Mechanical stress-induced reactive gliosis in the auditory nerve and cochlear nucleus.

OBJECT: Hearing levels following microsurgical treatment gradually deteriorate in a number of patients treated for vestibular schwannoma (VS), especially in the subacute postoperative stage. The cause of this late-onset deterioration of hearing is not completely understood. The aim of this study was to investigate the possibility that reactive gliosis is a contributory factor. METHODS: Mechanical damage to nerve tissue is a feature of complex surgical procedures. To explore this aspect of VS treatment, the authors compressed rat auditory nerves with 2 different degrees of injury while monitor

[The role of calcium in reaction of the heart for mechanical stress].

The article deals with the role of Ca2+ in four possible mechanisms of cardiac muscle response to mechanical stretch. First, the Ca2+ role is analysed in the changed contractive myocardium function under mechanical stretch. Second, discussed are the direct proofs of redistributing intercellular ions occurring at axial stretch of single cardiomyocytes. Given are the experimental proofs relating the role of Ca2+ and mechanical stretch. Third, considered is the change of electric activity in response to mechanical stretch. Fourthly, considered are the role of Ca2+ in expression and secretion of p

Regional mechanical properties and stress analysis of the human anterior lens capsule.

The lens capsule of the eye functions, in part, as a deformable support through which the ciliary body applies tractions that can alter lens curvature and corresponding refractive power during the process of accommodation. Although it has long been recognized that characterization of the mechanical properties of the lens capsule is fundamental to understanding this physiologic process as well as clinical interventions, prior data have been limited by one-dimensional testing of excised specimens despite the existence of multiaxial loading in vivo. In this paper, we employ a novel experimental a

Mechanical wall stress in abdominal aortic aneurysm: influence of diameter and asymmetry.

PURPOSE: Risk for rupture of an abdominal aortic aneurysm is widely believed to be related to its maximum diameter. From a biomechanical standpoint, however, risk is probably more precisely related to mechanical wall stress. Many abdominal aortic aneurysms are asymmetric (for example because of anterior bulging with posterior expansion limited by the vertebral column). The purpose of this work was to investigate the effect of maximum diameter and asymmetric bulge on wall stress. METHODS: Three-dimensional computer models of abdominal aortic aneurysms were generated. In one protocol, maximum d

Septins buffer actomyosin forces to protect the nucleus from genotoxic mechanical stress.

Invasively migrating cells thread their nucleus through confined interstitial spaces. How cells protect the nucleus from intracellular forces is poorly understood. Here, we show that the septin cytoskeleton buffers the actomyosin forces that have been shown to power nuclear movement. Septin filaments comprising SEPT9, a septin amplified in breast cancer, align with perinuclear actomyosin cables, which exhibit higher tensile stress during 3D confined migration through narrower pores. SEPT9 depletion amplifies actin stress during confined migration and after myosin II hyperactivation in nonmigra

Stress-correlated stereom organization in the echinoid tubercle reveals hierarchical mechanical integration.

Echinoid tests consist of a highly porous skeletal microstructure (stereom) optimized in terms of functional performance. One of the most mechanically demanding components of the test is the tubercle, which forms the mechanical interface between the test plates and spines and is subjected to complex loads generated by motion of the spine. However, the mechanical behaviour of tubercles under realistic loading conditions remains poorly explored. Here, we investigate the tubercle of Prionocidaris baculosa using high-resolution X-ray microscopy and finite-element analysis to simulate physiological

The intrinsic impact of mechanical stress on the maintenance of oocyte dormancy.

In the mammalian ovary, most oocytes remain dormant, and their dormant status plays a central role in maintaining the reservoir population of the female germ line. The equilibrium between the dormant and active states, the latter of which is responsible for producing mature oocytes, is therefore crucial for ensuring the sustained reproductive capability of females. We have previously reported that mechanical stress in the ovary plays a crucial role in oocyte dormancy. However, the mechanism underlying this relation remains unclear. Here, we demonstrated that the mechanical stress is directly t

Stability of arterial blood gas samples after delayed analysis and mechanical stress.

PURPOSE: To investigate the effect of prolonged time before analysis and mechanical manipulation on pre-analytical stability of biomarkers and the validity of blood gas analysis results. METHODS: We collected blood samples from 240 ICU patients from May 18, 2022 to March 31, 2023. Samples were analyzed immediately per standard operating procedure, then the syringes were kept at room temperature for 60 min, subjected to standardized mechanical forces (repeated drops) and analyzed again. Thirteen typical blood gas analytes were measured. Bland-Altman plots were prepared to assess differences be

Piezo1 at the crossroads: Mediating inflammation and mechanical stress in joint disorders.

Piezo1 is a mechanosensitive ion channel, which plays a pivotal role in translating mechanical stress into cellular signaling, thereby influencing inflammatory responses and tissue remodeling in joint disorders. Current evidences showed that Piezo1 functions as a mechanotransducer and amplifier of inflammation across joint disorders including osteoarthritis (OA), rheumatoid arthritis (RA), ankylosing spondylitis (AS), and intervertebral disc degeneration (IVDD). In OA, Piezo1 mediates chondrocyte apoptosis and matrix degradation via calcium influx, NF-κB/MAPK activation, and ferroptosis, formi

Oxidative Stress Related to Mechanical Heart Valves: A Pilot Cross-Sectional Study.

BACKGROUND: Valvular heart disease remains a major global health issue, with mechanical prosthetic heart valves (MPHVs) widely used in surgical valve replacement. However, these devices carry a risk of thrombosis, particularly in the mitral position. Several mechanisms may be involved in this risk, but the role of oxidative stress (OxS) remains unclear. Our aim was to assess the relationship between OxS impairment and platelet activation. METHODS: We analyzed data from a pilot, observational, monocentric study conducted at our anticoagulation clinic at Sapienza University of Rome, involving a

Functional outcome of cell seeded tracheal scaffold after mechanical stress in vitro.

Tracheal tissue engineering is still facing major challenges: realization of efficient vascularization and mechanical properties comparable to native trachea need to be achieved. In this study, we present a strategy for the manufacturing of a construct for tracheal tissue engineering by conditioning through cell seeding followed by mechanical stimulation in vitro. Scaffolds derived from porcine trachea decellularized with supercritical carbon dioxide were seeded with stem cells of different tissue sources and cultured in a bioreactor for 21 days under mechanical stimulation. Enhanced chondroge

Abnormal mechanical stress induced chondrocyte senescence by YAP loss-mediated METTL3 upregulation.

OBJECTIVES: Abnormal mechanical stress is the pivotal risk factor of temporomandibular joint osteoarthritis (TMJOA). This study investigated the pathogenic mechanism by which abnormal mechanical stress induced chondrocyte senescence. MATERIALS AND METHODS: Cellular senescence was investigated in the rodent model of unilateral anterior crossbite and in the chondrocytes subjected to mechanical overloading in vitro. The effects of Yes-associated protein (YAP) in chondrocyte senescence and its correlation with methyltransferase-like 3 (METTL3) and N6-methyladenosine (m6A) modification were evalua

The Piezo1 ion channel in glaucoma: a new perspective on mechanical stress.

Glaucomatous optic nerve damage caused by pathological intraocular pressure elevation is irreversible, and its course is often difficult to control. This group of eye diseases is closely related to biomechanics, and the correlation between glaucoma pathogenesis and mechanical stimulation has been studied in recent decades. The nonselective cation channel Piezo1, the most important known mechanical stress sensor, is a transmembrane protein widely expressed in various cell types. Piezo1 has been detected throughout the eye, and the close relationship between Piezo1 and glaucoma is being confirme

Cranioplasty with customized titanium and PEEK implants in a mechanical stress model.

Large skull defects as a result of craniectomies due to cerebral insults, trauma, or tumors create functional and aesthetic disturbances for the patient. Cranioplasty with implants in these cases are an alternative to autogenous bone transplantation. In our clinic, customized titanium or optima poly-ether-ether ketone (PEEK) implants are used to reconstruct craniectomy defects. To compare the two materials we investigated the structural changes of the implants fixed to a sintered polyamide skull model under mechanical stress in four simplified models. In a standard testing machine, the models

Mechanical stress-dependent transcriptional regulation of sarcolipin gene in the rodent atrium.

Sarcolipin, a homologue of phospholamban, regulates Ca2+ uptake through the interaction with sarcoplasmic reticulum Ca2+ ATPase (SERCA) and is predominantly expressed in the atrial muscle. Although the atrial chamber-specific expression of sarcolipin could be primarily regulated at the transcriptional level, the transcriptional regulation remains poorly understood. Since mechanical stress plays an important role in transcriptional regulation of a gene involved in cardiac hypertrophy and remodeling, we generated left-sided or right-sided pressure-overload models by transverse aortic constrictio

Apoptosis of periodontal ligament cells induced by mechanical stress during tooth movement.

The mechanical force generated during tooth movement creates compressed and cell-free areas in the periodontal membrane. The way in which periodontal ligament cells disappear at the compressed area during tooth movement remains unclear. In the present study we examined whether periodontal ligament cells undergo apoptosis by mechanical stress during tooth movement using the terminal deoxynucleotidyl transferase (TdT)-mediated dUTP-biotin nick-end-labelling method (TUNEL). TUNEL-positive stainings of periodontal ligament cells began to appear at the compressed areas 12 h after tooth movement, an
