BACKGROUND: Lactococcosis is a bacterial disease in fish caused by several Lactococcus species, including L. garvieae and L. petauri, with clinical signs frequently overlapping with those caused by other streptococcal species. This diagnostic overlap necessitates a prompt and precise diagnostic tool for guiding accurate treatment and control. To address this diagnostic challenge, we developed basic recombinase polymerase amplification (basic-RPA) and RPA combined with lateral flow dipstick (RPA-LFD) assays for the detection of lactococcosis-associated Lactococcus species.
RESULTS: Both method

Galectin (Gal) is a member of a family of β-galactoside-binding lectin. The members of this family play important roles in the recognition of carbohydrate ligands and in various other biological processes. In this study, we identified the gene encoding Gal-9 in Pagrus major (PmGal-9) and analyzed its expression in various tissues after pathogen challenge. Alignment analysis revealed that the two galactose-binding lectin domains of the deduced protein were highly conserved among all the teleosts. Phylogenetic analysis revealed that PmGal-9 is most closely related to the Gal-9 gene of gilthead s

Scavenger receptors (SRs) are a group of membrane-bound receptors that could bind to a variety of ligands including endogenous proteins and pathogens. SRs have been recognized to play vital roles in innate immune response against pathogen infection in both vertebrates and invertebrates. In this regard, one SmSCARA5 gene was captured in turbot (Scophthalmus maximus). The full-length SmSCARA5 transcript contains an open reading frame (ORF) of 1494 bp. SmSCARA55 showed both the highest identity and similarity to half-smooth tongue sole (Cynoglossus semilaevis), and a high degree of conservation o

Once thought to be a feature exclusive to lymphocyte-driven adaptive immunity, immune memory has also been shown to operate as part of the innate immune system following infection to provide an elevated host response to subsequent pathogenic challenge. This evolutionarily conserved process, termed 'trained immunity', enables cells of the innate immune system to 'remember' previous pathogen encounters and mount stronger responses to the same, or different, pathogens after returning to a non-activated state. Here we show that challenging larval zebrafish, that exclusively rely on innate immunity

Rapid and accurate diagnosis of infectious diseases in aquaculture is essential for preventing major economic and ecological losses. Traditional culture-based methods focus on isolation of individual pathogens, and often are burdened with extended processing times, particularly during investigations of polymicrobial infections. Application of Oxford Nanopore Technologies (ONT) sequencing offers a rapid, culture-independent workflow for the identification of bacterial and fungal pathogens directly from fish tissues. Swab and organ samples from four cases (1: Salmo spp.; 2: Cyprinus carpio; 3: S

Soft tissue bacterial infections following aquatic animal bites, stings, and minor injuries occur commonly and usually on the extremities in fishermen and beachgoers worldwide after freshwater and saltwater exposures. Louisiana has more tidal, saltwater, and brackish water shorelines (more than 7,000 miles) than any other state, including Alaska and Hawaii. As a result, Louisiana residents are often exposed to marine pathogens when fishing or working offshore or when enjoying Louisiana's miles of shorelines. Although many species of bacteria have been isolated from marine wounds, superficial s

The cDNA encoding of a complement factor D/adipsin and kallikrein-like serine protease, designated PoDAK, was isolated from the olive flounder Paralichthys olivaceus. PoDAK cDNA encodes a polypeptide with 277 amino acids containing conserved catalytic triad residues of serine proteases. The amino acid sequence of PoDAK showed high similarity to the kallikrein-like protein of medaka, mammalian adipsin/complement factor D and tissue kallikrein homolog, KT-14 of trout, complement factor D of zebrafish, and shared 31.6-36.8% homology with complement factor D/adipsin known from other species, inclu

Fish protein hydrolysate (FPH) has shown immense potential as a dietary protein supplement and immunostimulant in aquaculture, especially in Nile tilapia production. Four isoproteic diets (30% crude protein) were prepared by including FPH at varying percentages (0%, 0.5%, 1%, and 2%). Nile tilapia fed with FPH diets for 90 days, and their growth performance, feed utilization, blood biochemistry, liver and gut morphology, and resistance against Streptococcus iniae were investigated. The findings revealed that diets physical attributes such as pellet durability index and water stability were rem

Human contact with and consumption of fishes presents hazards from a range of bacterial zoonotic infections. Whereas many bacterial pathogens have been presented as fish-borne zoonoses on the basis of epidemiological and phenotypic evidence, genetic identity between fish and human isolates is not frequently examined or does not provide support for transmission between these hosts. In order to accurately assess the zoonotic risk from exposure to fishes in the context of aquaculture, wild fisheries and ornamental aquaria, it is important to critically examine evidence of linkages between bacteri

Cytotoxic reactive oxygen species (ROS), including hydroxyl radicals (.OH-), superoxide anions (.O2-), peroxides (.O22-), and nitrogen radicals (NO), are derivatives of hydrogen peroxide (H2O2). Catalase (CAT) is an immunologically important antioxidant enzyme that regulates host redox homeostasis by disintegrating cellular H2O2 into water and oxygen. The identified CAT homolog sequence from Scomber japonicus (SjCAT) comprised 1584 bp, encoding 527 amino acids in length with a molecular weight of 59.99 kDa, and an isoelectric point of 7.69. In silico analysis revealed that the SjCAT sequence e

The lactoferricin expressed in Bacillus subtilis is relatively low in yield, making it hard to apply in industrial settings. We constructed a six tandem repeat of lactoferricin cDNA driven by promoter PtrnQ. After transformation, two transformants P245 and P263 possessing a stable inheritance of plasmid and high expression of lactoferricin were selected. The bactericidal activities, 1 μl of aliquot of a total 5.5 ml of solution extracted from 5 ml of cultured P245 and P263, were equivalent to the efficacy of 238.25 and 322.7 ng of Ampicillin against Escherichia coli, respectively, and 366.4 an

Interleukin-20 (IL-20), a member of the IL-10 cytokine family, exhibits dual immunoregulatory roles in mammals, balancing both anti-inflammatory and pro-inflammatory responses. However, its function in antimicrobial immunity in teleost fish remains poorly characterized. In this study, we identified and functionally characterized an IL-20-like homolog in channel catfish (Ictalurus punctatus), designated IcIL-20. The IcIL-20 protein consists of 184 amino acids, contains six conserved cysteine residues, and displays structural features typical of the IL-10 family. Transcript analysis revealed con

CD28 is a co-stimulatory receptor that provides a critical second signal alongside T cell receptors for the activation of naive T cells. We characterized the CD28 gene of rock bream, which has a deduced amino acid sequence of 221 residues with an extracellular Ig-superfamily V domain, transmembrane region, and cytoplasmic tail. The conservation in domain structures and other motifs shows that it is highly likely that RbCD28 is a homologue of mammalian CD28 and may have related co-stimulatory functions. RbCD28 is constitutively expressed in most tissues that were analysed, with a relatively hig

This study evaluated the effects of dietary inclusion of fermented sesame oilcake (FSOC) on growth performance, antioxidant responses, immune parameters, bactericidal activity, and gut microbial composition in juvenile Nile tilapia, Oreochromis niloticus. FSOC was produced by aerobic solid-state fermentation using Saccharomyces cerevisiae. Fish (5-month-old, 13.31 ± 0.73 g, all-male) were randomly distributed into three treatment groups and fed diets containing 0% (control [CTL]), 10% (S-10), or 20% (S-20) FSOC for 8 weeks. Growth performance, feed conversion ratio, and survival were not signi

This study investigates the sequence characteristics, expression patterns, and immune functions of Nile tilapia Oreochromis niloticus CD94 (OnCD94), a C-type lectin (CTL) from the CTL V family, which also belongs to the natural killer cell receptor (NKCR) family. The Open Reading Frame (ORF) of OnCD94 is 681 bp and encodes 226 amino acids, lacks a signal peptide and contains a carbohydrate recognition domain (CRD) spanning amino acids 99-223. Multiple sequence alignment revealed that OnCD94 shares 17.39-91.59 % similarity with CD94 from other species. Phylogenetic analysis grouped fish CD94 in

Olive flounder (Paralichthys olivaceus) is the most valuable aquaculture species in Korea, corresponding to ~60% of its total production. However, infectious diseases often break out among farmed flounders, causing high mortality and substantial economic losses. Although some deleterious pathogens, such as Vibrio spp. and Streptococcus iniae, have been eradicated or contained over the years through vaccination and proper health management, the current disease status of Korean flounder shows that the viral hemorrhagic septicemia virus (VHSV), Streptococcus parauberis, and Miamiensis avidus are

Tumor necrosis factor receptor-associated factors (TRAFs), as key signal transducers, play a crucial role in both innate and adaptive immune responses. However, current research on the systematic identification of the TRAF family and its functional role in Siberian sturgeon (Acipenser baerii) is relatively unexplored. In this study, TRAF family members were first identified in A. baerii using comparative genomics and transcriptome analyses. To further determine the TRAF sequences of A. baerii, the open reading frame (ORF) sequences of the Abtraf1, Abtraf5, Abtraf6, and Abtraf7 were cloned. Bio

The mucosal surfaces constitute the first line of host defense against infection, and also serve as the dynamic interfaces that simultaneously mediate a diverse array of critical physiological processes, while in constantly contact with a wide range of pathogens. The lysozymes are considered as key components for innate immune response to pathogen infection with their strong antibacterial activities. But their activities in mucosal immune responses were always overlooked, especially for g-type lysozymes, whose expression patterns in mucosal tissues following bacterial challenge are still limit
