Association Between Prolonged Shedding of Zika Virus in Human Semen and Male Reproductive Tract Inflammation.

Zika virus (ZIKV) is a mosquito-borne flavivirus that causes congenital defects. Sexual transmission of ZIKV was confirmed in a recent epidemic; however, mechanisms behind ZIKV infection and persistence in the male reproductive tract (MRT) are unknown. Previously, we found that approximately 33% of men with symptomatic ZIKV infections shed ZIKV RNA in semen, and some men shed ZIKV RNA for >3 months. Here, we evaluated the semen of 49 ZIKV-infected men to identify immune factors correlating with long-term ZIKV shedding in semen and ZIKV-infected cell types in semen. We found that prolonged ZIKV

Reduction of avian influenza virus shedding by administration of Toll-like receptor ligands to chickens.

Avian influenza viruses (AIV) are of concern to the poultry industry. Outbreaks of AIV highlight the urgent need for effective control measures. Prophylactic strategies should be explored that rapidly elicit immunity against the virus. Toll-like receptors (TLRs) are innate immune molecules that can induce anti-viral responses, therefore the application of TLR ligands as prophylactic agents in chickens is gaining more attention. We hypothesized that treatment of chickens with TLR ligands reduces the shedding of AIV from infected birds. In addition, the effects of TLR ligand dose and route of ad

Extended viral shedding of a low pathogenic avian influenza virus by striped skunks (Mephitis mephitis).

BACKGROUND: Striped skunks (Mephitis mephitis) are susceptible to infection with some influenza A viruses. However, the viral shedding capability of this peri-domestic mammal and its potential role in influenza A virus ecology are largely undetermined. METHODOLOGY/PRINCIPAL FINDINGS: Striped skunks were experimentally infected with a low pathogenic (LP) H4N6 avian influenza virus (AIV) and monitored for 20 days post infection (DPI). All of the skunks exposed to H4N6 AIV shed large quantities of viral RNA, as detected by real-time RT-PCR and confirmed for live virus with virus isolation, from

Identification of novel rodent and shrew orthohepeviruses sheds light on hepatitis E virus evolution.

The family Hepeviridae has seen an explosive expansion in its host range in recent years, yet the evolutionary trajectory of this zoonotic pathogen remains largely unknown. The emergence of rat hepatitis E virus (HEV) has introduced a new public health threat due to its potential for zoonotic transmission. This study investigated 2 464 wild small mammals spanning four animal orders, eight families, 21 genera, and 37 species in Yunnan Province, China. Using broadly reactive reverse transcription-polymerase chain reaction (RT-PCR), we systematically screened the presence and prevalence of Orthoh

Ex Vivo Experiments Shed Light on the Innate Immune Response from Influenza Virus.

The innate immune response is recognized as a key driver in controlling an influenza virus infection in a host. However, the mechanistic action of such innate response is not fully understood. Infection experiments on ex vivo explants from swine trachea represent an efficient alternative to animal experiments, as the explants conserved key characteristics of an organ from an animal. In the present work we compare three cellular automata models of influenza virus dynamics. The models are fitted to free virus and infected cells data from ex vivo swine trachea experiments. Our findings suggest th

Effects of the HN Antigenic Difference between the Vaccine Strain and the Challenge Strain of Newcastle Disease Virus on Virus Shedding and Transmission.

Newcastle disease (ND) leading to heavy economic losses to the poultry industry worldwide is caused by Newcastle disease virus (NDV). Even though intensive vaccination programs have been implemented in many countries, virulent NDV can still be frequently isolated in well-vaccinated flocks. We compared the protection efficiency of LaSota and two sub-genotype VIId vaccines, NDV/AI4 and NDV O/AI4, in which NDV O/AI4 was constructed by replacing the hemagglutinin-neuraminidase (HN) gene of the vaccine strain NDV/AI4 with that from the variant NDV strain JS-14-12-Ch by the cross hemagglutination in

Disparate Kinetics of Viable Virus Shedding in Immunocompromised Patients: SARS-CoV-2 Versus Influenza Virus-A Prospective Real-World Cohort Study.

There are limited data comparing viral shedding between influenza and SARS-CoV-2 in immunocompromised patients. We prospectively enrolled 70 SARS-CoV-2 and 16 influenza patients, with 48 SARS-CoV-2 patients matched to 16 influenza patients by propensity score. Weekly collected respiratory samples were subjected to viral culture and antigen testing. In matched cohorts, the time to culture-confirmed viral clearance was shorter for influenza than SARS-CoV-2 (median, 4.5 vs. 29 days; adjusted hazard ratio [aHR] 27.5; P < .001), and antigen negativity occurred faster (3 vs. 23 days; aHR 19.2; P < .

A nonhuman primate model for genital herpes simplex virus 2 infection that results in vaginal vesicular lesions, virus shedding, and seroconversion.

The most commonly used animal models for evaluating the efficacy of HSV-2 candidate vaccines are mice and guinea pigs. While numerous HSV-2 vaccine candidates have been tested in these animals and were effective in reducing disease and mortality, these results did not predict the effectiveness of the vaccines in human trials. Infection of rhesus macaques rarely results in lesions or HSV-2 specific antibody responses. In seeking an animal model that better recapitulates human disease and that might be more predictive of the efficacy of prophylactic vaccines than mice and guinea pigs, we evaluat

Influenza A virus shedding reduction observed at 12 weeks post-vaccination when newborn pigs are administered live-attenuated influenza virus vaccine.

BACKGROUND: Influenza A virus in swine (IAV-S) causes an acute respiratory disease of swine which results in great economic losses. A bivalent H1N1 and H3N2, NS1-truncated live-attenuated IAV-S vaccine (LAIV, Ingelvac Provenza™ ) has recently become available. OBJECTIVE: Reduction of shedding during an outbreak in the nursery or finisher is an important parameter from an epidemiological control strategy; therefore, a laboratory efficacy study was conducted to evaluate nasal virus shedding when vaccinated pigs were challenged with either heterologous H1N2 or H3N2 strains 12 weeks post-vaccinat

Immunization of Domestic Ducks with Live Nonpathogenic H5N3 Influenza Virus Prevents Shedding and Transmission of Highly Pathogenic H5N1 Virus to Chickens.

Wild ducks are known to be able to carry avian influenza viruses over long distances and infect domestic ducks, which in their turn infect domestic chickens. Therefore, prevention of virus transmission between ducks and chickens is important to control the spread of avian influenza. Here we used a low pathogenic wild aquatic bird virus A/duck/Moscow/4182/2010 (H5N3) for prevention of highly pathogenic avian influenza virus (HPAIV) transmission between ducks and chickens. We first confirmed that the ducks orally infected with H5N1 HPAIV A/chicken/Kurgan/3/2005 excreted the virus in feces. All c

Effect of phylogenetic diversity of velogenic Newcastle disease virus challenge on virus shedding post homologous and heterologous DNA vaccination in chickens.

Newcastle disease (ND) is a highly devastating disease for the poultry industry as it causes high economic losses. In this present study, a DNA vaccine containing the F and HN surface antigens of a highly virulent Newcastle disease virus (NDV), NDV/1/Chicken/2005 (FJ939313), was successfully generated. Cell transfection test indicated that the vaccine expressed the F and HN genes in Hep-2 cells. The main objective of this study was to compare the extent of protection induced by DNA vaccination after homologous and heterologous NDV-challenge as determined by the amount of NDV shedding after cha

Cervical shedding of herpes simplex virus in human immunodeficiency virus-infected women: effects of hormonal contraception, pregnancy, and vitamin A deficiency.

Genital shedding of herpes simplex virus (HSV) results in frequent transmission of infection to sexual partners and neonates. In a cross-sectional study, cervical shedding of HSV DNA was detected in 43 (17%) cervical swab samples from 273 women seropositive for HSV-1, HSV-2, and human immunodeficiency virus type 1 (HIV-1). Cervical shedding of HSV was significantly associated with oral contraception (adjusted odds ratio [aOR], 4.5; 95% confidence interval [CI], 1.7-12.2), use of depo-medroxyprogesterone acetate (aOR, 3.2; 95% CI, 1.3-7.7), and pregnancy (aOR, 7.9; 95% CI, 2.0-31.7). Polymorphisms in the haemagglutinin gene influenced the viral shedding of pandemic 2009 influenza virus in swine.

Interactions between the viral surface glycoprotein haemagglutinin (HA) and the corresponding receptors on host cells is one important aspect of influenza virus infection. Mutations in HA have been described to affect pathogenicity, antigenicity and the transmission of influenza viruses. Here, we detected polymorphisms present in HA genes of two pandemic 2009 H1N1 (H1N1pdm09) isolates, A/California/04/2009 (Ca/09) and A/Mexico/4108/2009 (Mx/09), that resulted in amino acid changes at positions 186 (S to P) and 194 (L to I) of the mature HA1 protein. Although not reported in the published H1N1p

Cytomegalovirus and Epstein-Barr virus in breast milk are associated with HIV-1 shedding but not with mastitis.

BACKGROUND: Breast milk HIV-1 load is associated with clinical and subclinical mastitis, and both milk viral load and mastitis are associated with increased mother-to-child-transmission of HIV-1 through breastfeeding. Bacterial infections may cause clinical mastitis, but whether other copathogens common in HIV-1 infection are associated with subclinical mastitis or HIV-1 shedding is unknown. DESIGN: A cross-sectional study of HIV-1-infected breastfeeding women in Zimbabwe was performed to examine the relationship between a wide range of breast coinfections, mastitis, and HIV-1 shedding. The influence of seven environmental and physiological factors on duck plague virus shedding by carrier mallards.

Duck plague (DP) carrier mallards (Anas platyrhynchos) were subjected to seven environmental and physiological conditions in an attempt to stimulate DP virus shedding. The conditions were: food quality, social interaction, reproductive state, time dependency of food and water, noise, exercise, and sex of bird. Cloacal and oral swabs were taken daily for 10 days and assayed for DP virus content. The stimulated carrier ducks shed DP virus intermittently in amounts up to 10(8) ffus/swab/day (the highest 10-fold dilution still showing specific fluorescence). Unstimulated DP carrier ducks shed only
