Variations in the carbohydrate expression pattern and in lesions of the uterine horns of BALB/c mice infected with different Tritrichomonas foetus isolates.

Bovine tritrichomonosis, a sexually transmitted disease caused by the protozoan Tritrichomonas foetus, is characterized by producing reproductive alterations in cattle. Carbohydrates on the surface of the uterine epithelium are involved in the process of adhesion and colonization of the protozoan. The murine model has proved to be an inexpensive, practical and representative alternative to study the lesions produced in the natural host. For this work, during the first stage, 6-8 week old female BALB/c mice were inoculated with 24 different T. foetus isolates in order to classify them according

Emerging Trichomonad Infections in Companion Animals: Rapid Visual Detection of Pentatrichomonas hominis and Tritrichomonas foetus Using an RPA-CRISPR/Cas12a Assay.

Pentatrichomonas hominis (P. hominis) and Tritrichomonas foetus (T. foetus) are prevalent intestinal protozoa. P. hominis is associated with chronic diarrhea in humans and animals, whereas T. foetus causes gastrointestinal disease in companion animals and reproductive-tract infection in cattle. Rapid and accurate identification of these infections at the point-of-care (POC) is crucial for the diagnosis and effective management of zoonotic diseases. In this study, we developed two novel recombinase polymerase amplification (RPA) assays coupled with CRISPR/Cas12a detection. The dual-species assa

Comparing vaginal douche, cervicovaginal mucus, and uterine lavage for diagnosis of Tritrichomonas foetus in naive heifers exposed to a naturally infected bull.

OBJECTIVE: This study aimed to compare the effectiveness of quantitative PCR (qPCR) in detecting Tritrichomonas foetus from 3 sample types-vaginal douche, cervicovaginal mucus, and low-volume uterine lavage-in naïve heifers exposed to a naturally infected bull. METHODS: 11 naïve heifers were exposed for 30 days to a bull naturally infected with T foetus. Three sampling locations were used to collect diagnostic-material vaginal douche, cervical mucus aspirate, and low-volume uterine lavage and submitted for qPCR. Three sampling time periods were performed 21 days apart. RESULTS: 12 out of the

Comparative evaluation of a commercial Tritrichomonas foetus vaccine in infected and uninfected bulls and heifers during a 45-day natural breeding season.

Tritrichomonas foetus is a venereal protozoan parasite that causes significant reproductive losses in cattle. Bulls act as asymptomatic carriers, transmitting the pathogen to susceptible cows, leading to infertility, embryonic or fetal loss, and other reproductive disorders. With no FDA-approved treatments available, vaccination has become a key control strategy. This study evaluated the reproductive effects of a commercially available killed, whole-cell vaccine (TrichGuard®) administered to both heifers and T. foetus-positive bulls in a natural mating system. Heifers and bulls were randomly a

Noninferiority of cycle threshold values from 0.9% sterile saline compared with PBS as a collection medium for Tritrichomonas foetus RT-rtPCR testing.

Tritrichomonas foetus infection can cause reproductive losses and subsequent economic losses in cow-calf herds. Our objective was to determine if 0.9% sterile saline (saline) was noninferior to PBS as a transport medium for T. foetus reverse-transcription real-time PCR (RT-rtPCR) testing. Transport tubes were prepared with either 1.6 mL of PBS with preputial washing (n = 30) or 1.6 mL of saline with preputial washing (n = 30) and inoculated at 3 different concentrations of T. foetus organisms (high = 100 org/100 µL; moderate = 10 org/100 µL; low = 1 org/100 µL) for a total volume of 1.7 mL/tub

A noninferiority trial comparing the sensitivity and specificity of Tritrichomonas foetus RT-rtPCR using 0.9% sterile saline versus PBS as transport medium.

The reported limit of detection (LOD) for Tritrichomonas foetus reverse-transcription real-time PCR (RT-rtPCR) is one organism/extraction in PBS without prior culture. Our objective was to determine if 0.9% sterile saline (saline) was noninferior to PBS as a transport medium for T. foetus RT-rtPCR testing at the LOD. Each week for 10 wk, preputial washing samples were collected from known T. foetus-negative bulls, centrifuged, and combined into a single pellet that was evenly distributed into stock solutions for both media. The stock solutions were used to prepare sixty 1.3-mL samples of each

Detection of Tritrichomonas foetus by polymerase chain reaction in cultured isolates, cervicovaginal mucus, and formalin-fixed tissues from infected heifers and fetuses.

A rapid, reliable polymerase chain reaction (PCR) assay, originally developed for definitive laboratory identification of the bovine venereal pathogen Tritrichomonas foetus from cultures of male reproductive tract fluids, was used for testing the following: 1) cultured, geographically disparate trichomonad isolates, 2) formalin-fixed tissues from infected heifers and naturally infected fetuses, and 3) cervicovaginal mucus (CVM) from experimentally infected females. In 12 of 12 Western Hemisphere isolates of pathogenic T. foetus (isolated from outbreaks of clinical trichomoniasis or from screen

Comparison of the diagnostic sensitivity of a commercially available culture kit and a diagnostic culture test using Diamond's media for diagnosing Tritrichomonas foetus in bulls.

A number of different culture media have been described for use in the diagnosis of Tritrichomonas foetus infection in bulls, and recently, a commercial culture kit has become available. The objective of this study was to compare the sensitivity of 2 culture-based diagnostic tests for T. foetus in bulls. One test used a commercial kit for transport and culture of the samples. The other test used a thioglycollate transport medium (TFTM) for transport and a modified Diamond's medium (MDM) for culture of the samples. Twenty-one bulls infected with T. foetus were sampled repeatedly. On each sampli

Anti-Retroviral Lectins Have Modest Effects on Adherence of Trichomonas vaginalis to Epithelial Cells In Vitro and on Recovery of Tritrichomonas foetus in a Mouse Vaginal Model.

Trichomonas vaginalis causes vaginitis and increases the risk of HIV transmission by heterosexual sex, while Tritrichomonas foetus causes premature abortion in cattle. Our goals were to determine the effects, if any, of anti-retroviral lectins, which are designed to prevent heterosexual transmission of HIV, on adherence of Trichomonas to ectocervical cells and on Tritrichomonas infections in a mouse model. We show that Trichomonas Asn-linked glycans (N-glycans), like those of HIV, bind the mannose-binding lectin (MBL) that is part of the innate immune system. N-glycans of Trichomonas and Tritr

How laminin-1 can be recognized by the protozoan parasite Tritrichomonas foetus: possible role played by the extracellular matrix glycoprotein in both cytoadhesion and cytotoxicity exerted by the parasite.

The isoform 1 of the extracellular matrix glycoprotein Laminin is known to be an important ligand for some parasitic protozoa including Trichomonas vaginalis. The bovine parasite Tritrichomonas foetus seems to display a similar recognition process to laminin-1, as some amino acid sequences found in the LNS module of laminin-1 can also be recognized by this parasite. Which of the laminin-1 residing adhesion sequences are recognized by T. foetus, and the role played by such a protein-cell recognition process in both cytoadhesion and cytotoxicity exerted by the parasite are the subjects briefly r

High prevalence of Tritrichomonas foetus 'bovine genotype' in faecal samples from domestic pigs at a farm where bovine trichomonosis has not been reported for over 30 years.

Bovine venereal trichomonosis caused by the flagellate Tritrichomonas foetus is a notifiable disease in Australia. While, T. foetus is pathogenic in both cattle and cats, it has long been established that the same T. foetus colonises the stomach, caecum and nasal cavity of pigs without apparent clinical significance. Multi-locus genotyping grouped the non-pathogenic porcine T. foetus with the pathogenic 'bovine genotype', rather than with the 'feline genotype' T. foetus. Bovine trichomonosis is now uncommon due to wide-spread use of artificial insemination, however, whether T. foetus remains p

Metabolomic profiling and stable isotope labelling of Trichomonas vaginalis and Tritrichomonas foetus reveal major differences in amino acid metabolism including the production of 2-hydroxyisocaproic acid, cystathionine and S-methylcysteine.

Trichomonas vaginalis and Tritrichomonas foetus are pathogens that parasitise, respectively, human and bovine urogenital tracts causing disease. Using LC-MS, reference metabolomic profiles were obtained for both species and stable isotope labelling with D-[U-13C6] glucose was used to analyse central carbon metabolism. This facilitated a comparison of the metabolic pathways of T. vaginalis and T. foetus, extending earlier targeted biochemical studies. 43 metabolites, whose identities were confirmed by comparison of their retention times with authentic standards, occurred at more than 3-fold dif

Ronidazole in the treatment of trichomonad infections in cats.

Tritrichomonas foetus, a microscopic single-celled flagellated protozoan parasite, traditionally identified as a cause of reproductive disease in cattle, has been demonstrated as an important cause of diarrhea in cats. Until recently, an effective antimicrobial treatment for feline Tritrichomonas foetus had not been idnetified. Since recommended dosages of antimicrobial drugs have failed in cats infected with Tritrichomonas foetus and in vitro studies have revealed multiple drug resistance, investigations continue in the effort to find an effective treatment. One particular study by Dr.Jody Go

Bovine Trichomonosis Cases in the United States 2015-2019.

Tritrichomonas foetus is a sexually-transmitted protozoan parasite that causes early embryonic death in cattle. Tritrichomonas foetus is enzootic in the United States but is not a reportable disease at the national level. Thus, it is difficult to understand the prevalence and relative distribution of the disease for the purpose of developing appropriate control measures. In this study, a survey of state veterinarians was used to determine the number of reported cases in each state from 2015 to 2019. Our investigation revealed infections in 25 different states and a total of 3,817 reported case

Comparative sequence analysis of 5.8S rRNA genes and internal transcribed spacer (ITS) regions of trichomonadid protozoa.

The taxonomic situation in the genus Tritrichomonas is the subject of controversial discussion: potentially T. foetus and T. suis, the tritrichomonads from cattle and swine, respectively, could belong to the same species. In order to shed some light on this question, a molecular biological analysis was performed. The 5.8S rRNA gene and the flanking internal transcribed spacer regions (ITS1 and ITS2) of 12 different isolates of 3 Tritrichomonas species T. foetus, T. suis and T. mobilensis were enzymatically amplified by PCR and subcloned. Also, the corresponding regions of the trichomonads Tric

Diagnosis of trichomoniasis in 'virgin' bulls by culture and polymerase chain reaction.

The diagnostic test for Tritrichomonas foetus in bulls is microscopic examination of cultured preputial samples. Trichomonads other than T. foetus can be present in a preputial sample. Both a staining technique and a polymerase chain reaction assay were useful in differentiating between T. foetus and another trichomonad observed in samples from virgin bulls.

Investigations on the prevalence and potential pathogenicity of intestinal trichomonads in pigs using in situ hybridization.

In pigs, three different trichomonad species (Tritrichomonas foetus, Tetratrichomonas buttreyi and Tritrichomonas rotunda) have been described as commensals in the large intestine. The aim of this study was to gain further knowledge on the prevalence and pathogenicity of trichomonads in pigs by using a morphology-based approach. Chromogenic in situ hybridization (ISH) is a technique which allows direct localization of the protozoa in the intestinal tissue and correlation of the infection with pathologic changes. In the present study paraffin-wax embedded colon and ileum samples of 192 pigs wer
