Enzyme-Responsive Polymeric Drug Delivery Systems for the Treatment of Inflammatory Bowel Diseases: A Review.

Inflammatory bowel disease (IBD), including Crohn's disease and ulcerative colitis, is a chronic inflammatory disorder of the gastrointestinal tract that imposes an increasing global health burden. Conventional pharmacological treatments are often limited by systemic side effects and insufficient drug accumulation at inflamed intestinal sites. Enzyme-responsive polymeric drug delivery systems have emerged as a promising strategy to overcome these limitations by enabling site-specific and controlled drug release within the pathological microenvironment of the colon. This review summarizes recen

Synthesis and Light-Mediated Structural Disruption of an Azobenzene-Containing Helical Poly(isocyanide).

Helical poly(isocyanide)s are an important class of synthetic polymers possessing a static helical structure. Since their initial discovery, numerous examples of these helices have been fabricated. In this contribution, the synthesis of a chiral, azobenzene (azo)-containing isocyanide monomer is reported. Upon polymerization with nickel(II) catalysts, a well-defined circular dichroism (CD) trace is obtained, corresponding to the formation of a right-handed polymeric helix. The helical polymer, dissolved in chloroform and irradiated with UV light (365 nm), undergoes a cis to trans isomerization

Hydrophilic Polyethers Derived from Functional Epoxides: Beyond Poly(ethylene glycol).

Hydrophilic polymers constitute an essential section in polymer sciences and technologies. Poly(ethylene glycol) (PEG), a flagship hydrophilic polymer derived from the simplest epoxide, has attracted massive research interests and found wide-ranged applications from daily commodities to cutting-edge materials. In the meantime, limitations of PEG are increasingly noted, including hazardous nature of its monomer, unfavored crystallinity, lack of modification sites and stimuli-responsiveness, immunogenicity due to extensive uses, etc., which has heavily spurred explorations for PEG alternatives.

Smart Stimuli-responsive Nanogels: A Potential Tool for Targeted Drug Delivery.

Nanogels (NGs) are presently the focus of extensive research because of their special qualities, including minimal particle size, excellent encapsulating efficacy, and minimizing the breakdown of active compounds. As a result, NGs are great candidates for drug delivery systems. Cross-linked nanoparticles (NPs) called stimulus-responsive NGs are comprised of synthetic, natural, or a combination of natural and synthetic polymers. These NPs can swell in response to large amounts of solvent, but their structural makeup prevents them from dissolving. Furthermore, in response to (i) physical stimuli

Solvent- and guest-responsive self-assembly of Hamilton receptor tethered bis(merocyanine) dyes.

A novel supramolecular building block (8) that consists of a Hamilton receptor and two merocyanine dyes has been synthesized, and the self-assembly based on orthogonal hydrogen bonding and dipolar interactions has been studied in detail. Different self-assembled species, including oligomers, polymers, and inverted micelles could be observed upon variation of the solvent polarity and the concentration. Moreover, this system is highly responsive toward molecular stimuli such as merocyanine molecules with the barbituric acid motif that are bound by the Hamilton receptors. Detailed UV/Vis absorpti

Dynamic Ordering and Phase Segregation in Hydrogen-Bonded Polymers.

Hydrogen bonds (H-bonds) constitute highly relevant structural units of molecular self-assembly. They bridge biological and synthetic sciences, implementing dynamic properties into materials and molecules, not achieved via purely covalent bonds. Phase segregation on the other hand represents another important assembly principle, responsible for, e.g., cell compartimentation, membrane-formation, and microphase segregation in polymers. Yet, despite the expanding elegant synthetic strategies of supramolecular polymers, the investigation of phase behavior of macromolecules driven by H-bonding forc

Recent advances in hydrogels as strategy for drug delivery intended to vaginal infections.

Vaginal infections represent a clear women health problem due to the several issues as high recurrence rate, drug resistence and emergence of persistent strains. However, achieving improvements in therapeutic efficacy by using conventional formulations intended to vaginal drug delivery remains as a challenge due to anatomy and physiology of the vagina, since the secretion and renewal of vaginal fluids contribute to the removal of the dosage form. Hydrogels have been widely exploited aiming to achieve drug delivery directly into vaginal mucosa for local therapy due to their attractive features

Multi-stimuli responsive nanogel/hydrogel nanocomposites based on κ-carrageenan for prolonged release of levodopa as model drug.

This study describes the development of a novel biocompatible nanogel/hydrogel nanocomposite system for long-term delivery of bioactive molecules. To this aim, the nanogels were synthesized via radical polymerization of poly(N-isopropylacrylamide). The nanogels were introduced during the formation of a hydrogel network to produce nanogel/hydrogel soft nanocomposites, while the biocompatible hydrogel was based on poly (acrylic acid) grafted onto κ-carrageenan polysaccharide with entrapped magnetic iron oxide nanoparticles as crosslinker. In these systems, by using stimuli-responsive functional

Analysis of interaction between liposome membranes induced by stress condition: utilization of liposomes immobilized on indium tin oxide electrode.

NBD-cholesterol (NBD-Ch)-modified liposome was immobilized on indium tin oxide (ITO) electrode via the covalent binding method. The transfer of NBD-Ch between the immobilized liposomes and the target liposomes was observed by using a fluorescent microscope. The addition of liposome suspension co-incubated with alpha-chymotrypsin or stimuli-responsive polymer to the surface of the above ITO electrode, enhanced the liposome-liposome interaction, resulting in the promotion of NBD-Ch transfer. The apparent transfer rate constant of NBD-Ch was found to be correlated with the index for the liposome-

Ionically Counter-Programmable Phase-Separation of Superstable Star-Biopolyamide Coacervates for Electrothermally-Responsive Pseudosolid Smart Windows.

Thermoresponsive polymers are highly anticipated for passive thermochromic smart windows, yet the instability and too-high mobility of their light-scattering states for separated phases remain significant hurdles. To address these challenges, we report a design concept for pyrrolidone-based star-polyamides synthesized from itaconic acid and cadaverine, whose architecture provides exceptional sensitivity to external stimuli. In a stark departure from the classical Hofmeister series, Ca2+ initiates an anomalous salting-in effect that contrasts with the Na+-induced salting-out. DFT/MD calculation

Mucoadhesive properties of nanogels based on stimuli-sensitive glycosaminoglycan-graft-pNIPAAm copolymers.

Mucoadhesive formulations capable of situ gelation are promising for improving ocular drug delivery. Here we investigated two types of nanogels based on anionic glycosaminoglycans with grafted thermo-responsive poly(N-isopropylacrylamide) chains. One type of nanogels were formed by thermo-induced gelling of heparin-graft-poly(N-isopropylacrylamide) and chondroitin sulfate-graft-poly(N-isopropylacrylamide) copolymers. Another type of nanogels was based on the same copolymers, but terminal groups of thermosensitive macromolecular chains were modified to form covalent disulfide cross-links. All t

Thermoreversible Polymorph Transitions in Supramolecular Polymers of Hydrogen-Bonded Squaramides.

Hydrogen-bonded squaramide (SQ) supramolecular polymers exhibit uncommon thermoreversible polymorph transitions between particle- and fiber-like nanostructures. SQs 1-3, with different steric bulk, self-assemble in solution into particles (AggI) upon cooling to 298 K, and SQs 1 and 2, with only one dendronic group, show a reversible transformation into fibers (AggII) by further decreasing the temperature to 288 K. Nano-DSC and UV/Vis studies on SQ 1 reveal a concentration-dependent transition temperature and ΔH for the AggI-to-AggII conversion, while the kinetic studies on SQ 2 indicate the on

Synergistic effects of azobenzene and thiourea backbones in multiresponsive copolymers for sensing and adhesive technologies.

Stimuli-responsive polymers have garnered significant attention for their ability to adapt to environmental changes, offering applications in sensing, smart coatings, and adaptive devices. However, challenges remain in developing multifunctional polymers that combine dynamic responsiveness with robust mechanical properties. In this study, we design and synthesize multifunctional azobenzene-based copolymers, poly(thiourea triethylene glycol)-co-azobenzene (PTUEG3-co-Azo) copolymers, through a controlled polycondensation process to address these limitations.
