Photosynthetic Biomanufacturing in Mechanically Robust, 3D Printed Hydrogels.

Engineered living materials (ELMs) integrate synthetic polymers with engineered cells to create systems that sense, respond, and adapt to their environment. While promising as sustainable alternatives to traditional materials, ELMs remain underexplored for use with photoautotrophic organisms. In this study, we evaluate the viability of the cyanobacterium Synechococcus elongatus PCC 7942, which converts carbon dioxide into valuable chemicals using light energy, in three hydrogel matrices previously shown to support heterotrophic cells. S. elongatus remained viable and metabolically active only

Production of Bioplastic Compounds by Genetically Manipulated and Metabolic Engineered Cyanobacteria.

Direct conversion of carbon dioxide to valuable compounds is a desirable way to reduce the environmental burden and switch from fossil to renewable fuels. Cyanobacteria are photosynthetic bacteria that perform oxygenic photosynthesis and are able to produce valuable compounds from carbon dioxide in the air. Synechocystis and Synechococcus species, model unicellular cyanobacteria, can produce succinate and lactate, which are commodity chemicals used to generate bioplastics. Several cyanobacteria are also able to produce polyhydroxybutyrate, a biodegradable polyester that accumulates under nitro

A living therapeutic material to coordinate metabolic oxygenation and regenerative signaling in diabetic wounds.

Diabetic chronic wounds remain difficult to treat because hypoxia and insufficient regenerative signaling persist. Although live biotherapeutics offer a promising route to local oxygenation therapy, current methods largely support a single microbial function rather than coordinate microbial oxygenation with regenerative cues. Here, we report a hydrogel living therapeutic material in which a crosslinked hyaluronic acid matrix coordinates a living metabolic module of Synechococcus elongatus with a regenerative module of platelet-rich plasma (PRP). The matrix enables injectability and rapid gelat

Phosphorus Release and Regeneration Following Laboratory Lysis of Bacterial Cells.

The availability of phosphorus limits primary production in large regions of the oceans, and marine microbes use a variety of strategies to overcome this limitation. One strategy is the production of alkaline phosphatase (APase), which allows hydrolysis of larger dissolved organic phosphorus (DOP) compounds in the periplasm or at the cell surface for transport of orthophosphate into the cell. Cell lysis, driven by grazing and viral infection, releases phosphorus-containing cell components, along with active enzymes that could persist after lysis. The importance of this continued enzymatic acti

Evaluation of algal biofilms on indium tin oxide (ITO) for use in biophotovoltaic platforms based on photosynthetic performance.

In photosynthesis, a very small amount of the solar energy absorbed is transformed into chemical energy, while the rest is wasted as heat and fluorescence. This excess energy can be harvested through biophotovoltaic platforms to generate electrical energy. In this study, algal biofilms formed on ITO anodes were investigated for use in the algal biophotovoltaic platforms. Sixteen algal strains, comprising local isolates and two diatoms obtained from the Culture Collection of Marine Phytoplankton (CCMP), USA, were screened and eight were selected based on the growth rate, biochemical composition

Aerobic transformation of cadmium through metal sulfide biosynthesis in photosynthetic microorganisms.

BACKGROUND: Cadmium is a non-essential metal that is toxic because of its interference with essential metals such as iron, calcium and zinc causing numerous detrimental metabolic and cellular effects. The amount of this metal in the environment has increased dramatically since the advent of the industrial age as a result of mining activities, the use of fertilizers and sewage sludge in farming, and discharges from manufacturing activities. The metal bioremediation utility of phototrophic microbes has been demonstrated through their ability to detoxify Hg(II) into HgS under aerobic conditions.

Functional characterization of bicarbonate transporters from the cyanobacterial SbtA2 family and subsequent expression in tobacco.

Cyanobacteria rely on bicarbonate (HCO3-) as the primary inorganic carbon (Ci) source for photosynthesis in aquatic environments. To use this Ci source, cyanobacteria employ CO2-concentrating mechanisms that elevate cytoplasmic HCO3- via plasma membrane transporters, enhancing carboxylation by carboxysomal Rubisco. The sodium-dependent SbtA1 transporter family is well characterized in freshwater cyanobacteria, but the related SbtA2 family, prevalent in marine α-cyanobacteria, remains uncharacterized. Here, we report functional characterization of SbtA2 homologues from marine Synechococcus spp.

Integrated two-stage continuous system with pH-triggered CO2 delivery for sustainable algal wastewater-to-bioenergy production.

Microalgae offer high lipid content and significant carbon-neutral potential, yet inefficient bioprocess and carbon source management fundamentally hinder their scalability. Addressing these challenges, this study developed an innovative integrated two-stage continuous cultivation system for Synechococcus sp., combining pH-triggered CO2 delivery with wastewater nutrient recycling. Elucidating the relationships among growth, pH, and inorganic carbon, the precise CO2-based pH strategy eliminated salt stress, enhancing biomass productivity by 25 % over conventional chemical methods. The optimized

Glycogen Metabolism Supports Photosynthesis Start through the Oxidative Pentose Phosphate Pathway in Cyanobacteria.

Cyanobacteria experience drastic changes in their carbon metabolism under daily light/dark cycles. During the day, the Calvin-Benson cycle fixes CO2 and diverts excess carbon into glycogen storage. At night, glycogen is degraded to support cellular respiration. The dark/light transition represents a universal environmental stress for cyanobacteria and other photosynthetic lifeforms. Recent studies revealed the essential genetic background necessary for the fitness of cyanobacteria during diurnal growth. However, the metabolic processes underlying the dark/light transition are not well understo

Engineering a synthetic pathway in cyanobacteria for isopropanol production directly from carbon dioxide and light.

Production of alternate fuels or chemicals directly from solar energy and carbon dioxide using engineered cyanobacteria is an attractive method to reduce petroleum dependency and minimize carbon emissions. Here, we constructed a synthetic pathway composed of acetyl-CoA acetyl transferase (encoded by thl), acetoacetyl-CoA transferase (encoded by atoAD), acetoacetate decarboxylase (encoded by adc) and secondary alcohol dehydrogenase (encoded by adh) in Synechococcus elongatus strain PCC 7942 to produce isopropanol. Structure and dynamics of a multidomain nitric oxide synthase regulated by a C2 domain.

Nitric oxide synthase (NOS) is a widely studied multidomain redox enzyme that produces the key signaling molecule and cytotoxic agent nitric oxide (NO) for functions that range from mammalian vasodilation to prokaryotic antibiotic resistance. NOS enzymes from metazoans and cyanobacteria rely on dynamic associations of their oxygenase and coupled diflavin reductase domains that have largely evaded detailed structural characterization. Cryo-electron microscopy studies of a representative dimeric six-domain Synechococcus NOS reveal the architecture of the full-length enzyme, which contains an unu

Stages of biomolecular condensate formation in pro-β-carboxysome assembly.

Cyanobacteria have evolved a CO2-concentrating mechanism (CCM) in the form of a microcompartment with a proteinaceous shell called carboxysome, harbouring the photosynthetic enzyme Rubisco and carbonic anhydrase (CA). β-Carboxysome assembly proceeds by an inside-out process, in which Rubisco, CA and the shell adaptor protein ApN (also known as CcmN) first form the pro-carboxysome biomolecular condensate mediated by the scaffolding protein CM (also known as CcmM). How ApN assembles into the pro-carboxysome as a prerequisite for shell formation has remained unclear. Here we show that ApN is recr

DNA Repair Exhaustion Activates Orthocaspase-Mediated Cell Death and Promote Inclusive Fitness of Cyanobacterial Population Under Abiotic Stress.

Orthocaspases execute regulated cell death (RCD) in cyanobacteria, yet their post-stress activation and physiological relevance remain elusive. Here, we investigated this using heterologous expression of orthocaspases in Synechococcus elongatus PCC 7942. Orthocaspase-expressing (OE) strains, SynOC2_GFP and SynOC6_GFP, were generated using GFP-fused orthocaspases, AnaOC2 and AnaOC6. Catalytically inactive C/A variants were used to generate mutant-expressing (ME) controls, SynC140A_GFP and SynC134A_GFP. Under non-stress conditions, OE and ME strains accumulated orthocaspases without losing viabi

Rewiring photosynthetic carbon flow: Engineered cyanobacterial factories for sustainable carbohydrate production and carbon-negative biomanufacturing.

Cyanobacteria are promising platforms for light-driven carbon fixation and carbohydrate biosynthesis. However, optimization strategies that focus solely on carbon allocation are insufficient to achieve substantial improvements in yield and sustainability. Here, Synechococcus elongatus PCC 7942 was engineered to enhance sucrose production by simultaneously increasing total carbon input and reinforcing the artificial sink. The engineered strain secreted 5.821 g L-1 sucrose, which was 27.4 times higher than the wild-type. Transcriptomic analysis revealed upregulation of abundant genes involved in

pKa values and partition coefficients of nitroxide spin probes for membrane bioenergetics measurements.

Knowledge of pKa's is necessary to calculate intracellular/intravesicular pH values from nitroxide accumulation in cells or vesicles as detected with electron spin resonance (ESR) spectroscopy. pKa values were confirmed in lipid vesicles of known internal pH. To help select probes that do not accumulate in lipid membranes, octanol/buffer partition coefficients of uncharged nitroxides were determined. As an application of selected probes, pH gradients and internal aqueous volumes were analyzed in mitochondria (one internal compartment) and in the cyanobacterium Synechococcus 6311 (two internal

Characterising the structure of photosynthetic biofilms using fluid dynamic gauging.

A new configuration of the fluid dynamic gauging technique for measuring soft layers on surfaces was used to monitor the growth of a cyanobacterium, Synechococcus sp. WH 5701, on stainless steel (SS), glass and an indium tin oxide (ITO) on a polyethylene terephthalate (PET) substratum. The biofilm thickness increased steadily over 4 weeks and exhibited noticeable changes in microstructure and strength. The biofilms all exhibited a two-layer structure, with a compact layer next to the substratum and a loose layer above. Biofilms on ITO or SS exhibited cohesive failure when removed by fluid shea

Mutational analyses reveal PLP-independent functions at PipY, the cyanobacterial paradigm for pyridoxal-phosphate binding proteins.

Pyridoxal-phosphate binding proteins (PLPBP) are involved in the homeostasis of B6 vitamers and amino/keto acids, share a high degree of sequence conservation and are represented in all three domains of life. Despite the obligate presence of the catalyst cofactor PLP, attempts to show enzymatic activity have been unsuccessful. Instead, evidence of RNA binding activity has been provided for several members of the family. Here we use PipY, one of the few PLPBP members studied so far, as a model system to address the phenotypic impact in the cyanobacterium Synechococcus elongatus of mutations K26

The calcium-binding protein CSE links Ca2+ signaling with cell-cell communication in cyanobacteria.

Multicellular cyanobacteria have evolved sophisticated cell-cell communication machinery to exchange, synchronize, and coordinate the efforts of individual cells. Analogous to gap junctions that have traditionally been regarded as a eukaryotic feature, multicellular cyanobacteria coordinate their cell-cell communication via septal junctions (SJs). However, the signals that regulate cell-cell communication and septal-junction assembly are largely unknown. Lately, calcium signaling has been implicated in regulating cell junctions in eukaryotes. We recently discovered a Ca2+-binding protein, CSE,

Heat stress drives increases in phytoplankton cell size in future oligotrophic oceans.

Warming and nutrient limitation are major stressors that affect primary production in the ocean, with cascading impacts on the food web. Yet, we lack a mechanistic understanding of how phytoplankton manage multiple stressors and the implications of these responses for phytoplankton biogeography. By combining theory, proteome allocation modeling, and climate projections, we identified two potential competing strategies for multi-stressor growth: (1) nutrient efficient smaller cells or (2) heat-tolerant larger cells. We found that Prochlorococcus are more vulnerable in warmer, "heat-stressed", t
