Characterization of constituents by UPLC-MS and the influence of extraction methods of the seeds of Vernonia anthelmintica willd.: extraction, characterization, antioxidant and enzyme modulatory activities.

Vernonia anthelmintica Willd (VA) is a popular medicinal plant used in local and traditional medicine to manage various disorders. In order to explore the phytochemical profile, antioxidant and enzyme modulatory activities of extracts prepared from the seeds of VA, different extraction methodologies, including modern (accelerated-ASE, ultrasound-UAE, and tissue smashing-TSE extractions) and traditional (maceration and Soxhlet) extractions, were employed and their effects on the activities of the extracts were investigated. The chemical compounds of the extracts were qualitatively analyzed by u

The Effect of Volatile Oil from Vernonia anthelmintica Seeds on Melanin Synthesis in B16 Cells and Its Chemical Analysis by GC-QTOF-MS.

Vernonia anthelmintica Willd. seeds have been used in folk medicine for the treatment of leukoderma in Xinjiang, China, for more than 300 years. The promoting activities of its volatile oil (AVO) in melanogenesis and its chemical composition were investigated in this paper. The bioactivities of AVO were examined by melanin synthesis and tyrosinase activity assay in B16 cells. Using GC-QTOF-MS technology, each compound of AVO contains a single separated peak in GC and the retention indices of every GC peak were calculated by the retention times of C7~C30 n-alkanes that were injected at the same

Aqueous Vernonia amygdalina leaf extract in drinking water mitigates aflatoxin B1 toxicity in broilers: effects on performance, biomarker analysis, and liver histology.

This study evaluated aqueous Vernonia amygdalina leaf extract in drinking water as a mitigation strategy against Aflatoxin B1-induced toxicity in broilers, focusing on performance, haematology, serum biochemistry, pro-inflammatory cytokines, cellular stress markers, and liver histology. Two hundred and forty (240) day-old chicks (mixed sex), of the Cobb 500 breed were divided into four groups: control (CONT), AFB1-exposed (AFLB1), and two treatment groups (VE1AF and VE2AF) receiving 0.5 mg/kg AFB1 and Vernonia amygdalina aqueous extract at 1 g/L and 2 g/L, respectively. At 42 days, VE1AF and V

Investigating the effects and mechanisms of Vernonia anthelmintica (L.) willd., seed extracts on melanogenesis and vitiligo treatment based on multi-omics and network pharmacology.

ETHNOPHARMACOLOGICAL RELEVANCE: Vitiligo is a depigmentation disorder characterized by the loss of functional melanocytes, leading to skin and/or hair depigmentation. Vernonia anthelmintica (L.) Willd seed extracts have been traditionally used to treat pigmentation disorders in Central Asia. However, its pharmacological mechanism for vitiligo is still unknown. AIMS OF THE STUDY: In a previous study, we isolated the Extracts (encoded as CAM-Y7) from the Vernonia anthelmintica (L.) Willd seed. This research aims to understand the molecular basis of hyperpigmentation triggered by CAM-Y7. MATERI

BACKGROUND: Vitiligo is a long-term skin disease characterized by the loss of pigment in the skin. The current therapeutic approaches are limited. Although the anti-vitiligo mechanisms of Vernonia anthelmintica (L.) remain ambiguous, the herb has been broadly used in Uyghur hospitals to treat vitiligo. The overall objective of the present study aims to identify the potential lead compounds from Vernonia anthelmintica (L.) in the treatment of vitiligo via an oral route as well as the melanogenic mechanisms in the systematic approaches in silico of admetSAR and substructure-drug-target network-b

Identification of active compounds in Vernonia anthelmintica (L.) willd by targeted metabolome MRM and kaempferol promotes HaCaT cell proliferation and reduces oxidative stress.

Introduction: Vernonia anthelmintica (L.) Willd. is a traditional treatment for vitiligo in Xinjiang. However, its therapeutic mechanism remains unclear owing to its complex composition and limited research on its chemical profile. Methods: We employed a targeted metabolome approach, combining selective reaction monitoring/multiple response monitoring (SRM/MRM) with high-performance liquid chromatography and MRM mass spectrometry to quantitatively analyze the flavonoid constituents of Vernonia anthelmintica. We also used network pharmacology and molecular docking to identify potential vitiligo

Alkaloid extracts from Bitter leaf (Vernonia amygdalina) and Black nightshade (Solanum nigrum) inhibit phosphodiesterase-5, arginase activities and oxidative stress in rats penile tissue.

The erectogenic potential of alkaloids extracted from Bitter leaf (Vernonia amygdalina) and Black nightshade (Solanum nigrum) was investigated in this study. Fresh leaves obtained from Bitter leaf and Black night shade were air-dried, pulverized, and extracted for alkaloids. The inhibitory potential of the alkaloid extracts on arginase and phosphodiesterase-5 (PDE-5) activities in rats penile tissue was determined in vitro. The antioxidant properties were also evaluated and the constituent alkaloids quantified using GC-MS. The alkaloid extracts inhibited arginase (0-30.51 μg/ml) and PDE-5 (0-1

Protective Effects of Methanol Extract of Vernonia amygdalina (del.) Leaf on Aspirin-Induced Gastric Ulceration and Oxidative Mucosal Damage in a Rat Model of Gastric Injury.

This study investigated the quantitative polyphenolic constituents and gastroprotective effects of methanol extract of Vernonia amygdalina leaf (MEVA) against aspirin-induced gastric ulcer in rats. Ulceration was induced by 3 days' oral administration of aspirin (150 mg/kg body weight). Wistar rats were pretreated with cimetidine (reference drug) at a dose of 100 mg/kg body weight and MEVA at 200, 300, and 400 mg/kg body weight once daily for 28 days prior to ulcer induction. At the end of the experiment, gastric secretions, antioxidant status, and histopathological alteration were evaluated.

Isolation, structural elucidation and molecular docking studies against SARS-CoV-2 main protease of new stigmastane-type steroidal glucosides isolated from the whole plants of Vernonia gratiosa.

Phytochemical investigation of the whole plants of Vernonia gratiosa Hance. led in the isolation and identification of two new stigmastane-type steroidal glucosides (1-2), namely vernogratiosides A (1), and B (2). Their chemical structures were fully elucidated based on 1 D/2D NMR spectroscopic, HR-ESI-MS data analyses, and by producing derivatives by chemical reactions. The binding potential of the isolated compounds to replicase protein - main protease of SARS-CoV-2 were examined using the molecular docking simulations. Our results show that the isolated steroidal glucosides (1-2) bind to th

Crude Extract and Phenol-Rich Fractions from Vernonia amygdalina Leaves Ameliorates Streptozotocin-Induced Type 1 Diabetes in Rats by Mitigating Hepatic Injury, Dyslipidemia, and Production of Oxido-Inflammatory Markers.

Diabetes mellitus (DM) is a major disorder contributing to human mortality and morbidity globally. The use of medicinal plants in the management of diabetes is gaining global popularity due to their accessibility and cost-effectiveness. In this study, we evaluated the ameliorative potential of Vernonia amygdalina leaves crude extract (CE), free phenol (FP), and bound phenol (BP) fractions (50 mg/kg body weight) in a rat model of streptozotocin (STZ)-induced type 1 diabetes (T1DM).
