercetin-loaded squalene linoleic acid nanostructured lipid carriers were incorporated into a ZnSO₄ cross-linked hyaluronic acid nanogel (QS-NLCs-Zn-HA nanogel), developed as a potential therapeutic system for DES management. The nanogel was prepared via ionic cross-linking between cationic ZnSO₄ an

Mechanism-based inhibition of squalene epoxidase by phenothiazines for lipid metabolism disruption using repurposed antipsychotic drugs.

Squalene epoxidase (SQLE) is a crucial enzyme in the cholesterol-biosynthesis pathway and a promising target for treating cholesterol-related disorders. This study aimed to repurpose eighteen clinically approved phenothiazine antipsychotics as competitive SQLE inhibitors by integrating structure-based virtual screening, 200 ns molecular-dynamics simulations, MM/PBSA binding-energy calculations and in vitro enzyme assays. We first screened the 18 derivatives using molecular docking, structural/pharmacological diversity and ADMET analysis. Six compounds-ethopropazine, periciazine, piperacetazine

PP1-associated responses linked to HMGR-related changes during postharvest squalene accumulation in Camellia oleifera seeds.

Plants synthesize diverse isoprenoids that are essential for growth, development, and adaptation to environmental stress. In Camellia oleifera, squalene is a valuable minor constituent of seed oil, yet the regulatory processes linking postharvest stress to squalene accumulation remain poorly understood. Here, we investigated the relationship between protein phosphatase 1-associated responses and changes related to 3-hydroxy-3-methylglutaryl-coenzyme A reductase during postharvest squalene accumulation in Camellia oleifera seeds exposed to 35 °C and 95% relative humidity. Squalene content incre

Squalene alleviates cancer cachexia-induced muscle atrophy and adipose tissue wasting in CT26 tumor-bearing mice.

Cancer cachexia is a multifactorial metabolic syndrome characterized by progressive loss of body weight, skeletal muscle mass, and adipose tissue, primarily driven by chronic inflammation and systemic metabolic imbalance. Squalene (SQ), a naturally occurring triterpenoid abundant in shark liver oil, exhibits potent antioxidant and anti-inflammatory properties. In this study, we investigated the protective effects of SQ against cancer-induced skeletal muscle and adipose tissue wasting in CT26 tumor-bearing mice. Seven days after CT26 implantation, BALB/c mice were orally administered SQ (150 or

Enhanced Immune Responses Against Mycobacterium tuberculosis Through Heat-Killed BCG with Squalene-in-water Emulsion Adjuvant.

BACKGROUND: While clinical studies have documented sex differences in emotional, behavioral, and cognitive function of children with Attention Deficit and Hyperactive Disorder (ADHD), it is unknown if these sex differences are due to differences in referral and diagnosis or if they can be also seen when we screen a community sample for ADHD. If these sex differences exist in populations with a diagnosis history but cannot be seen in screening, then they are unfair, preventable, and due to gender (social processes in referral and diagnosis) rather than sex. AIM: Using the data from a community

Terbinafine Resistance of Trichophyton Clinical Isolates Caused by Specific Point Mutations in the Squalene Epoxidase Gene.

Terbinafine is one of the allylamine antifungal agents whose target is squalene epoxidase (SQLE). This agent has been extensively used in the therapy of dermatophyte infections. The incidence of patients with tinea pedis or unguium tolerant to terbinafine treatment prompted us to screen the terbinafine resistance of all Trichophyton clinical isolates from the laboratory of the Centre Hospitalier Universitaire Vaudois collected over a 3-year period and to identify their mechanism of resistance. Among 2,056 tested isolates, 17 (≈1%) showed reduced terbinafine susceptibility, and all of these wer

Oxidative tryptophan modification by terpene- and squalene-hydroperoxides and a possible link to cross-reactions in diagnostic tests.

Hydroperoxides can act as specific haptens and oxidatively modify proteins. Terpene hydroperoxides trigger unusually high frequencies of positive skin reactions in human patients if tested at high concentrations. It is unknown whether this is due to specific hapten formation. Here, we show that both terpene hydroperoxides and the endogenous hydroperoxide formed from squalene can oxidatively modify tryptophan. Oxidative modifications of Trp were recently postulated to explain cross-sensitization between unrelated photosensitizers. Current observations may extend this hypothesis: Oxidative event

Identification of oxidation products of squalene in solution and in latent fingerprints by ESI-MS and LC/APCI-MS.

An investigation was carried out to identify oxidation products of squalene (SQ) in latent fingerprints. Oxidation products of SQ incubated in solution with Rose Bengal as a photooxidizer were isolated by semipreparative HPLC-UV and identified by direct infusion ESI-MS and flow injection APCI-MS. Squalene hydroperoxides ranging from squalene monohydroperoxide (SQ-[OOH]) to SQ-[OOH]5 were identified together with SQ epoxide. SQ-[OOH] was the main oxidation product. An LC/APCI-MS method was developed and used to monitor the fate of SQ in solution and in latent fingerprints and the formation of S

pH-induced transition of sponge-phase nanoparticles to micelles: The case of Leu-enkephalin-squalene lipopeptide prodrug.

Self-assembled bioconjugates into nanoparticles (NPs) represent a powerful strategy to enhance drug efficacy. In the case of Leu-enkephalin-squalene (LENK-SQ) bioconjugate, self-assembly leads to spherical NPs internally organized into a sponge phase, which exhibit significant analgesic activity, in contrast to the free Leu-enkephalin (LENK) peptide, whose therapeutic use is severely limited by its rapid degradation and poor bioavailability. However, the presence of an ionizable group in the LENK peptide part raises questions regarding the structural evolution of these nano-assemblies under ph

MF59-like adjuvant containing yeast-derived squalene enhances the humoral immune response to cell-derived influenza vaccine.

The aims of this study were to assess the adjuvant properties of an MF59-like adjuvant containing yeast-derived squalene (MF59-like YD) in a cell-based quadrivalent influenza vaccine (QIV) and to investigate the potential mechanisms of action. MF59-like adjuvants containing either yeast-derived or shark-derived squalene were incorporated into QIV formulations. Antigen-specific immune responses in mouse serum were evaluated via enzyme-linked immunosorbent assays (ELISAs), hemagglutination inhibition (HI) assays, and microneutralization (MN) assays. The effects and mechanisms of action of the ad

A multifunctional nanotheranostic: hyaluronic acid-squalene nanoclusters for radiosensitization, ferroptosis, and Mn/Fe-based dual MR imaging.

Ferroptosis, a non-apoptotic form of cell death, has shown promise in overcoming resistance to conventional therapies that rely on apoptosis. The combination of ferroptosis inducers with radiation therapy has the potential to improve therapeutic outcomes. However, radiation therapy (RT) alone often requires additional non-toxic radiosensitizers to enhance its efficacy, as the lack of oxygen in hypoxic tumor regions leads to radioresistance. In this study, we synthesized hyaluronic acid and squalene nanoclusters embedded with manganese ferrite nanoparticles to enhance radiosensitization and fer

Pharmacokinetics and biodistribution of squalene-containing emulsion adjuvant following intramuscular injection of H5N1 influenza vaccine in mice.

Squalene is a component of oil-in-water emulsion adjuvants developed for potential use in some influenza vaccines. The biodistribution of the squalene-containing emulsion adjuvant (AddaVax™) alone and as part of complete H5N1 vaccine was quantified in mechanistically and toxicologically relevant target tissues up to 336 h (14 days) following injection into quadriceps muscle. At 1 h, about 55% of the intramuscularly injected dose of squalene was detected in the local quadriceps muscles and this decreased to 26% at 48 h. Twenty-four hours after the injection, approximately 5%, 1%, and 0.6% of th

From fruit to function: Squalene synthase driven triterpenoids biosynthesis in Gardenia jasminoides for food-active ingredient discovery.

The medicinal and edible plant Gardenia jasminoides is attracting attention as a source of natural food-active components. Integrated GC-MS/LC-MS metabolomics revealed that G. jasminoides fruits specifically accumulate bioactive triterpenoids, including the GRAS antioxidant squalene (220 ng/mg FW) and methyl oleanonate, a compound with reported anticancer (PPARγ agonist) activity. As the central biosynthetic hub, GjSQS showed fruit-predominant expression, strongly correlating with triterpenoid abundance. Functional validation confirmed its ER localization and efficient in vitro conversion of F

Molecular analysis of squalene epoxidase gene mutations in Trichophyton rubrum from clinical onychomycosis samples in South Korea.

Trichophyton rubrum is the primary causative pathogen of onychomycosis, and terbinafine remains the first-line treatment for both oral and topical therapy. However, increasing reports of terbinafine resistance-associated mutations in T. rubrum from India, Europe, and other parts of Asia have raised global concern. This study aimed to investigate mutations in the squalene epoxidase (SQLE) gene in T. rubrum from onychomycosis samples in South Korea. Between August 2021 and November 2023, toenail specimens from patients with KOH-positive fungal microscopy were collected from six hospitals. Fungal
