A Chirality-Converted Bacteriolytic Dodecapeptide Regulates Vibrio-Induced Polymicrobial Infection and Ameliorates Invasion-Associated Gut Microbiota Disequilibrium.

The polymicrobial infections caused by Vibrio species, especially multidrug-resistant isolates, are posing increasing threats to the health of coastal residents. Herein, a novel dodecapeptide, denoted as D-zp37, was designed and synthesized, aiming to combat these notorious pathogens. Experimental results proved that the simple chirality conversion of the parent peptide, zp37, could significantly boost the antibacterial activity and proteolytic stability. To a specific cephalosporin-resistant Vibrio alginolyticus, the minimal inhibitory concentration value of D-zp37 was as low as 0.5 µm. Beyon

Machine learning in understanding environmental variability of vibriosis in coastal waters.

Vibrio spp. comprise ecologically significant bacteria that thrive in warm, moderately saline water, and their incidence and proliferation are strongly influenced by environmental factors. In recent years, Vibrio spp. infections have been reported more frequently and over a greater geographical area along the US eastern seaboard. This study provides an analysis of Vibrio spp. infections, notably caused by Vibrio alginolyticus, Vibrio cholerae non-O1/non-O139, Vibrio fluvialis, Vibrio mimicus, V. parahaemolyticus, and V. vulnificus, extracted from the Centers for Disease Control and Prevention'

Diversity and pathogenic potential of vibrios isolated from Abrolhos Bank corals.

We performed the first taxonomic characterization of vibrios and other culturable microbiota from apparently healthy and diseased Brazilian-endemic corals at the Abrolhos reef bank. The diseases affecting corals were tissue necrosis in Phyllogorgia dillatata, white plague and bleaching in Mussismilia braziliensis and bleaching in Mussismilia hispida. Bacterial isolates were obtained from mucus of 22 coral specimens originated from the Abrolhos Bank (i.e. Itacolomis reef, Recife de Fora reef and Santa Barbara Island) in 2007. Vibrios counts in the water and coral mucus were approximately 104 cf

Gut microbiota analysis of Blenniidae fishes including an algae-eating fish and clear boundary formation among isolated Vibrio strains.

Some marine fishes are algae-feeding, and the microorganisms in their digestive tracts produce carbohydrate hydrolyzing enzymes such as agarose and fucosidase, which are potentially interesting resource for new functional enzymes. The purpose of this study was to establish a method for identifying and utilizing characteristic bacteria from the intestines of two algae-eating fish species: Andamia tetradactylus, which exclusively eats algae on the rock surface, and stellar rockskipper Entomacrodus stellifer, which feeds on both algae and invertebrates. We tested the species composition of the in

Transcriptome analysis of Catarina scallop (Argopecten ventricosus) juveniles treated with highly-diluted immunomodulatory compounds reveals activation of non-self-recognition system.

Marine bivalve hatchery productivity is continuously challenged by apparition and propagation of new diseases, mainly those related to vibriosis. Disinfectants and antibiotics are frequently overused to prevent pathogen presence, generating a potential negative impact on the environment. Recently, the use of highly diluted compounds with immunostimulant properties in marine organisms has been trailed successfully to activate the self-protection mechanisms of marine bivalves. Despite their potential as immunostimulants, little is known about their way of action. To understand their effect, a co

Seasonal Prevalence of Enteropathogenic Vibrio and Their Phages in the Riverine Estuarine Ecosystem of South Bengal.

Diarrheal disease remains an unsolved problem in developing countries. The emergence of new etiological agents (non-cholera vibrios) is a major cause of concern for health planners. We attempted to unveil the seasonal dynamics of entero-pathogenic Vibrios in Gangetic riverine-estuarine ecosystem. 120 surface water samples were collected for a period of one year from 3 sampling sites on the Hooghly river. Five enteropathogenic Vibrio species, V. cholerae (35%), V. parahaemolyticus (22.5%), V. mimicus (19.1%), V. alginolyticus (15.8%) and V. vulnificus (11.6%), were present in the water samples.

[Studies on halophilic vibrios causing a food poisoning outbreak in the city of Vladivostok].

V. parahaemolyticus and V. alginolyticus strains isolated from patients during an outbreak of an acute enteric disease in Vladivostok in 1997 were studied. All strains were found to possess typical taxonomic signs. V. parahaemolyticus isolated from humans had direct heat stable haemolysin exotoxin. The overwhelming majority of these strains belonged to serovar O3K6. Among the cultures under study 7 phage types were determined: phage types 1, 2, 7, 10 in 8 V. parahaemolyticus strains and phage types 2, 4. 5. 7 in 5 V. alginolyticus strains. The diagnostic halophilic phage lyzed vibrios in 30.2%

Complicated suppurative otitis media in a Greek diver due to a marine halophilic Vibrio sp.

Halophilic vibrios are distinct from non-cholera vibrios and have been recognized increasingly as potentially pathogenic bacteria in extraintestinal infections. A case of suppurative chronic otitis media in a Greek diver with Vibrio alginolyticus recovered from an ear drainage culture, is reported. The patient received appropriate antimicrobial therapy and her hearing improved significantly after a tympanoplasty type I was performed. The association of halophilic Vibrio spp. infections with prolonged seawater contact, particularly in subtropical climates, is discussed. In swimmers with extensi

Molecular characterization and gene expression of pattern recognition receptors in brown-marbled grouper (Epinephelus fuscoguttatus) fingerlings responding to vibriosis infection.

The pathogen recognition system involves receptors and genes that play a crucial role in activating innate immune response in brown-marbled grouper (Epinephelus fuscoguttatus) as a control agent against various infections including vibriosis. Here, we report the molecular cloning of partial open reading frames, sequences characterization, and expression profiles of Pattern Recognition Receptors (PRRs) in brown-marbled grouper. The PRRs, namely pglyrp5, tlr5, ctlD, and ctlE in brown-marbled grouper, possess conserved domains and showed shared evolutionary relationships with other fishes, humans

Reeler Domain-Containing Proteins Involved in the Antibacterial Immunity of Shrimp Litopenaeus vannamei.

Like other invertebrates, Litopenaeus vannamei lacks adaptive immunity and relies mainly on innate immunity for defense against foreign pathogens. In this study, three distinct Reeler domain-containing molecules were discovered in L. vannamei, designated as LvReeler1, LvReeler2, and LvReeler3. Analysis of tissue-specific expression patterns indicated that LvReeler1 showed predominant expression in the stomach, whereas LvReeler2 and LvReeler3 demonstrated peak transcriptional activity within gill tissues. The expression of these molecules was induced by Vibrio parahaemolyticus. In vivo interfer

Distribution, Prevalence, and Antibiotic Susceptibility Profiles of Infectious Noncholera Vibrio Species in Malaysia.

BACKGROUND: The noncholera Vibrio spp. which cause vibriosis are abundantly found in our water ecosystem. These bacteria could negatively affect both humans and animals. To date, there is a paucity of information available on the existence and pathogenicity of this particular noncholera Vibrio spp. in Malaysia in comparison to their counterpart, Vibrio cholera. METHODS: In this study, we extracted retrospective data from Malaysian surveillance database. Analysis was carried out using WHONET software focusing noncholera Vibrio spp. including Vibrio parahaemolyticus, Vibrio vulnificus, Vibrio f

Vibrio Community Structure Shapes the Diversity of Carbenicillin-Hydrolysing Class A β-Lactamase Circulating in European Coastal Environments.

Coastal environments are increasingly recognised as reservoirs of known antibiotic resistance genes (ARGs), but are less frequently identified as sources of novel ARGs. Here, we investigated class A β-lactamases circulating in European coastal environments used for oyster farming. We examined their diversity, function, and the ecological factors associated with their geographic distribution and environmental dynamics. A high diversity of carbenicillinases was detected in the culturable microbiome of European oysters. The Harveyi and Splendidus clades were key Vibrio lineages structuring the ge

Rapid on-site and simultaneous detection of four foodborne Vibrio species in seafood using a microfluidic RPA-CRISPR/Cas12a platform.

Foodborne Vibrio species commonly contaminate seafood and often co-occur, complicating rapid on-site detection. We developed a centrifugal microfluidic chip integrating one-pot RPA-CRISPR/Cas12a for simultaneous detection of four key species: V. parahaemolyticus, V. vulnificus, V. cholerae, and V. alginolyticus. In this system, target-specific RPA amplification generates amplicons that activate Cas12a-mediated trans-cleavage of fluorescent ssDNA reporters, producing target-dependent fluorescence signals for pathogen identification. The chip has eight units, each with four reaction chambers wit

Bacteria can rotate while body tethered to a solid surface.

The attachment of bacteria to solid surfaces has been studied primarily through the modes of pili or flagella tethering. We report on a common feature of tethering in pililess strains of three species of monotrichous bacteria-Vibrio alginolyticus, Pseudomonas aeruginosa, and Caulobacter crescentus-namely, that they may become tethered to the surface by their cell body rather than by a flagellum. These tethered bacteria rotate in alternating directions about a pivot point located under the cell body. Using high-intensity dark-field microscopy, we observed that, in most cases, the flagellum of a

Outer membrane vesicles in Vibrio species: Roles in biofilm formation and pathogenesis.

Outer membrane vesicles (OMVs) have been increasingly recognized as common mediators of bacterial physiology in Gram-negative bacteria, including Vibrio species. The degree and function of OMV production can differ among strains and even within a single species. The secretion of OMVs is a prevalent trait among many Vibrio species, particularly in pathogenic organisms such as Vibrio cholerae, Vibrio vulnificus, and Vibrio parahaemolyticus. The OMVs released by these organisms are often associated with infection, transport of virulence factors into host cells, defense against stress, biofilm for
