INTRODUCTION: Solid organ transplant patients require appropriate immunosuppression to sufficiently control the allorecognition of the graft. Two tests, the QuantiFERON Monitor (QFM) and the Torque teno virus load (TTVL) provide an option to monitor the strength of immunosuppression.
MATERIALS AND METHODS: TTVL and QFM were simultaneously determined in kidney transplant patients. Clinical data, microbiological and histopathological findings were collected from the patients' medical records.
RESULTS: 128 TTVL and QFM values were quantified in 107 patients. 69 patients (54%) had recurrent infe

Torque teno virus (TTV) is a recognized marker of immune competence, but its relevance in severe bacterial infections is unclear. We evaluated prevalence, viral load, and clinical relevance of TTV viremia in complicated Staphylococcus aureus infection (CSAI). In this ancillary analysis of two prospective pharmacokinetic cohort studies at a Swiss university hospital, stored plasma samples were tested for TTV DNA using TTV R-GENE PCR, and cytokines were measured by multiplex immunoassay. Associations between TTV load, inflammatory biomarkers, and clinical outcomes (including 30-day mortality) we

Graft inflammation and fibrosis are associated with progressive fibrosis and potential graft loss after liver transplantation, but diagnosis relies on invasive biopsies. Torque teno virus (TTV) may be a noninvasive marker of functional immunity, however its association with these changes remains unclear. In this Nordic multicenter cohort study, plasma samples collected with biopsies were analyzed for TTV load, and biopsies were scored for inflammation and fibrosis. Associations and performance of TTV versus tacrolimus were evaluated using logistic regression and ROC curves. TTV was categorized

Following lung transplantation (LuTx), some patients attain good long-term health, enabling participation in demanding activities, including high-altitude mountaineering. High-altitude exposure induces substantial physical stress through hypoxia, exhaustion and ultraviolet (UV) radiation, potentially compromising immunocompetence. Levels of immunosuppression and Torque Teno virus (TTV), a persistent, non-pathogenic DNA virus associated with immunocompetence after LuTX, were measured during an expedition to Mount Jebel Toubkal (4167 m), evaluating the effects of high-altitude exposure, physical

BACKGROUND: Hepatitis E virus (HEV) affects immunocompromised individuals. Unlike immunocompetent hosts, who rarely develop chronic infection, up to two-thirds of immunocompromised patients progress to chronicity. Ribavirin is the recommended antiviral therapy, yet predictors of sustained virological response (SVR) remain unclear. Torque Teno Virus (TTV), a marker of immunosuppression, has been proposed as a potential predictor of viral clearance.
OBJECTIVES: We evaluated ribavirin treatment outcomes and adverse effects in immunocompromised patients with HEV infection, and assessed whether TT

BACKGROUND: Torque teno virus (TTV) viral loads may reflect patients' net state of immunosuppression and identify patients at risk of infections and rejection early after solid organ transplantation. However, its role in long-term kidney recipients (KTRs) is less clear.
METHODS: This single-centre prospective cohort study evaluated the association between TTV viral load at recruitment and severe infections requiring hospitalisation within 12 months as the primary outcome in kidney recipients > 1-year post-transplant and on stable immunosuppression for > 3 months. Participants were followed up

BACKGROUND: Torque teno virus (TTV) load has emerged as a potential surrogate marker of net immunosuppression in solid organ transplantation. However, the clinical significance of pre-transplant TTV levels for early outcomes after lung transplantation remains unclear.
METHODS: In this multicenter cohort of 334 adult first-time lung transplant recipients (2015-2022), plasma samples were collected immediately prior to transplantation. The primary endpoint was infection within 3 months. One hundred healthy individuals served as controls for TTV comparisons. Secondary outcomes were acute rejectio

Torque Teno virus (TTV) is a ubiquitous virus that causes chronic infection in humans with unknown clinical consequences. Here, we investigated the influence of TTV infection on HCV direct-acting antiviral (DAA) efficacy in HIV/HCV coinfected and HCV monoinfected patients as controls. Of 92 study patients, 79.3% were TTV DNA positive; untreated patients exhibited a significantly higher proportion of TTV DNA-positivity vs. sustained virological response (SVR) patients (100.0% vs. 65.2%, p < 0.001), while TTV positivity was not significant in DAA failure patients vs. SVR patients despite HIV/HCV

Porcine torque teno sus virus (TTSuV) has been suggested as a co-factor for the development of porcine circovirus-associated diseases. However, the pathogenic role of TTSuV is still inconclusive, and the target cell and tissue tropism of this virus are also ambiguous. In the present study, a multi-strained pooled probe-based in situ hybridization was established to detect the nucleic acids of TTSuV1 and TTSuV2 in the tissue. The strategy of using polymerase chain reaction-derived digoxigenin-labeled multi-strained pooled probe, instead of single-strained probe or oligonucleotide, was to overco

BACKGROUND: The non-pathogenic Torque Teno Virus (TTV) is currently being investigated as a functional biomarker for the degree of immunosuppression. A reliable assay for quantification of TTV is extremely important. The primary objective of this study was to assess the reliability and agreement between two quantitative methods, an in-house PCR assay and the commercial TTV R-GENE, for the detection of TTV viral loads. The secondary objectives included testing of the in-house assay by performing external quality assessment (EQA) and assessing the TTV prevalence in 101 Danish blood donors.

Myalgic encephalomyelitis/chronic fatigue syndrome (ME/CFS) is a chronic disorder classified by the WHO as postviral fatigue syndrome (ICD-11 8E49 code). Diagnosing ME/CFS, often overlapping with fibromyalgia (FM), is challenging due to nonspecific symptoms and lack of biomarkers. The etiology of ME/CFS and FM is poorly understood, but evidence suggests viral infections play a critical role. This study employs microarray technology to quantitate viral RNA levels in immune cells from ME/CFS, FM, or co-diagnosed cases, and healthy controls. The results show significant overexpression of the Torq

Torque teno virus (TTV) is a nonenveloped, single-stranded, circular DNA virus of the family of Anelloviridae. The first contact with TTV usually occurs in early childhood, followed by persistent infection in bone marrow and lymphocytes. Increased levels of TTV-DNA are found in the serum in various states of immune deficiency. The objective of this study was to assess if monitoring of TTV viremia after allogeneic hematopoietic stem cell transplantation (allo-HSCT) is a predictive marker for immune-related clinical complications. In a retrospective study, 2054 whole-blood samples from 123 patie

Atopic dermatitis is a common inflammatory skin disease affecting up to 10% of adults. Whether atopic dermatitis exhibits a strong systemic inflammatory signature remains debated. We characterized peripheral whole-blood alterations in adults with moderate-to-severe atopic dermatitis undergoing treatment with dupilumab or baricitinib therapy. Blood samples were collected at weeks 0, 4, and 16. Whole-blood proteins were measured using Olink, transcriptomic profiling was performed by RNA sequencing, and torque teno virus plasma levels were quantified by qPCR. During targeted atopic dermatitis the

OBJECTIVE: The Torque Teno virus (TTV), a member of virus genus Anellovirus has been shown to be commonly present in humans, yet without detectable pathogenicity. Recent studies imply that TTV may contribute to provoke autoimmune progresses in systemic lupus erythematosus and idiopathic inflammatory myopathies. We aimed to study the presence of TTV in a group of patients with autoimmune bullous diseases with a further goal to identify long-lasting foreign antigen, such as TTV as possible triggers of skin-specific autoimmunity.

Hepatitis B Virus (HBV) infection is a global health concern. HBV genotypes differ in disease potential and geographical distribution. These genotypes impact the diagnostic and preventive strategies. Furthermore, limited data are available on Torque Teno Virus (TTV) co-infections with HBV. Hence, this study evaluated HBV infection prevalence, genotype distribution, and the presence of TTV among HBV-positive patients. During the study period, a total of 1820 serum samples were collected and tested for HBsAg using ELISA method. Of this, 43 (2.36%) were HBsAg positive. These HBsAg-positive sample

Neutralizing antibodies (NAbs) have been proposed as biomarkers for stratifying BK polyomavirus (BKPyV) replication risk, while torque teno virus (TTV) viral load is recognized as a global immunomarker in kidney transplantation. This study investigates their combined utility to improve BKPyV DNAemia risk assessment in 187 patients within 12 months posttransplant. BKPyV DNAemia occurred in 19% of recipients. Donor seronegativity was protective, whereas in case of seropositive donor, replication risk varied according to recipient NAb titers. Indeed, patients with low NAbs faced a high risk of BK
