Identification and localization of telocyte-like cells in human umbilical cord stroma.

Human umbilical cord (hUC) connective tissue, known as Wharton's jelly, contains multipotent stromal cells (MSCs). Despite being considered the primary source of MSCs for regenerative purposes, hUC is one of the least studied fetal tissues. We conducted a detailed examination of normal hUCs (n = 16) and identified telocyte-like cells (TLCs) exhibiting both structural and phenotypical features similar to telocytes previously described in various tissues, including the placenta. TLCs were found to be concentrated around intervascular stromal clefts in the UC. These cells had thin, elongated bipo

Presence and significance of telocytes in cholelithiasis and biliary dilatation in benign biliary disorders.

Telocytes are closely associated with the regulation of tissue smooth muscle dynamics in digestive system disorders. They are widely distributed in the biliary system and exert their influence on biliary motility through mechanisms such as the regulation of CCK and their electrophysiological effects on smooth muscle cells. To investigate the relationship between telocytes and benign biliary diseases, such as gallbladder stone disease and biliary dilation syndrome, we conducted histopathological analysis on tissues affected by these conditions. Additionally, we performed immunohistochemistry and

Morphological characteristics and distribution identification of telocytes in Tibetan sheep testis and epididymis.

Telocytes (TCs) are a type of stromal cell discovered in the various organs of different animals and have many potential functions, including angiogenesis, signalling, and substance transport. However, the TCs have not been detected in the testis or epididymis of Tibetan sheep. This study investigated the position, characteristics, and distribution of TCs in the testis and epididymis of Tibetan sheep using transmission electron microscopy (TEM), toluidine blue staining, immunohistochemistry, and double immunofluorescence to elucidate their possible functions. TEM revealed that TCs were often f

Immunohistochemical and ultrastructural identification of telocytes in the lamina propria of human vaginal mucosa.

Since their relatively recent discovery, telocytes (TCs) have been described as peculiar cells strategically positioned in the stromal tissue component of multiple organ systems of the mammalian body including female reproductive organs (i.e., ovary, uterine tube, and uterus). Nevertheless, current knowledge of TCs in the vagina is very limited. The present study was therefore undertaken to investigate the existence and characteristics of TCs in the stromal tissue of human vaginal mucosa by means of immunohistochemistry, immunofluorescence confocal microscopy, and transmission electron microsc

Distribution and Ultrastructural Features of Telocytes in the Pars Distalis of the Rat Pituitary Gland.

Telocytes (TCs), a novel type of interstitial cells, are characterized by their smaller cellular body and extremely long, thin processes which are called telopodes (Tps). They have been described in multiple organs from diverse animals. Currently, the existence of TCs in rat pars distalis (PD) has remained unexplored. This investigation was undertaken to visualize the distribution and structural features of TCs in the PD using immunofluorescence (IF) and further validated by transmission electron microscopy (TEM). HE staining revealed the presence of interstitial cells in the peri-sinusoidal v

Regulatory roles of Osteopontin in lung epithelial inflammation and epithelial-telocyte interaction.

BACKGROUND: Lung epithelial cells play important roles in lung inflammation and injury, although mechanisms remain unclear. Osteopontin (OPN) has essential roles in epithelial damage and repair and in lung cancer biological behaviours. Telocyte (TC) is a type of interstitial cell that interacts with epithelial cells to alleviate acute inflammation and lung injury. The present studies aim at exploring potential mechanisms by which OPN regulates the epithelial origin lung inflammation and the interaction of epithelial cells with TCs in acute and chronic lung injury. METHODS: The lung disease sp

Telocytes are the physiological counterpart of inflammatory fibroid polyps and PDGFRA-mutant GISTs.

PDGFRA mutations in the gastrointestinal (GI) tract can cause GI stromal tumour (GIST) and inflammatory fibroid polyp (IFP). Hitherto no cell type has been identified as a physiological counterpart of the latter, while interstitial Cajal cells (ICC) are considered the precursor of the former. However, ICC hyperplasia (ICCH), which strongly supports the ICC role in GIST pathogenesis, has been identified in germline KIT-mutant settings but not in PDGFRA-mutant ones, challenging the precursor role of ICC for PDGFRA-driven GISTs. Telocytes are a recently described interstitial cell type, CD34+/PDG

Live-Cell Imaging of Telocyte-like Cells in Primary Liver Cell Cultures.

Telocytes are small mesenchymal cells with unknown hepatic functions. In primary cultures of rat hepatocytes and non-parenchymal liver cells (NPLC), we observed, at a very low incidence, small-sized, highly motile cells. Because these cells fulfill ultrastructural/morphological 'telocytes hallmarks', foremost their minuteness and the presence of very thin telopodia, we refer to them as hepatic telocyte-like cells (hTCLs). Unique among hepatic cells and beyond these telocyte characteristics is their high motility and pronounced cell plasticity. Surprisingly, hTCLs display an exceptional explora

Impairment in the telocyte/CD34+ stromal cell network in human rheumatoid arthritis synovium.

Telocytes (TCs)/CD34+ stromal cells have recently emerged as peculiar interstitial cells detectable in a variety of organs throughout the human body. TCs are typically arranged in networks establishing unique spatial relationships with neighbour cells and likely contributing to the maintenance of tissue homeostasis by both cell-to-cell contacts and releasing extracellular vesicles. Hence, TC defects are being increasingly reported in different pathologies, such as chronic inflammatory and fibrotic conditions. In this regard, TCs/CD34+ stromal cells have been shown to constitute an intricate in

Adaptive changes of telocytes in the urinary bladder of patients affected by neurogenic detrusor overactivity.

Urinary bladder activity involves central and autonomic nervous systems and bladder wall. Studies on the pathogenesis of voiding disorders such as the neurogenic detrusor overactivity (NDO) due to suprasacral spinal cord lesions have emphasized the importance of an abnormal handling of the afferent signals from urothelium and lamina propria (LP). In the LP (and detrusor), three types of telocytes (TC) are present and form a 3D-network. TC are stromal cells able to form the scaffold that contains and organizes the connective components, to serve as guide for tissue (re)-modelling, to produce tr

Telocytes/CD34+ Stromal Cells in the Normal, Hyperplastic, and Adenomatous Human Parathyroid Glands.

Telocytes/CD34+ stromal cells (TCs/CD34+ SCs) have been studied in numerous organs and tissues, but their presence and characteristics in the parathyroid glands have not been explored. Using immunological and ultrastructural procedures, we assess the location, arrangement, and behavior of TCs/CD34+ SCs in normal human parathyroids, during their development and in their most frequent pathologic conditions. In normal parathyroids, TCs/CD34+ SCs show a small somatic body and long thin processes with a moniliform aspect, form labyrinthine systems, connect other neighboring TCs/CD34+ SCs, vessels,

Oviductal Oxygen Homeostasis in Patients with Uterine Myoma: Correlation between Hypoxia and Telocytes.

Oxygen balance is crucial for angiogenesis, immunity, and tissue repair. The human oviduct is essential for reproductive function, and any imbalance in homeostasis leads to fertility disturbances and might be a reason for ectopic pregnancy development. Uterine myoma is a widespread benign tumour, which is often accompanied by infertility. Telocytes have been discussed in the contexts of motility, fibrosis development, and angiogenesis. We observed the oviducts from patients with and without uterine myoma, comparing the expression of HIF-1, HO, VEGF and its receptor, NOS, oestrogen, and progest

Identification of PDGFRα+ cells in uterine fibroids - link between angiogenesis and uterine telocytes.

INTRODUCTION: Telocytes (TCs), also called interstitial Cajal-like cells (ICLC), CD34+ cells or PDGFRα+ cells (platelet-derived growth factor receptor α positive cells), a new type of cell of mesenchymal origin, were described over one decade ago. The unique nature of these cells still deserves attention from the scientific community. Telocytes make homo- and heterocellular contact with myocytes, immunocytes and nerves, have their own immunohistochemical and secretome profiles and thus might regulate local regenerative processes including angiogenesis and fibrosis. The aim of our study was to

Lymphatic lacunae of the human eye conjunctiva embedded within a stroma containing CD34+ telocytes.

An accurate identification of telocytes (TCs) was limited because of the heterogeneity of cell types expressing the markers attributed to TCs. Some endothelial lineage cells also could fit within the pattern of TCs. Such endothelial cells could line conjunctival lacunae previously assessed by laser confocal microscopy. We have been suggested that an accurate distinction of TCs from endothelial cells in the human eye conjunctiva could be achieved by use of CD31, CD34 and D2-40 (podoplanin); and that the conjunctival lacunae are in fact lymphatic. We aimed as testing the hypothesis by an immunoh

Evaluation of Telocytes in the Ovary, Oviduct and Uterus of Kyrgyz Mares According to Pregnancy Status.

In the current study, we aimed to determine the distribution of telocytes in the ovary, oviduct and uterus of Kyrgyz mares depending on whether they were pregnant. We also studied the progression during the first 6 months of pregnancy. To this end, ovary, oviduct and uterus tissues from 53 Kyrgyz mares were obtained post-mortem. The mare tissue samples were divided into two groups: pregnant (n = 43) and non-pregnant (n = 10). Telocyte levels in the tissues were determined using light microscopic and immunohistochemical methods. Subsequently, the data acquired were evaluated based on whether th

Immunohistochemical study on the distribution of telocytes in the knee joint components in a rat osteoarthritis model.

[Purpose] Telocytes are stromal cells that participate in tissue homeostasis. Osteoarthritis is a common degenerative disorder of multiple joint components that causes inflammation; however, the distribution of telocytes in joint components and the impact of osteoarthritis on telocytes is unclear. Therefore, we aimed to clarify the distribution of the telocyte in the joint components and determine the effect of osteoarthritis on telocytes. [Participants and Methods] We divided 30 male rats into control and osteoarthritis groups and surgically induced osteoarthritis by destabilizing the medial

Structural Investigation of Epididymal Microvasculature and Its Relation to Telocytes and Immune Cells in Camel.

The vascular and perivascular cells, including telocytes (TCs) and immune cells, play an important role in male fertility. The current study intended to describe in detail the microvascular structures harboring special regulatory devices in addition to the interstitial cellular components of the one-humped camel epididymis. The samples were collected from 10 clinically healthy mature camels (Camelus dromedarius). The distribution and characteristics of TCs, peripheral blood vessels of the epididymis, and immune cells were investigated using the light, immunohistochemistry, immunofluorescence,

Aberrations in the female reproductive organs and a role of telocytes in a rat model of anorexia nervosa.

BACKGROUND: Anorexia nervosa is a widely prevalent eating disorder that o en leads to life-threatening complications. Since it mostly concerns females, many authors have focused on studying the reproductive system in anorexic women. Recently discovered telocytes may give a new insight into the pathophysiology of gynecological complications in these patients. MATERIAL AND METHODS: We adopted an animal model of anorexia nervosa induced by voluntary physical activity. Sixteen female Wistar rats were divided into two groups: control and activity-based anorexia. When the weight loss of activity-ba

Telocytes in normal and keratoconic human cornea: an immunohistochemical and transmission electron microscopy study.

Telocytes (TC) are typically defined as cells with telopodes by their ultrastructural features. Their presence was reported in the interstitium of various organs in vertebrates, including humans. However, no study has yet described the presence of TC in the human eye and in particular, within the stromal compartment of the cornea. To address this issue, samples of normal and pathologic (keratoconic) human corneas were tested by immunohistochemistry for CD34, platelet-derived growth factor receptor α (PDGFRα) and c-kit/CD117 or examined by transmission electron microscopy. We found that TC coex
