Sensory nerve endings in the hard palate and papilla incisiva of the rhesus monkey.

The sensory innervation of the hard palate of the rhesus monkey was studied by light and electron microscopy. The mucosa of the hard palate is subject to a particularly heavy mechanical load requiring functional specialisation of the horny epithelium in the form of thickenings - the papilla incisiva and eight pairs of rugae palatinae. A thin layer of firm connective tissue (lamina propria) attaches the mucosa to the periost of the hard palate. Sensory nerve fibres were found most abundantly in the papilla incisiva and first rugae palatinae. Their number decreases in an aboral direction. Five t

Small ileal neurofibroma causing intussusception in a non-neurofibromatosis patient.

Neurofibromas in the small intestine are usually accompanied by von Recklinghausen's disease (neurofibromatosis), and usually originate in the intramuscular plexus of Auerbach. We present here a solitary neurofibroma, which caused an ileocolic intussusception, originating in the submucosal plexus of Meissner in a non-neurofibromatosis patient. To our knowledge, there is no previous report of a neurofibroma originating in the plexus of Meissner. This condition was clearly confirmed by macroscopic and microscopic evaluation.

Segmental Hypoganglionosis of the Colon: A Case Report.

A 64-year-old woman was admitted to hospital with persistent abdominal pain. She had been hospitalized with similar symptoms on five occasions during a period of 2 years. Computed tomography revealed dilatation and fecal impaction from the ileum to the transverse colon. A barium enema and simultaneous ileus tube radiography showed a narrow segment of descending-sigmoid colon. Colonoscopy showed no mucosal change. Her symptoms did not improve with conservative therapy, so descending and sigmoid colectomy was performed. Histologic examination showed disappearance of ganglion cells; axon of Meiss

Changes in the Population Size of Calbindin D-28k-Immunoreactive Enteric Neurons in the Porcine Caecum under the Influence of Bisphenol A: A Preliminary Study.

Calbindin D-28k (CB) is a calcium-binding protein widely distributed in living organisms that may act as a calcium buffer and sensory protein. CB is present in the enteric nervous system (ENS) situated in the gastrointestinal tract, which controls the majority of activities of the stomach and intestine. The influence of various doses of bisphenol A (BPA)-a chemical compound widely used in plastics production-on the number and distribution of CB-positive enteric neuronal cells in the porcine caecum was investigated with an immunofluorescence technique. The obtained results showed that low dosag

Microvascular patterns of the hard palatine mucosa in the Japanese monkey (Macaca fuscata).

Detailed investigations were made of the microvasculature of the mucous membrane, especially the transverse palatine plicae, of the hard palate of the Japanese monkey (Macaca fuscata) utilizing the plastic injection method. The microvascular patterns obtained were compared with those of the cat and other mammals. The incisive papilla was located at the anterior end of the median line of the hard palate, and seven or eight transverse palatine plicae were observed symmetrically from this papilla posteriorly at similar intervals. Each plica arched bilaterally in a wide M. The heights of the plica

Abnormalities of the enteric nervous system in heterozygous endothelin B receptor deficient (spotting lethal) rats resembling intestinal neuronal dysplasia.

BACKGROUND: A homozygous mutation of the endothelin B receptor (EDNRB) gene in spotting lethal (sl/sl) rats leads to Hirschsprung's disease (HSCR) with long segmented aganglionosis. However, the effects on the development of the enteric nervous system (ENS) promoted by a heterozygous mutation of the EDNRB gene are not known. The present study aimed to describe and morphometrically assess the phenotypic abnormalities of the ENS in heterozygous (+/sl) EDNRB deficient rats in comparison with homozygous (sl/sl) EDNRB deficient and wild-type (+/+) rats. METHODS: The distal small intestine, caecum,

Morphologies, dimensions and targets of gastric nitric oxide synthase neurons.

We investigated the distributions and targets of nitrergic neurons in the rat stomach, using neuronal nitric oxide synthase (NOS) immunohistochemistry and nicotinamide adenine dinucleotide phosphate (NADPH) diaphorase histochemistry. Nitrergic neurons comprised similar proportions of myenteric neurons, about 30%, in all gastric regions. Small numbers of nitrergic neurons occurred in submucosal ganglia. In total, there were ~ 125,000 neuronal nitric oxide synthase (nNOS) neurons in the stomach. The myenteric cell bodies had single axons, type I morphology and a wide range of sizes. Five targets

Changes in the somatostatin (SOM)-like immunoreactivity within nervous structures of the porcine descending colon under various pathological factors.

This study reports on changes in the somatostatin-like immunoreactive (SOM-LI) nerve structures of the enteric nervous system (ENS) in the porcine descending colon, caused by chemically driven inflammation, proliferative enteropathy (PE), which is a "natural" inflammation with proliferative changes and nerve injury (axotomy). The distribution pattern of SOM-LI structures was studied using the immunofluorescence technique in the circular muscle layer, the myenteric (MP), outer submucous (OSP) and inner submucous plexuses (ISP) and also in the mucosal layer. Under physiological conditions SOM-LI

Myenteric and submucosal plexuses of the congenital aganglionosis rat (spotting lethal) as revealed by scanning electron microscopy.

A rat with a congenital aganglionosis develops a mega-small intestine and megacolon. This rat mutant has been used as an animal model for Hirschsprung's disease. The myenteric and submucosal plexuses of the rats were exposed by the trypsin-HCl digestion method and examined by scanning electron microscopy. In the control rats the myenteric and submucosal plexuses were present throughout the gut. However, in the mutant rats the myenteric plexus of duodenum was different from those of control rats; the mesh of plexuses varied both in size and shape. There were no myenteric or submucosal plexuses

Adult type mucolipidosis with beta-galactosidase and sialidase deficiency. Histological and biochemical studies.

A case of adult type mucolipidosis with beta-galactosidase and sialidase deficiency is described. This patient, a woman aged 20, had mental retardation, macular cherry-red spots, corneal clouding, gargoyle-like face, cerebellar ataxia, myoclonus and convulsions beginning at the age of 14. Bony deformities, vacuoles in the peripheral lymphocyte and foamy cells in the bone marrow were also noted. Biopsy study of the sural nerve and vermiform appendix disclosed many vacuoles in almost every kind of cells, although the accumulated substance in these vacuoles could not be characterized histochemica

Reassessment of rectal approach to neuropathology in childhood: review of 307 biopsies over 11 years.

A series of 93 rectal biopsies performed for diagnosis of suspected progressive neurometabolic disease between 1967 and 1973 is reviewed, and the results of this and of two previously published series totalling 307 biopsies are analysed. In GM1 and GM2 gangliosidosis, Batten's disease, and certain other neuronal storage diseases rectal biopsy is a reliable diagnostic alternative to brain biopsy. However, the need for biopsy has diminished with improvement in other diagnostic methods, particularly enzyme assay, the availability of which should determine the extent to which biopsy is used. It is

Zonal colonic aganglionosis, a variant of Hirschsprung's disease.

On a review of 15 cases of Hirschsprung's disease of the colon we have encountered 4 cases of an atypical variant. These 4 cases had a zone of aganglionosis of both intermyenteric and submucous plexuses in the sigmoid colon and, in 1 case, a further zone of aganglionosis in the rectum. Ganglion cells reappeared distal to the sigmoid zone of aganglionosis. A transition zone was not invariably present. The clinical features demonstrated a variability of presentation typical of the more usual form of the disease. The importance of atypical Hirschsprung's disease is discussed from a diagnostic and

Decreased tyrosine kinase C expression may reflect developmental abnormalities in Hirschsprung's disease and idiopathic slow-transit constipation.

BACKGROUND: Some patients with Hirschsprung's disease have refractory constipation following excision of aganglionic bowel, as do patients with idiopathic slow-transit constipation (STC). Gut motility depends on enteric neuronal development in response to expression of trophic factors and their receptors. Recent studies indicate the importance of neurotrophin 3 (NT-3) and its high-affinity receptor tyrosine kinase C (trk C) in enteric neuronal development. METHODS: Blinded quantitative immunohistochemical analysis of colon from patients with Hirschsprung's disease (aganglionic, hypoganglionic

The transcriptomes, connections and development of submucosal neuron classes in the mouse small intestine.

The enteric submucosal plexus regulates essential digestive functions, yet its neuronal composition remains incompletely understood. We identified two putative secretomotor neuron classes and a previously unrecognized submucosal intrinsic primary afferent neuron class through single-cell RNA sequencing in the mouse small intestine. Using viral-mediated labeling of each class, we uncovered their morphologies and neural projections in the submucosa-mucosa context, finding connections among all classes and an unexpected close association with enterochromaffin cells. Further transcriptome analysis

Is intestinal neuronal dysplasia a disorder of the neuromuscular junction?

Intestinal neuronal dysplasia (IND) is a malformation of the enteric plexus and clinically resembles Hirschsprung's disease. The pathogenesis of IND is not known. There is no information available regarding the nature of the neuromuscular junction in this disease. The authors examined immunohistochemically full-thickness biopsy specimens from 14 patients (aged 3 weeks to 26 months) with IND and 10 age-matched controls, using monoclonal antibodies to growth associated protein-43 (GAP-43), synaptophysin, and neural-cell adhesion molecule (NCAM) as neuromuscular junction markers. In the normal bo

Anencephaly-associated aganglionosis.

Autopsies of 12 consecutively born infants with anencephaly showed varying degrees of aganglionosis and lateralization defects in four of them. This seemingly regular occurrence of these three defects together suggests that they are caused by an aberration of blastogenesis that results in a polytopic field defect.

Nitric oxide synthase-containing nerve elements in the pylorus of the cat.

Combined nicotinamide adenine dinucleotide phosphate-diaphorase (NADPH-d) histochemistry, nitric oxide synthase (NOS) and vasoactive intestinal polypeptide (VIP) immunocytochemistry were used to study the distribution of NOS- and VIP-containing nerve elements in the feline pylorus. A large number of stained multipolar neurons was found in the myenteric plexus. However, some NADPH-d and NOS positive neurons were also observed in the submucous plexus and in the internal part of muscular layer. A few stained perikarya were found in the tunica mucosa, in a very close situation to the blood vessels

[Intestinal neuronal dysplasia type B: how do we understand it today?].

Intestinal neuronal dysplasia type B (IND B) is currently considered to be a subtle malformation of the submucosal plexus, leading to an increased proportion of over-sized ganglia and potentially accompanied by a mild, chronic gastrointestinal motility disturbance. The diagnosis of IND B is morphologically based and involves the demonstration of an increased proportion of giant ganglia in the submucous plexus related to the patient's age. Giant ganglia are physiologically frequent in the neonatal period. Therefore, IND B should not be diagnosed prior to 1 year of age. Morphological features of
