Toxicokinetic and Partial Mass Balance Assessment of 14C-Alpha Olefins in Rats.

Higher olefins are a class of alkenes widely used as intermediates in the production of essential consumer and industrial products. This radiolabel disposition and partial mass balance study investigated the distribution and excretion of four 14C-radiolabelled alpha higher olefins (i.e., 1-octene, 1-decene, 1-hexadecene, and 1-eicosene) in male Wistar rats following a single oral dose (100 mg/kg). Blood, liver, kidney, adipose tissue, urine, and faeces were collected and analysed for total 14C-derived radioactivity. Urinary elimination was rapid, with approximately 70% and 90% of total radioac

Experimental exposure of rat skin to methyl bromide: a toxicokinetic and histopathological study.

Methyl bromide was experimentally exposed to a 12 cm2 area of the back skin of Wistar rats for 30 s, and for 1, 3, and 5 min, and time courses of both changes in plasma bromide concentration and of histopathological changes were examined. To measure bromide ion, a head space gas chromatography was used. The concentration of plasma bromide ion showed a sharp increase immediately after the exposure in all exposed groups, reaching a peak level after 1 h, then decreased rapidly. The ion level gradually decreased after 72 h to 1 week, and returned to a normal level after 4 to 8 weeks. Calculating f

Accidental cannabis intoxication in two young children: clinical presentation and toxicokinetics - a case series.

Accidental exposure to edible cannabis products in children is an increasing public health concern. The clinical presentation is often nonspecific, which can delay diagnosis and lead to inappropriate management. Toxicological screening is therefore essential for accurate diagnosis and appropriate treatment. We report the toxicokinetic (TK) profiles of delta-9-tetrahydrocannabinol (THC) and cannabidiol (CBD) following unintentional ingestion of cannabis in two pediatric cases, aged 12 and 15 months, respectively. Plasma concentrations of THC and CBD were measured using a validated liquid chroma

Maternal-Fetal Transfer and Toxicokinetics of 2,2',5,5'-Tetrachlorobiphenyl, [14C]-PCB52, Following Intratracheal Administration.

Despite increased recognition of the adverse impacts of PCB exposure on human health, comprehensive risk assessments, particularly regarding inhalation exposure and effects on the developing fetus, are lacking. Out of all PCB congeners, lower-chlorinated PCBs have been more prevalent in indoor and outdoor atmospheres. Thus, we investigated in vivo toxicokinetics and placental transfer of radiolabeled [14C]-PCB52 (0.157 mg/kg administered intratracheally) in Sprague-Dawley rats at gestational day 11 ± 1. Following dosing, 99.4 ± 0.5% of the administered dose was distributed to the systemic circ

Biotransformation and toxicokinetics of 2-phenoxyethanol after oral exposure in humans: a volunteer study.

2-Phenoxyethanol (PhE) is an aromatic glycol ether and is used in a variety of functions and applications, e.g., as preservative in pharmaceuticals, cosmetic and personal care products, as biocide in disinfectants (e.g. human hygiene), or as a solvent in formulations (e.g. coatings, functional fluids). Despite its widespread use, little is yet known on its biotransformation and toxicokinetics in humans. Therefore, a pilot study was conducted with oral administration of PhE (5 mg/kg body weight) to five volunteers. Blood and urine samples were collected and analyzed for PhE and three of its pre

[Methylmercury exposure in the general population; toxicokinetics; differences by gender, nutritional and genetic factors].

Mercury is an environmental toxicant that causes numerous adverse effects on human health and natural ecosystems. The factors that determine the existance of adverse effects, as well as their severity are, among others: the chemical form of mercury (elemental, inorganic, organic), dosis, age, period of exposure, pathways of exposure and environmental, nutritional and genetic factors. In the aquatic cycle of mercury, once it has been deposited, it is transformed into methylmercury due to the action of certain sulphate-reducing bacteria, which bioaccumulates in the aquatic organisms and moves in

Toxicokinetics of the ciguatoxin P-CTX-1 in rats after intraperitoneal or oral administration.

Ciguatoxins are voltage-gated selective algal toxins responsible for ciguatera fish poisoning. In this study we evaluate the toxicokinetics of one of the most common ciguatoxins found in the Pacific, the P-CTX-1, in rat after an oral or intraperitoneal (ip) dose of 0.26 μg/kg body weight. We report levels of ciguatoxin activity assessed over time in blood, urine and feces, and at 4 days in liver, muscle and brain, using the functional in vitro N2A cytotoxicity assay. Following exposure, the ciguatoxin activity exhibited a rapid systemic absorption that was followed by a bi-exponential decline,

Sex differences in the toxicokinetics of inhaled solvent vapors in humans 2. 2-propanol.

The aim of this study was to evaluate possible sex differences in the inhalation toxicokinetics of 2-propanol vapor. Nine women and eight men were exposed on different occasions for 2 h during light physical exercise (50 W) to 2-propanol (350 mg/m3) and to clean air (control exposure). The level corresponds to the Swedish occupational exposure limit. 2-Propanol and its metabolite acetone were monitored up to 24 h after exposure in exhaled air, blood, saliva, and urine by headspace gas chromatography. Body fat and lean body mass were estimated from sex-specific equations using bioelectrical imp

[Effect of qualitatively varying nutrition on several parameters of aflatoxin B1 metabolism and toxicokinetics].

The influence of qualitatively different nutrition (physiological and that with lysine, methionine, threonine and vitamins A, B, C deficiencies) on certain toxicokinetic parameters of 3H-aflatoxin B1, after single and multiple (every day, during 5 days) intraperitoneal injections of this agent in a dose of 25 micrograms/animal, was studied in two series of experiments on growing male WAG rats with the initial bw 40-50 g. It has been proved that the main parameters of aflatoxin B1 toxicokinetics depend on the injection multiplicity and the nutrition character. It has been established that the q

Enantioselective toxicokinetics drive the differential toxicity of chiral 6PPD-quinone in rainbow trout.

The tire-derived contaminant N-(1,3-dimethylbutyl)-N'-phenyl-p-phenylenediamine quinone (6PPD-Q) is widespread in aquatic ecosystems and poses significant ecological risk. As a chiral compound, its enantiomers may exhibit differential toxicity, yet the underlying mechanisms remain limited. Here, we found that (+)-6PPD-Q was 2.4-fold more toxic to rainbow trout than (-)-6PPD-Q (p < 0.05). In addition, in vitro metabolism assays using liver S9 fraction showed that (+)-6PPD-Q was metabolized 2.5-fold faster than (-)-6PPD-Q. Semi-quantitative nontarget analysis revealed that hydroxylated metabolit

Dietary Aluminium Exposure and Human Health: Sources, Bioavailability, Toxicokinetics, and Health Risk Assessment.

Background/Objectives: Aluminium is a widespread environmental element and food contaminant to which the general population is continuously exposed, primarily through diet and drinking water. Although gastrointestinal absorption is generally low, bioavailability varies according to chemical form, food matrix, and interactions with dietary components. Prolonged exposure can nevertheless result in gradual tissue accumulation. This review summarises current evidence on dietary aluminium exposure, factors influencing its bioavailability, toxicokinetics, biological effects, gut microbiota interacti

Toxicokinetics, Biotransformation, Excretion, and Tissue Residue of Zearalenone-14-Glucoside in Male Sheep.

Zearalenone-14-glucoside (Z14G) is a common modified mycotoxin attracting considerable attention due to its high occurrence in feed and food, and its potential to transform into the parent form zearalenone (ZEN) in vivo. However, the toxicokinetic data in ruminants remain scarce. In this study, male sheep (n = 5) were orally administered a single dose of Z14G at 2 mg·kg-1 BW. Serum, rumen fluid, urine, and fecal samples were collected at designated time points, and tissue samples were obtained at slaughter 168 h post-dosing. Domoic acid toxicokinetics in Dungeness crabs: new insights into mechanisms that regulate bioaccumulation.

Domoic acid (DA) is an excitatory neurotoxic amino acid produced by several marine algal species and is the causative agent of amnesic shellfish poisoning. Profound differences in the toxicokinetics of DA have been identified in a wide variety of shellfish. We characterized the toxicokinetics of DA in Dungeness crabs (Metacarcinus magister) after oral and intravascular dosing (IV) using a variety of doses ranging from 0.1 to 20mg/kg. After a 1mg/kg oral dose, DA disappeared from the foregut within 2h and largely accumulated in the hepatopancreas, with hemolymph and other tissues having 100-100

Lethality, accumulation and toxicokinetics of aluminum in some tissues of male albino rats.

In the present work, the lethality percentiles including median lethal doses (LD50), accumulation, distribution and toxicokinetics of aluminum in the liver, kidney, intestine, brain and serum of male albino rats, following a single oral administration were studied throughout 1, 3, 7, 14 and 28 days. The estimated LD50 at 24 h was 3.45 g Al/kg body weight (b.wt.). The utilized dose of Al was 1/50 LD50 (0.07 g Al/kg b.wt.). Aluminum residues, in Al-treated rats, were significantly decreased in response to the experimental periods and were negatively correlated with time. In addition, the hepatic

Toxicokinetics of bisphenol A in pregnant DA/Han rats after single i.v. application.

Bisphenol A (BPA) is an important chemical in the production of epoxy resins and polycarbonate plastics, and basic monomers which are used for a variety of applications. Consumer exposure to BPA may be possible from migration of BPA from dental sealants or from polycarbonate or epoxy-lined food and drink containers. BPA is known to act as weak estrogen mimic in rodents, and there is a concern of adverse endocrine effects, especially from prenatal exposure to this potential 'endocrine disruptor'. To address this concern, we have studied the disposition and transplacental transfer of BPA in preg

The oral bioavailability and toxicokinetics of methylmercury in common loon (Gavia immer) chicks.

We compared the toxicokinetics of methylmercury in captive common loon chicks during two time intervals to assess the impact of feather growth on the kinetics of mercury. We also determined the oral bioavailability of methylmercury during these trials to test for age-related changes. The blood concentration-time curves for individuals dosed during feather development (initiated 35 days post hatch) were best described by a one-compartment toxicokinetic model with an elimination half-life of 3 days. The data for birds dosed following completion of feather growth (84 days post hatch) were best fi

Phototoxic retinal degeneration and toxicokinetics of sitafloxacin, a quinolone antibacterial agent, in mice.

We examined drug concentrations and the incidence of retinal degeneration in the eyes of albino BALB/c mice after a single intravenous administration of sitafloxacin plus a 4 h period of UVA irradiation. Retinal degeneration was induced at 40 mg/kg or more plus UVA irradiation, and there was little decrease in ocular sitafloxacin concentration under UVA irradiation. We then examined the incidence of retinal degeneration with various periods of UVA irradiation in BALB/c mice given a single intravenous administration of 40 mg/kg sitafloxacin. Retinal degeneration occurred in all the groups recei

Universal bioaccumulation, biomagnification, and toxicokinetic metrics based on equivalent aqueous concentration.

Diverse bioaccumulation and toxicokinetic parameters present a Babel-Tower challenge for cross-disciplinary researchers and various stakeholders alike as they struggle to formulate a holistic understanding of bioaccumulation phenomena from laboratory to field. This work attempts to resolve the challenge by proposing a universal framework for bioaccumulation, trophic magnification, and toxicokinetic metrics. The framework converts different concentrations and toxicokinetic parameters to aqueous equivalent concentration C⁎ and first-order toxicokinetic constant k⁎ using equilibrium distribution
