Electroacupuncture improves cyclophosphamide-induced bladder overactivity by reducing mechanotransduction in the rat urothelium.

OBJECTIVE: To investigate whether electroacupuncture (EA) therapy for overactive bladder (OAB) exerts its effect by modulating mechanosensitive channels in the urothelium, thereby improving bladder sensory function. METHODS: In this study, a rat model of OAB was created by using intraperitoneal injections of cyclophosphamide. We performed either EA or bladder perfusion with HC-067047 [a transient receptor potential vanilloid 4 (TRPV4) antagonist] and assessed the efficacy of electro-acupuncture in the treatment of OAB in rats viaurodynamic determination and Void spot assay. tissue morphology,

Age- and Urothelium-related Changes in Hydrogen Sulfide-induced Responses in Mouse Bladder.

The alterations in bladder function are associated with aging. Hydrogen sulfide (H 2 S) is a gaseous neurotransmitter that is synthesized in the urinary bladder and is suggested to regulate bladder smooth muscle tone. The effects of age and urothelium on the L-cysteine/H 2 S-induced relaxant responses were investigated in young (3-4 months) and aged (23-24 months) mice. The relaxant responses to endogenous H 2 S (L-cysteine) augmented in denuded urothelium bladder tissue in both age groups. However, the relaxant responses to exogenous H 2 S (sodium hydrogen sulfide: 1 μM - 3 mM) did not change

Comparative evaluation of commonly used catheters through histopathological changes induced in bladder urothelium.

There is growing concern about the toxic potential of chemicals coating various catheter materials, especially due to lack of international standards for toxicity testing. The present study evaluates the toxicity potential in terms of pathological changes induced by the catheters commonly used in practice, i.e. latex-, silicone- and Teflon-coated catheters. Teflon-coated catheters appear to be superior to others in the present study.

Renal pelvic neoplasms and atypical urothelium in patients with end-stage analgesic nephropathy.

In a series of 772 renal transplant patients, 84 had analgesic nephropathy (AN). Four of them had renal pelvic carcinoma. The incidence of atypical urothelial changes of the renal pelvis was studied in 56 AN patients, the majority nephrectomized before or shortly after the renal transplantation. Urothelial atypia, usually bilaterally, was found in 27 patients. Multiple sections resulted in an even higher incidence (8/9). No atypical changes were found in normal kidneys or in end-stage diseased kidneys with other diseases, or in chronically rejected renal allografts. These findings further stre

Primary Urothelium Carcinoma of the Distal Urethra in a Male: Case Report and Literature Review.

Male primary urethral urothelium carcinoma is a rare clinical case. Here, we detail a case of a 58-year-old man with primary urothelium carcinoma of the distal urethra treated in our hospital. The patient with a neoplasm inside the external urethral orifice for 2 years, which was previously diagnosed as condyloma acuminata, had received photodynamic therapy for 3 times, with initial symptoms of urinary stream bifurcation and dysuria. The exfoliative urine cytology showed negative. Cystoscopy showed a tumor growing around the distal urethra. Biopsy and immunohistochemistry revealed high-grade p

Protective effects of heparin and other sulfated glycosaminoglycans on crystal adhesion to injured urothelium.

Heparin was found to prevent calcium oxalate crystal adhesion to chemically injured rat bladder urothelium. Heparin was the only polysaccharide tested which restored injured urothelium to the functional status of normal, uninjured urothelium with respect to anticrystal adhesion properties. Other sulfated glycosaminoglycans, sulfated polysaccharides and monosaccharides were without restorative or protective effects. 3H-heparin was found to bind to injured urothelium but not to normal urothelium.

Cytology in the diagnosis and followup of transitional cell carcinoma of the urothelium: a review with a case series.

Acceptance of the value of urinary cytology in transitional cell carcinoma has been mixed. Over-all accuracy is only fair to good and the results in mass screening are poor. However, cytology can be valuable in the serial evaluation of selected patients by the collection of multiple specimens. It is an important adjunct in upper tract lesions, in carcinoma in situ and in the evaluation of the patient with unexplained hematuria. Herein we analyze patients with transitional cell carcinoma of the urothelium with special reference to the use of cytology irrigation specimens from the bladder and th

Mitochondrial viability in neurogenic bladder urothelium after sigmoidocolocystoplasty. Implications for persistent vesicoureteral reflux.

PURPOSE: We investigated whether inflammatory cell infiltration (ICI), fibrosis, and mitochondrial viability of the neurogenic bladder urothelium are involved in the mechanism of persistent vesicoureteral reflux (VUR) after sigmoidocolocystoplasty (SCP). METHODS: Bladder biopsies obtained 1994-2023 from 62 neurogenic bladder patients were examined by hematoxylin and eosin for ICI, Masson's trichrome for fibrosis, and immunofluorescence for urothelial growth differentiation factor 15 (GDF15; a mitochondrial stress-responsive cytokine) (positive/negative) and heat shock protein 60 (HSP60; a mit

MAFB-mediated CEBPA regulated human urothelium growth through Wnt/β-catenin signaling pathway.

MAFB is essential for regulating male-type urethral differentiation, and especially, its variation can contribute to hypospadias in mice. However, the potential mechanism is still unclear. Here we observed that the basic leucine zipper (bZIP) transcription factor MAFB and CCAAT/enhancer-binding protein alpha (CEBPA) could promote human urothelium SV-HUC-1 growth. Moreover, MAFB and CEBPA expression were reduced in the prepuce tissues of hypospadias patients. Based on transcriptome sequencing analysis and Western blot, MAFB knockdown was found to suppress CEBPA protein expression and repress Wn

Restoring the barrier of chronically damaged urothelium using chondroitin sulfate glycosaminoglycan-replenishment therapy.

PURPOSE OF REVIEW: This study aims to further understand the physiological mechanism of chondroitin sulfate treatment on the urinary bladder in cases of inflammation, by investigating the effect of chondroitin sulfate therapy on recovery of urothelial barrier in an in-vitro chronic injury model. RECENT FINDINGS: With inflammatory bladder conditions, the urothelial barrier seems decreased. Glycosaminoglycan (GAG) replacement therapy is supposed to help restore this barrier. Clinical studies on inflammatory bladder conditions are complicated because of the heterogeneous patient population, henc

Urothelial differentiation of human amniotic fluid stem cells by urothelium specific conditioned medium.

Human amniotic fluid stem cells (HAFSCs) have a high proliferative capacity and a good differentiation potential, and may thus be suitable for regenerative medicine. To date, urothelial differentiation mechanisms of HAFSCs are poorly understood. We have investigated the urothelial differentiation potential of HAFSCs so that they can be therapeutically applied to cure defective diseases of bladder. To induce the stem cell differentiation, HAFSCs were cultured in a bladder cancer-derived conditioned medium. After 2 weeks of culture, HAFSCs began to express the urothelial lineage-specific markers

Cellular localization of blood group antigens (including HLA markers) in human bladder urothelium.

Expression of A, B, H, Lewis, I, i, HLA class I and class II antigens was studied on 81 urinary bladder samples. Striking variation in the expression of HLA class II and to a lesser extent of HLA class I determinants was observed. Expression of at least five distinct A (or B) determinants, the synthesis of which is controlled by H, A, B, secretor and Lewis genes, was demonstrated by using a panel of reagents directed against different A, B and H epitopes. These results suggest that evaluation of blood group changes during tumoral processes requires the precise determination of the specificity

Some short-term cell kinetic effects of ionizing radiation on mouse bladder urothelium.

Early morphological changes and the pattern of reactive proliferation of the hairless mouse urinary bladder urothelium after irradiation are reported. Groups of female hairless mice were anaesthetized with sodium pentobarbital and irradiated over the bladder region with 0, 10, 20 and 30 Gy. Control groups were sham-treated. Short-term cell kinetic changes were monitored using incorporation of tritiated thymidine and flow cytometry. Only minor radiation-induced alterations in the cell kinetic pattern were recorded, and no significant histomorphological changes were seen. However, a marked incre

Lipoxygenase-5 Expression in Canine Urinary Bladder: Normal Urothelium, Cystitis and Transitional Cell Carcinoma.

Transitional cell carcinoma (TCC) is the most common canine urinary tract tumour and mimics human invasive TCC. Human TCCs overexpress lipoxygenase (LOX)-5 and the use of target inhibitors has proven effective in inhibiting neoplastic growth. In this study, we investigated the immunohistochemical expression of LOX-5 in normal canine urinary bladder, cystitis and TCC. The comparative expression of LOX-5, cyclo-oxygenase (COX)-1 and COX-2 among the three tissue groups was also examined. Biopsy samples from cases of cystitis and TCC were reviewed from 2012 to 2016; samples of histologically norma

Epigallocatechin gallate attenuates interstitial cystitis in human bladder urothelium cells by modulating purinergic receptors.

BACKGROUND: Epigallocatechin gallate (EGCG) has exhibited antitumor properties against bladder cancer. However, its effects in interstitial cystitis (IC) have not been investigated. METHODS: Here, we performed repeated cystoscopy and re-biopsy of bladder mucosa before and after intravesical irrigation of EGCG in eight patients diagnosed with IC based on clinical and histopathologic assessments. Six normal bladder tissue samples were obtained from age-, race-, and sex-matched asymptomatic control subjects. IC symptom index was used to compare the therapeutic effect in IC patients. Patient-deri

A mucoadhesive-to-penetrating nanomotors-in-hydrogel system for urothelium-oriented intravesical drug delivery.

Intravesical therapy (IT) is widely used to tackle various urological diseases. However, its clinical efficacy is decreased by the impermeability of various barriers presented on the bladder luminal surface, including the urinary mucus layer and the densely packed tissue barrier. In this study, we report a mucoadhesive-to-penetrating nanomotors-in-hydrogel system for urothelium-oriented intravesical drug delivery. Upon intravesical instillation, its poloxamer 407 (PLX) hydrogel gelated and adhered to the urothelium to prolong its intravesical retention. The urea afterwards diffused into the hy

Urinary ATP Levels Are Controlled by Nucleotidases Released from the Urothelium in a Regulated Manner.

Adenosine 5′-triphosphate (ATP) is released in the bladder lumen during filling. Urothelial ATP is presumed to regulate bladder excitability. Urinary ATP is suggested as a urinary biomarker of bladder dysfunctions since ATP is increased in the urine of patients with overactive bladder, interstitial cystitis or bladder pain syndrome. Altered urinary ATP might also be associated with voiding dysfunctions linked to disease states associated with metabolic syndrome. Extracellular ATP levels are determined by ATP release and ATP hydrolysis by membrane-bound and soluble nucleotidases (s-NTDs). It is
