Immunoinformatics based design of a multiepitope vaccine targeting Streptobacillus moniliformis for the prevention of rat-bite fever.

OBJECTIVES: To design and computationally evaluate a multiepitope-based vaccine candidate targeting antigenic proteins of Streptobacillus moniliformis using immunoinformatics approaches. METHODS: Three antigenic proteins, ATP-dependent zinc metalloprotease FtsH, signal peptidase I, and protein translocase subunit SecY, were selected based on antigenicity, non-allergenicity, and non-toxicity. B-cell and T-cell epitopes were predicted and screened for immunogenic potential and global population coverage. A multiepitope vaccine construct containing six MHC class I and six MHC class II epitopes w

Rat bite fever due to Streptobacillus notomytis complicated by meningitis and spondylodiscitis: a case report.

BACKGROUND: Only three other cases of rat bite fever caused by Streptobacillus notomytis in humans have been reported since this species was identified in 2015. Data specific to the differences in clinical features and geographic distribution between S. notomytis infection and S. moniliformis infection are scarce. All previous cases of human S. notomytis infection were reported from Japan. This is the first case of S. notomytis infection reported from outside of Japan. CASE PRESENTATION: A 72-year-old Thai woman was admitted to Siriraj Hospital (Bangkok, Thailand)-Thailand's largest universit

Case report: A fatal case of aortic and mitral valve endocarditis caused by Streptobacillus moniliformis.

BACKGROUND: Infective endocarditis (IE) secondary to rat-bite fever (RBF) is rare but potentially lethal. Rapid diagnosis is of utmost prognostic importance. However, the diagnosis of RBF is challenging because Streptobacillus moniliformis does not grow under conventional culture conditions. CASE SUMMARY: A 65-year-old male without previous cardiac history presented with sudden onset of balance problems and facial palsy. For 2 weeks, he had experienced intermittent fever and myalgia. Transoesophageal echocardiography (TOE) revealed severe mitral and aortic valve IE with aortic root abscess. T

Acute Tetraplegia Caused by Rat Bite Fever in Snake Keeper and Transmission of Streptobacillus moniliformis.

We report acute tetraplegia caused by rat bite fever in a 59-year old man (snake keeper) and transmission of Streptobacillus moniliformis. We found an identical characteristic bacterial pattern in rat and human samples, which validated genotyping-based evidence for infection with the same strain, and identified diagnostic difficulties concerning infection with this microorganism.

Phylogenetic and comparative genomics of the family Leptotrichiaceae and introduction of a novel fingerprinting MLVA for Streptobacillus moniliformis.

BACKGROUND: The Leptotrichiaceae are a family of fairly unnoticed bacteria containing both microbiota on mucous membranes as well as significant pathogens such as Streptobacillus moniliformis, the causative organism of streptobacillary rat bite fever. Comprehensive genomic studies in members of this family have so far not been carried out. We aimed to analyze 47 genomes from 20 different member species to illuminate phylogenetic aspects, as well as genomic and discriminatory properties. RESULTS: Our data provide a novel and reliable basis of support for previously established phylogeny from t

A spinal epidural abscess due to Streptobacillus moniliformis infection following a rat bite: case report.

The authors describe the case of a 40-year-old man suffering from an epidural abscess in the thoracic spine due to a rarely isolated pathogen, Streptobacillus moniliformis, the causative agent of rat bite fever. Besides diffuse abdominal pain, ataxia, paresthesia, hypesthesia, and enhanced reflexes of the lower extremities, the patient suffered from a decreased sensation of bladder filling. His history was also positive for a rat bite 6 weeks earlier. Magnetic resonance imaging showed an epidural, space-occupying lesion compressing the spinal cord at the vertebral levels of T6-8. Broad range polymerase chain reaction for diagnosis of rat-bite fever caused by Streptobacillus moniliformis.

An 11-year-old boy presented with fever, vomiting, rash, limping and blisters on his feet after a finger bite by his domestic rat. Although cultures from blood, cerebrospinal fluid and urine remained negative, broad range polymerase chain reaction amplification of a part of the 16S rRNA gene followed by sequencing allowed the detection and identification of Streptobacillus moniliformis in blister fluid, thus confirming the suspected clinical diagnosis of rat-bite fever.

Rat bite fever caused by Streptobacillus moniliformis in a child: human infection and rat carriage diagnosed by PCR.

A child owning pet rats developed an eruptive fever with blisters, polyarthritis, and spectacular desquamation of the hands. Streptobacillus moniliformis was identified after culture of the child's blister fluid and was detected in rat samples by molecular methods. Such detection in the pet of a human victim of rat bite fever has not been reported previously.

Infection in a Young Immunocompetent Male Caused by Streptobacillus felis, a Putative Zoonotic Microorganism Transmitted by Cats.

Rat bite fever (RBF) is predominantly caused by Streptobacillus moniliformis. We report a human infection with Streptobacillus felis. Clinical presentation was consistent with RBF, but serologic testing was negative for S moniliformis. Eventually, S felis-specific sequences were detected in skin lesions of the patient and in the oropharynx of local cats.

Streptobacillus felis, a member of the oropharynx microbiota of the Felidae, isolated from a tropical rusty-spotted cat.

Streptobacillus felis is a fastidious microorganism and a novel member of the potentially zoonotic bacteria causing rat bite fever. Since its description, this is the second isolation of S. felis in a diseased member of the Felidae. Interestingly, the strain from this study was isolated from a zoo held, rusty-spotted cat (Prionailurus rubiginosus), with pneumonia, thereby indicating a possible broader host range in feline species. A recent preliminary sampling of domestic cats (Felis silvestris forma catus) revealed that this microorganism is common in the oropharynx, suggesting that S. felis

Approved and novel strategies in diagnostics of rat bite fever and other Streptobacillus infections in humans and animals.

Rat bite fever (RBF), a worldwide occurring and most likely under-diagnosed zoonosis caused by Streptobacillus moniliformis, represents the most prominent disease of Streptobacillus infections. Recently, novel members have been described, from which a reservoir in rats and other animal species and a zoonotic potential can be assumed. Despite regularly published case reports, diagnostics of RBF continues to represent a 'diagnostic dilemma', because the mostly applied 16S rRNA sequence analysis may be uncertain for proper pathogen identification. Virtually nothing is known regarding prevalence i

Rat bite fever, a fatal case of Streptobacillus moniliformis infection in a 14-month-old boy.

Streptobacillus moniliformis is the primary cause of rat bite fever in North America. Children under 12 years of age are primarily infected, presenting with an acute syndrome of fever, rash, and polyarthritis. Common vectors include rats and mice. Transmission is predominantly from a bite or scratch, but contact with or ingestion of food contaminated with feces or saliva has also been reported. We report a fatal case of S. moniliformis infection in a 14-month-old-boy. At home, the child was exposed to filthy living conditions and the family had pet ferrets. Autopsy revealed a red-pink, mostly

Streptobacillus moniliformis mitral valve endocarditis and septic arthritis: the challenges of diagnosing rat-bite fever endocarditis.

Streptobacillus moniliformis, the cause of rat-bite fever (RBF) in the United States, has rarely been reported as a cause of infectious endocarditis. In the majority of previously reported cases, the diagnosis was clinically- based in patients with underlying valvular abnormalities in the setting of positive blood culture for Streptobacillus moniliformis. We report a case of native valve endocarditis secondary to Streptobacillus moniliformis in a woman with a mitral valve vegetation but negative blood cultures where the diagnosis was established using molecular diagnostics on the valvular tiss

Numerical analysis of electrophoretic protein patterns of Streptobacillus moniliformis strains from human, murine and avian infections.

A total of 31 cultures representing 22 different strains of Streptobacillus moniliformis has been characterised by unidimensional SDS-PAGE of cell proteins. The isolates were from sporadic and outbreak cases of human infection in the USA, UK and France, as well as mouse, turkey and rat isolates from various countries. The protein patterns, which contained 40-50 discrete bands, were highly reproducible and were used as the basis for a numerical analysis. Three clusters were obtained at the 86% similarity (S) level and these were further divided to give a total of seven clusters at the 90% S lev

Multiplex serology for Streptobacillus moniliformis and other 'rat bite fever-like' microorganisms for seroprevalence studies in rodents.

Rat bite fever (RBF) is a zoonotic disease caused primarily by Streptobacillus (S.) moniliformis. Norway or brown rats (Rattus [R.] norvegicus) are the natural host for S. moniliformis and carry the bacterium in the nasopharynx without clinical disease. Transmission to humans often occurs through rat bites or scratches, but also through contact with the excreta of infected rats. Although human infections with S. moniliformis occur worldwide, they are rarely diagnosed. For decades, S. moniliformis was the only known member of the genus Streptobacillus. In recent years, however, four additional
