A key volatile infochemical that elicits a strong olfactory response of the predatory mite Neoseiulus californicus, an important natural enemy of the two-spotted spider mite Tetranychus urticae.

Herbivore-induced plant volatiles (HIPVs) emitted from lima bean leaves infested with the two-spotted spider mites Tetranychus urticae strongly attract the predatory mites Neoseiulus californicus. Among these HIPVs, methyl salicylate and linalool can attract the predators. Three green-leaf volatiles (GLVs) of (Z)-3-hexen-1-ol, (Z)-3-hexenyl acetate and (E)-2-hexenal, found in the odor blends from T. urticae-infested leaves and physically damaged leaves, can also attract the predators. To search for a strong predator attractant, the olfactory responses of N. californicus to each synthetic compo

Mechanism of action of soybean oil in arthropods: mortality results from tracheal infiltration and obstruction of gas exchange structures.

Oils are widely used in arthropod pest control, yet their modes of action remain poorly understood and often differ across studies. To investigate the mechanism of action of a commercial soybean oil formulation, we evaluated its physical and toxicological effects on three arthropod species across several developmental stages: adults and nymphs of Uroleucon sonchi (L.) (Hemiptera: Aphididae), adults and eggs of Tetranychus urticae Koch (Trombidiformes: Tetranychidae), and larvae and eggs of Plutella xylostella (L.) (Lepidoptera: Plutellidae). Adults, immature stages, and eggs were either topica

Microbial diversity and cytoplasmic incompatibility in Tetranychus urticae Spanish populations.

The two-spotted spider mite, Tetranychus urticae Koch (Acari: Tetranychidae), is an important citrus pest worldwide. Global populations of T. urticae harbour Wolbachia and other bacterial species, some of which are considered endosymbionts. The genus Wolbachia encompasses strict intracellular bacteria that cause several types of reproductive distortions in their hosts, such as male feminization, thelytokous parthenogenesis, cytoplasmic incompatibility, or male killing. The presence of different Wolbachia strains can affect the mite's reproductive parameters, which can have important implicatio

Knockout of nAChR subunits in spider mites and their phytoseiid predators confers spinosyn cross-resistance and reveals a conserved mode of action in mites.

Spinosyns are allosteric modulators of nicotinic acetylcholine receptors (nAChRs) which in insects specifically target subunit α6. However, their mode of action in mites and compatibility with phytoseiid predators remain unclear. We combined phylogenetics with CRISPR/Cas-based reverse genetics to test whether α6-like subunits mediate spinosyn toxicity in mites and to assess prospects for resistance breeding in phytoseiids. The phylogenetic analysis identified seven α and three β subunits in multiple phytoseiids and in Tetranychus urticae. A single phytoseiid subunit clustered within the insect

Synthesis and Anti-insect Activity of Triazole-Containing Amide Derivatives.

The outbreak of crop diseases and insect pests, especially hemipteran piercing insects, poses a major threat to global food security. This study was designed to synthesize a series of novel flonicamid (FL) derivatives using an active substructure splicing strategy. Their insecticidal activities were evaluated against key pests, including Myzus persicae, Bemisia tabaci and Tetranychus urticae. The LC50 values of compounds 3ea, 3fa, 3ga, and 5cab against B. tabaci were 69.8 mg/L, 186 mg/L, 91.9 mg/L, and 112 mg/L, respectively. For M. persicae, the LC50 values of compounds 3aa, 5gaa, and 5fab we

Discovery of Acaricidal, Insecticidal, and Fungicidal Candidates Inspired by Natural Ethyl Cinnamate Compounds Isolated from Polygonum orientale L.

As a continuation of our research on the pesticide development of Polygonum orientale L., the chemical constituents of the seeds of P. orientale were systematically investigated. Eleven natural compounds (PO-1 to PO-11) were isolated from the EtOAc extract of P. orientale. Notably, compound PO-9 and its dimeric compound PO-10 were first isolated from P. orientale and possessed excellent acaricidal activity against Tetranychus cinnabarinus. With PO-9 and PO-10 as the lead compounds, two series of cinnamate derivatives were further synthesized, and their acaricidal, insecticidal, and fungicidal

Analysis of the relationship between P-glycoprotein and abamectin resistance in Tetranychus cinnabarinus (Boisduval).

Abamectin is an effective acaricide and widely used in the control of Tetranychus cinnabarinus. With the increase of control failures, it is however important to clarify the resistance mechanism to improve the control of this mite. P-glycoprotein (Pgp) is an ATP-dependent drug efflux pump for xenobiotic compounds and is involved in multidrug resistance. In this study, the results showed that verapamil, the specific inhibitor of Pgp, could enhance the lethal effect of abamectin on mites, and this effect is more enhanced in abamectin-resistant strain (AbR, mortality increased 74.51%) than that i

Efficacy and activity prediction by molecular topology of new drugs against the Tetranychus urticae plague.

Tetranychus urticae Koch is an important pest affecting citrus, for which biological control has not yet been achieved; therefore, acaricides are commonly used instead. The goal of the work reported in this paper was to measure the efficacy of different new compounds--uracil derivatives--on this mite and conduct a quantitative structure-activity relationship (QSAR) study based on the results obtained, in order to set up a model capable of predicting the acaricidal activity of further new compounds. Some of the tested new products proved highly effective against T. urticae. Besides, topological

Variation in herbivory-induced volatiles among cucumber (Cucumis sativus L.) varieties has consequences for the attraction of carnivorous natural enemies.

In response to herbivory by arthropods, plants emit herbivory-induced volatiles that attract carnivorous enemies of the inducing herbivores. Here, we compared the attractiveness of eight cucumber varieties (Cucumis sativus L.) to Phytoseiulus persimilis predatory mites after infestation of the plants with herbivorous spider mites (Tetranychus urticae) under greenhouse conditions. Attractiveness differed considerably, with the most attractive variety attracting twice as many predators as the least attractive variety. Chemical analysis of the volatiles released by the infested plants revealed si

Jasmonic acid is a key regulator of spider mite-induced volatile terpenoid and methyl salicylate emission in tomato.

The tomato (Lycopersicon esculentum) mutant def-1, which is deficient in induced jasmonic acid (JA) accumulation upon wounding or herbivory, was used to study the role of JA in the direct and indirect defense responses to phytophagous mites (Tetranychus urticae). In contrast to earlier reports, spider mites laid as many eggs and caused as much damage on def-1 as on wild-type plants, even though def-1 lacked induction of proteinase inhibitor activity. However, the hatching-rate of eggs on def-1 was significantly higher, suggesting that JA-dependent direct defenses enhanced egg mortality or incr

Effects of a short-term heat stress on the functional response of Scolothrips longicornis to Tetranychus urticae.

Biological control employing predatory insects represents an environmentally sustainable approach for suppressing two-spotted spider mite populations in both greenhouse and open-field cropping systems. In the present study, a laboratory experiment was performed to assess the functional response and predation efficiency of the predatory thrips Scolothrips longicornis (Thysanoptera: Thripidae) feeding on eggs of the two-spotted spider mite, Tetranychus urticae (Acari: Tetranychidae), with particular emphasis on the influence of short-term heat stress. Functional response assays were conducted ov

Design, Synthesis, Acaricidal Activity, Structure-Activity Relationship, and Molecular Docking of Novel Piperidine Amine Derivatives Containing a Pyrimidine Motif.

Tetranychus cinnabarinus, a prominent mite in agricultural crops, has developed escalating resistance as a result of extensive and repeated chemical pesticide application. Consequently, it remains a quite urgent goal to identify novel structures with high efficacy to reduce resistance. In this study, a series of novel piperidine pyrimidinamine derivatives were designed and synthesized by employing the intermediate derivatization method (IDM) and then screened for their acaricidal activity. Bioassays indicated that most of the target compounds exhibited some acaricial activities against T. cinn

Plant, pest and predator interplay: tomato trichomes effects on Tetranychus urticae (Koch) and the predatory mite Typhlodromus (Anthoseius) recki Wainstein.

Trichomes are well-known efficient plant defense mechanisms to limit arthropod herbivory, especially in Solanaceae. The present study aims to evaluate the impact of trichome types on the development, survival and dispersal of Tetranychus urticae, and the phytoseiid predatory mite Typhlodromus (Anthoseius) recki. Six Solanum lycopersicum cultivars and two wild Solanum species, S. cheesmaniae and S. peruvianum, presenting contrasting densities and types of trichomes, were considered. Cultivars and species were characterized by counting each trichome type on leaves, petioles and stems. Mites stuc

Contrasting defence mechanisms against spider mite infestation in cyanogenic and non-cyanogenic legumes.

Understanding the complex interactions between plants and herbivores is essential for improving crop resistance. Aiming to expand the role of cyanogenesis in plant defence, we investigated the response of the cyanogenic Phaseolus lunatus (lima bean) and the non-cyanogenic Phaseolus vulgaris (common bean) to Tetranychus urticae (spider mite) infestation. Despite mite infesting both legumes, leaf damage infringed by this feeder was reduced in lima bean. Comparative transcriptome analyses revealed that both species exhibited substantial metabolic and transcriptional changes upon infestation, alth

Isolation and Characterization of New 16-Membered Macrolides from the aveA3 Gene Replacement Mutant Strain Streptomyces avermitilis TM24 with Acaricidal and Nematicidal Activities.

Polyketides represent an important class of biologically active and structurally diverse compounds found in nature. They are biosynthesized from acyl CoA precursors by polyketide synthases (PKSs). The use of combinatorial biosynthesis to form hybrid PKSs is considered to be an excellent approach for the development of novel polyketides. Here, 10 new 16-membered macrolide compounds were isolated from the broth of the genetically engineered strain Streptomyces avermitilis TM24, in which the PKS gene aveA3 was seamlessly replaced by the milbemycin PKS gene milA3. Their structures were elucidated

Photo-enzymatic repair of UVB-induced DNA damage in the two-spotted spider mite Tetranychus urticae.

Ambient ultraviolet-B (UVB) radiation induces lethal effects in the two-spotted spider mite Tetranychus urticae, whereas photoreactivation by irradiation with ultraviolet-A and visible light (VIS) plays an important role to increase survival of mites irradiated by UVB. The physiological mechanisms and ecological significance of photoreactivation in terrestrial arthropods have not been shown clearly. We verified the biological impact and accumulation of DNA lesions by UVB irradiation and the repair of them by photoreactivation in T. urticae larvae. Survival of UVB-irradiated larvae decreased wi

Significance of terpenoids in induced indirect plant defence against herbivorous arthropods.

Many plants respond to herbivory by arthropods with an induced emission of volatiles such as green leaf volatiles and terpenoids. These herbivore-induced plant volatiles (HIPVs) can attract carnivores, for example, predators and parasitoids. We investigated the significance of terpenoids in attracting herbivores and carnivores in two tritrophic systems where we manipulated the terpenoid emission by treating the plants with fosmidomycin, which inhibits one of the terpenoid biosynthetic pathways and consequently terpenoid emission. In the 'lima bean' system, volatiles from spider-mite-infested f

Phenotypic plasticity of cyanogenesis in lima bean Phaseolus lunatus-activity and activation of beta-glucosidase.

Cyanogenesis, the release of toxic HCN from damaged plant tissues, is generally considered as a constitutive plant defense. We found phenotypic plasticity of cyanogenesis in young leaves of lima bean Phaseolus lunatus based on increased activity of the beta-glucosidase in response to herbivore attack. Two aspects of plant cyanogenesis have to be considered in ecological analyses: (1) the cyanogenic potential (HCNp), which indicates the total amount of cyanide-containing compounds present in a given tissue, and (2) the cyanogenic capacity (HCNc), representing the release of HCN per unit time. A novel spider venom peptide from the predatory mite Neoseiulus barkeri shows lethal effect on phytophagous pests.

The long-term use of pesticides in the field, and the high fertility and adaptability of phytophagous mites have led to resistance problems; consequently, novel safe and efficient active substances are necessary to broaden the tools of pest mite control. Natural enemies of arthropods typically secrete substances with paralytic or lethal effects on their prey, and those substances are a resource for future biopesticides. In this study, two putative venom peptide genes were identified in a parasitic mite Neoseiulus barkeri transcriptome. Recombinant venom NbSP2 peptide injected into Tetranychus
