The fine tuning of proliferation and neurogenesis, neuronal migration and differentiation and connectivity underlies the proper development of the cerebral cortex. Mutations in genes involved in these processes are responsible for neurodevelopmental disorders, such as cortical dysgeneses, which are usually associated with severe mental retardation and epilepsy. Over the past few years, the importance of cytoskeleton components in cellular processes crucial for cortical development has emerged from a body of functional data. This was reinforced by the association of mutations in the LIS1 and DC

Eukaryotic cells have internal scaffolding of microtubules cytoskeleton that gives them their characteristic shapes. We analyzed by immuno-fluorescence the shift and distribution of tubuline during in vitro bull sperm nuclei swelling by the action of heparin-reduced glutathione physiological decondensing agents. Sperm tubulin display a pattern that shows tubulin fluorescence all over the head, leaving the acrosome tip devoid of tubulin. In the second stage we can observe that the acrosomal zone is practically devoid of fluorescence and a net of fluorescent microtubules that seems to be anchore

A role for charge-based interactions in protein stability at the monomer or dimer level is well known. We show here that such interactions can also be important for the higher-order structures of microtubule assembly. Alkali metal chlorides increase the rate of polymerization of pure tubulin driven by either taxol or dimethyl sulfoxide. The effect is cation selective, exhibiting a sequence Na+ > K+ > Li+ > Cs+, with optimal concentrations for Na+ at approximately 160 mM. Hofmeister anion effects are additive with these rate stimulations. Sodium is less potent than guanidinium ion stimulation r

Tubulin represents a highly promising anticancer target and several inhibitors have been approved for cancer therapy. However, drug resistance and dose-dependent toxicity have limited clinical application. Recently, some novel pyrimidine derivatives have emerged as tubulin inhibitors, such as Lexibulin and Verubulin, which could provide a promising avenue for discovering highly effective anticancer agents. This review covers a comprehensive overview of structure-activity relationships associated with biological activity, as well as clinical development. Furthermore, we highlight the challenges

Glioblastoma (GBM) is the most common primary malignant brain tumor in the central nervous system, characterized by high invasiveness, incidence, and mortality. Tubulin folding cofactor B (TBCB) is a microtubule-associated regulator implicated in microtubule plus-end organization and related structural features; however, its role and underlying mechanisms in GBM remain unclear. This study aimed to elucidate the function and mechanisms of TBCB in GBM cell proliferation and migration. Analyses of public databases (GEO, TCGA, CGGA) and clinical GBM specimens revealed that TBCB was significantly u

Microtubules have long been considered an ideal target for anticancer drugs because of the essential role they play in mitosis, forming the dynamic spindle apparatus. In this work, we reported the discovery of a novel series of homoisoflavonoids analogues bearing 3-benzylidene-6,7,8-trimethoxy-chroman-4-one moiety as tubulin polymerization inhibitors. The cytotoxic activity of the synthesized compounds was evaluated against two human cancer cell lines including A-375 and HL-60. Among them, compound B6 was identified as a potent tubulin polymerization inhibitor with effective anticancer potency

In the brain of patients with Alzheimer's disease (AD), the number and length of microtubules (MTs) are significantly and selectively reduced. MTs are involved in a wide range of cellular functions, and defects of the microtubular system have emerged as a unifying hypothesis for the heterogeneous and variable clinical presentations of AD. MTs orchestrate their numerous functions through the spatiotemporal regulation of the binding of specialised microtubule-associated proteins (MAPs) and molecular motors. Covalent posttranslational modifications (PTMs) on the tubulin C-termini that protrude at

The structure, dynamic behavior, and spatial organization of microtubules are regulated by microtubule-associated proteins. An important microtubule-associated protein is the protein Tau, because its microtubule interaction is impaired in the course of Alzheimer's disease and several other neurodegenerative diseases. Here, we show that Tau binds to microtubules by using small groups of evolutionary conserved residues. The binding sites are formed by residues that are essential for the pathological aggregation of Tau, suggesting competition between physiological interaction and pathogenic misfo

Tubulin glutamylation is a post-translational modification that occurs predominantly in the ciliary axoneme and has been suggested to be important for ciliary function. However, its relationship to disorders of the primary cilium, termed ciliopathies, has not been explored. Here we mapped a new locus for Joubert syndrome (JBTS), which we have designated as JBTS15, and identified causative mutations in CEP41, which encodes a 41-kDa centrosomal protein. We show that CEP41 is localized to the basal body and primary cilia, and regulates ciliary entry of TTLL6, an evolutionarily conserved polygluta

Tubulin is one of the molecular components that regulate cytosketal structure relating to cell differentiation, invasion, and metastasis in cancer. Recently, glu-tubulin, in which the C-terminal tyrosine of α-tubulin is removed by tubulin carboxypeptidase, overexpression has been reported in malignant tumors of the mammary gland immunohistochemically. We identified 147 cases accessioned in the Department of Pathology, International University of Health and Welfare Hospital from 2003 to 2009. Of the 78 malignant tumor cases, staining for glu-tubulin was observed in 56 (71.8%), and only 22 cases

Polymicrogyria is a relatively common but poorly understood defect of cortical development characterized by numerous small gyri and a thick disorganized cortical plate lacking normal lamination. Here we report de novo mutations in a beta-tubulin gene, TUBB2B, in four individuals and a 27-gestational-week fetus with bilateral asymmetrical polymicrogyria. Neuropathological examination of the fetus revealed an absence of cortical lamination associated with the presence of ectopic neuronal cells in the white matter and in the leptomeningeal spaces due to breaches in the pial basement membrane. Tubulin ligands known to be toxic to certain organisms or cells were tested for their ability to inhibit proliferation of trypanosomes in culture. Tubulin was purified from Trypanosoma brucei brucei or rat brain by poly-L-lysine affinity chromatography and used in binding studies in order to compare the binding of [3H]mebendazole to trypanosome and mammalian tubulin. All the compounds tested in culture inhibited trypanosome proliferation in a concentration-dependent manner. The concentration required to inhibit trypanosome proliferation by 50 or 90% (IC50 or IC90) in 24 hr was determined for e

We have studied the localization and affinity of methyl mercury hydroxide (MeHg) binding sites on microtubules. There is one class of binding sites for MeHg on tubulin, a high affinity class with fifteen sites. MeHg binds to tubulin stoichiometrically within microtubules, and does not induce microtubule disassembly at this low binding ratio. MeHg binds in microtubules either in the presence or absence of free tubulin subunits but free subunits act as uncompetitive inhibitors for MeHg binding to the polymer. These stoichiometric polymer surface binding sites for MeHg apparently do not interfere

We used three antitubulin antibodies to localize Dictyostelium tubulin subunits on two-dimensional polyacrylamide gels by Western blotting. All three antibodies, a polyclonal antibody against sea urchin alpha- and beta-tubulin and two monoclonal antibodies against yeast alpha-tubulin, recognize the same set of polypeptides with a molecular weight of 55,000 while focusing at a pH far more basic than all other tubulins. Each antibody specifically stains the microtubule system of slime mold amoebae by indirect immunofluorescence. The microtubule system can be isolated as a major component of the

Long, unique cytoskeleton-like structures resembling microtubules of higher eukaryotes were discovered in Mycoplasmoides gallisepticum (former taxonomy name Mycoplasma gallisepticum), one of the smallest prokaryotes. Identifying the structural components of these formations is of great interest. These structures may contribute to the motility of M. gallisepticum cells. Since proteins involved in motility are potential targets for new antibacterial therapies, studying them is crucial. M. gallisepticum causes chronic respiratory diseases in poultry, leading to significant economic losses. Theref

The β-tubulin gene is essential for reproductive development, especially for male fertility, in different insects including Bombyx mori and Drosophila melanogaster. Targeting reproductive genes such as β-tubulin offers a promising approach to pest control that is more sustainable than chemical pesticides. However, there is limited research on the functional role of β-tubulin in Plutella xylostella, a highly damaging pest of vegetable crops. In the present study, we first identified and cloned the β-tubulin gene in P. xylostella (Pxβtubulin-1). Pxβtubulin-1 protein contains two conserved domain

Microtubules are critical components of the cytoskeleton that are extensively involved in various cellular and biological processes. The execution of these functions is intricately linked to post-translational modifications of tubulin. Post-translational modifications of tubulin include acetylation, tyrosination, de-tyrosination, glutamylation, SUMOylation, and so on. These modifications are closely associated with a wide range of biological processes. Accumulating evidence indicates that aberrant microtubule modifications are implicated in various diseases, including cancer, Alzheimer's disea

Colchicine site inhibitors of tubulin polymerization function as antiproliferative anticancer agents, with certain inhibitors demonstrating a dual mechanism of action as tumor-selective vascular disrupting agents (VDAs). Our previous studies yielded potent benzosuberene-based colchicine site inhibitors of tubulin polymerization. To expand structure-activity correlations, the seven-membered fused ring and the pendant ring were modified to generate a series of new compounds, 11 being strong inhibitors (IC50 ≤ 5 μM) of tubulin polymerization. Structural modifications introduced a second benzylic

Microtubules (MTs) play a vital role as key components of the eukaryotic cytoskeleton. The phylum Apicomplexa comprises eukaryotic unicellular parasitic organisms defined by the presence of an apical complex which consists of specialized secretory organelles and tubulin-based cytoskeletal elements. One apicomplexan parasite, Toxoplasma gondii, is an omnipresent opportunistic pathogen with significant medical and veterinary implications. To ensure successful infection and widespread dissemination, T. gondii heavily relies on the tubulin structures present in the apical complex. Recent advances

BACKGROUND: Vascular dementia (VaD) and Alzheimer's disease (AD) are the two most common forms of dementia. Although these two types of dementia have different etiologies, they share some similarities in their pathophysiology, such as neuronal loss and decreased levels of tau protein. We hypothesize that these can have an impact upon the molecular changes in tubulin, precede the neuronal cell loss, and lead to changes in cytoskeletal associated proteins, as documented in both VaD and AD.
OBJECTIVE: We characterized different isotypes of tubulin together with their posttranslational modificati
