Altered expression of occludin and tight junction formation in psoriasis.

In simple epithelia, tight junctions are well developed and have barrier and fence functions. On the other hand, tight junctions are less developed in stratified epithelia. In the rodent epidermis, only maculae occludentes (i.e. focal strands or spot tight junctions) are observed in the most superficial zone of the granular cell layer. Occludin is an integral membrane protein, and is localized at tight junctions in simple epithelia. In normal epidermis, occludin is expressed at the maculae occludentes in the granular cell layer, indicating that it is associated with keratinocyte differentiatio

ROS-Triggered Disassembly of Nanoparticle-Networked Hydrogels Enables Dynamic Gating of Corneal Tight Junctions.

The blood-ocular barrier has always been a major challenge in the treatment of eye disease, such as fungal keratitis (FK). Herein, we report a reactive oxygen species (ROS)-triggered disassembly of nanoparticle-networked hydrogels enabling dynamic modulation of corneal tight junctions for enhanced drug bioavailability. The voriconazole (VCZ)-loaded fluorinated chitosan nanoparticles (FCS NPs) are incorporated as dynamic crosslinking nodes within a network formed by phenylboronic acid-modified alginate (SA-PBA) and poly vinyl alcohol (PVA), featuring a macroporous backbone interconnected by web

ARHGEF18 is a flow-responsive exchange factor controlling endothelial tight junctions and vascular leakage.

The shear stress resulting from blood flow is a major regulator of endothelial cell (EC) biology and morphology. Rho protein-mediated cytoskeleton remodeling is an early and essential step of EC responses to flow. However, how Rho protein signaling is controlled by shear stress remains unclear. Here we demonstrate that phosphorylation, activity, and expression of the Rho nucleotide exchange factor (RhoGEF) ARHGEF18 in ECs are modulated by the magnitude of shear stress. When phosphorylated, ARHGEF18 interacts with tight junctions; participates in EC elongation, alignment, and migration; and all

Disruption of Tight Junctions in Intestinal Epithelial Cells by Toxic Advanced Glycation End-Products.

Intestinal epithelial cells (IECs) play a crucial role in forming a protective barrier and regulating the absorption of substances passing through the small intestine. Disrupting the epithelial barrier function can result in intestinal diseases such as inflammatory bowel disease. Glyceraldehyde (GA)-derived advanced glycation end-products (AGEs) (toxic AGEs, TAGE) are AGEs formed by the nonenzymatic Maillard reaction. Although AGEs have been implicated in intestinal barrier breakdown, the associated mechanism remains underexplored. In this study, the effects of accumulated TAGE in IECs were in

Enhancing Paracellular Permeability of Airway Epithelium by Opening Tight Junctions via Osmo-Mechanical Stimulation.

Pulmonary drug delivery offers significant advantages for local and systemic therapy by directly targeting the lungs, minimizing off-target effects and avoiding first-pass metabolism. However, the airway epithelium, which serves as a protective barrier, presents significant challenges for effective drug delivery. Tight junctions (TJs) between epithelial cells and mucociliary clearance hinder drug absorption, especially for high-molecular-weight drugs. To address this, various TJ modulators, such as chelators and surfactants, have been explored but their safety concerns limit clinical applicati

Protective Effects of Baicalin on Peritoneal Tight Junctions in Piglets Challenged with Glaesserella parasuis.

Glaesserella parasuis (G. parasuis) causes inflammation and damage to piglets. Whether polyserositis caused by G. parasuis is due to tight junctions damage and the protective effect of baicalin on it have not been examined. Therefore, this study aims to investigate the effects of baicalin on peritoneal tight junctions of piglets challenged with G. parasuis and its underlying molecular mechanisms. Piglets were challenged with G. parasuis and treated with or without baicalin. RT-PCR was performed to examine the expression of peritoneal tight junctions genes. Immunofluorescence was carried out to

Larazotide acetate induces recovery of ischemia-injured porcine jejunum via repair of tight junctions.

Intestinal ischemia results in mucosal injury, including paracellular barrier loss due to disruption of tight junctions. Larazotide acetate (LA), a small peptide studied in Phase III clinical trials for treatment of celiac disease, regulates tight junctions (TJs). We hypothesized that LA would dose-dependently hasten recovery of intestinal ischemic injury via modulation of TJs. Ischemia-injured tissue from 6-8-week-old pigs was recovered in Ussing chambers for 240-minutes in the presence of LA. LA (1 μM but not 0.1 μM or 10 μM) significantly enhanced transepithelial electrical resistance (TER)

Lipopolysaccharide disrupts the milk-blood barrier by modulating claudins in mammary alveolar tight junctions.

Mastitis, inflammation of the mammary gland, is the most costly common disease in the dairy industry, and is caused by mammary pathogenic bacteria, including Escherichia coli. The bacteria invade the mammary alveolar lumen and disrupt the blood-milk barrier. In normal mammary gland, alveolar epithelial tight junctions (TJs) contribute the blood-milk barrier of alveolar epithelium by blocking the leakage of milk components from the luminal side into the blood serum. In this study, we focused on claudin subtypes that participate in the alveolar epithelial TJs, because the composition of claudins

The blood-testis and blood-epididymis barriers are more than just their tight junctions.

The terms blood-testis barrier (BTB) or blood-epididymis barrier (BEB), are often described as Sertoli cell-Sertoli cell tight junctions (TJs) or TJs between the epithelial cells in the epididymis, respectively. However, in reality, the BTB and BEB are much more complex than just the TJ. The focus of this minireview is to remind readers that the complete BTB and BEB are comprised of three components: anatomical, physiological, and immunological. The TJs form the anatomical (physical) barrier that restricts passage of molecules and cells from entering or exiting the lumen. The physiological bar

Aldosterone and tight junctions: modulation of claudin-4 phosphorylation in renal collecting duct cells.

Aldosterone classically modulates Na transport in tight epithelia such as the renal collecting duct (CD) through the transcellular route, but it is not known whether the hormone could also affect paracellular permeability. Such permeability is controlled by tight junctions (TJ) that form a size- and charge-selective barrier. Among TJ proteins, claudin-4 has been highlighted as a key element to control paracellular charge selectivity. In RCCD2 CD cells grown on filters, we have identified novel early aldosterone effects on TJ. Endogenous claudin-4 abundance and cellular localization were unalte

Expression and function of tight junctions in the crypt epithelium of human palatine tonsils.

The human palatine tonsils have surface and crypt stratified epithelium and may be initiated via the epithelium to mount immune responses to various presenting antigens. Here we investigated the expression and function of tight junctions in the epithelium of human palatine tonsils from patients with tonsillar hypertrophy or recurrent tonsillitis. Occludin, ZO-1, JAM-1, and claudin-1, -3, -4, -7, -8, and -14 mRNAs were detected in tonsillar hypertrophy. Occludin and claudin-14 were expressed in the uppermost layer of the tonsil surface epithelium, whereas ZO-1, JAM-1, and claudin-1, -4, and -7

Tight junctions of the human ciliary epithelium: regional morphology and implications on transepithelial resistance.

The tight junctions of the human ciliary epithelium have been studied using the freeze-fracture technique with particular regard to regional differences in the tight junction networks and implications on the transepithelial resistance. The tight junctions of the non-pigmented epithelium always form continuous networks and consist of from two to more than 20 superimposed strands (mean 3 to 4), suggesting that relatively tight and leaky sites of the paracellular pathway coexist within the same cell perimeter. The tight junction morphology is more complex in the anterior pars plicata than in the

Effects of tight junctions on biomechanical response of blood-brain barrier under stable cavitation.

The rising prevalence of central nervous system (CNS) diseases has imposed substantial social and economic burdens on healthcare systems. The blood-brain barrier (BBB), a highly selective physiological barrier in the CNS, severely restricts the delivery of most therapeutic agents to the brain, thereby limiting treatment efficacy. Stable cavitation of microbubbles induced by focused ultrasound (FUS) offers a promising strategy for transiently and non-invasively opening the BBB, holding significant clinical potential. However, the underlying biophysical mechanisms remain challenging for understa

Barrier Function and Tight Junctions in Tyrosine Kinase Inhibitor-Induced Diarrhoea: A Scoping Review.

INTRODUCTION: Tyrosine Kinase Inhibitors (TKIs) are widely used in modern cancer treatment and have significantly improved outcomes for many patients. However, their use is frequently limited by gastrointestinal side effects, particularly diarrhoea. Although diarrhoea may appear mild in otherwise healthy individuals, it can substantially impair the quality of life of cancer patients and disrupt treatment continuity. In more severe cases, high-grade diarrhoea may lead to dehydration, malabsorption, fatigue and treatment interruption. METHODS: This scoping review systematically synthesised avai

Soluble adenylyl cyclase regulates tight junctions of human intestinal epithelial cells via claudin-4.

Although cAMP is a major intracellular messenger, studies on modulating effects of cAMP on tight junctions in epithelial cells are very limited. In this study, we characterized a bidirectional and prominent regulation of the tight junction by soluble adenylyl cyclase (sAC) in human intestinal epithelial (Caco2Bbe1) cells. Pharmacological inhibition and knockdown of sAC substantially increased TEER, whereas overexpression of sAC reciprocally decreased TEER. Mechanistically, EPAC1 was the sole cAMP effector responsible for the regulation of TEER by sAC. Pharmacological inhibition or knockdown of

The role and mechanism of tight junctions in the regulation of salivary gland secretion.

Tight junctions (TJs) are cell-cell interactions that localize at the most apical portion of epithelial/endothelial cells. One of the predominant functions of TJs is to regulate material transport through paracellular pathway, which serves as a selective barrier. In recent years, the expression and function of TJs in salivary glands has attracted great interest. The characteristics of multiple salivary gland TJ proteins have been identified. During salivation, the activation of muscarinic acetylcholine receptor and transient receptor potential vanilloid subtype 1, as well as other stimuli, pro

Claudin-9 constitutes tight junctions of folliculo-stellate cells in the anterior pituitary gland.

The anterior pituitary gland regulates growth, metabolism, and reproduction by secreting hormones. Folliculo-stellate (FS) cells are non-endocrine cells located among hormone-producing cells in the anterior pituitary glands. They form follicular lumens, which are sealed by tight junctions (TJs). Although FS cells are hypothesized to contribute to fine-tuning of endocrine cells, little is known about the exact roles of FS cells. Here, we investigated the molecular composition of TJs in FS cells. We demonstrated that occludin is a good marker for TJs in the pituitary gland and examined the struc

Tight and adherens junctions in the ovine uterus: differential regulation by pregnancy and progesterone.

In species with noninvasive implantation by conceptus trophectoderm, fetal/maternal communications occur across the endometrial epithelia. The present studies identified changes in junctional complexes in the ovine endometrium that regulate paracellular trafficking of water, ions, and other molecules, and the secretory capacity of the uterine epithelia. Distinct temporal and spatial alterations in occludin, tight junction protein 2, and claudin 1-4 proteins were observed in the endometrium of cyclic and early pregnant ewes. Dynamic changes in tight junction formation were characterized by an a
