Characterization of prostaglandin E2 binding to uterine membranes from baboon, rabbit and tree shrew (Tupaia belangeri).

Membrane preparations (100,000 g pellet) of rabbit, baboon and tree shrew (Tupaia belangeri) uteri were studied for binding of [3H]prostaglandin E2 ([3H]PGE2). Unbound [3H]PGE2 was separated by filtration through Whatman GF/F filters. Non-specific binding was determined by the amount of radioactivity associated with the filters in the presence of a 100-fold excess of radioinert PGE2. PGE2 bound to membranes could be displaced by some other prostaglandin (PG) molecules: PGE1, 16,16-dimethyl-PGE2, PGA1 and PGF2 alpha, but not by 6-keto-PGF1 alpha, PGD2 or arachidonic acid. No PGE2 binding was de

A Golgi study of the medial geniculate body in the tree shrew (Tupaia glis).

The subdivisions of the medial geniculate body in Tupaia recognized in previous connectional and cytoarchitectural studies are identified in Golgi-impregnated material. They may be distinguished by the organization of the neuropil, especially the dendrites, and, in many cases, by differences in the neurons. The ventral nucleus contains tufted cells with disc-shaped dendritic fields which are aligned to form laminae. The caudomarginal and deep dorsal nuclei have less tufted, less precisely arranged cells with longer, thin appendages. Neurons in the suprageniculate and dorsal nuclei are similar

Tree shrew immune cell atlas identifies NR1H3⁺ tissue macrophages with conserved anti-inflammatory function.

Rodents are widely used in immunology but do not always recapitulate human immune functions. The tree shrew (Tupaia belangeri) is phylogenetically closer to primates than rodents and may help bridge this gap, yet its immune system has not been comprehensively characterised at single-cell resolution. Here, we present a single-cell transcriptomic atlas of the tree shrew immune system, profiling 39 cell types across 12 tissues. We uncover human-like tonsillar structures and two transcriptionally distinct splenic macrophage subsets: an NR1H3+ immunoregulatory subset and an IRF8+ pro-inflammatory s

Unexpected Retinal Abnormalities in the Cone-Dominant Northern Tree Shrew.

The cone-dominant northern tree shrew has been used in a wide range of vision research studies. Given the similarity of their visual system to primates, there have been extensive in vivo and ex vivo studies of the visual system from retina to cortex. Tree shrews have long been used to study myopia and more recently, experimental models of glaucoma and diabetic retinopathy have become available. They are also amenable to various noninvasive imaging methods-previous studies have established their retinal structure and function through scanning laser ophthalmoscopy, optical coherence tomography,

Regional Specialization of Pyramidal Neuron Morphology and Physiology in the Tree Shrew Neocortex.

The mammalian cerebral cortex is divided into different areas according to their function and pattern of connections. Studies comparing primary visual (V1) and prefrontal cortex (PFC) of primates have demonstrated striking pyramidal neuron (PN) specialization not present in comparable areas of the mouse neocortex. To better understand PFC evolution and regional PN specialization, we studied the tree shrew, a species with a close phylogenetic relationship to primates. We defined the tree shrew PFC based on cytoarchitectonic borders, thalamic connectivity and characterized the morphology and ele

Chronic psychosocial stress induces morphological alterations in hippocampal pyramidal neurons of the tree shrew.

The effect of sustained psychosocial stress on the morphology of hippocampal pyramidal neurons was analysed in male tree shrews after 14, 20, and 28 days of social confrontation. A variety of physiological changes such as constantly elevated levels of urinary cortisol and norepinephrine and reduced body weight, which are indicative of chronic stress were observed in the subordinate, but not in the dominant males. Light microscopic analysis of Nissl-stained hippocampal sections showed that the staining intensity of the nucleoplasm in the CA1 and CA3 pyramidal neurons was increased after prolong

Tree shrew model of early diabetic retinopathy reveals microvascular dysfunction and identifies phosphoserine aminotransferase 1 as a novel therapeutic target.

BACKGROUND: Diabetic retinopathy (DR) is the most significant manifestation of diabetic microangiopathy. The existing tree shrew model of DR has dysfunctional retinal short wave sensitivity (SWS) cones and retinal ganglion cells, but it remains unclear whether the retinal microvessels are also compromised. In this study, we established a tree shrew diabetes model to investigate the characteristics of retinal microvascular disease observed in early human DR. METHODS: A high-fat and high-sugar diet combined with streptozotocin was used to establish the tree shrew diabetes model. After 20 weeks

Fatty acid synthase promotes Hantaan virus infection through lipogenic regulation in the tree shrew model.

Hantaan virus (HTNV) is the principal etiological agent of hemorrhagic fever with renal syndrome (HFRS) in Asia. To date, however, progress in defining mechanisms of host-pathogen interaction has been limited by the absence of an appropriate animal model. In the present study, the tree shrew ( Tupaia belangeri) was shown to be permissive to HTNV infection and to recapitulate key renal pathology and immunological features observed in patients with HFRS. Following intranasal challenge, viral replication occurred predominantly in the kidney, spleen, and lung, accompanied by tubular dilation, glom

Morphological and Molecular Distinctions of Parallel Processing Streams Reveal Two Koniocellular Pathways in the Tree Shrew DLGN.

In the mammalian visual system, three functionally distinct parallel processing streams extend from the retina to the visual thalamus and then to the visual cortex: magnocellular (M), parvocellular (P), and koniocellular (K). Tree shrews (Tupaia belangeri), a preprimate species, provide an advantageous model to study the K pathway in isolation because, while M and P pathways remain mixed in Lamina 1 (L1), L2, L4, and L5 of the lateral geniculate nucleus (LGN), L3 and L6 receive strictly K-input from the contralateral eye. Additionally, K-input laminae selectively receive glutamatergic axons fr

Neutralizing monoclonal antibodies protect against human adenovirus type 55 infection in transgenic mice and tree shrews.

Re-emerging human adenovirus type 55 (HAdV55) has become a significant threat to public health due to its widespread circulation and the association with severe pneumonia, but an effective anti-HAdV55 agent remains unavailable. Herein, we report the generation of macaque-derived, human-like monoclonal antibodies (mAbs) protecting against HAdV55 infection with high potency. Using fluorophore-labelled HAdV55 virions as probes, we isolated specific memory B cells from rhesus macaques (Macaca mulatta) that were immunized twice with an experimental vaccine based on E1-, E3-deleted, replication-inco

Transformation of receptive field properties from lateral geniculate nucleus to superficial V1 in the tree shrew.

Tree shrew primary visual cortex (V1) exhibits a pronounced laminar segregation of inputs from different classes of relay neurons in the lateral geniculate nucleus (LGN). We examined how several receptive field (RF) properties were transformed from LGN to V1 layer 4 to V1 layer 2/3. The progression of RF properties across these stages differed markedly from that found in the cat. V1 layer 4 cells are largely similar to the the LGN cells that provide their input, being dominated by a single sign (ON or OFF) and being strongly modulated by sinusoidal gratings. Some layer 4 neurons, notably those

A novel tree shrew model of lipopolysaccharide-induced acute respiratory distress syndrome.

INTRODUCTION: Acute respiratory distress syndrome (ARDS) is a leading cause of respiratory failure, with substantial attributable morbidity and mortality. The small animal models that are currently used for ARDS do not fully manifest all of the pathological hallmarks of human patients, which hampers both the studies of disease mechanism and drug development. OBJECTIVES: To examine whether the phenotypic changes of primate-like tree shrews in response to a one-hit lipopolysaccharides (LPS) injury resemble human ARDS features. METHODS: LPS was administered to tree shrews through intratracheal

Experimental myopia increases and scleral crosslinking using genipin inhibits cyclic softening in the tree shrew sclera.

PURPOSE: Myopia progression is thought to involve biomechanical weakening of the sclera, which leads to irreversible deformations and axial elongation of the eye. Scleral crosslinking has been proposed as a potential treatment option for myopia control by strengthening the mechanically weakened sclera. The biomechanical mechanism by which the sclera weakens during myopia and strengthens after crosslinking is not fully understood. Here, we assess the effect of lens-induced myopia and exogenous crosslinking using genipin on the inelastic mechanical properties of the tree shrew sclera measured by

Opioidergic innervation of the tree shrew pineal gland: an immunohistochemical study.

The tree shrew (Tupaia glis) has been described as a missing link relating primate to insectivore stock. The pineal gland of the tree shrew consists of a superficial pineal and a deep pineal, which are connected by a long and slender pineal stalk. A monoclonal antibody against leu-enkephalin was used in an immunohistochemical investigation of the tree shrew pineal gland. A moderate innervation of leu-enkephalin immunoreactive nerve fibers has been demonstrated in both superficial and deep pineal gland of the tree shrew. The density of the nerve fibers was slightly higher in the superficial pin

Myopia induction in animals following alteration of the visual input during development: a review.

In this review the effects of changes in the quality of the visual environment on the development of myopia during eye growth in various mammalian and avian species are described. The effect of changes in the light/dark cycle on myopia development has been studied only in the avian eye, mainly that of the domestic fowl. In the eyes of chicks reared from hatching to maturity under continuous illumination, the following findings were reported: myopia, astigmatism, increases in axial length and equatorial width, shallow anterior chamber, increase in corneal diameter, reduction of corneal curvatur

Comparative infectivity of EBV subtypes in tree shrews (Tupaia belangeri).

BACKGROUND: The Epstein-Barr Virus (EBV) has a high global prevalence and is strongly associated with the pathogenesis of diverse diseases. Types 1 (T1) and 2 (T2) represent the two major EBV subtypes. However, their infectivity and pathogenic mechanisms remain unclear. We aimed to explore the differences between T1 and T2 EBV infections using the tree shrew model. METHODS: T1 and T2 EBVs were purified from the supernatant of EBV-producing cells lines B95-8 and P3HR-1, respectively cell line. Eighteen tree shrews (evenly divided by sex) were randomly assigned to three groups (n = 6). Each ani

Tree shrew cytochrome P450 2E1 is a functional enzyme that metabolises chlorzoxazone and p-nitrophenol.

Cytochromes P450 (CYPs or P450s) are important enzymes for drug metabolism. Tree shrews are non-primate animal species used in various fields of biomedical research, including infection (especially hepatitis viruses), depression, and myopia. A recent tree shrew genome analysis indicated that the sequences and the numbers of P450 genes are similar to those of humans; however, P450s have not been adequately identified and analysed in this species. In this study, a novel CYP2E1 was isolated from tree shrew liver and was characterised in comparison with human, dog, and pig CYP2E1. Tree shrew CYP2E1

Chronic clomipramine treatment reverses core symptom of depression in subordinate tree shrews.

Chronic stress is the major cause of clinical depression. The behavioral signs of depression, including anhedonia, learning and memory deficits, and sleep disruption, result from the damaging effects of stress hormones on specific neural pathways. The Chinese tree shrew (Tupaia belangeri chinensis) is an aggressive non-human primate with a hierarchical social structure that has become a well-established model of the behavioral, endocrine, and neurobiological changes associated with stress-induced depression. The tricyclic antidepressant clomipramine treats many of the core symptoms of depressi

Seroprevalence of rickettsial infection in commensal rodents and shrews trapped in the Bangkok Metropolitan Area.

Murine typhus and scrub typhus are important human rickettsial diseases in Thailand. Small mammals, including many species of rodents and shrews, serve as the reservoir host of rickettsial diseases. Rickettsia typhi can be transmitted to humans by fleas causing murine typhus, while infection with Orientia tsutsugamushi causing scrub typhus in humans is transmitted by chiggers. The prevalence of rickettsial infection depends on the geographic area. The seroprevalence of antibody to R. typhi and O. tsutsugamushi was studied in commensal rodents and shrews trapped in markets in the Bangkok Metrop
