Biochemical composition of lymphocytic choriomeningitis virus interfering particles.

Lymphocytic choriomeningitis (LCM) was interfering particles were enriched relative to infectious virions by ultracentrifugation in a shallow gradient made of Urografin. Electrophoretic analysis revealed that they lacked the small ('S') 23S RNA as well as GP-1 and GP-2 of the infectious virion and also lacked a newly characterized glycoprotein of apparent mol. wt. 85 x 10(3); instead, they contained a novel glycoprotein with mol. wt. 65 x 10(3).

Pathogenesis of temperature-sensitive mutants of Sindbis virus in the embryonated egg. III. Autologous, homologous, and heterologous interference.

Two temperature-sensitive (ts) mutants of Sindbis virus able to synthetize RNA, but not two ts mutants defective in viral RNA synthesis, reduced to lethality of the superinfecting heat-resistant parent strain of Sindbis virus in embryonated chicken eggs as compared with results in controls. Autologous interference (of mutant with parent) was observed under a variety of conditions. Time-course studies in one selected system showed that lower titers of superinfecting parent virus were detected in doubly infected eggs than in controls eggs infected with only parent virus. Homologous interference

Experimental mixed infection with two tick-borne viruses and interferon-mediated interference.

Tick-borne encephalitis virus (TEV), a Flavivirus, and Lipovník virus (LV), a member of the Kemerovo group and complex and possible member of the Rioviridae, were used to infect chick embryo cell (CEC) cultures. LV reproduction was inhibited by preinfection with TEV, mainly in aged cultures. In young CEC cultures, TEV production was stimulated slightly and interferon was sometimes depressed by this superinfection; on the contrary, in aged cultures the superinfecting LV stimulated interferon also when infective LV was not produced because of interference. Double staining by the fluorescent anti

Stage-dependent efficacy of long double-stranded RNA administered after white spot syndrome virus infection in shrimp.

White spot syndrome virus (WSSV) causes catastrophic losses in shrimp aquaculture, yet no antiviral interventions are available once infection has begun. In this study, we evaluated the stage-dependent efficacy of RNA interference (RNAi) by administering long double-stranded RNA (dsRNA) targeting the WSSV ribonucleotide reductase 2 (rr2) gene at defined time points following viral challenge in Penaeus vannamei. Delivery of dsRNA during the peri-infection period (0-12 h post-infection, hpi) resulted in near-complete protection, consistent with interference prior to extensive viral establishment

Intracellular interactions shape antiviral resistance outcomes in poliovirus via eco-evolutionary feedback.

Resistance evolution can undermine antiviral treatment. However, targeting antivirals to shared viral proteins could inhibit resistance evolution if susceptible viruses sensitize resistant ones during cellular coinfection. Pocapavir, a poliovirus capsid inhibitor, uses this sociovirological interference strategy. While susceptible viruses substantially suppress pocapavir resistance in cell culture, a pocapavir clinical trial found widespread resistance and limited clearance time improvements in treated participants. Here, to reconcile these findings, we present an intrahost eco-evolutionary mo

Interference between avian corona and influenza viruses: The role of the epithelial architecture of the chicken trachea.

Respiratory viral infections are among the major causes of disease in poultry. While viral dual infections are known to occur, viral interference in chicken airways is mechanistically hardly understood. The effects of infectious bronchitis virus (IBV) infection on tissue morphology, sialic acid (sia) expression and susceptibility of the chicken trachea for superinfection with IBV or avian influenza virus (AIV) were studied. In vivo, tracheal epithelium of chickens infected with IBV QX showed marked inflammatory cell infiltration and loss of cilia and goblet cells five days post inoculation. Pl

Oligonucleotide-Based Approaches to Inhibit Dengue Virus Replication.

Dengue fever is one of the most common viral infections affecting humans. It is an expanding public health problem, particularly in tropical and subtropical regions. No effective vaccine or antiviral therapies against Dengue virus (DENV) infection are available. Therefore, there is a strong need to develop safe and effective therapeutic strategies that can reduce the burden and duration of hospitalizations due to this life-threatening disease. Oligonucleotide-based strategies are considered as an attractive means of inhibiting viral replication since oligonucleotides can be designed to interac

Varicella-zoster virus immune evasion.

CD4+ and CD8+ T cells play dual roles in varicella-zoster virus (VZV) pathogenesis. The first role is to deliver the virus to cutaneous sites during primary VZV infection, permitting replication at these sites and the successful transmission of the virus to other susceptible individuals. The second contribution of T cells is to provide the critical antigen-specific adaptive immunity needed to stop viral replication and maintain VZV latency in sensory ganglia. The equilibrium between VZV and the host can be predicted to be served by immune evasion mechanisms in at least two important ways, incl

Viruses and Antiviral Responses of Nematodes.

Nematodes are ubiquitous metazoans that occupy diverse ecological niches, exposing them to a wide range of viruses and positioning them as both viral hosts and vectors. Free-living nematodes, particularly Caenorhabditis elegans and Caenorhabditis briggsae, provide powerful genetic models for studying antiviral immune mechanisms. Natural infections with noda-like viruses have revealed powerful antiviral defense strategies in Caenorhabditis species, including RNA interference, the Intracellular Pathogen Response, Signal Transducers and Activators of Transcription (STAT)-like transcriptional regu

Dual interference with novel genes mfgl2 and mTNFR1 ameliorates murine hepatitis virus type 3-induced fulminant hepatitis in BALB/cJ mice.

Our studies and those of many others have implicated hepatocyte necrosis and apoptosis mediated by fibrinogen-like protein-2 (fgl2) prothrombinase and tumor necrosis factor receptor (TNFR) in the development of fulminant viral hepatitis, a disease with a mortality rate greater than 80% in cases lacking immediate organ transplantation. This study was designed to explore the efficacy of dual short hairpin RNA (shRNA) interference with fgl2 and TNFR1 in the treatment of murine hepatitis virus strain 3 (MHV-3)-induced fulminant hepatitis in mice. RNA-centered regulation in SFTSV infection: mechanisms, host interactions, and translational implications.

Severe fever with thrombocytopenia syndrome virus (SFTSV) is an emerging tick-borne pathogen that causes substantial morbidity and high case fatality in severe cases, representing a major public health concern in East Asia. Although protein-mediated mechanisms of SFTSV replication and immune evasion have been extensively studied, accumulating evidence indicates that viral infection is also influenced by RNA-centered regulatory processes. These processes include viral RNA transcription and replication, epitranscriptomic modification, host RNA-binding proteins, RNA interference-related pathways,

Identifying Restriction Factors for Hemorrhagic Fever Viruses: Dengue and Junín.

Host restriction factors are cellular components that interfere with viral multiplication. They are up-regulated and expressed upon viral infection and in consequence their activity is specific. So far several important restriction factors have been described against diverse viruses. The cellular antiviral mechanisms defined include proteins with the ability to interfere with early steps of viral replication and others that have been shown to block viral morphogenesis. However, other strategies by which the antiviral action is exerted still remain elusive. An additional interesting matter is h

Inhibition of monkeypox virus replication by RNA interference.

The Orthopoxvirus genus of Poxviridae family is comprised of several human pathogens, including cowpox (CPXV), Vaccinia (VACV), monkeypox (MPV) and Variola (VARV) viruses. Species of this virus genus cause human diseases with various severities and outcome ranging from mild conditions to death in fulminating cases. Currently, vaccination is the only protective measure against infection with these viruses and no licensed antiviral drug therapy is available. In this study, we investigated the potential of RNA interference pathway (RNAi) as a therapeutic approach for orthopox virus infections usi

Oncornavirus-like protein expression in human prostatic tissue.

A sensitive competition radioimmunoassay using 125 I-labelled p 30 interspecies antigen, antiserum specific to the interspecies antigen of the feline leukaemia virus, and aqueous tissue extracts from prostate was used to examine benign hyperplastic prostates for the presence of protein components able to complete with the interspecies viral antigens. Six of 20 prostatic nodular hyperplastic tissues were competitive in radioimmunoassay with the 125 I-labelled viral antigen for binding sites on the antiviral antibodies. These findings suggest the presence of oncornavirus-like proteins in prostat

Wedelolactone inhibits human cytomegalovirus replication by targeting distinct steps of the viral replication cycle.

Wedelolactone (WDL) is a coumestan present in the plants Eclipta prostrata and Wedelia calendulacea which are used for treatment of a multitude of health problems in traditional medicine. It has previously been shown that WDL exerts antiviral activity against human immunodeficiency virus and hepatitis C virus. In this study, we investigated the effect of WDL on lytic human cytomegalovirus (HCMV) infection. We demonstrate a strong interference with HCMV replication as analyzed in multi-round replication settings. A more detailed analysis of the underlying mechanisms revealed that WDL acts at tw

Interference between the viruses of tick-borne encephalitis complex in the Ixodes ricinus ticks.

The interference of two closely related viruses of the tick-borne encephalitis complex (TBE) in Ixodes ricinus ticks has been observed. 42.9 to 65.0% of dually infested ticks developed virus neutralization antibodies in mice without signs of illness, which contrasts with the control ticks infected with 198 strain only. Other explanations of this interference could include alteration of viral receptors on host cells or induction of antiviral activity in the infected ticks and/or other vectors such as mosquitoes.

Cellular resistance to arbovirus infection.

When an arbovirus enters an arthropod in an infected blood meal, several mechanisms may interact to affect its life cycle and ultimate transmissibility. Intrinsic absolute failure in the establishment of infection must be contrasted with infection that is successfully established but is variably modulated in its viral yield throughout the vector's life-span. Degrees of vertebrate host resistance make this modulation a central factor in determining whether an arthropod is an important vector in nature; moreover, human intervention that affects modulating mechanisms may become a basis for diseas
