Chromatin accessibility directly governs flavonoid biosynthesis and indirectly orchestrates cannabinoid production in Cannabis.

Secondary metabolites in hemp enhance its pharmaceutical and nutraceutical value, yet the epigenetic regulatory network underlying secondary metabolite biosynthesis remains poorly understood in hemp. Here, we profiled the inflorescences of two cultivars with different trichome density by integrating metabolomics, transcriptomics and ATAC-seq. Multi-omics data revealed pronounced differences in metabolites (491 differentially accumulated metabolites (DAMs)), transcripts (8343 differentially expressed genes (DEGs)), and chromatin accessibility (11376 different accessibility genes, (DAGs)) betwee

Cellular engineering of Artemisia annua and Artemisia dubia with the rol ABC genes for enhanced production of potent anti-malarial drug artemisinin.

BACKGROUND: Malaria is causing more than half of a million deaths and 214 million clinical cases annually. Despite tremendous efforts for the control of malaria, the global morbidity and mortality have not been significantly changed in the last 50 years. Artemisinin, extracted from the medicinal plant Artemisia sp. is an effective anti-malarial drug. In 2015, elucidation of the effectiveness of artemisinin as a potent anti-malarial drug was acknowledged with a Nobel prize. Owing to the tight market and low yield of artemisinin, an economical way to increase its production is to increase its co

Taxonomic significance of seed macro-micromorphology of Turkish Alcea L. (Malvaceae) through light microscopy and scanning electron microscopy.

Seed morphological properties of 19 taxa belonging to the genus Alcea L. (Malvaceae) distributed in Turkey were investigated using a light microscope and scanning electron microscope to identify their characters and to evaluate their diagnostic value. The seeds are reniform with a rounded apex and base, reniform in shape, and light to dark brown, grayish-brown, or blackish-brown in color. The seed length ranges from 2.22 to 6.5 mm and seed width from 1.72 to 6.5 mm. The indumentum at the ventral and dorsal regions of the seed differs in density. Three types of seed coat ornamentations were obs

Evolutionary diversification of acyl-CoA synthetases underpins hydrophobic barrier formation across diverse tomato tissues and beyond.

The transition of plants from aquatic to terrestrial environments required effective barriers against water loss and UV damage. The plant cuticle, a hydrophobic barrier covering aerial surfaces, emerged as a critical innovation, yet how its biosynthesis is regulated in specialized structures remains poorly understood. This study identifies two long-chain acyl-CoA synthetases, SlLACS1 and SlLACS2, that exhibit both distinct and overlapping functions in cuticle formation across tomato tissues. These genes show striking specificity in different trichome types: SlLACS1 functions in type I/IV trich

Effects of arbuscular mycorrhiza and phosphorus application on artemisinin concentration in Artemisia annua L.

Annual wormwood (Artemisia annua L.) produces an array of complex terpenoids including artemisinin, a compound of current interest in the treatment of drug-resistant malaria. However, this promising antimalarial compound remains expensive and is hardly available on the global scale. Synthesis of artemisinin has not been proved to be feasible commercially. Therefore, increase in yield of naturally occurring artemisinin is an important area of investigation. The effects of inoculation by two arbuscular mycorrhizal (AM) fungi, Glomus macrocarpum and Glomus fasciculatum, either alone or supplement

Genome-wide identification and characterisation of bHLH transcription factors in Artemisia annua.

BACKGROUND: A. annua (also named Artemisia annua, sweet wormwood) is the main source of the anti-malarial drug artemisinin, which is synthesised and stored in its trichomes. Members of the basic Helix-Loop-Helix (bHLH) family of transcription factors (TFs) have been implicated in artemisinin biosynthesis in A. annua and in trichome development in other plant species. RESULTS: Here, we have systematically identified and characterised 226 putative bHLH TFs in A. annua. All of the proteins contain a HLH domain, 213 of which also contain the basic motif that mediates DNA binding of HLH dimers. Of

Specialized structures and developmental mechanisms of secondary metabolite-synthesizing organs and tissues in medicinal plants.

Specialized structures in medicinal plants underpin the spatial regulation of secondary metabolism, determining the biosynthesis, accumulation, and storage of pharmacologically active compounds. Specialized structures, such as glandular trichomes, roots, rhizomes, laticifer, heartwood, and so on, have evolved distinct developmental programs and metabolic regulatory networks, enabling efficient synthesis, storage, and secretion of bioactive compounds. Understanding how these tissues originate, differentiate, and coordinate metabolism is essential not only for elucidating the molecular basis of

Proteomic analysis of Artemisia annua--towards elucidating the biosynthetic pathways of the antimalarial pro-drug artemisinin.

BACKGROUND: MS-based proteomics was applied to the analysis of the medicinal plant Artemisia annua, exploiting a recently published contig sequence database (Graham et al. (2010) Science 327, 328-331) and other genomic and proteomic sequence databases for comparison. A. annua is the predominant natural source of artemisinin, the precursor for artemisinin-based combination therapies (ACTs), which are the WHO-recommended treatment for P. falciparum malaria. RESULTS: The comparison of various databases containing A. annua sequences (NCBInr/viridiplantae, UniProt/viridiplantae, UniProt/A. annua,

Larvicidal efficacy of Varronia curassavica leaf phytochemicals against Aedes aegypti: anatomical, histological and cytotoxic analyses.

Aedes aegypti, the primary vector of diseases like Dengue, Zika, Yellow Fever, and Chikungunya, presents a significant public health challenge, particularly in tropical regions. With rising resistance to conventional insecticides, plant-derived bioinsecticides have emerged as a promising alternative. This study assessed the larvicidal potential of phytochemicals from Varronia curassavica, a species native to the Atlantic Forest. Microscopic analyses revealed various trichome types on V. curassavica leaves, including glandular trichomes containing essential oils, phenolic substances, lipids, al

Reproductive biology of Trichocentrum pumilum: an orchid pollinated by oil-collecting bees.

The reproductive biology, reward production and pollination mechanism of Trichocentrum pumilum were studied in a gallery forest in the interior of the State of São Paulo, southeast Brazil. The floral visitors and pollination mechanism were recorded, and experimental pollinations were carried out in order to determine the breeding system of this species. Trichocentrum pumilum blooms in spring. Each paniculate inflorescence bears an average of 85 flowers that present a central yellow callus and finger-like trichomes on the lateral lobes of the lip. A lipoidal substance is produced and stored amo

Poplar coma morphogenesis and miRNA regulatory networks by combining ovary tissue sectioning and deep sequencing.

Poplar coma, commonly referred to as "seed hairs", is a tuft of trichomes attached to the seed coat that helps seed dispersal. However, they can also trigger health impacts for humans, including sneezing, shortness of breath, and skin irritation. Despite efforts to study the regulatory mechanism of herbaceous trichome formation, poplar coma remains poorly understood. In this study, we showed that the epidermal cells of the funiculus and placenta are the origin of poplar coma based on observations of paraffin sections. Small RNA (sRNA) and degradome libraries were also constructed at three stag

Inhibition of lipoxygenase and prostaglandin endoperoxide synthase by anacardic acids.

C22:1 omega 5-anacardic acid was found to be a good inhibitor of both potato lipoxygenase and ovine prostaglandin endoperoxide synthase with approximate IC50's of 6 and 27 microM, respectively. Very similar inhibition was seen with the crude exudate, rich in omega 5-anacardic acids, from glandular trichomes of an arthropod-resistant strain of geranium, Pelargonium xhortorum. The saturated anacardic acid (C22:0 sat), abundant in the trichome exudate of susceptible strains, was nearly as inhibitory toward both prostaglandin endoperoxide synthase and lipoxygenase as the omega 5-unsaturated compou

Leaf Epidermal Micromorphology of the Korean Angelica (Apiaceae) and Its Taxonomic Implications.

The genus Angelica L. (Apiaceae) is mainly distributed throughout the Northern Hemisphere and comprises approximately 110 species. Although several molecular phylogenetic studies have been conducted on this genus, the leaf micromorphology has not been sufficiently explored. In this study, we examined the leaf micromorphological characteristics of all the 16 Korean Angelica taxa using light and scanning electron microscopy. The results revealed considerable variation in the morphology of stomatal complexes, epidermal cells, and trichomes. Leaf epidermal cells were generally irregular in shape,

Functional divergence of plant SCAR/WAVE proteins is determined by intrinsically disordered regions.

Dynamic actin cytoskeleton reorganization enables plant developmental processes requiring polarized transport such as root hair and leaf trichome formation. The SCAR/WAVE complex plays a crucial role in regulating these dynamics through ARP2/3-mediated actin branching. SCAR/WAVE genes occur as small families across a wide range of plant species, but whether and how they fulfill different functions remains unclear. We use a systematic chimera approach to define the differential functionality of two closely related Medicago truncatula SCAR proteins in plant development. We show that SCAR/WAVE co

Pesticide droplet interfacial behavior and drying deposition characteristics on apple leaf surfaces at different growth stages: role of nonionic surfactant self-assembly.

The physicochemical properties of pesticide solutions can be effectively altered via surfactant addition, which significantly influences the wetting, bouncing behavior and deposition efficiency of pesticide droplets. We combined high-speed imaging, contact angle measurements, and infrared thermography to track droplet impact, spreading, and drying on apple leaves at three growth stages-flower bud, young fruit, and fruit enlargement. Deep-focus microscopy observations revealed that leaf surfaces at different growth and developmental stages possess trichomes of varying densities, which exert a n

Population diversity analyses provide insights into key horticultural traits of Chinese native thymes.

Chinese native thymes (CNTs) in the genus Thymus (family Lamiaceae) are rich in bioactive terpenes, which exert antiviral, anti-inflammatory, antioxidation, immunological, and antimicrobial effects. Plants exhibit morphological variation, including erect-type and creeping-type growth forms; however, the molecular mechanisms underlying important horticultural traits have not been determined. Here, we collected 39 CNTs providing strategic plant resources for studies of lignin, terpenoids, and glandular trichomes of thymes. Using resequencing data as well as phenotypic, metabonomic, phylogenetic,

Capillarity Constructed Open Siphon for Sustainable Drainage.

Siphon is an effective method to transfer liquid from a higher to a lower level, which has many applications in hygienic design, clinical apparatus, and hydraulic engineering. Traditional operation requires energy to overcome gravity and establish flow in a closed system. Achieving sustainable high flux siphon drainage without energy input remains a challenge due to viscous dissipation. Here, an unexpected open siphon behavior on the South American pitcher plant Heliamphora minor consisting of trichomes covered pitcher and a wedge-shaped sheath is examined. NAPP and PIRP encode subunits of a putative wave regulatory protein complex involved in plant cell morphogenesis.

The ARP2/3 complex is an important regulator of actin nucleation and branching in eukaryotic organisms. All seven subunits of the ARP2/3 complex have been identified in Arabidopsis thaliana, and mutation of at least three of the subunits results in defects in epidermal cell expansion, including distorted trichomes. However, the mechanisms regulating the activity of the ARP2/3 complex in plants are largely unknown. In mammalian cells, WAVE and WASP proteins are involved in activation of the ARP2/3 complex. WAVE1 activity is regulated by a protein complex containing NAP1/HEM/KETTE/GEX-3 and PIR1

HOMEODOMAIN-LIKE shapes chromatin architecture and rewires leaf epidermal patterns.

During development, cells balance proliferation and differentiation to generate diverse cell types, yet how these transitions are fine-tuned remains unclear. In the Arabidopsis leaf epidermis, stomata form through regulated cell division, whereas pavement cells and trichomes arise through differentiation, making this tissue a tractable system for identifying modulators of developmental decisions. Here, we identified a previously uncharacterized homeodomain-like protein HDL, that is required for chromocenter organization through interactions with the histone deacetylase HDA6 and the methyltrans
