Synthesis and biological evaluation of 1,3,4-thiadiazole derivatives as type III secretion system inhibitors against Xanthomonas oryzae.

Xanthomonas oryzae pv. oryzae (Xoo) infection directly leads to a severe disease known as leaf blight, which is a major cause of yield loss of rice. Use of traditional bactericides has resulted in severe resistance in pathogenic bacteria. A new approach screening compounds that target the virulence factors rather than killing bacterial pathogens is imperative. In gram-negative bacteria, the type III secretion system (T3SS) is a conserved and significant virulence factor considered as a target for drug development. Therefore, we designed and synthesized a new series of 5-phenyl-2-furan carboxyl

Genomic fingerprinting of epidemic and endemic strains of Stenotrophomonas maltophilia (formerly Xanthomonas maltophilia) by arbitrarily primed PCR.

Arbitrarily primed PCR (AP-PCR) was used to type 64 clinical isolates of Stenotrophomonas maltophilia from 60 patients and the hands of one nurse. Forty-seven different patterns were observed, most patients having isolates with unique genomic fingerprints. A single pattern, however, was obtained from six of eight patients involved in an intensive care nursery outbreak, confirming the suspected nosocomial transmission of this microorganism. This strain was also found in four other patients hospitalized at the same time but in different units. AP-PCR typing results had a good correlation with th

Clinical isolate of a Xanthomonas maltophilia strain producing L-1-deficient and L-2-inducible beta-lactamases.

Xanthomonas maltophilia produces two inducible beta-lactamases, L-1 and L-2, and resists the antimicrobial activity of beta-lactam antibiotics including carbapenems. L-1 has carbapenemase activity and L-2 is a cephalosporinase. It has been suggested that these beta-lactamases share regulatory components. We isolated a recent clinical X. maltophilia strain susceptible to carbapenems and resistant to almost all the other beta-lactam antibiotics tested. beta-Lactamase induction with cefotaxime showed that the clinical isolate had low-level expression of L-1 beta-lactamase but remained inducible f

A-to-I mRNA Editing in a Ferric Siderophore Receptor Improves Competition for Iron in Xanthomonas oryzae pv. oryzicola.

Iron is an essential element for the growth and survival of pathogenic bacteria; however, it is not fully understood how bacteria sense and respond to iron deficiency or excess. In this study, we show that xfeA in Xanthomonas oryzae pv. oryzicola senses extracytoplasmic iron and changes the hydrogen bonding network of ligand channel domains by adenosine-to-inosine (A-to-I) RNA editing. The frequency of A-to-I RNA editing during iron-deficient conditions increased by 76.87%, which facilitated the passage of iron through the XfeA outer membrane channel. When bacteria were subjected to high iron

Diversity and Geographical Structure of Xanthomonas citri pv. citri on Citrus in the South West Indian Ocean Region.

A thorough knowledge of genotypic and phenotypic variations (e.g., virulence, resistance to antimicrobial compounds) in bacteria causing plant disease outbreaks is key for optimizing disease surveillance and management. Using a comprehensive strain collection, tandem repeat-based genotyping techniques and pathogenicity assays, we characterized the diversity of X. citri pv. citri from the South West Indian Ocean (SWIO) region. Most strains belonged to the prevalent lineage 1 pathotype A that has a wide host range among rutaceous species. We report the first occurrence of genetically unrelated,

Novel genes involved in the regulation of pathogenicity factor production within the rpf gene cluster of Xanthomonas campestris.

The synthesis of extracellular enzymes and extracellular polysaccharide (EPS) in Xanthomonas campestris pathovar campestris (Xcc) is subject to co-ordinate regulation by a cluster of genes called rpf (for regulation of pathogenicity factors). These genes are located within a 21.9 kb region of the chromosome isolated as the cosmid clone pIJ3020. The genes in the left-hand section of this region of the chromosome have previously been characterized. This paper reports on the genes in the right-hand section and on the phenotypes of mutants with transposon insertions in these genes. Sequence analys

The ROS-triggered antibacterial mechanism of zinc oxide nanoparticles against Xanthomonas citri subsp. citri for citrus canker management.

Citrus canker, caused by Xanthomonas citri subsp. citri (Xcc), severely impacts citrus production. This study developed a novel zinc oxide nanoparticles ROS-triggered generation system (ZnO NPs) as an effective bactericidal agent against Xcc. The ZnO NPs exhibited higher antibacterial activity than the commercially available pesticide zinc thiazole. The minimum inhibitory concentration (MIC) of ZnO NPs (104.5 μg/mL) is 7.42 times higher than that of 104.5 μg/mL zinc thiazole. Additionally, 1 MIC of ZnO NPs and 1 MIC of zinc thiazole (690.5 μg/mL) exhibited similar therapeutic efficiency, with

Integrated transcriptomic and phenotypic analysis reveals the multi-target antibacterial mechanism of sulforaphane against Xanthomonas oryzae pv. oryzicola.

Plant pathogenic bacteria represent a significant threat to global crop production, thereby necessitating the development of novel and sustainable bactericides. Sulforaphane (SFN), a natural isothiocyanate derived from cruciferous vegetables, is known to possess diverse biological activities including antimicrobial properties. This study provides a comprehensive evaluation of the antibacterial efficacy and underlying mechanisms of SFN against Xanthomonas oryzae pv. oryzicola (Xoc), the pathogen responsible for rice bacterial leaf streak. In vitro assays demonstrated that SFN possesses broad-sp

Xanthomonas campestris pv. campestris: Current Knowledge and Emerging Biological Control Strategies Using Phyllosphere- and Rhizosphere-Associated Bacillus spp.

Xanthomonas campestris pv. campestris, the causal agent of black rot, is one of the most important bacterial pathogens affecting Brassicaceae crops worldwide. Its seed-borne nature, multiple routes of dissemination, and limited availability of resistant cultivars make black rot difficult to manage and underscore the importance of early and reliable pathogen detection. This review summarizes current knowledge on the epidemiology of X. campestris pv. campestris, with emphasis on advances in conventional and molecular approaches used for its detection and characterization, as well as on the curre

Selectivity and Activity of Benzene-1,2,4-triol and its Dimers as Antimicrobial Compounds Against Xanthomonas citri subsp. citri.

Citrus canker, caused by the bacterium Xanthomonas citri subsp. citri, is one of the main threats to citrus fruit production. Several phenolic compounds active against X. citri have been described in recent years. Benzene-1,2,4-triol is a bio-based phenolic compound that has shown high potential as a scaffold for the synthesis of new anti-X. citri compounds. However, benzene-1,2,4-triol is prone to oxidative dimerization. We evaluated the antibacterial activity of benzene-1,2,4-triol, its oxidized dimers, and analogous compounds. Benzene-1,2,4-triol has a low inhibitory concentration against X

Xanthomonas campestris Employs the RND Family Pump HepABCD for Phenolic Acid Efflux and Enhancing Viability and Antioxidant Activity.

Xanthomonas campestris pv. campestris (Xcc) is the causal pathogen of black rot in cruciferous plants. Upon infection, Xcc triggers the accumulation of some phenolic acids in the host plants. The mechanism by which Xcc copes with these defense compounds remains unclear. Here, we revealed that Xcc exports benzoic acid, cinnamic acid, and their monohydroxylated derivatives through the resistance-nodulation-division family efflux pump HepABCD. This efflux system influenced glutathione and catalase levels in the wild-type strain XC1, affecting cell viability and modulating ROS levels. We identifie

Protocatechuic acid, ferulic acid and relevant defense enzymes correlate closely with walnut resistance to Xanthomonas arboricola pv. juglandis.

BACKGROUND: Juglans regia L. is an important nut tree that has a wide range of distribution in temperate regions of the world. In some walnut orchards, walnut blight can become a problematic disease that affects the growth of walnut trees. To explore the correlation between biochemical response and walnut resistance, we inoculated four walnut cultivars with Xanthomonas arboricola pv. juglandis (Xaj). The walnut cultivars were, namely, 'Xiangling', 'Xiluo 2', 'Yuanfeng' and 'Xifu 2'. Total phenol content (TPC) and total flavonoid content (TFC) were measured, whereby nine major phenolic compound

The Xanthomonas RaxH-RaxR Two-Component Regulatory System Is Orthologous to the Zinc-Responsive Pseudomonas ColS-ColR System.

Genome sequence comparisons to infer likely gene functions require accurate ortholog assignments. In Pseudomonas spp., the sensor-regulator ColS-ColR two-component regulatory system responds to zinc and other metals to control certain membrane-related functions, including lipid A remodeling. In Xanthomonas spp., three different two-component regulatory systems, RaxH-RaxR, VgrS-VgrR, and DetS-DetR, have been denoted as ColS-ColR in several different genome annotations and publications. To clarify these assignments, we compared the sensor periplasmic domain sequences and found that those from Ps

Genetic locus encoding functions involved in biosynthesis and outer membrane localization of xanthomonadin in Xanthomonas oryzae pv. oryzae.

Xanthomonadins are membrane-bound, brominated, aryl-polyene pigments specific to the genus Xanthomonas. We have characterized a genetic locus (pig) from Xanthomonas oryzae pv. oryzae which contains four open reading frames (ORFs) that are essential for xanthomonadin production. Three of these ORFs are homologous to acyl carrier proteins, dehydratases, and acyl transferases, suggesting a type II polyketide synthase pathway for xanthomonadin biosynthesis. The fourth ORF has no homologue in the database. For the first time, we report that a putative cytoplasmic membrane protein encoded in the pig

Effectiveness of tailocins produced by Pseudomonas fluorescens SF4c in controlling the bacterial-spot disease in tomatoes caused by Xanthomonas vesicatoria.

The development of alternatives for the use of chemical pesticides for plant disease control is the present-day and ongoing challenge for achieving sustainable agriculture. Pseudomonas fluorescens SF4c, native strain from wheat, produces tailocins (phage-tail-like bacteriocins) with antimicrobial activity against several phytopathogenic strains. We thus investigated the efficacy of foliar application of these bacteriocins to control the bacterial-spot disease in tomato caused by Xanthomonas vesicatoria Xcv Bv5-4a. The disease severity and incidence index were reduced by 44 and 36%, respectivel

Genome-Guided TaqMan qPCR for Detection of Xanthomonas albilineans and Quantitative Interpretation of Pathogen Abundance in Sugarcane Leaf Scald.

Sugarcane leaf scald, caused by Xanthomonas albilineans, is difficult to diagnose because latent infections often precede symptom development, allowing infected planting material to disseminate the pathogen unnoticed. Although quantitative PCR (qPCR) has substantially improved pathogen detection, most available assays target conserved ribosomal regions, which can limit species-level discrimination and offer little guidance for interpreting pathogen abundance. Comparative genomic analysis identified Xal_000736 as a candidate species-specific target for assay development. A TaqMan qPCR assay tar

Antibacterial agents to control of pathogenic Xanthomonas arboricola pv. juglandis on walnut fruits and plants by polygodial derivatives formulations.

BACKGROUND: Walnut bacterial blight, caused by Xanthomonas arboricola pv. juglandis (Xaj), leads to significant yield losses in walnut production, which is a key agricultural sector in Chile. Conventional control methods based on copper have limitations owing to environmental concerns, phytotoxicity, and bacterial resistance. Plant-derived compounds such as polygodial offer a promising alternative as a result of their potent antibacterial properties. This study aimed to develop and evaluate the antimicrobial efficacy of nanoemulsions containing drimanic compounds, specifically polygodial (1) a
