The role of taurine in male reproduction: Physiology, pathology and toxicology.

Taurine, a sulfur-containing amino acid, has a wide range of biological effects, such as bile salt formation, osmotic regulation, oxidative stress inhibition, immunomodulation and neuromodulation. Taurine has been proved to be synthesized and abundant in male reproductive organs. Recently, accumulating data showed that taurine has a potential protective effect on reproductive function of male animals. In physiology, taurine can promote the endocrine function of the hypothalamus-pituitary-testis (HPT) axis, testicular tissue development, spermatogenesis and maturation, delay the aging of testic

In vitro and in vivo Antiallergic Effects of Taurine on Allergic Rhinitis.

BACKGROUND: The current treatment for allergic rhinitis (AR) is inadequate. OBJECTIVE: The present study aimed to investigate the therapeutic effect of taurine on AR and to identify the underlying molecular mechanisms. METHODS: The serum level of the antioxidant enzyme extracellular superoxide dismutase (SOD3) was determined in AR patients and in healthy controls. The antiallergic inflammatory effects of taurine were evaluated in a dinitrophenyl-human serum albumin (DNP-HSA)-stimulated human mast cell line (HMC-1) and in an ovalbumin (OVA)-induced AR mouse model. RESULTS: Clinically, a redu

A preclinical study to model taurine pharmokinetics in the undernourished rat.

Malnutrition is a common feature of chronic and acute diseases, often associated with a poor prognosis, including worsening of clinical outcome, owing, among other factors, to dysfunction of the most internal organs and systems affecting the absorption, metabolism and elimination of drugs and nutrients. Taurine is involved in numerous biological processes and is required in increased amounts in response to pathological conditions. The aim of this study was to describe the behaviour of taurine in well-nourished (WN) rats and to analyse the influence of protein-energy undernutrition on the pharm

Taurine deficiency is a cause of vigabatrin-induced retinal phototoxicity.

OBJECTIVE: Although vigabatrin irreversibly constricts the visual field, it remains a potent therapy for infantile spasms and a third-line drug for refractory epilepsies. In albino animals, this drug induces a reduction in retinal cell function, retinal disorganization, and cone photoreceptor damage. The objective of this study was to investigate the light dependence of the vigabatrin-elicited retinal toxicity and to screen for molecules preventing this secondary effect of vigabatrin. METHODS: Rats and mice were treated daily with 40 and 3mg vigabatrin, respectively. Retinal cell lesions were

Is taurine beneficial in reducing risk factors for diabetes mellitus?

Taurine is a semiessential amino acid, and its deficiency is involved in retinal and cardiac degenerations. In recent years, it was found that diabetes mellitus (DM) is associated with taurine, and many in vivo experimental studies showed that taurine administration is able to reduce the alterations induced by DM in the retina, lens, and peripheral nerve, although its effects on diabetic kidney are dubious. Interestingly, long-term taurine supplementation reduces the mortality rate in diabetic rats. The mechanisms by which taurine exerts beneficial effects in DM are discussed below. Recently,

Taurine in submandibular gland of the rat: effect of muscarinic drugs.

Taurine exerts a number of actions in mammalian cells, including regulation of ion transport and osmoregulation. The production and secretion of saliva involve transepithelial ion transport, thereby making the plasma-like primary saliva hypotonic before secretion. Therefore, it is plausible to suggest modulation of salivary taurine by muscarinic agents that affect salivary gland function. One of the objectives of this study was to determine tissue content and localization of taurine in the submandibular gland of the rat. Further, we determined whether treatment with muscarinic drugs that eithe

The cytoprotective role of taurine in exercise-induced muscle injury.

Intense exercise is thought to increase oxidative stress and damage muscle tissue. Taurine is present in high concentration in skeletal muscle and may play a role in cellular defenses against free radical-mediated damage. The aim of this study was to determine if manipulating muscle levels of taurine would alter markers of free radical damage after exercise-induced injury. Adult male Sprague-Dawley rats were supplemented via the drinking water with either 3% (w/v) taurine (n = 10) or the competitive taurine transport inhibitor, beta-alanine (n = 10), for one month. Controls (n = 20) drank tap

Immunoreactivity for taurine in the cochlea: its abundance in supporting cells.

Taurine is the second most abundant free amino acid in the brain where its osmoregulatory function is well established. Taurine-deprived kittens show retinal pathology leading to blindness. In the inner ear, taurine has been reported to be the most abundant free amino acid although its role in inner ear function is not known. Immunohistochemistry was employed here to investigate the localisation of taurine in normal cochleae of the guinea pig compared with two different conditions: experimentally induced endolymphatic hydrops and after oral administration of glycerol. In normal cochleae, by li

Immunocytochemical demonstration of taurine in the retina and pineal organ of the pigeon.

Taurine-like immunoreactivity (TLI) is found in the pigeon retina at high levels within cones, in select cells of the inner nuclear layer and in sublaminae of the inner plexiform layer. At the ultrastructural level a high density of immuno-gold particles in visualized in the ellipsoid, but not in the paraboloid area of the inner segment. It is present in perikarya and the presynaptic endings of cones, but not in postsynaptic cell processes. The immuno-gold particles are not associated directly with the synaptic vesicles. In the pineal organ TLI was confined to the so-called dark pinealocytes w

Endogenous synthesis of taurine and GABA in rat ocular tissues.

The endogenous production of taurine and gamma-aminobutyric acid (GABA) in rat ocular tissues was investigated. The activities of taurine-producing enzyme, cysteine sulfinic acid decarboxylase (CSAD), and GABA-synthesizing enzyme, glutamic acid decarboxylase (GAD), were observed in the retina, lens, iris-ciliary body and cornea. The highest specific activity of CSAD was in the cornea and that of GAD in the retina. The discrepancy between CSAD activity and taurine content within the ocular tissues indicates that intra- or extraocular transport processes may regulate the concentration of taurine

Distribution of taurine in the crystalline lens of vertebrate species and in cataractogenesis.

Marked heterogeneity was observed in the distribution of taurine in different regions of the lens in various species. In general low taurine pools were observed in the nucleus of all species except frog and human. The distribution of taurine in human senile cataractous lenses at different stages of maturation showed decreased contents in all the regions except capsule epithelium as compared to the normal human lenses. This decrease is progressive upto the 'mature' stage of cataract. In rat lenses with galactose cataracts taurine contents decreased by about 83-94% of the normal values in the eq

Taurine transporter SLC6A6 expression promotes mesenchymal stromal cell function.

Mesenchymal stromal cell (MSC) differentiation is critical for the development, maintenance, and repair of bone tissue. MSCs also play a key role in regulating self-renewal and differentiation of normal hematopoietic and leukemic stem cells. Our prior work has identified a key role of taurine produced by bone marrow osteolineage cells in supporting the growth of taurine transporter (TauT or Slc6a6) expressing leukemia cells. Here, we analyze multiple murine non-hematopoietic bone marrow single-cell RNA-sequencing datasets and discover that TauT expression is enriched in MSCs in vivo. Although

Taurine ameliorates viral encephalitis by restoring PRKN-mediated mitophagy.

Mitophagy is a selective type of autophagy that removes damaged mitochondria to maintain mitochondrial homeostasis and regulate the antiviral immune response. Despite increasing evidence that herpes simplex virus type 1 (HSV-1) infection causes mitochondrial damage, the regulatory mechanisms governing mitochondrial homeostasis and its biological implications in the context of HSV-1 infection and viral encephalitis remain unclear. In our recent work, we find that HSV-1 infection causes the accumulation of damaged mitochondria via defective mitophagy in vitro and in brain tissue of mice. The vir

Taurine Deficiency in Tissues Aggravates Radiation-Induced Gastrointestinal Syndrome.

Ionizing radiation administered for cancer treatment or from nuclear plant accidents are two common causes of radiation exposure. Ionizing radiation exposure generates reactive oxygen species and free radicals, which cause oxidative stress. We previously reported that taurine contributes to the recovery from radiation-induced injuries, suggesting its potential as a radioprotector and radiation mitigator. However, the effect of taurine on radiation-induced gastrointestinal syndrome remains poorly understood. The aim of this study was to examine the effect of taurine tissue depletion on radiatio

The molecular targets of taurine confer anti-hyperlipidemic effects.

Hyperlipidemia, an independent risk factor for atherosclerosis, is regarded as a lipid metabolism disorder associated with elevated plasma triglyceride and/or cholesterol. Genetic factors and unhealthy lifestyles, such as excess caloric intake and physical inactivity, can result in hyperlipidemia. Taurine, a sulfur-containing non-essential amino acid, is abundant in marine foods and has been associated with wide-ranging beneficial physiological effects, with special reference to regulating aberrant lipid metabolism. Its anti-hyperlipidemic mechanism is complex, which is related to many enzymes

Whole-Blood Taurine Concentrations in Cats With Intestinal Disease.

BACKGROUND: Increased delivery of taurine-conjugated bile acids to the distal bowel can lead to dysbiosis resulting in colitis in mouse models of inflammatory bowel disease. A similar situation also could occur in cats with intestinal disease and might therefore result in decreased whole-body taurine concentration. HYPOTHESIS/OBJECTIVES: To determine whether whole-blood taurine concentrations are decreased at the time of diagnosis in cats with intestinal disease and to correlate concentrations with clinical and laboratory variables. ANIMALS: Twenty-one cats with chronic inflammatory enteropa

Taurine ameliorates stress-induced degeneration of the urinary bladder.

We studied the potential effects of taurine, a free radical scavenger, on chronic water avoidance stress (WAS)-induced degeneration of the mucosa of the urinary bladder in experimental rats. Wistar albino rats were exposed to WAS for 2h/day, for 5 days (WAS group). Before exposing them to WAS, taurine (50mg/kg) (WAS+taurine group) was injected intraperitonally into the animals. Samples of urinary bladder were then investigated by light and scanning electron microscopy. Lipid peroxidation and gluthathione levels were also measured in the urinary bladder. In the WAS-only group, inflammatory cell
