Dynamic physiological and molecular changes in gastric ulcer healing achieved by melatonin and its precursor L-tryptophan in rats.

Following induction of gastric ulcer in rats by serosal application of acetic acid, local mucosal necrosis ensues accompanied by a reduction in mucosal microcirculation and by almost immediate expression of inducible nitric oxide (NO) synthase (iNOS) and proinflammatory cytokines. Daily application of melatonin (20 mg/kg) or l-tryptophan (100 mg/kg) accelerates ulcer healing by affecting the cyclooxygenase-2 (COX-2)-prostaglandin (PG) system with excessive production of protective PG, especially in later period of ulcer healing. Furthermore, expression of hypoxia inducible factor, vascular-end

Mechanisms of the Eimeria tenella growth inhibitory activity induced by concanavalin A and reticuloendotheliosis virus supernatants with interferon gamma activity in chicken macrophages and fibroblasts.

Pretreatment of chicken bone marrow macrophages and embryo fibroblasts with supernatants containing chicken interferon gamma (IFN-gamma) for 24 hr prior to inoculation inhibited intracellular Eimeria tenella replication, measured by [3H] uracil incorporation. The supernatants (Sns) were obtained from culture of lymphoblastoid cells transformed by a reticuloendotheliosis virus (REV) and chicken splenocytes stimulated with concanavalin A (Con A). The mechanisms of the E. tenella growth inhibitory activity induced by Sn REV and Sn Con A in chicken macrophages and fibroblasts were studied. Additio

A unique substrate specificity of PonAAS2, an aromatic aldehyde synthase, involved in phytohormone auxin biosynthesis in a gall-inducing sawfly Euura sp. "Pontania".

The aromatic aldehyde synthase (AAS), PonAAS2, from the gall-inducing sawfly has been identified as a biosynthetic enzyme for indole-3-acetic acid (IAA), a key molecule of the plant hormone auxin, which is thought to play a role in gall induction. Unlike other insect AASs that convert Dopa, PonAAS2 uniquely converts l-tryptophan (Trp) into indole-3-acetaldehyde, a precursor of IAA. In this study, an examination of AAS enzymes from various insect species revealed that the ability to convert Trp has been acquired in only a very limited taxonomic group. Comparative analysis between PonAAS2 and Dj

[The metabolic basis of the hyperphenylalaninemias and tyrosinemia].

The hyperphenylalaninemias are caused by the defect of either phenylalanine hydroxylase (PAH) or tetrahydrobiopterin (BH4) cofactor. The former is diagnosed as phenylketonuria (PKU) or benign hyperphenylalaninemia, based on the serum phenylalanine values. The latter, so called malignant hyperphenylalaninemia, includes three enzyme defects, dihydropteridine reductase (DHPR), 6-pyruvoyl tetrahydropterin synthase (PT PS) and guanosine triphosphate cyclohydrolase (GTP-CH). Excess phenylalanine and its metabolites cause brain damage before 6 years of age. Deficiency of BH4 impairs two other hydroxy

Quantitative analysis of the proteome and protein oxidative modifications in primary human coronary artery endothelial cells and associated extracellular matrix.

Vascular endothelial cells (ECs) play a key role in physiology by controlling arterial contraction and relaxation, and molecular transport. EC dysfunction is associated with multiple pathologies. Here, we characterize the cellular and extracellular matrix (ECM) proteomes of primary human coronary artery ECs, from multiple donors, and oxidation/nitration products formed on these during cell culture, using liquid chromatography-mass spectrometry. In total ∼9900 proteins were identified in cells from 3 donors, with ∼7000 proteins per donor. Of these ∼5300 were consistently identified, indicating

Mechanism of auranofin toxicity: Inhibition of tetrahydrobiopterin metabolism.

Auranofin is a gold-containing anti-inflammatory drug used for the treatment of rheumatoid arthritis. Recent studies indicate that auranofin targets cellular antioxidants including the thioredoxin system and the ubiquitin-proteasome system, which can result in cellular oxidative stress. In the present studies, we identified a novel mechanistic site of action of auranofin, namely tetrahydrobiopterin (BH4) biosynthesis. BH4 is an essential cofactor required for aromatic amino acid hydroxylases, nitric oxide synthase and alkylglycerol monooxygenase, enzymes that produce mediators important in reg

Sulfonated peptide alleviates hypertensive renal injury via antioxidant, anti-inflammatory, and renal metabolomics.

Excessive dietary salt intake is a well-established risk factor for hypertension, with numerous underscoring its significant role in the pathogenesis of cardiovascular diseases. Dietary management is widely recognized as essential for both the prevention and treatment of hypertension. This study aimed to evaluate the preventive efficacy of a sulfonated peptide, Leucyl-glycyl-asparaginyl-glycyl-cysteinylsulfonic acid-proline (Leu-Gly-Asn-Gly-Cya-Pro, SLP), against high-salt-induced hypertension and renal injury, thereby providing new insights into hypertension prevention strategies. Integrated analysis explores the contrasting detoxification mechanisms of DIMBOA and flavonoids on imazethapyr in wheat plants.

The widespread use of herbicides poses escalating threats to ecosystem stability and food safety, underscoring the urgent need to elucidate plant-based detoxification mechanisms. However, the contrasting roles of secondary metabolites like DIMBOA and flavonoids in mitigating herbicide toxicity in wheat remain unclear. Research indicates that DIMBOA primarily acts as a defense metabolite, reducing reactive oxygen species (ROS) accumulation through iron chelation and reinforcing structural defenses, but its growth recovery is limited due to metabolic competition with auxin biosynthesis. In contr

A single diiron enzyme catalyses the oxidative rearrangement of tryptophan to indole nitrile.

Nitriles are uncommon in nature and are typically constructed from oximes through the oxidative decarboxylation of amino acid substrates or from the derivatization of carboxylic acids. Here we report a third nitrile biosynthesis strategy featuring the cyanobacterial nitrile synthase AetD. During the biosynthesis of the eagle-killing neurotoxin, aetokthonotoxin, AetD transforms the 2-aminopropionate portion of 5,7-dibromo-L-tryptophan to a nitrile. Employing a combination of structural, biochemical and biophysical techniques, we characterized AetD as a non-haem diiron enzyme that belongs to the

Lithium inhibits tryptophan catabolism via the inflammation-induced kynurenine pathway in human microglia.

Despite its decades' long therapeutic use in psychiatry, the biological mechanisms underlying lithium's mood-stabilizing effects have remained largely elusive. Here, we investigated the effect of lithium on tryptophan breakdown via the kynurenine pathway using immortalized human microglia cells, primary human microglia isolated from surgical specimens, and microglia-like cells differentiated from human induced pluripotent stem cells. Interferon (IFN)-γ, but not lipopolysaccharide, was able to activate immortalized human microglia, inducing a robust increase in indoleamine-2,3-dioxygenase (IDO1

Development of a drug delivery vehicle protein exhibiting high binding affinity and low leakage of the anti-cancer drug SN-38.

Drug leakage from delivery vehicles is a major limitation of drug delivery systems (DDSs) for cancer chemotherapy because premature release of loaded drugs reduces therapeutic efficacy and increases off-target toxicity. We previously developed a DDS for the poorly water-soluble anti-cancer drug SN-38 using lipocalin-type prostaglandin D synthase (L-PGDS). To suppress drug leakage, in this study we generated an L-PGDS mutant (M94W-M145W) with enhanced binding affinity for SN-38 by introducing amino acid substitutions into the ligand-binding cavity. Docking simulations identified residues involv

Interplay between IDO1 and iNOS in human retinal pigment epithelial cells.

Human retinal pigment epithelial (hRPE) cells form a selectively permeable monolayer between the neural retina and the highly permeable choroidal vessels. Thus, hRPE cells bear important regulatory functions and are potential targets of pathogens in vivo. Endogenous bacterial endophthalmitis (EBE) is frequently caused by infections with the Gram-positive bacterium Staphylococcus aureus (S. aureus). Upon microbial infection, interferon gamma (IFN-γ), a major cytokine of the adaptive immune response, induces a broad spectrum of effector molecules, such as the tryptophan-degrading enzyme indoleam

Metabolism of tetrahydrobiopterin: its relevance in monoaminergic neurons and neurological disorders.

(6R)-L-erythro-5,6,7,8-tetrahydrobiopterin (BH4) is an essential cofactor for aromatic amino acid hydroxylases, such as phenylalanine hydroxylase (PAH), tyrosine hydroxylase (TH), tryptophan hydroxylase, and nitric oxide synthase, which catalyze physiologically important reactions in mammals. The biosynthesis and metabolism of BH4 is usually studied mostly in the liver and only slightly in the brain, as the BH4 level in the liver is relatively high because BH4 is required for the reaction of PAH. We found that GTP (guanosine triphosphate) cyclohydrolase I, an enzyme for the biosynthesis of BH4

Free heme pool and activity of key enzyme of heme synthesis in the rat liver under action of agents affecting reduced glutathione level.

The decrease of GSH level in the rat liver was found to be accompanied by an increase of tryptophan 2,3-dioxygenase (TDO) heme saturation during first hours after HgCl2, phenylhydrazine (Ph) injection or rhabdomyolysis (the coefficient of correlation -0.978). The activity of the key enzyme of heme synthesis--5-aminolevulinate synthase (ALAS) was 2.5-fold increased in the first hours after Ph injection and rhabdomyolysis. Glutathione injection in vivo as well as CdCl2 caused the increase of GSH content and the inhibition of ALAS. The coefficient of correlation for GSH content and ALAS activity

Genome mining of Trp-anthranilic acid cyclic dipeptides in Aspergillus driven by analysis of NRPS adenylation domains.

Aspergillus fungi are rich in secondary metabolites, yet their biosynthetic potential remains underexplored. Nonribosomal peptides (NRPs), an important class of natural products with diverse biological activities, are synthesized by nonribosomal peptide synthetases (NRPSs). In this study, we performed a large-scale bioinformatic analysis of NRPS adenylation (A) domains from 1,162 Aspergillus genomes to predict their substrate specificities. Using AdenylPred, we analyzed 40,684 A-domain sequences and found that large phenyl-derivative amino acids and small hydrophobic amino acids are the predom

Multiple roles of haem in cystathionine β-synthase activity: implications for hemin and other therapies of acute hepatic porphyria.

The role of haem in the activity of cystathionine β-synthase (CBS) is reviewed and a hypothesis postulating multiple effects of haem on enzyme activity under conditions of haem excess or deficiency is proposed, with implications for some therapies of acute hepatic porphyrias. CBS utilises both haem and pyridoxal 5'-phosphate (PLP) as cofactors. Although haem does not participate directly in the catalytic process, it is vital for PLP binding to the enzyme and potentially also for CBS stability. Haem deficiency can therefore undermine CBS activity by impairing PLP binding and facilitating CBS de

Antibacterial, Anti-Inflammatory and Anti-Melanogenic Effects of a Novel Peptide.

BACKGROUND: Acne-induced post-inflammatory hyperpigmentation (PIH) is driven by Cutibacterium acnes-mediated inflammation and dysregulated melanogenesis; however, effective treatments targeting both processes remain limited. OBJECTIVE: This study aimed to evaluate the anti-bacterial, anti-inflammatory, and anti-melanogenic effects of a newly made 2-isopropyl-5-methylphenol-conjugated valine-tryptophan dipeptide (2IP5MP-VW). METHODS: IPMP-VW conjugates were synthesized and screened for antibacterial activity against C. acnes. The effects of 2IP5MP-VW on inflammation and melanogenesis were ass

Identification of stable reference genes for quantitative gene expression analysis in the duodenum of meat-type ducks.

Quantitative polymerase chain reaction (qPCR) is an important method to detect gene expression at the molecular level. The selection of appropriate housekeeping genes is the key to accurately calculating the expression level of target genes and conducting gene function studies. In this study, the expression of eight candidate reference genes, glyceraldehyde-3-phosphate dehydrogenase (GAPDH), beta-actin (β-actin), 18S ribosomal RNA (18S rRNA), hydroxymethylbilane synthase (HMBS), hypoxanthine phosphoribosyltransferase 1 (HPRT1), TATA box binding protein (TBP), ribosomal protein L13 (RPL13), and
