Correlation between enterotoxigenicity, tested by different ELISA-techniques, antibiotic resistance patterns and phage groups of Staphylococcus aureus strains.

A group of 596 Staphylococcus aureus strains isolated from various clinical sources or implicated in food poisoning was investigated for enterotoxins A and B (SEA and SEB) production. The conventional ELISA techniques (competitive and sandwich ELISA) were compared with a newly developed avidin-biotin ELISA in their ability to detect the enterotoxins. The avidin-biotin system was not remarkably influenced by SPA up to 10 micrograms/ml. A semi-quantitative competitive ELISA for the detection of staphylococcal protein A (SPA) in culture supernatants was carried out in parallel. The strains isolat

Characterization and antibacterial efficacy of the broad-host-range phage P108 against biofilm-forming and methicillin-resistant Staphylococcus aureus.

Staphylococcus aureus (S. aureus) is a widely distributed opportunistic pathogen capable of causing a variety of serious infections, with its antibiotic resistance becoming increasingly prevalent. As natural bactericidal agents, bacteriophages (phages) have emerged as promising therapeutic alternatives to antibiotics. In this study, we isolated a lytic phage P108 capable of targeting methicillin-resistant S. aureus (MRSA) strains. Phage P108 features an icosahedral head with a diameter of approximately 84.7 nm and a contractile tail measuring about 221.5 nm in length. It has a latent period of

A versatile toolset for Staphylococcus aureus gene inactivation enables testing of anti-phage defense systems in native genetic backgrounds.

BACKGROUND: Functional gene analysis of Staphylococcus aureus across different strains requires the development of genetic engineering tools. In studies on phage therapy, such analysis is especially important for understanding the genomic differences between strains, underlying the differences in sensitivity to phages. Meanwhile, difficulties in introducing DNA constructs for genetic manipulations into non-laboratory S. aureus strains hinder the elucidation of staphylococcal anti-phage defense strategies. RESULTS: We used a transducing S. aureus bacteriophage and an E. coli -S. aureus shuttle

Isolation and characterization of a Staphylococcus aureus-lytic jumbo phage LY01 and its local therapeutic efficacy in a porcine dermatitis model.

Porcine dermatitis caused by Staphylococcus aureus (S. aureus) has garnered increasing attention due to the escalating antibiotic resistance, necessitating the development of alternative therapeutic strategies. In this study, a S. aureus-lytic jumbo phage, designated as LY01, was isolated from swine farm effluent and subsequently characterized. Key findings revealed that the bacteriophage exhibited lytic activity against S. aureus with broad host specificity. LY01 possesses a short latent period (30 min), thermal stability (30°C-50°C), and pH stability (pH 5-9). Phylogenetic analysis based on

Phage-derived endolysin prevents dissemination of methicillin resistant Staphylococcus aureus during oral co-infection with Candida albicans.

The oral cavity is colonized by many species of bacteria and fungi without causing serious infections of hosts. However, upon loss of homeostasis, some of these colonizers have the potential to invade the host and cause life-threatening infections. The fungus Candida albicans and the gram-positive bacterium Staphylococcus aureus represent well-studied examples. In immunocompromised mice, oral co-colonization with these two species can result in dissemination of S. aureus through the body, causing S. aureus bacteremia and subsequent sepsis. In the proof-of-principle experiments described here,

First Use of Phage Therapy in Canada for the Treatment of a Life-Threatening, Multidrug-Resistant Staphylococcus epidermidis Periprosthetic Joint Infection.

We describe the first use of phage therapy in Canada for the treatment of a life-threatening periprosthetic joint infection (PJI), with successful outcome. PJI is a devastating complication of joint replacement surgery, with high morbidity and mortality. Our patient presented with early sepsis from a chronic recalcitrant multidrug-resistant (MDR) Staphylococcus epidermidis hip PJI which had repeatedly failed standard therapy. She had previously undergone 10 operations of the right hip, and only three weeks after completing a prolonged course of daptomycin following her most recent hip revision

The zebrafish embryo model: unveiling its potential for investigating phage therapy against methicillin-resistant Staphylococcus aureus infection.

Staphylococcus aureus is a pathogenic bacterium responsible for a broad spectrum of infections, including cutaneous, respiratory, osteoarticular, and systemic infections. It poses a significant clinical challenge due to its ability to develop antibiotic resistance. This resistance limits therapeutic options, increases the risk of severe complications, and underscores the urgent need for new strategies to address this threat, including the investigation of treatments complementary to antibiotics. The evaluation of novel antimicrobial agents often employs animal models, with the zebrafish embryo

Efficacy of phage cocktail AB-SA01 therapy in diabetic mouse wound infections caused by multidrug-resistant Staphylococcus aureus.

BACKGROUND: Diabetic foot ulcer (DFU) is a serious complication of diabetes mellitus. Antibiotic-resistant Staphylococcus aureus is frequently isolated from DFU infections. Bacteriophages (phages) represent an alternative or adjunct treatment to antibiotic therapy. Here we describe the efficacy of AB-SA01, a cocktail of three S. aureus Myoviridae phages, made to current good manufacturing practice (cGMP) standards, and which has undergone two phase I clinical trials, in treatment of multidrug-resistant (MDR) S. aureus infections. RESULTS: Wounds of saline-treated mice showed no healing, but e

Temperate bacteriophage SapYZUs7 alters Staphylococcus aureus fitness balance by regulating expression of phage resistance, virulence and antimicrobial resistance gene.

Temperate bacteriophages are crucial for maintaining the pathogenicity and fitness of S. aureus, which also show promise as a biocontrol agent for S. aureus. However, the fitness benefit and cost of lysogeny by S. aureus temperate phages and their underlying mechanisms remain unexplored. In this study, phage resistance, virulence, antimicrobial resistance (AMR), transcriptome, and metabolome of phage SapYZUs7 lysogenic and non-lysogenic S. aureus strains were compared. Whole-genome analysis revealed that SapYZUs7 harbouring smaII associated with a single-protein MazF-like antiphage system coul

Development and mouse model evaluation of a new phage cocktail intended as an alternative to antibiotics for treatment of Staphylococcus aureus-induced bovine mastitis.

Bovine mastitis is a prevalent infectious disease in dairy herds worldwide, resulting in substantial economic losses. Staphylococcus aureus is a major cause of mastitis in animals, and its antibiotic resistance poses challenges for treatment. Recently, renewed interest has focused on the development of alternative methods to antibiotic therapy, including bacteriophages (phages), for controlling bacterial infections. In this study, 2 lytic phages, vB_SauM_JDYN (JDYN) and vB_SauM_JDF86 (JDF86), were isolated from the cattle sewage effluent samples collected from dairy farms in Shanghai. The 2 ph

An Endogenous Staphylococcus aureus CRISPR-Cas System Limits Phage Proliferation and Is Efficiently Excised from the Genome as Part of the SCCmec Cassette.

CRISPR-Cas is an adaptive immune system that allows bacteria to inactivate mobile genetic elements. Approximately 50% of bacteria harbor CRISPR-Cas; however, in the human pathogen Staphylococcus aureus, CRISPR-Cas loci are less common and often studied in heterologous systems. We analyzed the prevalence of CRISPR-Cas in genomes of methicillin-resistant Staphylococcus aureus (MRSA) strains isolated in Denmark. Only 2.9% of the strains carried CRISPR-Cas systems, but for strains of sequence type ST630, over half were positive. All CRISPR-Cas loci were type III-A and located within the staphyloco

Draft genomes of six methicillin-resistant Staphylococcus aureus in a Philippine tertiary hospital with insights on putative antimicrobial and phage resistance mechanisms.

The genomes of six methicillin-resistant Staphylococcus aureus (MRSA) revealed CC5(ST5)-SCCmec-IVc-spa-t105-PVL+ (n = 3), CC5(ST6)-SCCmec-IVc-spa-t10002 (n = 1), CC5(ST5)-SCCmec-IVa-spa-t002 (n = 1), and CC8(ST8)-SCCmec-IVa-spa-t008-PVL+ (n = 1) genotypes. All possessed multiple antibiotic resistance genes (putative mecA and blaZ, and dfrG for cotrimoxazole-resistant strains), intact staphylococcal prophages, and putative antiphage systems. The results could aid in MRSA infection management and control.

Antibiotic susceptibility and phage typing of methicillin-resistant and -sensitive Staphylococcus aureus clinical isolates at three periods during 1991-1997.

The aim of the study presented here was to evaluate the antibiotic susceptibility of Staphylococcus aureus over a 7-year period. A total of 2,122 clinical isolates of Staphylococcus aureus were collected from hospitalized patients at 3-year intervals during the period 1991-1997. The prevalence of methicillin-resistant isolates was 41.6%, 38.2% and 36% in 1991, 1994 and 1997, respectively; all of these isolates were sensitive to vancomycin. Over the study period, resistance to pristinamycin and fusidic acid increased slightly and resistance to imipenem, rifampicin and amikacin increased greatly

Pathogens in the bay: environmental Staphylococcus saprophyticus strains mirror clinical counterparts in virulence, biofilm formation, antimicrobial resistance, pathogenicity, and phage susceptibility.

Staphylococcus saprophyticus is a leading cause of urinary tract infections, yet little is known about its environmental reservoirs and traits outside clinical settings. This study aimed to compare clinical and environmental S. saprophyticus strains isolated from a polluted coastal ecosystem in Brazil, assessing their virulence-associated traits, antimicrobial resistance, biofilm formation, and susceptibility to bacteriophages. Forty strains (20 clinical, 20 environmental) were characterized using GTG5-PCR, virulence gene screening, antibiotic susceptibility testing, biofilm assays, exposure t
