Current influenza vaccines including live attenuated influenza virus (LAIV) provide suboptimal protection against drift and potential pandemic strains. We hypothesized that supplementing LAIV with a highly conserved antigenic target M2 ectodomain (M2e) would confer cross-protection by inducing humoral and cellular immune responses to conserved antigenic targets. Intranasal vaccination with LAIV (A/Netherlands/602/09, H1N1) supplemented with tandem repeat M2e containing virus-like particles (M2e5x VLP) induced M2e- and virus-specific antibodies. Upon heterosubtypic virus challenge, M2e5x VLP-su

Tick-borne encephalitis virus (TBEV) is an important tick-borne pathogen that poses as a serious public health concern. The coverage and immunogenicity of the currently available vaccines against TBEV are relatively low; therefore, it is crucial to develop novel and effective vaccines against TBEV. The present study describes a novel strategy for the assembly of virus-like particles (VLPs) by co-expressing the structural (core/prM/E) and non-structural (NS2B/NS3Pro) proteins of TBEV. The efficacy of the VLPs was subsequently evaluated in C57BL/6 mice, and the resultant IgG serum could neutrali

BACKGROUND: Zika virus (ZIKV), a mosquito-borne flavivirus, is a re-emerging virus that constitutes a public health threat due to its recent global spread, recurrent outbreaks, and infections that are associated with neurological abnormalities in developing fetuses and Guillain-Barré syndrome in adults. To date, there are no approved vaccines against ZIKV infection. Various preclinical and clinical development programs are currently ongoing in an effort to bring forward a vaccine for ZIKV.
METHODOLOGY/PRINCIPLE FINDINGS: We have developed a ZIKV vaccine candidate based on Virus-Like-Particles

Neuraminidase is the second major surface antigen on influenza virus. We investigated the immunogenicity and cross protective efficacy of virus-like particle containing neuraminidase derived from 2009 pandemic H1N1 influenza virus (N1 VLP) in comparison with inactivated split influenza vaccine. Immunization of mice with N1 VLP induced antibody responses specific for virus and cross-reactive neuraminidase inhibition activity whereas an inactivated split vaccine induced strain-specific hemagglutination inhibition activity. N1 VLP-immunized mice developed cross protective immunity against antigen

UNLABELLED: Influenza virus infections are a global public health problem, with a significant impact of morbidity and mortality from both annual epidemics and pandemics. The current strategy for preventing annual influenza is to develop a new vaccine each year against specific circulating virus strains. Because these vaccines are unlikely to protect against an antigenically divergent strain or a new pandemic virus with a novel hemagglutinin (HA) subtype, there is a critical need for vaccines that protect against all influenza A viruses, a so-called "universal" vaccine. Here we show that mice w

Rift Valley fever virus (RVFV), a member of the family Bunyaviridae, regularly accounts for large and severe outbreaks among humans and livestock in Africa and Arabia. Therefore, safe and efficient vaccines are highly needed. Here, we report the production of recombinant virus-like particles (VLPs) that, in addition to their similarity to RVFV particles, are able to express the viral nucleocapsid (N) gene. A single inoculation of 1 × 10(6) of these N-VLPs was sufficient to protect 100% of mice from infection with a lethal dose of 1 × 10(5) PFU of RVFV. Our study demonstrates that N-VLPs can be

There is intense interest in the design and use of vaccine strategies against influenza to enhance protective immune responses in the elderly. To address the need for improved influenza vaccines for the aged, two inflammatory adjuvants, Imject(®) alum (a stimulator of the Nod-like receptor, Nalp3) and poly I:C (a toll-like receptor type 3 ligand), were used during vaccination with novel influenza virus-like particles (VLP). Adult (4 month old) or aged (24 month old) mice were vaccinated with VLPs alone or in combination with adjuvant. VLP-vaccinated adult mice were protected from a lethal infl

Lyme disease (LD) is the most prevalent vector-borne disease in the United States, impacting ∼476,000 individuals annually with increasing incidence. Prevention relies on personal protective measures such as insecticides and tick checks, underscoring the need for new preventatives such as vaccines. In this work, the importance of vaccine platform in LD vaccine development efforts was evaluated using non-lipidated OspA as model antigen. Recombinant OspA conjugated to CRM197 (EcoCRM OspA), a virus-like particle vaccine utilizing SpyTag and SpyCatcher (OspA-SpyVLP), and an mRNA-based vaccine cons

Equine influenza (EI) is a highly contagious acute respiratory disease of wild and domesticated horses, donkeys, mules, and other Equidae. EI is caused by the Equine Influenza virus (EIV), is endemic in many countries and outbreaks still have a severe impact on the equine industry globally. Conventional EI vaccines are widely used, but a need exists for a platform that facilitates prompt manufacturing of a highly immunogenic, antigenically matched, updated vaccine product. Here we developed a plant-produced bivalent EI virus-like particle (VLP) vaccine candidate which lacks the viral genome an

BACKGROUND: Nano-emulsions with immunogenic properties can be incorporated into vaccines to act as an adjuvant where they can enhance the immune response of a given vaccine. Analytically, studying vaccine antigens, such as Virus-Like Particles (VLPs), in the presence of adjuvants, like nano-emulsions, is very challenging as they are both heterogenous nano species of similar sizes but very different physiochemical properties. Therefore, typical analysis of nanoparticles using separation approaches such as Size Exclusion Chromatography (SEC) and Field-Flow Fractionation (FFF) is difficult due to

Foot-and-mouth disease (FMD) remains a major threat to global livestock health, and current inactivated vaccines face limitations in biosafety and cross-protection. Virus-like particle (VLP) -based vaccines offer a safer alternative, however, their production is often hindered by the cytotoxicity of the viral 3C protease. In this study, we developed a modified 3C protease with reduced cytotoxicity, enabling efficient expression of FMDV VLPs via an internal ribosome entry site (IRES)-based system. To enhance immunogenicity, the swine IgG Fc fragment (sFc) was genetically fused to the VLP capsid

The global incidence of dengue, which is caused by dengue virus (DENV) infection, has grown dramatically in recent decades and secondary infection with heterologous serotype of the virus may cause severe symptoms. Efficacious dengue vaccines should be able to provide long-lasting immunity against all four DENV serotypes simultaneously. In this study, we constructed a novel vaccine platform based on tetravalent dengue virus-like particles (DENV-LPs) in which envelope (E) protein carried a FLAG tag sequence at the position located not only in the exterior loop on the protruding domain but outsid

Rabies is a viral infection of the central nervous system for which vaccination is the only treatment possible. Besides preexposure, vaccination is highly recommended for people living in endemic areas, veterinarians, and laboratory workers. Our group has developed rabies virus-like particles (RV-VLPs) with immunogenic features expressed in mammalian cells for vaccine applications. In this chapter the methods to obtain and characterize a stable HEK293 cell line expressing RV-VLPs are detailed. Further, analytical ultracentrifugation steps to purify the obtained VLPs are developed, as well as w

Respiratory syncytial virus (RSV) remains a major cause of severe respiratory disease in infants and older adults, and despite recent advances in prefusion F-based vaccines and monoclonal antibody prophylaxis, additional vaccine strategies remain of interest. Virus-like particle (VLP) platforms offer a promising approach by mimicking the native virion structure while maintaining an excellent safety profile. In this study, we evaluated the immunogenicity and protective efficacy of RSV-VLPs containing prefusogenic F, G and matrix (M) proteins, administered intramuscularly with or without adjuvan

We aimed to study the stability of a Beta (β)-SARS-CoV-2 virus-like particle (VLP) vaccine in a series of preliminary experiments using selective stabilizing excipients. β-SARS-CoV-2 VLPs were produced and purified using established methodologies. The thermostability of VLPs was tested at 4°C and -30°C in the presence or absence of stabilizers polysorbate 80, sorbitol, or L-histidine in the presence of a physiological NaCl concentration of 137 mM. The integrity of VLPs was assessed using ELISA, Western immunoblot, and dynamic light scatter (DLS). β-SARS-CoV-2 VLPs were stable at 4°C for 14 day

Zika virus (ZIKV) is an arthropod-borne Orthoflavivirus that caused a major outbreak in the Americas in 2015-16. In Brazil, up to 46% of ZIKV positive pregnancies resulted in congenital Zika syndrome (CZS). CZS is characterized by a wide range of neurologic birth defects and miscarriage in up to 7.6% of affected pregnancies. With no current licensed ZIKV vaccines, we sought to evaluate a Zika virus-like particle (VLP) vaccine candidate in a rhesus macaque (RM) pregnancy model. VLPs were produced in mammalian cells expressing the pre-membrane-envelope region of the Asian-lineage ZIKV strain PRV

Background/Objectives: The dengue virus remains endemic in over 100 countries, transmitted by mosquito bites. Current management relies on supportive care, as no highly effective vaccine or approved antiviral exists. The CYD-TDV (Dengvaxia®) vaccine, licensed since 2015 with around 60% efficacy, raises the risk of severe dengue in seronegative children. The newer "Qdenga" vaccine offers up to 80% efficacy after a year but provides suboptimal protection against DENV-3 in seronegative individuals. Over the past two decades, virus-like particles (VLPs) have gained attention as safe, replication-i

BACKGROUND: A systemic evaluation of the antigenic differences of the H7 influenza hemagglutinin (HA) proteins, especially for the viruses isolated after 2016, are limited. The purpose of this study was to investigate the antigenic differences of major H7 strains with an ultimate aim to discover H7 HA proteins that can elicit protective receptor-binding antibodies against co-circulating H7 influenza strains.
METHOD: A panel of eight H7 influenza strains were selected from 3,633 H7 HA amino acid sequences identified over the past two decades (2000-2018). The sequences were expressed on the sur

Coxsackievirus A10 (CVA10) has emerged worldwide as one of the main pathogens of hand, foot, and mouth disease (HFMD) in recent years. However, there is currently no commercial vaccine available to prevent CVA10 infection. Here we report the development of a recombinant virus-like particle (VLP) based candidate vaccine for CVA10. Co-expression of the capsid protein precursor P1 and the protease 3CD of CVA10 in Pichia pastoris resulted in cleavage of P1 into three capsid subunit proteins VP0, VP1, and VP3. These three subunit proteins co-assembled into CVA10 VLPs, which were visualized as spher
