Cadmium (Cd) is a toxic pollutant widely present in the environment that can enter the human body through diet and inhalation, posing a serious threat to ecosystems and human health. Cd exposure can cause toxicity in multiple organs, but the mechanism underlying Cd-induced pulmonary fibrosis remains incompletely understood. This study investigated the molecular mechanisms of Cd-induced pulmonary injury using both in vivo (porcine lung tissue) and in vitro (MLE-12 cells) experiments. The results showed that Cd exposure led to mitochondrial damage, iron deposition, decreased antioxidant capacity

Cryptosporidium parvum is one of several Cryptosporidium spp. that cause the parasitic infection cryptosporidiosis. Cryptosporidiosis is a diarrheal infection that is spread via the fecal-oral route and is commonly caused by contaminated drinking water. Triosephosphate isomerase is an enzyme that is ubiquitous to all organisms that perform glycolysis. Triosephosphate isomerase catalyzes the formation of glyceraldehyde 3-phosphate from dihydroxyacetone phosphate, which is a critical step to ensure the maximum ATP production per glucose molecule. In this paper, the 1.55 Å resolution crystal stru

Triokinase/FMN cyclase (TKFC) is a bifunctional enzyme that catalyzes both ATP-dependent triose phosphorylation and the formation of riboflavin 4',5'-cyclic phosphate (cFMN) from FAD, thereby linking fructose and flavin metabolism through a single protein. Structural modeling supports a model of TKFC as a conformationally dynamic homodimer in which interacting K and L domains cooperate through composite intersubunit active sites to support both kinase and cyclase reactions. Beyond its classical role in fructose metabolism, TKFC has emerged as a determinant of fructose-derived carbon partitioni

Methylglyoxal (MGO), a highly reactive 1,2-dicarbonyl, is generated by all three principal pathways of advanced glycation end product (AGE) synthesis in type 2 diabetes mellitus (T2DM)-the Hodge, Namiki, and Wolff pathways-and by the non-enzymatic degradation of glycolytic triose phosphates. It is the principal substrate of the glutathione-dependent GLO1/GLO2 glyoxalase system and the main source of the hydroimidazolone-1 (MG-H1) adduct, and it directly modifies intracellular proteins across multiple tissues. This clinically oriented narrative review synthesizes dicarbonyl chemistry, glyoxalas

To date, genotyping data on giardiasis have not been available in the Czech Republic. In this study, we characterized 47 human isolates of Giardia intestinalis from symptomatic as well as asymptomatic giardiasis cases. Genomic DNA from trophozoites was tested by PCR-sequence analysis at three loci (β-giardin, glutamate dehydrogenase and triose phosphate isomerase). Sequence analysis showed assemblages A and B in 41 (87.2%) and six (12.8%) isolates, respectively. Two of the 41 assemblage A samples were genotyped as sub-assemblage AI, and 39 were genotyped as sub-assemblage AII. Four previously

Little is known about the genetic diversity of the protozoan parasite, Giardia duodenalis, infecting humans in Queensland, Australia. The present study typed 88 microscopically Giardia-positive isolates using assemblage-specific primers at the triose phosphate isomerase (tpi) gene and sequenced a subset of isolates at the glutamate dehydrogenase (gdh) gene (n = 30) and tpi locus (n = 27). Using the tpi-assemblage specific primers, G. duodenalis assemblage A and assemblage B were detected in 50% (44/88) and 38·6% (34/88) of samples, respectively. Mixed infections with assemblages A and B were i

Giardia intestinalis is a cosmopolitan protozoan parasite that can infect a range of animals, including dairy cattle. As information regarding the prevalence and genotyping of G. intestinalis infection in dairy cattle in northwestern China is limited, 2,945 feces samples from 1,224 dairy cattle in Gansu Province and from 1,614 in Ningxia Hui Autonomous Region (NXHAR) were examined between December 2012 and March 2014. The overall prevalence of G. intestinalis was 3.63% (107/2,945), with 2.63% and 4.38% in Gansu and NXHAR, respectively. Logistic regression analysis showed region, age and season

Paracoccidioides species are dimorphic fungi and are the etiologic agents of paracoccidioidomycosis, which is a serious disease that involves multiple organs. The many tissues colonized by this fungus suggest a variety of surface molecules involved in adhesion. A surprising finding is that most enzymes in the glycolytic pathway, tricarboxylic acid (TCA) cycle and glyoxylate cycle in Paracoccidioides spp. have adhesive properties that aid in interacting with the host extracellular matrix and thus act as 'moonlighting' proteins. Moonlighting proteins have multiple functions, which adds a dimensi

The functional significance of a non-conserved cysteine residue (C86) proximal to the interfacial region of Rhipicephalus microplus triosephosphate isomerase (RmTIM) was investigated through systematic substitution with aspartic acid (C86D), lysine (C86K), and alanine (C86A) via site-directed mutagenesis. Kinetic analyses revealed substantial perturbations in enzymatic parameters for the C86D and C86K variants, with marked alterations in maximal velocity (Vmax), substrate affinity (Km), catalytic turnover (kcat), and catalytic efficiency (kcat/Km). Thermodynamic destabilization was universally

Background/Objectives: Triosephosphate Isomerase (TPI) is a glycolytic enzyme known to be associated with TPI deficiency, a severe form of childhood-onset glycolytic enzymopathy associated with hemolytic anemia, neuromuscular impairment and early death. Most often the disease results from the common TPI1E105D mutation, which can be either homozygous or compound heterozygous with another allele. Methods: We purified TPIR5G protein, studied mutant protein biochemistry, established and characterized TPIR5G/f.s.patient cells, and investigated newly identified compounds for their efficacy in vitro

Triosephosphate isomerase (TPI) is best known as a glycolytic enzyme that interconverts the 3-carbon sugars dihydroxyacetone phosphate (DHAP) and glyceraldehyde-3-phosphate (G3P). TPI is an essential enzyme that is required for the catabolism of DHAP and a net yield of ATP from anaerobic glucose metabolism. Loss of TPI function results in the recessive disease TPI Deficiency (TPI Df). Recently, numerous lines of evidence suggest the TPI protein has other functions beyond glycolysis, a phenomenon known as moonlighting or gene sharing. Here we review the numerous functions ascribed to TPI, inclu

Trypanosoma cruzi (T. cruzi) is a parasite that affects humans and other mammals. T. cruzi depends on glycolysis as a source of adenosine triphosphate (ATP) supply, and triosephosphate isomerase (TIM) plays a key role in this metabolic pathway. This enzyme is an attractive target for the design of new trypanocidal drugs. In this study, a ligand-based virtual screening (LBVS) from the ZINC15 database using benzimidazole as a scaffold was accomplished. Later, a molecular docking on the interface of T. cruzi TIM (TcTIM) was performed and the compounds were grouped by interaction profiles. Subsequ

Triosephosphate isomerase deficiency constitutes a rare autosomal recessive disorder, characterized by hemolytic anemia, neurodegeneration, and recurrent bacterial infections. It is the most severe glycolytic enzyme defect associated with progressive neurologic dysfunction. Patients with various inherited triosephosphate isomerase deficiency gene mutations were identified. The most frequent is a Glu104Asp mutation, manifested in homozygous and compound heterozygous states. The mutation Val231Met is very rare. We describe a second triosephosphate isomerase-deficient patient homozygous for the V

The formose reaction is a unique chemical reaction for the preparation of saccharides from formaldehyde, a single carbon compound. We applied zeolite materials as heterogeneous catalysts to the formose reaction. The simple addition of Linde type A zeolite containing calcium ions (Ca-LTA) to an aqueous solution of formaldehyde and glycolaldehyde produced saccharides at room temperature. A quantitative analysis performed by high-performance liquid chromatography revealed that triose, tetrose, pentose, and hexose saccharides were produced with few byproducts. Ca-LTA was recovered from the reactio

Schistosomiasis is recognized as a tropical disease of considerable public health importance, but domestic livestock infections due to Schistosoma japonicum, S. bovis, S. mattheei and S. curassoni are often overlooked causes of significant animal morbidity and mortality in Asia and Africa. In addition, whereas schistosomiasis japonica is recognized as an important zoonosis in China and the Philippines, reports of viable schistosome hybrids between animal livestock species and S. haematobium point to an underappreciated zoonotic component of transmission in Africa as well.
