A New Cyst Nematode Heterodera luodingensis n. sp. (Nematoda: Heteroderinae) from Rice in China.

A unique cyst nematode population (Heterodera spp.) was collected from rice roots in Luoding County, Guangdong Province, China. Morphological and molecular analyses revealed that it is significantly different from all previously described cyst nematode species. It is described as Heterodera luodingensis n. sp. and classified in the Cyperi group. H. luodingensis n. sp. is characterized by its lemon-shaped cyst with a prominent terminal vulval cone that is ambifenestrate with abundant bullae and a relatively short vulval slit, measuring 31.3 (24.4 to 38.7) μm in length. The second-stage juvenile

Transcriptome-Based Comparative Expression Profiling of Sweet Potato during a Compatible Response with Root-Knot Nematode Meloidogyne incognita Infection.

M. incognita, a root-knot nematode (RKN), infects the roots of several important food crops, including sweet potato (Ipomoea batatas Lam.), and severely reduces yields. However, the molecular mechanisms underlying infection remain unclear. Previously, we investigated differential responses to RKN invasion in susceptible and resistant sweet potato cultivars through RNA-seq-based transcriptome analysis. In this study, gene expression similarities and differences were examined in RKN-susceptible sweet potato cultivars during the compatible response to RKN infection. Three susceptible cultivars in

Nematode-Suppressive Potential of Digestates to Meloidogyne incognita and Heterodera schachtii.

Management of plant-parasitic nematodes uses host plant resistance, crop rotation, cultural methods, and nematicide applications. Host plant resistance is tedious to develop, and crop rotation and cultural methods are challenging to use. Environmental and human health concerns render sole reliance on chemical nematode suppression nonsustainable. Previously, digestate from anaerobically fermented maize silage suppressed Heterodera schachtii in Beta vulgaris crops. Here, seven digestates were investigated for nematode suppressive potential: liquid dairy manure digestate (LDMD), liquid dairy manu

Mutations in Acetylcholinesterase2 (ace2) increase the insensitivity of acetylcholinesterase to fosthiazate in the root-knot nematode Meloidogyne incognita.

The root-knot nematode Meloidogyne incognita causes severe damage to continuously cropping vegetables. The control of this nematode relies heavily on organophosphate nematicides in China. Here, we described resistance to the organophosphate nematicide fosthiazate in a greenhouse-collected resistant population (RP) and a laboratory susceptible population (SP) of M. incognita. Fosthiazate was 2.74-fold less toxic to nematodes from RP than that from SP. Quantitative real-time PCR revealed that the acetylcholinesterase2 (ace2) transcription level in the RP was significantly higher than that in the

Heterodera schachtii nematodes interfere with aphid-plant relations on Brassica oleracea.

Aboveground and belowground herbivore species modify plant defense responses differently. Simultaneous attack can lead to non-additive effects on primary and secondary metabolite composition in roots and shoots. We previously found that aphid (Brevicoryne brassicae) population growth on Brassica oleracea was reduced on plants that were infested with nematodes (Heterodera schachtii) prior (4 weeks) to aphid infestation. Here, we examined how infection with root-feeding nematodes affected primary and secondary metabolites in the host plant and whether this could explain the increase in aphid dou

[The participation of salicylic and jasmonic acids in genetic and induced resistance of tomato to Meloidogyne incognita (Kofoid and White, 1919)].

Salicylic (SA) and jasmonic (JA) acids are the best known mediators of signal systems in plants. In this investigation the participation and character of interactions between SA- and JA-signals under the induced and genetic resistance of plants to nematodes was investigated on the model system tomato (Lycopersicon esculentum) and the root-knot nematode Meloidogyne incognita. This study demonstrates that application of JA and SA to tomato foliage induces systemic effects that suppress root-knot nematode infestation, inhibition of nematode reproduction, and also increased activity of LOX and PAL

Biogenic Synthesis and Nematicidal Potency of Highly Stable Ag and Fe Nanoparticles From Krascheninnikovia ceratoides and Elaeagnus angustifolia.

Managing root-knot nematodes, particularly Meloidogyne incognita, is a significant difficulty in sustainable agriculture because of the environmental toxicity of synthetic chemical nematicides. This study uses leaf extracts of Krascheninnikovia ceratoides (KC) and Elaeagnus angustifolia (EA) to synthesize silver (KC-AgNPs) and iron (EA-FeNPs) nanoparticles in a fast and environmentally friendly manner. The products were characterized by XRD, SEM, FTIR, and UV-visible spectroscopy, and their UV-visible spectral profiles were monitored for 30 days and under pH and temperature stress. XRD-derived

Germplasm Evaluation in Chickpea for Resistance to Plant Parasitic Nematodes.

Cicer arietinum L. (chickpea) is the second most widely cultivated food legume crop after common bean worldwide. Chickpea is an important source of dietary protein for human beings. It also helps in improvement of soil fertility by adding atmospheric N2 (ca. 60 kg/ha) through biological nitrogen fixation. Plant parasitic nematodes (PPNs) are one of the important biotic factors, which affects the crop production and productivity worldwide. PPNs cause severe yield losses estimated to the tune of US$100-173 billion worldwide. PPNs not only feed on host plants, but also responsible for inducing wo

Nematicidal Potential of Ceriops tagal Extracts and Authentic Compounds against Cyst Nematode Heterodera zeae.

Heterodera zeae, a cyst nematode, is a serious pest for economically crucial crops. Six extracts of Ceriops tagal were evaluated against infectious larvae of H. zeae through a glass cavity block assay, causing significant mortality to the larvae. The lower layer of dichloromethane extract (CTDCL) caused 97% mortality (LC50 = 1.64 ppm) at 1% concentration after 72 h. GC-MS and ESI-HRMS analyses of the extracts resulted in the identification of seventy-eight constituents. The activity of the extracts may be attributed to the strong synergistic interactions among the plant constituents. Moreover,

Pratylenchus coffeae Effector PcENG2 Targets the Maize Chitinase ZmCHI5 to Promote Parasitism.

Pratylenchus coffeae, a damaging root-lesion nematode, secretes effectors to manipulate host processes and promote parasitism. Here, we characterize PcENG2, a β-1,4-endoglucanase localized in the nematode esophageal gland cells and secreted during infection. PcENG2 expression is upregulated during the early parasitic stage of P. coffeae. PcENG2 promotes cell wall and callose degradation, as demonstrated by immunolocalization and immunoelectron microscopy. It also suppresses host reactive oxygen species (ROS) production and enhances susceptibility to P. coffeae when ectopically expressed in Ara

Orchid mycorrhizal fungus from the Brazilian Cerrado exhibits biocontrol activity against Meloidogyne javanica.

Waitea circinata, an orchid-associated mycorrhizal fungus, was evaluated for the first time as a biological control agent against the root-knot nematode Meloidogyne javanica. The fungus was originally isolated from the roots of Epidendrum nocturnum, a rupicolous orchid to the Brazilian Cerrado. In vitro assays demonstrated that W. circinata parasitized nematode eggs, with parasitism rates of 13-15% and characteristic emergence of septate hyphae with right-angle branching. Microscopy revealed eggshell degradation, arrested embryogenesis, and juvenile deformation. A mycelial suspension (MSF) of

DNA-based assessment of root lesion nematode infections in cereal roots.

Root lesion nematodes (RLN) of the genus Pratylenchus are causing significant damage in cereal production worldwide. Due to climate change and without efficient and environment-friendly treatments, the damages through RLNs are predicted to increase. Microscopic assessments of RLNs in the field and the greenhouses are time-consuming and laborious. As a result, cereal breeders have mostly ignored this pest. We present a method measuring RLN in infected cereal roots using a standardized PCR approach. Publicly available Pratylenchus neglectus primer combinations were evaluated. An optimal primer c

Root architecture plasticity in response to endoparasitic cyst nematodes is mediated by damage signaling.

Plant root architecture plasticity in response to biotic stresses has not been thoroughly investigated. Infection by endoparasitic cyst nematodes induces root architectural changes that involve the formation of secondary roots at infection sites. However, the molecular mechanisms regulating secondary root formation in response to cyst nematode infection remain largely unknown. We first assessed whether secondary roots form in a nematode density-dependent manner by challenging wild-type Arabidopsis plants with increasing numbers of cyst nematodes (Heterodera schachtii). Next, using jasmonate-re

Management of root lesion nematode, Pratylenchus delattrei in crossandra using oil cakes.

Selected oil cakes, neem, castor and mahua, were tried independently and in combination with a chemical nematicide (carbofuran 3G) for the management of Pratylenchus delattrei in crossandra under glass house conditions. The neem oil cake was effective compared to other oil cakes used and there was a synergistic effect when the neemcake was coupled with carbofuran 3G in the management of Pratylenchus delattrei. The treatment resulted in better establishment of seedlings, and with increased plant bio-mass and flower yield.

The potato cyst nematode Globodera pallida overcomes major potato resistance through selection on standing variation at a single locus.

Globodera pallida poses a major threat to potato production, with management strategies primarily relying on genetic resistance. However, increasing virulence in field populations across Western Europe raises major concerns for G. pallida control. To investigate the evolutionary mechanisms driving this rise in virulence, we propagated 13 field populations on 30 commercial potato varieties. Our findings indicate that the genetic basis of resistance in potatoes is small, with the major resistance conferred by GpaV from Solanum vernei. The wide application of GpaVvrn has led to continuous selecti

Dual Function of Itaconic Acid From Trichoderma citrinoviride Against Meloidogyne incognita.

Root-knot nematodes (Meloidogyne spp.) cause catastrophic yield losses in global agriculture. This study identified itaconic acid (IA), through comparative metabolomic analysis (the study of small molecules in biological systems), as a key virulence-related metabolite produced by the fungus Trichoderma citrinoviride Snef1910. In vitro assays demonstrated potent nematicidal activity of IA, with an LC50 value of 243.4 mg/liter against J2s of M. incognita and 78.4% egg-hatching inhibition at 800 mg/liter, performance that is comparable with the structural analog trans-aconitic acid. Pot and green

Globodera pallida virulence on major potato resistance has a common genetic basis across Western Europe.

The potato cyst nematode Globodera pallida poses a major threat to potato production in Western Europe. Current management strategies largely depend on the use of potato varieties carrying the genetic resistance GpaVvrn. However, reports from multiple West-European countries indicate a steady rise in virulence against GpaVvrn-containing potato varieties, raising serious concerns about G. pallida control. Although recent studies have resolved the genetic basis of virulence in two Dutch G. pallida populations, it remains unclear how conserved this genetic adaptation is in populations from differ

Nano-emulsion of garlic oil extracts: development, characterization and nematicidal efficacy against Meloidogyne incognita on tomato.

The root-knot nematode, Meloidogyne incognita, causes serious economic losses in agricultural production worldwide. Although traditional nematicides have proven effective, their utility is becoming increasingly restricted by regulations due to risks to ecosystems and public health. Consequently, plant-based extracts are gaining popularity as an eco-friendly alternative for nematode control. However, the‍‌‍‍‌ limitations of using plant extracts necessitate the development of novel ‍‌‍‍‌formulations. Therefore, garlic oil was extracted with ethanol or ethyl acetate and analyzed for composition b

Pratylenchus brasiliensis sp. nov. (Nematoda: Pratylenchidae): An Emerging Threat to Soybean in an Important New Agricultural Frontier in Brazil.

Root-lesion nematodes (Pratylenchus spp.) are significant agricultural pests worldwide. From January 2021 to January 2023, 365 root and rhizosphere soil samples were collected in key soybean-producing regions, revealing a new species, Pratylenchus brasiliensis sp. nov., in Brazil's agricultural frontier. Identification employed a polyphasic approach, integrating microscopy, morphometric analyses, and multigene phylogenetics. Molecular species delineation targeted the D2-D3 expansion segments of the 28S rRNA gene, the internal transcribed spacer (ITS) rRNA region, and cytochrome oxidase I (COXI

Transgenic overexpression of GmAPC7-CT improves seed yield and reduces susceptibility to soybean mosaic virus and Meloidogyne incognita in soybean.

Stable transgenic soybean lines overexpressing the GmAPC7-CT gene have demonstrated increased seed yield and reduced susceptibility to the soybean mosaic virus and Meloidogyne incognita. The Anaphase-Promoting Complex subunit 7 (APC7) is a core structural component of the anaphase-promoting complex or cyclosome (APC/C). The terminal region of this AtAPC7 gene has been shown in Arabidopsis thaliana to accumulate more transcripts than the full-length gene. The AtAPC7-CT gene (terminal region of the AtAPC7) encodes a protein with significant homology to a tobacco viral replication inhibitor (IVR)
