Trichoderma Biofertilizer Links to Altered Soil Chemistry, Altered Microbial Communities, and Improved Grassland Biomass.

In grasslands, forage and livestock production results in soil nutrient deficits as grasslands typically receive no nutrient inputs, leading to a loss of grassland biomass. The application of mature compost has been shown to effectively increase grassland nutrient availability. However, research on fertilization regime influence and potential microbial ecological regulation mechanisms are rarely conducted in grassland soil. We conducted a two-year experiment in meadow steppe grasslands, focusing on above- and belowground consequences of organic or Trichoderma biofertilizer applications and pot

Phosphogypsum biotransformation by aerobic bacterial flora and isolated Trichoderma asperellum from Tunisian storage piles.

Aerobic microorganisms able to grow on phosphogypsum (PG), characterized by heavy metals accumulation and high acidity were investigated by enrichment cultures. The PG was used at different concentrations, varying from 20 to 200 g/L in the enrichment culture medium supplemented with compost and Tamarix roots. This treatment reduced COD and heavy metals PG concentration. An efficient isolated fungus, identified by molecular approach as Trichoderma asperellum, was able to grow on PG as the sole carbon and energy sources at the different experimented concentrations, and to increase the culture me

Docking-guided and phosphomimetic rational engineering of glucose-insensitive xylose transporters from Trichoderma reesei.

BACKGROUND: The engineering of Saccharomyces cerevisiae for the use of xylose is fundamental to improving fermentation performance in the production of second-generation ethanol (2G) via pentose fermentation. For this, one of the main strategies involves expressing heterologous xylose transporters to ensure efficient uptake of this sugar. However, due to the intrinsic non-specificity of sugar transporters, competition occurs between sugars (e.g., xylose and glucose), leading to reduced pentose transport efficiency and lower ethanol productivity. This study aimed to develop and characterize sug

Synergistic transcriptional modules in Trichoderma asperellum enhance glutathione detoxification to counteract fungal pathogen toxins.

Trichoderma fungi are potent biocontrol agents. However, their defence mechanisms against pathogen-derived toxins remain poorly understood. We identified two synergistic transcription factor modules in T. asperellum that orchestrate the detoxification of cytotoxic secondary metabolites from the poplar blight pathogen Alternaria alternata. Overexpression of the central regulator TasMYB46 reduced disease lesion area by approximately 22% and was associated with decreased pathogen-induced reactive oxygen species (ROS) accumulation. Mechanistically, TasMYB46 directly activates the glutathione S-tra

Peptide-mediated stabilization of Trichoderma reesei Cel7A for enhanced thermal stability in bioethanol production.

Improving the thermal stability of Trichoderma reesei Cel7A, a key cellulase for bioethanol production, is crucial for reducing costs and enhancing efficiency under industrial conditions. While Cel7A functions optimally at moderate temperatures (40-50 °C), its activity diminishes at higher ranges (50-80 °C). This study integrates Molecular Dynamics (MD) simulations, Machine Learning (ML) predictions, and Molecular Mechanics/Generalized Born Surface Area (MM/GBSA) free energy calculations to develop a peptide-mediated stabilization strategy. Analysis of root mean square fluctuations (RMSF) at 3

Expression of Trichoderma spp. endochitinase gene improves red rot disease resistance in transgenic sugarcane.

Sugarcane (Saccharum spp.)is an economically useful crop grown globally for sugar, ethanol and biofuel production. The crop is vulnerable to fungus Colletotrichum falcatum known to cause red rot disease. The pathogen hydrolyses stalk parenchyma cells where sucrose is accumulated resulting in upto 75% losses in sugar recovery. In this study, transgenic sugarcane having resistance against red rot was developed by introducing Trichoderma spp. endochitinase following Agrobacterium mediated transformation. The transgene introduction and expression in genetically modified plants were verified throug

Structural basis of strict substrate recognition of l-lysine α-oxidase from Trichoderma viride.

l-Lysine oxidase (LysOX) is a FAD-dependent homodimeric enzyme that catalyzes the oxidative deamination of l-lysine to produce α-keto-ε-aminocaproate with ammonia and hydrogen peroxide. LysOX shows strict substrate specificity for l-lysine, whereas most l-amino acid oxidases (LAAOs) exhibit broad substrate specificity for l-amino acids. Previous studies of LysOX showed that overall structural similarity to the well-studied snake venom LAAOs. However, the molecular mechanism of strict specificity for l-lysine was still unclear. We here determined the structure of LysOX in complex with l-lysine

Cellulase production by Trichoderma reesei when cultured on xylose-based media supplemented with sorbose.

The ability of L-sorbose to stimulate cellulase production In shake flask culture of Trichoderma reesei was examined in mineral salts media (initial pH 5.0) containing either 1.0% D-xylose, 1.0% cellulose, and/or 0.1, 0.3, or 0.5% L-sorbose. When sorbose was the only carbon source, growth was limited, little substrate was utilized, pH increased, and cellulase activity was not apparent. The other carbon sources promoted good growth, pH dropped sharply to 2.5-3.0, substrate was utilized rapidly, and cellulase activity was detected. After three weeks of fermentation, twice as much cellulase activ

Multilocus Identification of Indigenous Trichoderma Isolates and Their Biocontrol Mechanisms Against Macrophomina in Northern Australia.

Charcoal rot, caused by the pathogen Macrophomina, is becoming an increasing challenge in Australia's northern cropping systems, with few effective management options available. The use of non-indigenous biocontrol agents raises ecological and regulatory concerns, which highlights the need to identify locally adapted microbial antagonists. In this study, indigenous Trichoderma isolates were collected from rhizosphere soils across Queensland and northern New South Wales and characterised using multilocus sequencing (ITS, tef-1α, rpb2) coupled with phylogenetic analysis. Twenty-six isolates were

Molecular insights into the mutualism that induces iron deficiency tolerance in sorghum inoculated with Trichoderma harzianum.

Iron (Fe) deficiency is a common mineral stress in plants, including sorghum. Although the soil fungus Trichoderma harzianum has been shown to mitigate Fe deficiency in some circumstances, neither the range nor mechanism(s) of this process are well understood. In this study, high pH-induced Fe deficiency in sorghum cultivated in pots with natural field soil exhibited a significant decrease in biomass, photosynthetic rate, transpiration rate, stomatal conductance, water use efficiency, and Fe-uptake in both the root and shoot. However, the establishment of T. harzianum colonization in roots of

Ethanolic Extract of the Fungus Trichoderma asperelloides Induces Ultrastructural Effects and Death on Leishmania amazonensis.

The Trichoderma genus comprises several species of fungi whose diversity of secondary metabolites represents a source of potential molecules with medical application. Because of increased pathogen resistance and demand for lower production costs, the search for new pharmacologically active molecules effective against pathogens has become more intense. This is particularly evident in the case of American cutaneous leishmaniasis due to the high toxicity of current treatments, parenteral administration, and increasing rate of refractory cases. We have previously shown that a fungus from genus Tri

Production of trichodiene by Trichoderma harzianum alters the perception of this biocontrol strain by plants and antagonized fungi.

Trichothecenes are phytotoxic sesquiterpenic mycotoxins that can act as virulence factors in plant diseases. Harzianum A (HA) is a non-phytotoxic trichothecene produced by Trichoderma arundinaceum. The first step in HA biosynthesis is the conversion of farnesyl diphosphate to trichodiene (TD), a volatile organic compound (VOC), catalysed by a sesquiterpene synthase encoded by the tri5 gene. Expression of tri5 in the biocontrol strain Trichoderma harzianum CECT 2413 resulted in production of TD in parallel with a reduction of ergosterol biosynthesis and an unexpected increase in the level of sq
