Pax3 transcripts in melanoblast development.

Larvae of the South African clawed frog (Xenopus laevis) can regenerate the telencephalon, which consists of the olfactory bulb and the cerebrum, after it has been partially removed. Some authors have argued that the telencephalon, once removed, must be reconnected to the olfactory nerve in order to regenerate. However, considerable regeneration has been observed before reconnection. Therefore, we have conducted several experiments to learn whether or not reconnection is a prerequisite for regeneration. We found that the olfactory bulb did not regenerate without reconnection, while the cerebru

The Gli3 hypomorphic mutation Pdn causes selective impairment in the growth, patterning, and axon guidance capability of the lateral ganglionic eminence.

Previous studies have defined a requirement for Sonic hedgehog (Shh) signaling in patterning the ventral telencephalon, a major source of the neuronal diversity found in the mature telencephalon. The zinc finger transcription factor Gli3 is a critical component of the Shh signaling pathway and its loss causes major defects in telencephalic development. Gli3 is expressed in a graded manner along the dorsoventral axis of the telencephalon but it is unknown whether Gli3 expression levels are important for dorsoventral telencephalic patterning. To address this, we used the Gli3 hypomorphic mouse m

Is calcium deficiency related to thiamine-dependent neuropathy in pigeon?

In convulsive pigeons on a diet of polished rice, a significant decrease of thiamine and calcium in the central nervous system rather than in peripheral nervous system was observed. In subcellular fractions of telencephalon in these same pigeons, thiamine and calcium levels decreased in the myelin-membrane and synaptosomal fractions. When radioactive thiamine was injected into the pieon during a convulsion, radio-activity was most prominent in the telencephalon and in subcellular fractions, in the myelin-membrane and synaptosomal fractions of telencephalon immediately after recovery from the c

Identification and functional evidence of GABAergic neurons in parts of the brain of adult zebrafish (Danio rerio).

The distribution of GABA-containing neurons was studied in the brain of the adult zebrafish by Nissl staining and immunohistochemistry. GABA immunoreactivity (GABA-IR) was demonstrated in parts of the brain such as olfactory bulb (OB), telencephalon, tectum stratum, and in the hypothalamus. GABA-IR appeared in the area where Nissl-stained cell bodies were abundant. The internal cellular layer of the OB was most densely stained by Nissl staining, and also showed a high level of GABA-IR. The telencephalon and the hypothalamus revealed a similar pattern to the OB in terms of Nissl staining and GA

Afferent and efferent connections of the nucleus prethalamicus in the yellowfin goby Acanthogobius flavimanus.

The nucleus prethalamicus (PTh) receives fibers from the optic tectum and then projects to the dorsal telencephalon in the yellowfin goby Acanthogobius flavimanus. However, it remained unclear whether the PTh is a visual relay nucleus, because the optic tectum receives not only visual but also other sensory modalities. Furthermore, precise telencephalic regions receiving prethalamic input remained unknown in the goby. We therefore investigated the full set of afferent and efferent connections of the PTh by direct tracer injections into the nucleus. Injections into the PTh labeled cells in the

Proliferative Capacity of Adult Mouse Brain.

We studied cell proliferation in the postnatal mouse brain between the ages of 2 and 30 months and identified four compartments with different densities of proliferating cells. The first identified compartment corresponds to the postnatal pallial neurogenic (PPN) zone in the telencephalon; the second to the subpallial postnatal neurogenic (SPPN) zone in the telencephalon; the third to the white matter bundles in the telencephalon; and the fourth to all brain parts outside of the other three compartments. We estimated that about 3.4 million new cells, including 0.8 million in the subgranular zo

Brain Evolution: Intelligence without a Cortex.

Birds are capable of high level cognition even though their telencephalon is organized into nuclear groups rather than a six-layered cortex as in the mammalian brain. New data show that, despite their different macroscale organization, the circuitries of avian and mammalian telencephalon are fundamentally similar and may therefore carry out equivalent computations.

Distribution of the mRNA encoding the four dopamine D(1) receptor subtypes in the brain of the european eel (Anguilla anguilla): comparative approach to the function of D(1) receptors in vertebrates.

Four subtypes of D(1) dopamine receptors are expressed in the brain of the European eel (Anguilla anguilla), an elopomorph teleost. To correlate this molecular multiplicity with specific localisation and functions, the distribution of the D(1) receptor transcripts was analysed by in situ hybridisation. The four D(1) receptor transcripts exhibit largely overlapping expression territories. In telencephalon, they are found in the olfactory bulb and the dorsal telencephalon (except its lateral part) but are most abundant in the subpallial areas. More caudally, the entopeduncular nucleus, preoptic

Aprosopia/holoprosencephaly in a stillborn puppy: when the face predicts the brain.

In a litter of three puppies, one was stillborn and had facial and brain defects. Fusion of the maxilla and mandible and absence of the face were observed. The forebrain (telencephalon and the diencephalon) was reduced in size and fused, and the telencephalic longitudinal fissure, olfactory bulbs, and optic nerves were absent (Figures 6 and 7). Lissencephaly was observed in the telencephalon and cerebellum. A diagnosis of aprosopia/holoprosencephaly was made.

Oncogenic fusions converge on shared mechanisms in initiating astroblastoma.

Chromosomal rearrangements and gene fusions are the initial events in the development of many cancers. Astroblastoma (ABM), a brain cancer of unknown cellular origin and challenging to treat, is associated with diverse in-frame gene fusions, including MN1-BEND2 and MN1-CXXC5 (refs. 1,2). However, it remains unclear whether these gene fusions contribute to tumorigenesis. Here we show in mice that these two ABM-associated fusions converge on similar molecular activities and initiate malignancy specifically in ventral telencephalon neural progenitors. BEND2 and CXXC5 recognize similar DNA motifs,

Cerebral inducible nitric oxide synthase protein expression in microglia, astrocytes and neurons in Trypanosoma brucei brucei-infected rats.

To study the anatomo-biochemical substrates of brain inflammatory processes, Wistar male rats were infected with Trypanosoma brucei brucei. With this reproducible animal model of human African trypanosomiasis, brain cells (astrocytes, microglial cells, neurons) expressing the inducible nitric oxide synthase (iNOS) enzyme were revealed. Immunohistochemistry was achieved for each control and infected animal through eight coronal brain sections taken along the caudorostral axis of the brain (brainstem, cerebellum, diencephalon and telencephalon). Specific markers of astrocytes (anti-glial fibrill

Expression of extracellular matrix molecules in the embryonic rat olfactory pathway.

Primary olfactory neurons arise from placodal neuroepithelium that is separate from the neuroepithelial plate that forms the neural tube and crest. The axons of these neurons course along a stereotypical pathway and invade the rostral telencephalic vesicle where they induce the formation of the olfactory bulb. In the present study we examined the expression of several extracellular matrix constituents during formation of the olfactory nerve pathway in order to identify putative developmentally significant molecules. Double-label immunofluorescence was used to simultaneously map the trajectory

The hem of the embryonic cerebral cortex is defined by the expression of multiple Wnt genes and is compromised in Gli3-deficient mice.

In the developing vertebrate CNS, members of the Wnt gene family are characteristically expressed at signaling centers that pattern adjacent parts of the neural tube. To identify candidate signaling centers in the telencephalon, we isolated Wnt gene fragments from cDNA derived from embryonic mouse telencephalon. In situ hybridization experiments demonstrate that one of the isolated Wnt genes, Wnt7a, is broadly expressed in the embryonic telencephalon. By contrast, three others, Wnt3a, 5a and a novel mouse Wnt gene, Wnt2b, are expressed only at the medial edge of the telencephalon, defining the

Whole-brain atlas of the wall lizard, Hemidactylus flaviviridis: A histo-anatomical perspective.

The current study provides the first histological whole-brain atlas of the lizard, Hemidactylus flaviviridis. Macroscopic analysis of the lizard brain demonstrates its gross morphology, including various fissures and major divisions, forebrain, midbrain, and hindbrain. Serial coronal sections stained with cresyl violet provided a detailed insight into the architecture of the lizard brain with a well-defined ventricular system featuring lateral ventricles in the telencephalon, third ventricle in the diencephalon, and fourth ventricle in the rhombencephalon. The forebrain comprised telencephalon

[Histochemical study of the cholinesterases of various formations of the vertebrate endbrain].

Histochemical studies have been made on true (AChE) and false (BChE) cholinesterases in different parts of the telencephalon of amphibians, reptiles and mammals. The data obtained indicate that the enzymic localization depends on both the level of the development of the brain and phylogenetic age of its individual formations. In the telencephalon of Ambystoma mexicanum, high concentration of AChE was found in vascular-capillary network, in other investigated amphibians (Triturus vulgaris, Rana temporaria) diffuse distribution of AChE of different intensity was observed in ancient formations of

LHRH neuronal subtypes have multiple origins in chickens.

Previous studies indicate that LHRH neurons of the septal-preoptic area originate in the olfactory epithelium. In the current study, we found that LHRH neurons entering the telencephalon from the olfactory nerve were not continuous with LHRH neurons in the thalamus. In addition, ablation of the olfactory placode eliminated LHRH neurons in the telencephalon but did not eliminate LHRH neurons in the thalamus. We propose that there are at least two sites of LHRH neuron production, the olfactory epithelium and the diencephalon.

Forebrain organization in elasmobranchs.

It has long been known that many elasmobranch fishes have relatively large brains. The telencephalon, in particular, has increased in size in several groups, and as a percent of total brain weight, it is as large as in some mammals. Little is known, however, about the organization, connections, and functions of the telencephalon in elasmobranchs. Early experimental studies indicated that olfaction does not dominate the telencephalon and that other sensory modalities are represented, particularly in the pallium. We have investigated the intrinsic and extrinsic connections of the telencephalon i

Distribution and relative abundance of neurons in the pigeon forebrain containing somatostatin, neuropeptide Y, or both.

Immunohistochemical studies in several mammalian species and in red-eared turtles have shown that somatostatin (SS) and neuropeptide Y (NPY) co-occur in a substantial proportion of the telencephalic neurons containing either. To explore further the possibility that telencephalic neurons co-containing SS and NPY may be evolutionarily conserved among amniotes, we determined the distribution and co-occurrence of SS and NPY in forebrain neurons in pigeons. Single-label immunohistochemical studies revealed the presence of overlapping populations of SS+ neurons and NPY+ neurons in most of the major

Development of Thalamo-Telencephalic Circuits in a Reptile.

Neuronal circuits are a core component required for proper brain function and organization. How these circuits are built remains incompletely understood. The vast majority of studies on forebrain development have investigated mammals. Whether the described observations found in mammals apply to other vertebrates is unknown. The present report examined the development of forebrain circuits that interconnect the dorsal thalamus and the telencephalon in Alligator mississippiensis. A tracer, neurobiotin, which is transported in both an anterograde and retrograde fashion, was used. Four circuits we

The pattern of c-fos activation in the CNS is related to behavior in the mudskipper, Periophthalmus cantonensis.

The effects of three types of behavior on c-fos activation in different brain regions of the mudskipper, Periophthalmus cantonensis, were studied by immunocytochemistry. Animals were divided into four groups: The control group did not undergo any specific treatment. The second group consisted of animals that were agitated for an hour with a glass rod at an irregular speed. The third group contained animals showing aggressive behavior during an hour of monitoring, i.e. documented territorial hostility by raising of dorsal fins and pursuit of intruders. In contrast to these three groups, which h
