Pulmonary cold sensing through Club cells triggers adipose thermogenesis to ameliorate obesity.

Cold exposure is a well-known trigger for thermogenesis, primarily through skin cold-sensitive neurons activating adipose metabolism via the central nervous system (CNS). However, whether the lungs-also exposed to cold air-function as a cold sensor and regulate metabolism remains unknown. Here, using CC10-Cre- and adeno-associated virus (AAV)-based mouse models, we uncover a pulmonary cold sensing system that drives thermogenesis, establishing a peripheral thermoregulatory mechanism. Specifically, we identify KCNK2 channels in Club cells within the bronchial epithelium as primary pulmonary col

Growth hormone enhances mitochondria biogenesis and endows mitochondrial thermogenesis in murine adipocytes.

Growth hormone (GH) can reduce the size of white adipocytes in vivo and in vitro. This catabolic effect was previously believed to be related to the breakdown of stored triglycerides into free fatty acids. However, little is known about its effects on mitochondria in adipose or adipocytes. We thus examined effects of GH on mitochondrial number change and metabolic changes in fat of diet induced obese mice as well as 3T3-L1 adipocytes. We first found in vitro GH increase the density of mitochondria in 3T3-L1 adipocytes. We figured out that GH enhanced the expression of mitochondria biogenesis r

Perinatal Vitamin D Deficiency Enhances Brown Adipose Tissue Thermogenesis in Weanling Rats.

Perinatal vitamin D (Vit. D) deficiency (VDD) disrupts the development of key tissues involved in metabolic regulation, including the endocrine pancreas, white adipose tissue, and skeletal muscle. Brown adipose tissue (BAT), essential for thermoregulation and energy homeostasis, may also be affected, but the impact of perinatal VDD on BAT physiology remains unclear. In this study, forty female Wistar rats were fed either a standard AIN93G diet (1000 IU Vit. D3/kg; control group, CT) (n = 20) or a modified AIN93G diet lacking Vit. D (VDD group) (n = 20) for six weeks prior to conception and thr

Mild Hyperthermia-Induced Thermogenesis in the Endoplasmic Reticulum Defines Stress Response Mechanisms.

Previous studies reported that a mild, non-protein-denaturing, fever-like temperature increase induced the unfolded protein response (UPR) in mammalian cells. Our dSTORM super-resolution microscopy experiments revealed that the master regulator of the UPR, the IRE1 (inositol-requiring enzyme 1) protein, is clustered as a result of UPR activation in a human osteosarcoma cell line (U2OS) upon mild heat stress. Using ER thermo yellow, a temperature-sensitive fluorescent probe targeted to the endoplasmic reticulum (ER), we detected significant intracellular thermogenesis in mouse embryonic fibrobl

Adipose tissue-specific ablation of PGC-1β impairs thermogenesis in brown fat.

Impaired thermogenesis observed in mice with whole-body ablation of peroxisome proliferator-activated receptor-γ coactivator-1β (PGC-1β; officially known as PPARGC1B) may result from impaired brown fat (brown adipose tissue; BAT) function, but other mechanism(s) could be involved. Here, using adipose-specific PGC-1β knockout mice (PGC-1β-AT-KO mice) we aimed to learn whether specific PGC-1β ablation in adipocytes is sufficient to drive cold sensitivity. Indeed, we found that warm-adapted (30°C) mutant mice were relatively sensitive to acute cold exposure (6°C). When these mice were subjected t

Central Neural Circuits Orchestrating Thermogenesis, Sleep-Wakefulness States and General Anesthesia States.

Great progress has been made in specifically identifying the central neural circuits (CNCs) of the core body temperature (Tcore), sleep-wakefulness states (SWs), and general anesthesia states (GAs), mainly utilizing optogenetic or chemogenetic manipulations. We summarize the neuronal populations and neural pathways of these three CNCs, which gives evidence for the orchestration within these three CNCs, and the integrative regulation of these three CNCs by different environmental light signals. We also outline some transient receptor potential (TRP) channels that function in the CNCs-Tcore and

PITPNC1 promotes the thermogenesis of brown adipose tissue under acute cold exposure.

Brown adipose tissue (BAT) plays an essential role in non-shivering thermogenesis. The phosphatidylinositol transfer protein, cytoplasmic 1 (PITPNC1) is identified as a lipid transporter that reciprocally transfers phospholipids between intracellular membrane structures. However, the physiological significance of PITPNC1 and its regulatory mechanism remain unclear. Here, we demonstrate that PITPNC1 is a key player in thermogenesis of BAT. While Pitpnc1-/- mice do not differ with wildtype mice in body weight and insulin sensitivity on either chow or high-fat diet, they develop hypothermia when

Oral Contraceptive Pill Alters Acute Dietary Protein-Induced Thermogenesis in Young Women.

OBJECTIVE: There is much interest in the role of dietary protein for weight control. However, there remains a need to characterize individual determinants of the thermogenic effects of protein. This study aimed to investigate the influence of menstrual cycle phase and the combined, monophasic oral contraceptive pill on the thermogenic response to a standardized high-protein (HP) versus normal-protein (NP) meal. METHODS: Following an overnight fast, resting energy expenditure (EE) was measured in 16 healthy young women (8 taking and 8 not taking the pill) and 8 men for 30 minutes pre ingestion

Disinhibition of lower midbrain neurons enhances non-shivering thermogenesis in anesthetized rats.

UNLABELLED: The hypothesis that the lower midbrain, specifically in and around the retrorubral field (RRF) and/or rubrospinal tract (rs), contains a tonic inhibitory mechanism on non-shivering thermogenesis (NST) was examined in urethane-anesthetized rats. It has been proposed that removal of the tonic inhibitory mechanism in the lower midbrain causes body temperature increase through disinhibition-induced activation of the central sympathetic nervous system. The present experiments were carried out to examine whether and where the proposed midbrain region contains cell bodies that tonically i

Weight loss induced by chronic phenylpropanolamine: anorexia and brown adipose tissue thermogenesis.

The effects of chronic treatment with phenylpropanolamine (PPA) on body weight (BW), food intake (FI), and water intake (WI) and interscapular brown adipose tissue (IBAT) thermogenesis in adult male Sprague-Dawley rats were evaluated in 2 experiments. In Experiment 1, rats were treated (IP) twice daily (0900 and 2100 hr) for 12 days with either saline or 5, 10, or 20 mg/kg dl-PPA. Rats treated with 20 mg/kg dl-PPA exhibited significant decreases in both FI and BW but not WI. Basal IBAT temperature was slightly increased in chronic 20 mg/kg dl-PPA rats and there was no evidence of tolerance to

Pharmacological inhibition of CLPP stabilizes LETMD1 to boost thermogenesis and alleviate obesity.

Obesity poses a severe global health burden, and activating adipose thermogenesis offers a promising therapeutic avenue. However, the contribution of mitochondrial proteostasis to thermogenic regulation remains elusive. Here, we identify mitochondrial protease CLPP as a negative regulator of adipose thermogenesis. Pharmacological suppression of CLPP activity enhances thermogenic programmes in adipocytes. Mechanistically, CLPP interacts with LETMD1 and promotes its degradation at the post-translational level, while LETMD1 stabilization is required for the thermogenic effects induced by CLPP sup

CDK4/6 are necessary for UCP1-mediated thermogenesis of white adipose tissue.

AIMS: In white adipose tissue (WAT) the cell cycle regulators CDK4 and CDK6 (CDK4/6) promote adipogenesis and maintain the adipocyte mature state. Here we aimed to investigate their role in the Ucp1-mediated thermogenesis of WAT depots and in the biogenesis of beige adipocytes. MAIN METHODS: We treated mice with the CDK4/6 inhibitor palbociclib at room temperature (RT) or cold and analyzed thermogenic markers in the epididymal (abdominal) and inguinal (subcutaneous) WAT depots. We also assessed the effect of in vivo palbociclib-treatment on the percentage of beige precursors in the stroma vas

The mitochondrial calcium uniporter engages UCP1 to form a thermoporter that promotes thermogenesis.

Uncoupling protein 1 (UCP1)-mediated adaptive thermogenesis protects mammals against hypothermia and metabolic dysregulation. Whether and how mitochondrial calcium regulates this process remains unclear. Here, we show that mitochondrial calcium uniporter (MCU) recruits UCP1 through essential MCU regulator (EMRE) to form an MCU-EMRE-UCP1 complex upon adrenergic stimulation. This complex formation increases mitochondrial calcium uptake to accelerate the tricarboxylic acid cycle and supply more protons that promote uncoupled respiration, functioning as a thermogenic uniporter. Mitochondrial calci

Body composition, inflammation and thermogenesis in pathways to obesity and the metabolic syndrome: an overview.

According to the World Health Organization, overweight and obesity are defined as abnormal or excessive fat accumulation that presents a risk to health. What constitutes 'abnormal' fat accumulation in this definition is not specified, but this most likely represents a consensus term that encapsulates the importance of adipose tissue dysfunctions, rather than solely excess fat per se, in the pathogenesis of disease entities of the metabolic syndrome, particularly type 2 diabetes and cardiovascular diseases. Over the past decades, such abnormalities in fat accumulation have been linked to a patt

Shivering thermogenesis in the pigeon: the effects of activity, diurnal factors, and feeding state.

Shivering (electromyographic activity of the pectoral muscle), oxygen consumption, and body temperature were measured from undisturbed pigeons for periods of several weeks, and segments from the midparts of each phase of the light-dark cycle were compared at various ambient temperatures and feeding regimes. Behavior was recorded with a video camera. None of the observed types of behavior (e.g., walking, preening, feeding, drinking, pecking, defecation) induced spurious electrical activity in the pectoral muscle. On the other hand, none of these behaviors directly inhibited ongoing shivering. T

Sympathetic mechanisms in diet-induced thermogenesis: modification by ciclazindol and anorectic drugs.

1 The sympathetic noradrenergic activation of brown adipose tissue and the biochemical mechanisms involved in diet-induced thermogenesis were studied in rats. 2 A close correlation was found between brown adipose tissue Na+, K+-adenosinetriphosphatase (Na+, K+-ATPase) activity in vitro and in vivo measurements of resting oxygen consumption (VO2). The effects of noradrenaline on in vitro NA+, K+-ATPase activity in brown adipose tissue and in vivo VO2 could be mimicked by a variety of agents. These included beta-adrenoceptor agonists and agents known to induce the release of noradrenaline or inh

The contribution of increased thermogenesis to the effect of anorectic drugs on body composition in mice.

The study investigated whether changes in body composition of normal and genetically obese C57BL/6J (ob/ob) mice caused by the anorectic drugs phentermine, diethylpropion, fenfluramine, and mazindol are entirely due to reduced food intake. Mice were dosed daily (25 mg/kg po) for 28 days after which time carcass composition was determined. Compared to controls fed at libitum, reductions in food intake were for phentermine, 7%; fenfluramine, 17%; diethylpropion, 17%, whereas reduction in body lipid content were for phentermine, 16%; mazindol, 18%; fenfluramine, 8%; diethylpropion, 10%. Since die

Diazepam in labour: its metabolism and effect on the clinical condition and thermogenesis of the newborn.

Following the administration of diazepam to mothers in labour the clinical effects, changes in thermogenesis, and metabolism of the drug in the newborn were observed under carefully controlled conditions. A total maternal dose of 30 mg or less in the 15 hours before delivery had little effect on the infants' state. Larger doses, however, were accompanied by low Apgar scores at birth, apnoeic spells, hypotonia, reluctance to feed, and an impaired metabolic response to a cold stress. Measurement of plasma levels of diazepam and its active metabolite showed that both products were detectable in s

Junctophilin-2-orchestrated calcium signalosome regulates brown adipocyte thermogenesis and energy metabolism.

Dysfunctional adipocyte calcium handling is implicated in obesity and thermogenesis. Junctophilins (JPs) stabilize calcium microdomain junctions between the plasma membrane and endoplasmic reticulum, but whether JPs are required for adipocyte function is not known. We show that JP2 is enriched in thermogenic brown adipose tissue (BAT) relative to other fat depots and is downregulated under conditions of nutrient overload. Conditional knockdown of JP2 in adipocytes, and more selectively in BAT, exacerbates cold intolerance and susceptibility to diet induced obesity. Mechanistically, JP2-deplete
