Putative volatile biomarkers of bovine tuberculosis infection in breath, skin and feces of cattle.

Bovine tuberculosis (bTB) is an infectious disease with significant impact on animal health, public health and international trade. Standard bTB screening in live cattle consists in injecting tuberculin and measuring the swelling at the place of injection few days later. This procedure is expensive, time-consuming, logistically challenging, and is not conclusive before performing confirmatory tests and additional analysis. The analysis of the volatile organic compounds (VOCs) emitted by non-invasive biological samples can provide an alternative diagnostic approach suitable for bTB screening. I

Vaccination Strategies in a Potential Use of the Vaccine against Bovine Tuberculosis in Infected Herds.

Bovine tuberculosis (bTB) is a disease of cattle that represents a risk to public health and causes severe economic losses to the livestock industry. Recently, one of the strategies recommended for reducing the prevalence of the disease in animals is the use of the BCG vaccine, alone or in combination with proteins. It has been shown that the vaccine elicits a strong immune response, downsizes the number of animals with visible lesions, and reduces the rate of infection as well as the bacillary count. This paper, based on scientific evidence, makes suggestions about some practical vaccination

Study on supplemental test to improve the detection of bovine tuberculosis in individual animals and herds.

BACKGROUND: Bovine tuberculosis (bTB), is a worldwide disease caused by Mycobacterium bovis (M. bovis). The success of bTB eradication and control programs is based on early detection and the removal of reactors from a herd thus routine testing and cull strategy have been applied globally. Since the late nineteenth century, the Tuberculin Skin Test (TST) has been the primary antemortem test available to support bTB eradication campaigns. Due to the TST limitations in terms of Se and Sp, the credibility of the diagnosis is frequently questioned given the occurrence of false-positive and false-n

Evaluation of cocktails with recombinant proteins of Mycobacterium bovis for a specific diagnosis of bovine tuberculosis.

The Delayed type hypersensitivity skin test (DTH) and interferon-gamma assay are used for the diagnosis of bovine tuberculosis (TBB). The specificity of these diagnoses, however, is compromised because both are based on the response against purified protein derivative of Mycobacterium bovis (PPD-B). In this study, we assessed the potential of two cocktails containing M. bovis recombinant proteins: cocktail 1 (C1): ESAT-6, CFP-10 and MPB83 and cocktail 2 (C2): ESAT-6, CFP-10, MPB83, HspX, TB10.3, and MPB70. C1, C2, and PPD-B showed similar response by DTH in M. bovis-sensitized guinea pigs. Imp

Bovine tuberculosis: effect of the tuberculin skin test on in vitro interferon gamma responses.

Bovine tuberculosis (bTB) is a disease of zoonotic and economic importance. In many countries, control is based on test and slaughter policies and/or abattoir surveillance. For testing, cell mediated immune- (CMI-) based assays (i.e., tuberculin skin test (TST) supplemented by the interferon gamma (IFN-gamma) assay) are the primary surveillance and disease control tests for bTB. The combined use of the in vivo and in vitro CMI assays to increase overall sensitivity has raised the question of whether the IFN-gamma response is influenced by injection of purified protein derivatives (PPDs) for TS

Development of a skin test for bovine tuberculosis for differentiating infected from vaccinated animals.

The tuberculin skin test has been used for the diagnosis of bovine and human tuberculosis (TB) for over a hundred years. However, the specificity of the test is compromised by vaccination with the Mycobacterium bovis-derived vaccine strain bacille Calmette-Guérin (BCG). Since current promising vaccines against bovine TB are based on heterologous prime-boost combinations that include BCG, there is a need for diagnostic tests for differentiating infected from vaccinated animals (DIVA). The application of antigens such as ESAT-6 and CFP-10 for DIVA has so far been realized largely through their a

Is interleukin-4delta3 splice variant expression in bovine tuberculosis a marker of protective immunity?

Splice variants of the interleukin-4 (IL-4) cytokine gene have been described for humans, mice, and cattle. IL-4 splice variants have been shown to inhibit IL-4-mediated cellular responses and thus act as IL-4 antagonists. Recent work has highlighted the possibility of a correlation between IL-4 splice variants and protection against clinical tuberculosis. In this study we investigated the potential role of IL-4 splice variants IL-4delta2 and IL-4delta3 in cattle with bovine tuberculosis, using quantitative real-time reverse transcription-PCR. For this analysis we used naturally exposed tuberc

Temporal and seasonal trends of bovine tuberculosis in the Irish cattle population (2008-2024).

BACKGROUND: Bovine tuberculosis (bTB) has substantial impacts on the Irish cattle industry. Between 2016 and 2024, its burden increased in Irish cattle. As an initial step towards understanding drivers of this increase, we explore long-term trends in measures of bTB burden and cattle demographics in Ireland. METHODS: Using data from 2008 to 2024, we describe changes in cattle management and demographics over time, as well as measures of bTB burden at herd- and animal-level. A negative binomial regression model was implemented to explore the association between year and herd type and the count

A retrospective study of bovine tuberculosis at the municipal abattoir of Bauchi State, Northeastern Nigeria.

BACKGROUND AND AIM: Bovine tuberculosis (bTB) still remains a major zoonotic bacterial disease affecting livestock and humans worldwide. The disease remains a poorly managed tropical disease in most developing countries of the world; where in addition to productivity losses and significance in international trade, it posed a major public health threat to both humans and animals. A retrospective study was designed to investigate the occurrence of bTB lesions at Bauchi municipal abattoir. MATERIALS AND METHODS: The study utilized abattoir records spanning a period of 10 years (2004-2013). The r

Severity of bovine tuberculosis is associated with co-infection with common pathogens in wild boar.

Co-infections with parasites or viruses drive tuberculosis dynamics in humans, but little is known about their effects in other non-human hosts. This work aims to investigate the relationship between Mycobacterium bovis infection and other pathogens in wild boar (Sus scrofa), a recognized reservoir of bovine tuberculosis (bTB) in Mediterranean ecosystems. For this purpose, it has been assessed whether contacts with common concomitant pathogens are associated with the development of severe bTB lesions in 165 wild boar from mid-western Spain. The presence of bTB lesions affecting only one anatom

New generation vaccines and delivery systems for control of bovine tuberculosis in cattle and wildlife.

Advances in the understanding of protective immune responses to tuberculosis are providing opportunities for the rational development of improved vaccines for bovine tuberculosis. Protection requires activation of macrophages through stimulation of a Th 1 type immune response. Ideally, a vaccine for cattle should induce protection without causing animals to react in a tuberculin test when exposed to Mycobacterium bovis. A number of new tuberculosis vaccines including attenuated M. bovis strains, killed mycobacteria, protein and DNA vaccines have been developed and many of these are being asses

Exposure of wild boar to Mycobacterium tuberculosis complex in France since 2000 is consistent with the distribution of bovine tuberculosis outbreaks in cattle.

The Eurasian wild boar (Sus scrofa) is increasingly considered as a relevant actor in the epidemiology of animal tuberculosis (TB). Therefore, monitoring TB in wild boar becomes a key tool for establishing comprehensive control schemes for this disease. To estimate the exposure of free living wild boar to Mycobacterium tuberculosis complex (MTC) in France, a bovine-purified protein derivative based ELISA was used to test 2,080 archived serum samples of hunter-harvested animals in 58 French "départements". Two cut-off values were used for diagnostic interpretation: 0.2, recommended by the manuf

Bovine Tuberculosis in a Rural Greek Area: Within-Herd Prevalence and Risk Factors in 30 Tuberculosis Skin Test-Positive Herds.

Bovine tuberculosis (bTB) is a zoonotic disease of major importance for cattle health, farm productivity, and public health. This study aimed to characterize the epidemiological features and assess their associations with farm characteristics of bovine tuberculosis-positive herds in a high-prevalence area of Greece. Apparent animal-level positivity for MBTC infection was evaluated in cattle from 30 infected farms in the Regional Unit of Florina, and patterns associated with positivity were explored. A total of 1464 animals were examined using the single intradermal tuberculin test, of which 56

Improving Bovine Tuberculosis Surveillance Through Risk-Based Prioritization of Slaughterhouse-Triggered Trace-Back Investigations.

Slaughterhouse detection of lesions compatible with bovine tuberculosis represents a key passive surveillance component in Santa Catarina, Brazil, yet subsequent trace-back investigations often fail to identify infected farms. This study developed a quantitative framework to prioritize epidemiological investigations by estimating the probability of infection associated with each farm connected to PCR-confirmed cases. Using official movement records and historical diagnostic data, we reconstructed the lifetime contact networks of slaughtered cattle presenting confirmed Mycobacterium bovis lesio

Comparative Evaluation of ELISA Tests for Bovine Tuberculosis Detection: Implications for Improved Disease Control.

Accurate diagnosis of bovine tuberculosis (bTB) remains a critical challenge for disease control and eradication programs. While cell-mediated immune (CMI) response tests such as the comparative cervical intradermal tuberculin test (CCITT) are widely used, their sensitivity is limited, especially in later stages of infection. Antibody-based assays, such as enzyme-linked immunosorbent assays (ELISAs), may offer complementary detection and improve case identification. This study aimed to comparatively evaluate two indirect ELISA tests for the detection of antibodies against Mycobacterium bovis i

Bovine tuberculosis model validation against a field study of badger vaccination with selective culling.

Bovine tuberculosis (bTB) is a costly disease in Britain and Ireland shared by cattle and badgers (Meles meles), and to reduce the infection in cattle to low levels some form of badger management is considered necessary. We compare the results of a badger field trial where test-positive badgers are culled, and test-negative badgers vaccinated (a TVR approach) with the results of the simulation model originally used to predict the effect of the trial in Northern Ireland. Initial model results depended strongly on whether social perturbation occurred in the badger population following culling, a

Histological observations of bovine tuberculosis in lung and lymph node tissues from British deer.

Deer are recognized as hosts of Mycobacterium bovis and assessing the role of wild cervids in perpetuating tuberculosis among cattle has motivated extensive research on several continents. In this paper, the histopathology of lymph node and lung tuberculous granulomas in M. bovis positive British deer is presented. The overall aim was to seek further insights into the potential for onward transmission from infected deer to other species, including cattle. Samples were obtained from an extensive survey of wild mammals in South-West England and from statutory tuberculosis surveillance. M. bovis

Host transcriptional responses identify putative diagnostic biomarkers for bovine tuberculosis in cattle and buffalo.

Bovine tuberculosis (bTB), caused by Mycobacterium bovis, remains a major zoonosis impacting livestock productivity and public health. To characterize species-specific immune responses and identify transcriptional biomarkers of infection, we quantified the expression of 45 immune-related genes in peripheral blood from naturally infected cattle and Mediterranean buffalo. Differential and multivariate analyses revealed distinct species- and state-specific transcriptional signatures. Sixteen genes were significantly modulated in cattle, with Canonical Discriminant Analysis (CDA) defining a promis
