Steroid-associated hip joint collapse in bipedal emus.

In this study we established a bipedal animal model of steroid-associated hip joint collapse in emus for testing potential treatment protocols to be developed for prevention of steroid-associated joint collapse in preclinical settings. Five adult male emus were treated with a steroid-associated osteonecrosis (SAON) induction protocol using combination of pulsed lipopolysaccharide (LPS) and methylprednisolone (MPS). Additional three emus were used as normal control. Post-induction, emu gait was observed, magnetic resonance imaging (MRI) was performed, and blood was collected for routine examina

Obstructive sleep apnea of obese adults: pathophysiology and perioperative airway management.

Collapsible pharyngeal airway size is determined by interaction between structural properties of the pharyngeal airway and neural regulation of the pharyngeal dilating muscles. Obesity seems to have two distinct mechanical influences on the pharyngeal airway collapsibility. First, obesity increases soft tissue surrounding the pharyngeal airway within limited maxillomandible enclosure occupying and narrowing its space (pharyngeal anatomical imbalance). Second, obesity, particularly central obesity, increases visceral fat volume decreasing lung volume. Pharyngeal wall collapsibility is increased

Protein structure and coupled solvent dynamics in α-synuclein fibrils under controlled confinement.

The formation of amyloid fibrils by the intrinsically disordered protein α-synuclein in human brain neurons and extracellular tissue is a hallmark of Parkinson disease. Although the stable protein β-structure in α-synuclein fibrils has been determined at high resolution, the status of the dynamically disordered domains and their responses to in vivo confinement and small-molecule interactions remain elusive. We use electron paramagnetic resonance spectroscopy of the spin probe, TEMPOL, colocalized with α-synuclein fibrils, and temperature-controlled, ice boundary confinement to characterize th

Arm position and collapsibility of infraclavicular axillary vein during voluntary breathing: An ultrasound-guided observational study.

BACKGROUND: Arm abduction influences cross-sectional area of the infraclavicular axillary vein, yet the effect of arm abduction on collapsibility of the vein has not been quantified. Decrease in collapsibility of the axillary vein can enable successful cannulation and can decrease injury to underlying vital structures. METHODS: The infraclavicular axillary vein was scanned in 70 patients close to the clavicle with a high-frequency linear transducer in arm adducted position (Point A), after arm abduction at the initial probe position (Point A') and after tracing the vein medially close to clav

Tissue remodelling in pulmonary fibrosis.

Many lung diseases result in fibrotic remodelling. Fibrotic lung disorders can be divided into diseases with known and unknown aetiology. Among those with unknown aetiology, idiopathic pulmonary fibrosis (IPF) is a common diagnosis. Because of its progressive character leading to a rapid decline in lung function, it is a fatal disease with poor prognosis and limited therapeutic options. Thus, IPF has motivated many studies in the last few decades in order to increase our mechanistic understanding of the pathogenesis of the disease. The current concept suggests an ongoing injury of the alveolar

Vertebroplasty and kyphoplasty for the treatment of thoracic fractures in osteoporotic patients: a finite element comparative analysis.

BACKGROUND: Vertebral compression fractures occur in the thoracolumbar junction, causing the collapse of the vertebral body. For their treatment, vertebroplasty and kyphoplasty are used, but it is still unknown which technique is to be preferred. METHODS: Finite element models of the thoracic spine were developed to evaluate the outcomes of vertebroplasty and kyphoplasty. A mild and severe collapse of T10 treated with vertebroplasty or kyphoplasty was studied. Stresses on the endplates and intradiscal pressures were extrapolated to determine the stress distribution in the adjacent structures.

Analysis of early stage osteonecrosis of the human femoral head and the mechanism of femoral head collapse.

We explored the mechanism of early stage osteonecrotic femoral head collapse by analyzing and comparing different regions in human osteonecrotic femoral head samples. Eight osteonecrotic femoral heads (ARCO II-III) were obtained from patients undergoing total hip arthroplasty. Bone structure was observed and evaluated by micro-computed tomography (CT) scans and pathology. Osteoblast and osteoclast activities were detected by tartrate-resistant acid phosphatase, alkaline phosphatase, and immunofluorescent staining. Some trabeculae had microfractures in the subchondral bone and necrotic region,

How effective are contrastive learning-based approaches for activity-cliff prediction?

Activity cliffs, defined as structurally similar molecules with vastly different properties represent a fundamental challenge in modern day drug discovery for property prediction models. While Graph Neural Networks (GNNs) have advanced molecular property prediction, they inherently struggle with this problem due to representation collapse and node over smoothening. In this work, we evaluate the efficacy of incorporating various contrastive learning-based loss functions, including the Supervised Contrastive (SupCon) configurations into several GNN backbones to navigate this structure-activity l

Obesity and obstructive sleep apnoea: mechanisms for increased collapsibility of the passive pharyngeal airway.

Epidemiological evidence suggests there are significant links between obesity and obstructive sleep apnoea (OSA), with a particular emphasis on the importance of fat distribution in the development of OSA. In patients with OSA, the structure of the pharyngeal airway collapses. A collapsible tube within a rigid box collapses either due to decreased intraluminal pressure or increased external tissue pressure (i.e. reduction in transmural pressure), or due to reduction in the longitudinal tension of the tube. Accordingly, obesity should structurally increase the collapsibility of the pharyngeal a

Dissipative structures and interstitial gel phase transition: Prigogine's internal time as a framework for understanding chronic disease.

The interstitial matrix of multicellular organisms - a polyanionic hydrogel embedded within connective tissue - undergoes volume phase transitions (VPT) between a swollen phase and a collapsed phase, as described by Tanaka (1978) for synthetic gels and extended to biological systems by Verdugo and others. We propose that this phase transition constitutes the mesoscale physical mechanism by which thermodynamic closure occurs in living tissue: the collapsed gel physically isolates embedded cells from the thermal, osmotic, and ionic gradients that drive entropy export, thereby attenuating the dis

Thermoresponsive lignin-based polyelectrolyte complexes for the preparation of spherical nanoparticles: Application in pesticide encapsulation.

Lignin-based nanoparticles hold tremendous potential for various applications. This study proposes an innovative and straightforward method for the synthesis of spherical hybrid lignin nanoparticles (hy-LNPs) with a tunable pore structure. The approach involves blending lignin with 20 wt% polyamide-epichlorohydrin, resulting in the formation of thermoresponsive lignin-based polyelectrolyte complexes. Upon heating to 80 °C, the complexes undergo self-assembly into uniform spherical nanoparticles, achieving a minimum polydispersity index (PDI) as low as 0.08. The study reveals that nanoparticle

Failure Analysis of the Tree Column Structures Type AlSi10Mg Alloy Branches Manufactured by Selective Laser Melting.

AlSi10Mg alloy branches were fabricated by selective laser melting (SLM), and the branches were employed to evaluate their effect on the mechanical properties. When the porous branches were compressed along its building direction, the tree column structures-type AlSi10Mg alloy branches collapsed twice, which had typical elastic, shear, collapse, and densification stages. The compressive stress concentration at the interface between the support and the porous body caused the fracture of the tree column structures-type AlSi10Mg alloy branches. The fracture surface indicated that the prepared tre

The Use of the Proximal Hamate as an Autograft for Proximal Pole Scaphoid Fractures: Clinical Outcomes and Biomechanical Implications.

Scaphoid proximal pole fractures remain a surgical challenge because of high propensity for nonunion, osteonecrosis, and ultimately carpal collapse. Options for management of nonsalvageable proximal pole fractures include non-vascularized bone grafts, vascularized pedicled bone grafts, free vascularized bone flaps, and rib cartilage grafts. The proximal pole of the hamate can also serve as a replacement arthroplasty in the setting of proximal pole scaphoid nonunions with collapse, bone loss, and/or osteonecrosis. This novel graft addresses shortcomings of other graft choices by providing a loc

Mechanical behavior and failure mode of body-centered cubic, gyroid, diamond, and Voronoi functionally graded additively manufactured biomedical lattice structures.

Given the capability to produce parts with complex geometries, powder bed fusion using a laser beam (PBF-LB), one of several additive manufacturing techniques, is becoming increasingly prevalent in both research and industry. Advances in the development of biomedical lattice structures show a trend in the use of functional gradients for greater customization and adjustment of mechanical properties according to the demands. This study analyzed four biomedical potential lattice structures (regular and graded) manufactured using PBF-LB in Ti6Al4V alloy. X-ray computed microtomography results demo

Anatomical determinants of upper airway collapsibility in obstructive sleep apnea: A systematic review and meta-analysis.

Upper airway (UA) collapsibility is one of the key factors that determine the severity of obstructive sleep apnea (OSA). Interventions for OSA are aimed at reducing UA collapsibility, but selecting the optimal alternative intervention for patients who fail CPAP is challenging because currently no validated method predicts how anatomical changes affect UA collapsibility. The gold standard objective measure of UA collapsibility is the pharyngeal critical pressure (Pcrit). A systematic literature review and meta-analysis were performed to identify the anatomical factors with the strongest correla

Basal ganglia components have distinct computational roles in decision-making dynamics under conflict and uncertainty.

The basal ganglia (BG) play a key role in decision-making, preventing impulsive actions in some contexts while facilitating fast adaptations in others. The specific contributions of different BG structures to this nuanced behavior remain unclear, particularly under varying situations of noisy and conflicting information that necessitate ongoing adjustments in the balance between speed and accuracy. Theoretical accounts suggest that dynamic regulation of the amount of evidence required to commit to a decision (a dynamic "decision boundary") may be necessary to meet these competing demands. Thro

[Histopathological evaluation of cirrhosis reversal].

A normal liver can develop cirrhosis through long-term and repeated stimulation from various etiologies. Histological manifestations like the collapse of hepatic lobular structure (including microvascular structure) and the formation of pseudolobules can lead to portal hypertension and even decompensated cirrhosis. More and more evidence suggests that effective etiological treatment can not only delay but also reverse the progression of cirrhosis. The mechanism of cirrhosis reversal mainly includes the degradation of extracellular matrix, hepatocyte regeneration, and hepatic lobular remodeling

Structure-Dynamics-Property Relationship Reveals the Origin of Porous Integrity in Imine-Linked Covalent Organic Frameworks.

Molecular flexibility encoded in low-frequency lattice dynamics is increasingly recognized as a key determinant of the properties of porous materials, particularly metal-organic frameworks, but remains largely unexplored in covalent organic frameworks (COFs). Here, we establish a structure-dynamics-property relationship for chemically related COFs by combining three-dimensional electron diffraction, low-frequency vibrational spectroscopy, and first-principles calculations. Using 3D electron diffraction, we determined the crystal structure of the collapsed reduced framework COF-300-AR, revealin
