Combination of glycyrrhizic acid and compound probiotics alleviates deoxynivalenol-induced damage to weaned piglets.

Deoxynivalenol (DON) can affect health and growth performance of pigs, resulting in significant economic losses in swine production. The aim of this study was to investigate the effect of glycyrrhizic acid combined with compound probiotics, i.e. Enterococcus faecalis plus Saccharomyces cerevisiae (GAP) on improving growth performance, intestinal health and its fecal microbiota composition change of piglets challenged with DON. A total of 160 42-day-old weaned piglets (Landrace × Large White) were used and the experimental period was 28 d. The results showed that supplementing GAP in the diet s

Deoxynivalenol: Emerging Toxic Mechanisms and Control Strategies, Current and Future Perspectives.

Deoxynivalenol (DON) is the most frequently present mycotoxin contaminant in food and feed, causing a variety of toxic effects in humans and animals. Currently, a series of mechanisms involved in DON toxicity have been identified. In addition to the activation of oxidative stress and the MAPK signaling pathway, DON can activate hypoxia-inducible factor-1α, which further regulates reactive oxygen species production and cancer cell apoptosis. Noncoding RNA and signaling pathways including Wnt/β-catenin, FOXO, and TLR4/NF-κB also participate in DON toxicity. The intestinal microbiota and the brai

Global distribution, toxicity to humans and animals, biodegradation, and nutritional mitigation of deoxynivalenol: A review.

Deoxynivalenol (DON) is one of the main types of B trichothecenes, and it causes health-related issues in humans and animals and imposes considerable challenges to food and feed safety globally each year. This review investigates the global hazards of DON, describes the occurrence of DON in food and feed in different countries, and systematically uncovers the mechanisms of the various toxic effects of DON. For DON pollution, many treatments have been reported on the degradation of DON, and each of the treatments has different degradation efficacies and degrades DON by a distinct mechanism. The

ZEA and DON inhibited inflammation after L. monocytogenes infection and induced ribosomal hyperfunction.

The complex microbial community in food environment is a major problem of human or animal health and safety. Mycotoxins and food-borne bacteria can both induce inflammation in the body and cause a series of changes in biological functions. In this study, mice were gavaged with low doses of ZEA, DON, or ZEA + DON, and then infected with L. monocytogenes. A cytokine microarray, including 40 inflammation-related serum cytokines, and proteomics were used to verify the effects of ZEA, DON, and ZEA + DON on the host inflammation and biological function after L. monocytogenes infection. The results s

Lactobacillus plantarum metabolites reduce deoxynivalenol toxicity on jejunal explants of piglets.

The deterioration of food and feed stuffs and toxic intestinal effects due to fungal colonization and concomitant production of mycotoxins is an increasing concern. The development of fungi resistance to many commonly used chemical preservatives adds further alarm. Therefore, effective detoxification methods would be useful in counteracting this problem. Biotransformation/adsorption of mycotoxins by lactic acid bacteria and their metabolites is a promising approach to minimize the deleterious effects of mycotoxins. The objective of the present study was to evaluate the beneficial effects of La

Down-regulation of the endoplasmic reticulum chaperone GRP78/BiP by vomitoxin (Deoxynivalenol).

The mechanisms by which trichothecene mycotoxins cause immunological effects in leukocytes such as cytokine up-regulation, aberrant IgA production, or apoptotic cell death are not fully understood. In the present study, mRNA differential display analysis was used to evaluate changes in gene expression induced by the trichothecene vomitoxin (VT or deoxynivalenol) in a T-cell model, the murine EL-4 thymoma, that was stimulated with phorbol 12-myristate 13-acetate (PMA) and ionomycin (ION). Ten differentially expressed fragments of cDNA were isolated and sequenced and three of these were identifi

Public health impacts of foodborne mycotoxins.

Mycotoxins are toxic and carcinogenic metabolites produced by fungi that colonize food crops. The most agriculturally important mycotoxins known today are aflatoxins, which cause liver cancer and have also been implicated in child growth impairment and acute toxicoses; fumonisins, which have been associated with esophageal cancer (EC) and neural tube defects (NTDs); deoxynivalenol (DON) and other trichothecenes, which are immunotoxic and cause gastroenteritis; and ochratoxin A (OTA), which has been associated with renal diseases. This review describes the adverse human health impacts associate

Acute lead arsenate poisoning in beef cattle in Uruguay.

We describe and illustrate lesions in an outbreak of lead arsenate poisoning in beef cattle that ingested pesticide residues stored in an abandoned building of a former orange orchard. Of 70 exposed cattle, 14 had diarrhea, paresis, ataxia, recumbency, and/or seizures. Ten of the affected animals died after a clinical course of 12-18 h. Pathologic findings in 3 steers included extensive necrohemorrhagic, ulcerative rumenitis, omasitis, and abomasitis; lymphocytolysis in lymphoid organs; and nephrosis. Hepatic arsenic and lead levels in cases 1-3 were 20, 24, and 31 ppm, and 8.3, 25, and 9.4 pp

Effects of fusariotoxins in the equine species.

Fumonisins, trichothecenes and zearalenone are the most commonly occurring Fusarium mycotoxins in cereal grains and animal feed. In this review, the toxicity of these mycotoxins in horses is considered with particular reference to recent data on specific and proposed syndromes. Compared to other animal species, very little information is available on the adverse effects of fusariotoxins in horses. Fumonisin B(1) (FB(1)) is the causative agent of leukoencephalomalacia, which is typified by depression, aimless circling, head pressing, paresis, ataxia, blindness and death. FB(1) has also been sho

Deoxynivalenol drives liver injury progression by dysregulating core molecular networks: integrated multi-omics, network toxicology and molecular docking analysis.

BACKGROUND: Deoxynivalenol (DON), a prevalent food-borne mycotoxin, increasingly recognized as a potent driver in the progression of chronic liver disease to cirrhosis and hepatocellular carcinoma (HCC); however, its systematic role is unclear. This study aims to decode the pathogenic networks of DON through an integrated multi-omics and toxicological framework. METHODS: We integrated transcriptomic datasets from public repositories (GSE139602 and GSE25097) and single-cell RNA-seq data (GSE136103 and GSE149614) with toxicogenomics data. Analytical approaches included differential expression a

Mycotoxins: Emerging toxic mechanisms, and unanswered research questions.

Recently, a series of toxic mechanisms have been explored in mycotoxins. Emerging evidence show that mycotoxins may induce human neurodegenerative diseases (ND); however, this idea is still unproven. Besides to identify this hypothesis, some questions, for example, how the mycotoxins induce this disease and what the molecular mechanism is, as well as whether the brain-gut axis is involved in this context, should be answered. Very recent studies further reported an "immune evasion" mechanism in trichothecenes; moreover, hypoxia seems to play important function in this process; nevertheless, whe

Venturicidin A Is a Potential Fungicide for Controlling Fusarium Head Blight by Affecting Deoxynivalenol Biosynthesis, Toxisome Formation, and Mitochondrial Structure.

Fusarium graminearum, which causes Fusarium head blight (FHB) in cereals, is one of the most devastating fungal diseases by causing great yield losses and mycotoxin contamination. A major bioactive ingredient, venturicidin A (VentA), was isolated from Streptomyces pratensis S10 mycelial extract with an activity-guided approach. No report is available on antifungal activity of VentA against F. graminearum and effects on deoxynivalenol (DON) biosynthesis. Here, VentA showed a high antagonistic activity toward F. graminearum with an EC50 value of 3.69 μg/mL. As observed by scanning electron micro

Resveratrol Alleviated T-2 Toxin-Induced Liver Injury via Preservation of Nrf2 Pathway and GSH Synthesis.

T-2 toxin is a trichothecene mycotoxin and is considered as an extremely inevitable pollutant with potent hepatotoxicity. However, the approach to alleviation of T-2 toxin-triggered hepatotoxicity has been recognized as a serious challenge. Resveratrol (Res) is a polyphenol natural product isolated from various plant species, but its protective effect against T-2 toxin hepatotoxicity and detailed mechanism remains obscure. In the present study, the effect of Res against the hepatotoxicity was evaluated, and the underlying mechanisms were further revealed in mice. Functionally, Res inhibited li

Listeriosis in diacetoxyscirpenol-treated mice.

Mice were treated with the trichothecene mycotoxin diacetoxyscirpenol (DAS) and subsequently were inoculated intraperitoneally with Listeria monocytogenes. The effect of the mycotoxin on the course of the infection was monitored by observing the resultant mortality and the bacterial content of the spleens from inoculated mice. Mice given 3 mg of DAS/kg of body weight, PO, at days -2 and -1 before inoculation had increased mortality and splenic Listeria counts. In these mice, thymus weights were reduced, and lymphocytes were depleted from the thymus cortex and from splenic lymphoid follicles an

T-2 toxin as an emetic factor in moldy corn.

Extracts of Fusarium poae (NRRL 3287) grown either on sterile corn at 8 C or in Richards solution at room temperature were shown to have emetic activity in pigeons at nonlethal concentration under conditions of oral and intravenous administration. The causative agent was found to be T-2 toxin (3-hydroxy-4,15-diacetoxy-8-[3-methylbutyryloxy]-12,13-epoxy-Delta(9)-trichothecene). Oral and intravenous mean toxic dose values for this compound were found to be 0.72 and 0.15 mg/kg, respectively, as compared with an oral mean lethal dose of 2.75 mg/kg. The fact that T-2 toxin causes emesis at nonletha

Alcoholic beer fermentation: impact of glutathione and peroxidase fortification on deoxynivalenol reduction.

The use of contaminated cereals in fermentation processes can elicit a marked response from yeasts in the presence of mycotoxins, as indicated by the production of defense compounds such as peroxidase (PO) and glutathione (GSH) to reduce mycotoxin damage. The objective of this study was to investigate the effect of fortifying GSH and PO levels in alcoholic fermentation as a strategy for reduce concentration deoxynivalenol (DON) during the brewing process. The QuEChERS method was validated for quantifying the mycotoxin and estimating reduction. Fermentation was conducted in a reactor containing

Structure-function characterization of an aldo-keto reductase involved in detoxification of the mycotoxin, deoxynivalenol.

Deoxynivalenol (DON) is a mycotoxin, produced by filamentous fungi such as Fusarium graminearum, that causes significant yield losses of cereal grain crops worldwide. One of the most promising methods to detoxify this mycotoxin involves its enzymatic epimerization to 3-epi-DON. DepB plays a critical role in this process by reducing 3-keto-DON, an intermediate in the epimerization process, to 3-epi-DON. DepBRleg from Rhizobium leguminosarum is a member of the new aldo-keto reductase family, AKR18, and it has the unusual ability to utilize both NADH and NADPH as coenzymes, albeit with a 40-fold

Constitutively active microglial populations limit anorexia induced by the food contaminant deoxynivalenol.

Microglia are involved in neuroinflammatory processes during diverse pathophysiological conditions. To date, the possible contribution of these cells to deoxynivalenol (DON)-induced brain inflammation and anorexia has not yet been evaluated. DON, one of the most abundant trichothecenes found in cereals, has been implicated in mycotoxicosis in both humans and farm animals. DON-induced toxicity is characterized by reduced food intake, weight gain, and immunological effects. We previously showed that exposure to DON induces an inflammatory response within the hypothalamus and dorsal vagal complex
