Janus-Structured Microgel Barrier with Tissue Adhesive and Hemostatic Characteristics for Efficient Prevention of Postoperative Adhesion.

Postoperative adhesion (POA) is a common and serious complication following various types of surgery. Current physical barriers either have a short residence time at the surgical site with a low tissue attachment capacity or are prone to undesired adhesion formation owing to the double-sided adhesive property, which limits the POA prevention efficacy of the barriers. In this study, Janus-structured microgels (Janus-MGs) with asymmetric tissue adhesion capabilities are fabricated using a novel bio-friendly gas-shearing microfluidic platform. The anti-adhesive side of Janus-MGs, which consists o

Adhesion GPCR GPR56 Expression Profiling in Human Tissues.

Despite the immense functional relevance of GPR56 (gene ADGRG1) in highly diverse (patho)physiological processes such as tumorigenesis, immune regulation, and brain development, little is known about its exact tissue localization. Here, we validated antibodies for GPR56-specific binding using cells with tagged GPR56 or eliminated ADGRG1 in immunotechniques. Using the most suitable antibody, we then established the human GPR56 tissue expression profile. Overall, ADGRG1 RNA-sequencing data of human tissues and GPR56 protein expression correlate very well. In the adult brain especially, microglia

Selective adhesion of nanoparticles to inflamed tissue in gastric ulcers.

PURPOSE: Gastrointestinal deposition of nanoparticles was examined after oral administration to mice suffering from an experimental gastric ulcer model. Local drug delivery could reduce side effects and would be a distinct improvement compared to existing therapeutic approaches, e.g. in the local therapy of Helicobacter pylori. METHODS: A gastric ulcer was induced to Swiss mice by acetic acid injection. Fluorescent polystyrene particles with a nominal size of 50, 200, and 750 nm were administered orally for 3 or 5 days and particle adhesion in the gastrointestinal tract analyzed. RESULTS: In

Intraocular use of hydrogel tissue adhesive in rabbit eyes.

PURPOSE: To evaluate the safety and efficacy of newly developed hydrogel glue to treat rhegmatogenous retinal detachments in in vitro and in vivo studies. METHODS: In the in vitro study, the solid hydrogel glue was soaked in a balanced salt solution (BSS). The pH was measured periodically, and the dissolution time was recorded. In the in vivo study in six albino rabbits, 0.1 ml of hydrogel glue was injected into the right vitreous cavity, and 0.1 ml BSS was injected into the left vitreous, as the control. Clinical, electroretinography (ERG) and histological examinations were performed. Retina

A congenital orbital defect with periorbital-dural tissue adhesion.

This case demonstrates a previously unreported congenital orbital deformity. The patient was born with a unilateral exophthalmos. The etiology of the defect was demonstrated by CT scan as a convex bowing of the right superomedial orbital wall behind the axis of the eye. At surgery, this convexity was revealed to be a small bony defect through which the periorbital tissue and dura mater adhered. This sort of orbital deformity, although subtle, must be considered in the differential diagnosis of congenital unilateral exophthalmos.

[1st report on the use of an autogenous tissue adhesive in otorhinolaryngology].

An autogenous tissue glue has been developed at the ENT Department of the University Clinic Graz. It is prepared from the patient's own plasma, and has the following advantages: There is no danger of transmitting infectious diseases, there are no problems with storage, temperature and expiration date. The glue is produced at room temperature and is cheap. The production and application of this autologous tissue glue are described and a first report is given on initial experiences.

Percutaneous embolization of bony pelvic neoplasms with tissue adhesive.

Eight patients with tumors of the bony pelvis underwent embolization with isobutyl-2-cyanoacrylate (IBCA). Five patients had primary bone tumors, of which 2 were malignant and 3 were benign; 3 patients had metastases to the bony pelvis from the thyroid gland, kidney, and femur, respectively. Embolization was performed to minimize blood loss during resection of a giant-cell tumor in one patient and insertion of a hip prosthesis in another who had metastatic renal carcinoma. It was also done prior to scheduled surgery in one of the patients with aneurysmal bone cyst, but healing was sufficient t

Injectable pullulan hydrogel for the prevention of postoperative tissue adhesion.

Methods for reducing and preventing postoperative abdominal adhesions have been researched for decades; however, despite these efforts, the formation of postoperative peritoneal adhesions is continuously reported. Adhesions cause serious complications such as postoperative pain, intestinal obstruction, and infertility. Tissue adhesion barriers have been developed as films, membranes, knits, sprays, and hydrogels. Hydrogels have several advantages when used as adhesion barriers, including flexibility, low tissue adhesiveness, biodegradability, and non-toxic degraded products. Furthermore, compa

An orthopedic tissue adhesive for targeted delivery of intraoperative biologics.

Tissue adhesives can bind together damaged tissues and serve as tools to deliver and localize therapeutics to facilitate regeneration. One emerging therapeutic trend in orthopedics is the use of intraoperative biologics (IOB), such as bone marrow (BM) and platelet-rich plasma (PRP), to stimulate healing. Here, we introduce the application of the biomaterial chondroitin sulfate succinimidyl succinate (CS-NHS) to deliver IOB in a hydrogel adhesive. We demonstrate the biomaterial's ability to bind various tissue types and its cellular biocompatibility with encapsulated human mesenchymal stem cell

Adhesion molecules mediating recruitment of monocytes to inflamed tissue.

Three families of cell-surface proteins are largely responsible for the adherence of leukocytes to cells and matrices: integrins, immunoglobulin (Ig)-related molecules and selectins. Blood monocytes express beta 1 integrins VLA-4, -5 and -6 and beta 2 integrins CD11a/CD18, CD11b/CD18 and CD11c/CD18. These cells also express the Ig-related molecules ICAM-1, -2 and -3, ligands for the beta 2 integrins. In addition, monocytes express L-selectin and the oligosaccharides Lex and sialyl Lex, ligands for the endothelial selectins E- and P-. In vitro studies with blocking antibodies have identified ad

Bacterial adhesion to oral tissues: a model for infectious diseases.

The majority of bacteria which colonize humans display sharp host and tissue tropisms; consequently, relatively little is known about how they initiate colonization on mucosal surfaces. The mouth has a variety of features which have enabled it to serve as a useful model for the discovery of basic principles of host-parasite interactions occurring in mucosal environments. Early studies demonstrated that indigenous bacteria attach to surfaces of the mouth in a highly selective manner; attachment was often observed to correlate with colonization. These studies led to the recognition that bacteria

[Effect of vitamin A on the adhesive interaction of cells in epithelial tissue].

Both the increasing and decreasing contents of vitamin A in rats induced a lowering of cell adhesion in the skin multilayer epithelium, and that of their packing density. However, the same stimulus did not alter cell adhesion in the small intestinal monolayer epithelium. The data obtained show that vitamin A may serve as a factor modulating intercellular adhesive interactions with tissue specificity.

Sodium hyaluronate and tissue adhesive in treating corneal perforations.

The injection of sodium hyaluronate through perforation sites prior to tissue adhesive application has extended the current use of this modality to large perforations and perforations in which intraocular tissue is incarcerated. Three clinical cases are described in which the conjoint therapy was used, and four monkey eyes were studied clinically and histopathologically to test for adverse drug interactions.

Tissue adhesive arrests stromal melting in the human cornea.

The direct early application of cyanoacrylate adhesive to a prepared ulcer bed and adjacent basement membrane, followed by placement of a bandage lens, gave good results in ten patients with corneal ulceration. The patients had ulceration with keratitis sicca, herpes keratitis, and other surface diseases. Progressive melting was arrested in all cases, and concurrent adjunctive therapies were used as indicated. The technique is quick and simple.

Foreign body reaction after usage of tissue adhesives for skin closure: a case report and review of the literature.

INTRODUCTION: The usage of tissue adhesives such as 2-octyl cyanoacrylate is a standard tool for closure of superficial wounds of the body. The benefits using 2-octyl cyanoacrylate are well known and appreciated by surgeons world wide. Especially in superficial or small wounds in children where no surgical procedure is wanted this technique is commonly used. However, so far the risks and possible complications after using 2-octyl cyanoacrylate are not sufficiently published and discussed. CASE REPORT: We report a case of foreign body reaction after 3 weeks in a 39-year-old female patient afte
