Interactions of salicylate, dietary zinc, and genetic strain in teratogenesis in rats.

The influence of dietary zinc concentration on salicylate teratogenesis was studied in Wistar and Sprague-Dawley rats. Females were fed purified diets containing 0.4, 4.5, 9, 100, or 1,000 micrograms zinc/gm diet, or a stock diet (Purina Rat Chow) from day zero to day 21 of gestation, when they were killed and the fetuses were examined. On day 9, rats were given saline or 250, 500, or 750 mg sodium salicylate/kg body weight by gavage. Increasing drug dose caused increased frequency of malformed or resorbed fetuses, while increasing dietary zinc reduced the teratogenic effects of salicylate, bu

Concurrent dietary administration of D-SAL and ethanol diminishes ethanol's teratogenesis.

BACKGROUND: SAL (SALLRSIPA) is a peptide fragment of activity-dependent neurotrophic factor. Both L- and D-SAL diminish ethanol's pathogenesis, however, the D-peptide is protease resistant, and can therefore be effectively administered in a diet. The present study tested the hypothesis that D-SAL provided in a liquid diet containing ethanol will prevent ethanol-induced teratogenicity in mice. METHODS: Following an ethanol acclimation period, female C57Bl/6J mice were withdrawn from the ethanol, bred, and then returned during gestational days (GD) 7 and 8 to a control liquid diet or one contai

Clinical and experimental studies linking oxidative metabolism to phenytoin-induced teratogenesis.

The research presented in this article concerns the proposed mechanism of phenytoin-induced teratogenicity that focuses on oxidative metabolites as sources of reactive species in clinical studies and by testing paradigms in animal models. The clinical aspect involved determining whether at-risk fetuses could be detected prenatally on the basis of low or deficient epoxide hydrolase activity. In 19 pregnancies monitored by amniocentesis, we predicted an adverse outcome in four infants on the basis of low enzyme activity. When examined neonatally, all four infants had the dysmorphic features of t

Teratogenesis and low copper status resulting from D-penicillamine in rats.

The teratogenicity of D-penicillamine was studied using the Sprague-Dawley rat. Fetuses from rats fed diets containing 0.83% D-penicillamine during pregnancy had a high incidence of malformations. Maternal and fetal tissue copper levels were significantly lower in the D-penicillamine group than in controls, suggesting that the teratogenicity of the drug may be due in part to the induction of copper deficiency by its chelating properties.

Kissing as an evolutionary adaptation to protect against Human Cytomegalovirus-like teratogenesis.

Mouth to mouth sexual kissing is seen in more than 90% of human cultures. Various theories have been put forward to account for this but none offer a full explanation within an evolutionary framework. As mouth to mouth sexual kissing exposes each participant to the diseases of the other, it must confer significant benefit. Human Cytomegalovirus (HCMV) is a ubiquitous infection that carries a severe teratogenic risk if primary infection is acquired during certain critical periods. As HCMV is present in salivary gland epithelial cells and sheds from periodontitis induced lesions, female inoculat

Perinatal programming and functional teratogenesis: impact on body weight regulation and obesity.

It is increasingly accepted that alterations of the intrauterine and early postnatal nutritional, metabolic, and hormonal environment may cause predispositions for the development of diseases in later life. Studies in the offspring of diabetic mothers have decisively contributed to this perception. Alterations of the fetal and neonatal environment which offspring of diabetic mothers 'experience' seem to program a disposition to develop obesity, diabetes mellitus and Syndrome X-like alterations throughout later life. Underweight at birth is also suggested to lead to an increased risk of Syndrom

The influence of dietary folate supplementation on the incidence of teratogenesis in zinc-deficient rats.

Two studies were conducted to investigate the possibility that pteroylmonoglutamic acid supplementation would alleviate teratogenesis in zinc-deficient rats. Pregnant rats of the Wistar strain were fed on Zn-deficient (less than 0.5 mg Zn/kg) or Zn-supplemented (75 or 95 mg Zn/kg) diets from mating until day 18.5 of gestation. The basal level of pteroylmonoglutamic acid added to all diets (0.56 mg/kg) was supplemented with 30-200 mg/kg in selected diets. Dietary Zn deprivation resulted in fetal resorption, fetal growth retardation and reduced concentrations of Zn in fetuses and maternal plasma

An exploration of the role of hydroxyurea injection time in fetal growth and teratogenesis in rats.

Pregnant Wistar rats were injected with hydroxyurea (HU) intraperitoneally (IP) t one of several 6-hour intervals on days 9-10.75 to study the role of circadian growth variations in the teratologic response of the fetuses. Two stocks of rats were studied and the results in each compared. The fetal response to HU, as observed at day 21, was not detectably modified by circadian fetal growth variation. No correlations between hour of HU administration and fetal weight, placental weight, resorption, or total malformation rates were found. Cyclic variations in the incidence of hydronephrosis and le

Teratogenesis of acetazolamide in the CBA/J and SWV strains of mice. I. Teratology.

SWV mice were totally resistant to the teratogenic and embryolethal actions of acetazolamide. The time of maximal sensitivity to acetazolamide-induced ectrodactyly in the CBA/J strain was the middle of day 10; the dose response at this time was studied. Comparison of the responses of the two strains and reciprocal hybrids indicated that sensitivity is a property of the embryo and is not maternally mediated. SWV mice were also resistant to dichlorphenamide which suggests they may be resistant to many or even all teratogenic carbonic anhydrase inhibitors.

Quantitative trait locus mapping for ethanol teratogenesis in BXD recombinant inbred mice.

BACKGROUND: Individual differences in susceptibility to the detrimental effects of prenatal ethanol (EtOH) exposure have been demonstrated. Many factors, including genetics, play a role in susceptibility and resistance. We have previously shown that C57BL/6J (B6) mice display a number of morphological malformations following an acute dose of EtOH in utero, while DBA/2J (D2) mice are relatively resistant. Here, we present the results of quantitative trait locus (QTL) mapping for EtOH teratogenesis in recombinant inbred strains derived from a cross between B6 and D2 (BXD RIs). METHODS: Pregnant

Zika Virus: A Systematic Review of Teratogenesis, Congenital Anomalies, and Child Mortality.

Zika virus infection (ZIKV) was one of the most catastrophic epidemics. ZIKV in nonpregnant women is mild and sometimes asymptomatic. However, infection during pregnancy leads to congenital malformations in the fetus, while maternal signs of infection are preceded by a rash. The maternal-fetal infection begins with a rash that occurs early during pregnancy. The most severe pathologies were related to the first trimester of gestation, including microcephaly, musculoskeletal, genitourinary, craniofacial, ocular, and pulmonary manifestations. The prognosis may not be encouraging. Herd immunity in

Xenopus embryos to study fetal alcohol syndrome, a model for environmental teratogenesis.

Vertebrate model systems are central to characterize the outcomes of ethanol exposure and the etiology of fetal alcohol spectrum disorder (FASD), taking advantage of their genetic and morphological closeness and similarity to humans. We discuss the contribution of amphibian embryos to FASD research, focusing on Xenopus embryos. The Xenopus experimental system is characterized by external development and accessibility throughout embryogenesis, large clutch sizes, gene and protein activity manipulation, transgenesis and genome editing, convenient chemical treatment, explants and conjugates, and

Fishing for Fetal Alcohol Spectrum Disorders: Zebrafish as a Model for Ethanol Teratogenesis.

Fetal Alcohol Spectrum Disorders (FASD) describes a wide array of ethanol-induced developmental defects, including craniofacial dysmorphology and cognitive impairments. It affects ∼1 in 100 children born in the United States each year. Due to the pleiotropic effects of ethanol, animal models have proven critical in characterizing the mechanisms of ethanol teratogenesis. In this review, we focus on the utility of zebrafish in characterizing ethanol-induced developmental defects. A growing number of laboratories have focused on using zebrafish to examine ethanol-induced defects in craniofacial,

Embryonic oxidative stress as a mechanism of teratogenesis with special emphasis on diabetic embryopathy.

Reactive oxygen species (ROS) are involved in the etiology of numerous diseases including cardio-vascular diseases and diabetes mellitus. There is evidence that several teratogens affect the developing embryo by increasing its oxidative stress and, because of its relatively weak antioxidant defense, especially at the early stages of organogenesis, result in severe embryonic damage. This mechanism seems to operate in diabetes-induced embryonic damage as well as in the mechanism of teratogenicity caused by ionizing radiation, hypoxia, alcohol and cocaine use and cigarette smoking. We studied the

Teratogen update: carcinogenesis and teratogenesis associated with exposure to diethylstilbestrol (DES) in utero.

Exposure of the human fetus to physician-prescribed diethylstilbestrol and other synthetic estrogens (collectively referred to as "DES") led to an important iatrogenic epidemic. In the United States alone, at least four million fetuses and their mothers had a substantial exposure to these estrogens now known to be mild carcinogens and potent teratogens. Mothers exposed to DES may have a somewhat higher risk of breast cancer than women who were not exposed. The sequelae of in utero exposure of daughters include clear cell adenocarcinoma of the vagina and cervix, various gross anomalies of the g

In vivo evaluation of teratogenesis and cytogenetic changes following methylmercuric chloride treatment.

Mercury is a major environmental pollutant and a proven teratogen in man and animals. Its teratogenicity and effects on fetal chromosomes were investigated in mice. Various dose levels of methylmercuric chloride (MMC) were administered via an intragastric tube to pregnant ICR Swiss/Webster mice on day 9 of gestation. On day 18 of gestation the animals were killed and the fetuses were removed. Fetal lung and liver tissues were processed for cytogenetic studies. Fetuses were also fixed in Bouin's solution for subsequent teratological examination by using Wilson's technique. Mercury levels were d

Teratologic studies on the Himalayan rabbit: new aspects of thalidomide-induced teratogenesis.

The aim of our study was to determine the period of maximum sensitivity for the induction of characteristic malformations with thalidomide (TH) in Himalayan rabbits. TH was administered orally in different doses (50, 100, 150 and 200 mg/kg) four times at 24-h intervals starting at 192 h of gestation. The malformations affected various organs: renal defects (dysplasia) and limb anomalies (dysmelia)--which had never occurred spontaneously in this strain--appeared as dose-dependent effects of the drug. By administering single doses of TH (200 and 300 mg/kg body wt) between hours 192 and 264 of ge
