Effect of hydroxypropyl-beta-cyclodextrin on the solubility, stability and in-vitro release of ciprofloxacin for ocular drug delivery.

Eye drops in the form of an aqueous solution with a lower viscosity are preferred for local administrations in ophthalmology. In ophthalmic formulations, cyclodextrins (CDs) are frequently used in recent years in order to increase water solubility, stability and bioavailability of an active substance and decrease an irritation to the eye. The scope of the present study was to investigate the influence of hydroxypropyl-beta-cyclodextrin (HPCD) on the solubility, stability and in vitro release of ciprofloxacin (CIP). According to the phase solubility studies, A(L) type solubility curve was obtai

High shear mixing granulation of ibuprofen and beta-cyclodextrin: effects of process variables on ibuprofen dissolution.

The aims of the study were to evaluate the effect of high shear mixer (HSM) granulation process parameters and scale-up on wet mass consistency and granulation characteristics. A mixer torque rheometer (MTR) was employed to evaluate the granulating solvents used (water, isopropanol, and 1:1 vol/vol mixture of both) based on the wet mass consistency. Gral 25 and mini-HSM were used for the granulation. The MTR study showed that the water significantly enhanced the beta-cyclodextrin (beta CD) binding tendency and the strength of liquid bridges formed between the particles, whereas the isopropanol

Properties of cholesterol-reduced butter made with beta-cyclodextrin and added evening primrose oil and phytosterols.

The present study was carried out to examine changes in the chemical and sensory properties of butter in which the cholesterol was reduced and to which evening primrose oil (EPO) and phytosterols were added. Crosslinked beta-cyclodextrin (beta-CD) made from adipic acid was used, and approximately 90% of the cholesterol was removed. The color measurement values "L" and "a" were significantly different between the control (butter with no beta-CD treatment and no added EPO and phytosterols) and treatment A (butter treated with 10% crosslinked beta-CD); however, the color values for "L" and "a" we

In vitro and in vivo evaluation of an amylobarbitone/hydroxypropyl-beta-cyclodextrin complex prepared by a freeze-drying method.

The complex formation of amylobarbitone (AMB) with 2-hydroxypropyl-beta-cyclodextrin (HP-beta-CD) was investigated in aqueous solution and in the solid state. The apparent stability constant for complex formation (Kc) calculated by phase solubility and spectral shift methods was 524 M-1 and 568 M-1, respectively. The stoichiometric molar ratio of the complex was estimated to be 1:1 and the solubility of AMB in water was increased about 3 fold. The solid dispersion system of AMB/HP-beta-CD in 1:1 molar ratio was prepared by a freeze-drying method. Differential scanning calorimetry (DSC), x-ray

[The separation of three basic drugs with carboxymethyl-poly-beta-cyclodextrin as chiral additive by capillary electrophoresis].

Five cyclodextrin derivatives, namely phosphate ester beta-CD, carboxymethyl-beta-CD(CM-beta-CD), bis-[6-oxygen-(beta-carboxymethyl-succinic acid-4-ester)] beta-CD(F-CM-beta-CD), beta-CD polymer(P-beta-CD), carboxymethyl-poly-beta-CD (CD-P-beta-CD), were used as chiral selector for separation of three basic drugs, lobeline, thiopendonesodium, flunarizine by capillary zone electrophoresis (CZE). All the five cyclodextrins have chiral separation ability to lobeline. P-beta-CD and CM-P-beta-CD have chiral recognition to thiopentonesodium and flunarizine. The results indicate that via optimizing s

Innovative approaches for beetroot (Beta Vulgaris L.) aqueous extraction by cyclodextrins and its use to alleviate ethanol induced gastric ulcer in rats.

One of the most common chronic conditions of peptic ulcer is gastric ulcer (GU), which is recognized as a high-risk illness in the present-day lifestyle. Originating from Asia and Europe, Beetroot (Beta vulgaris L.) is packed with high amounts of bioactive compounds like betanin and phenolics. These contribute to its nutritional value and overall health benefits. In this work, varying concentrations (0.5% to 7% w/v) of beta-cyclodextrin (β-CD) and hydroxypropyl-beta-cyclodextrin (HP-β-CD) were used to improve the efficiency of extracting beetroot components with water, using both stirring and

Properties and Encapsulation Capabilities of Beta-Cyclodextrin Modified with (Hydroxymethyl or 2-Hydroxyethyl)-1,2,3-Triazole Purified Electrochemically.

Two beta-cyclodextrins (β-CD) modified with 2-hydroxyethyl-1,2,3-triazole or hydroxymethyl-1,2,3-triazole groups are synthesized utilizing copper catalyzed 1,3-Huisgen click cycloaddition reaction. A simple electrochemical purification process is employed to remove unwanted copper from the desired products affording up to 88% removal in 10 h. Physicochemical analysis of both purified modified β-CDs (Modβ-CDs) reveales marked differences caused just by the additional methyl group, not only between their physicochemical properties (such as melting point and hydrophilicity) but also in their capa

Host-guest stoichiometry affects the physicochemical properties of beta-cyclodextrin/ferulic acid inclusion complexes and films.

An inclusion system of embedding ferulic acid into β-cyclodextrin (FACD) with different host-guest stoichiometries was prepared by a co-precipitation method. Then, the physicochemical properties and release kinetics of the FACD were evaluated. The results of thermal analysis, X-ray diffraction, scanning electron microscopy, and Fourier transform infrared spectroscopy (FTIR) proved the successful embedding of FA into the β-cyclodextrin matrix. Four mathematical models were applied to adjust the ferulic acid release profile and identify preferential kinetics. The results of physicochemical prope

Direct administration of 2-Hydroxypropyl-Beta-Cyclodextrin into guinea pig cochleae: Effects on physiological and histological measurements.

2-Hydroxypropyl-Beta-Cyclodextrin (HPβCD) can be used to treat Niemann-Pick type C disease, Alzheimer's disease, and atherosclerosis. But, a consequence is that HPβCD can cause hearing loss. HPβCD was recently found to be toxic to outer hair cells (OHCs) in the organ of Corti. Previous studies on the chronic effects of in vivo HPβCD toxicity did not know the intra-cochlear concentration of HPβCD and attributed variable effects on OHCs to indirect drug delivery to the cochlea. We studied the acute effects of known HPβCD concentrations administered directly into intact guinea pig cochleae. Our n

Production of beta-maltooligosaccharides of alpha- and delta-tocopherols by Klebsiella pneumoniae and cyclodextrin glucanotransferase as anti-allergic agents.

The glycosylation of alpha- and delta-tocopherols using Klebsiella pneumoniae and cyclodextrin glucanotransferase (CGTase) was investigated. K. pneumoniae converted alpha- and delta-tocopherols into the corresponding beta-glucosides in 10 and 8% yield, respectively. CGTase glycosylated alpha-tocopheryl beta-glucoside to alpha-tocopheryl beta-maltoside (51%) and alpha-tocopheryl beta-maltotrioside (35%). On the other hand, delta-tocopheryl beta-glucoside was converted into the corresponding beta-maltoside (45%) and beta-maltotrioside (29%) by CGTase. The beta-glucoside of alpha-tocopherol, and

Preparation and enantioseparation characteristics of three chiral stationary phases based on modified beta-cyclodextrin for liquid chromatography.

In order to study the effect of the nature and the length of the spacer, three mixed 10-undecenoate/phenylcarbamate derivatives of beta-cyclodextrin have been prepared and linked to allylsilica gel by means of a radical reaction. The chiral recognition ability of the resulting materials, when used as liquid chromatography chiral stationary phases (CSPs), was evaluated using heptane and either 2-propanol or chloroform as organic mobile-phase modifiers. A large variety of racemic compounds have been separated successfully on these CSPs (mainly pharmaceuticals and herbicides). Optimization of the

Monolithic silica columns with chemically bonded beta-cyclodextrin as a stationary phase for enantiomer separations of chiral pharmaceuticals.

An enantioselective silica rod type chiral stationary phase (CSP) is presented; a novel combination of the well known enantiomer separation properties of beta-cyclodextrin and the unique properties concerning the flow behavior of silica monoliths. Two different synthesis routes are described, and it was found that the in situ modification of a plain silica rod column turned out to be the best. The chromatographic behaviour of the beta-cyclodextrin silica rod was studied and compared with a very similar commercially available beta-cyclodextrin bonded particulate material (ChiraDex). Even if the

Effect of (hydroxypropyl)-beta-cyclodextrin on flux of morphine, fentanyl, sufentanil, and alfentanil through the spinal meninges of monkey.

Previous studies have demonstrated that (hydroxypropyl)-beta-cyclodextrin behaves as a slow-release reservoir when used as a vehicle for intrathecal administration of opioids. The goal of the current investigation was to determine if (hydroxypropyl)-beta-cyclodextrin might serve as a slow-release vehicle for epidural opioid administration as well. An in vitro diffusion cell model was used to determine the flux of morphine, fentanyl, alfentanil, and sufentanil through the spinal meninges of Macaque nemestrina monkeys in the absence or presence of varying concentrations of (hydroxypropyl)-beta-c

Micelles via self-assembly of amphiphilic beta-cyclodextrin block copolymers as drug carrier for cancer therapy.

We developed intelligent, star-shaped amphiphilic β-cyclodextrin (β-CD) co-polymer nanocarriers to circumvent the poor drug loading and water-solubility of β-CD. The secondary hydroxyl groups of β-CD were methylated to improve solubility, and the primary hydroxyl groups were conjugated with mPEG-b-PCL-SH through disulfide linkage to amplify the hydrophobic cavity and enhance the stability of the nanocarrier. A series of amphiphilic β-CD block copolymers (CCPPs) differing in molecular weights were synthesized that could self-assemble into core-shell nanospheres measuring 50-70 nm in water. The
