Inborn errors of proline metabolism.

l-Proline concentration is primarily related to the balance of enzymatic activities of proline dehydrogenase [proline oxidase (POX)] and Delta-1-pyrroline-5-carboxylate (P5C) reductase. As a result, P5C plays a pivotal role in maintaining the concentration of proline in body fluids and inborn errors of P5C metabolism lead to disturbance of proline metabolism. Several inborn errors of proline metabolism have been described. Hyperprolinemia type I (HPI) is a result of a deficiency in POX. The POX gene (PRODH) is located on chromosome 22 (22q11.2) and this region is deleted in velo-cardio-facial

Exploring the functional and metabolic effects of adding garra fish meal to a plant-based broiler chicken diet.

The present study evaluated the metabolic and functional effects of adding garra meal to a broiler chicken diet. Three hundred twenty Sasso-breed day-old chicks were randomly assigned to four dietary treatments with either 0, 10, 20 or 30% garra meal added on top of formulated starter and grower basal diets. The experiment lasted for 42 days. Feed intake and body weight gain increased at the starter and grower phases of broilers with garra meal addition (P < 0.05). Broiler chickens fed 30% garra meal were more efficient in converting feed to body weight and yielded the highest carcass weight (

Alpha-ketoglutarate protects against myocardial infarction via FTO-mediated anti-inflammatory macrophage activation.

Ischemic heart disease lacks optimal therapies targeting post-infarction inflammation and remodeling. The role of TCA cycle metabolites in modulating macrophage-driven cardiac inflammation remains unclear. This study hypothesized that AKG supplementation attenuates cardiac dysfunction by regulating macrophage activation via TCA cycle replenishment and FTO-dependent epigenetic mechanisms. Myocardial infarction was induced in male C57BL/6 mice and macrophage-specific FTO knockout mice via left anterior descending artery ligation. Mice received AKG supplementation. Techniques included echocardiog

Mitochondria-Targeting Polymeric Micelles for Intervertebral Disc Degeneration Alleviation via Coordinated Cascade Energetic Intervention.

Mitochondrial dysfunction plays a pivotal role in intervertebral disc degeneration (IVDD) by disrupting mitochondrial bioenergetic balance, which leads to impaired ATP synthesis, accumulation of ROS, degradation of the extracellular matrix, and degeneration of nucleus pulposus cells. Despite advances in single or multiple therapeutic strategies, a more comprehensive and coupled cascade regulation strategy that fundamentally restores mitochondrial function is urgently needed, but rarely reported. Mitochondrial bioenergetics is cascade-regulated by the interplay between the OXPHOS and the TCA cy

Fructose reprogrammes glutamine-dependent oxidative metabolism to support LPS-induced inflammation.

Fructose intake has increased substantially throughout the developed world and is associated with obesity, type 2 diabetes and non-alcoholic fatty liver disease. Currently, our understanding of the metabolic and mechanistic implications for immune cells, such as monocytes and macrophages, exposed to elevated levels of dietary fructose is limited. Here, we show that fructose reprograms cellular metabolic pathways to favour glutaminolysis and oxidative metabolism, which are required to support increased inflammatory cytokine production in both LPS-treated human monocytes and mouse macrophages. Krebs-cycle-deficient hereditary cancer syndromes are defined by defects in homologous-recombination DNA repair.

The hereditary cancer syndromes hereditary leiomyomatosis and renal cell cancer (HLRCC) and succinate dehydrogenase-related hereditary paraganglioma and pheochromocytoma (SDH PGL/PCC) are linked to germline loss-of-function mutations in genes encoding the Krebs cycle enzymes fumarate hydratase and succinate dehydrogenase, thus leading to elevated levels of fumarate and succinate, respectively1-3. Here, we report that fumarate and succinate both suppress the homologous recombination (HR) DNA-repair pathway required for the resolution of DNA double-strand breaks (DSBs) and for the maintenance of

Radioresistant Cervical Cancers Are Sensitive to Inhibition of Glycolysis and Redox Metabolism.

Highly glycolytic cervical cancers largely resist treatment by cisplatin and coadministered pelvic irradiation as the present standard of care. In this study, we investigated the effects of inhibiting glycolysis and thiol redox metabolism to evaluate them as alternate treatment strategies in these cancers. In a panel of multiple cervical cancer cell lines, we evaluated sensitivity to inhibition of glycolysis (2-deoxyglucose, 2-DG) with or without simultaneous inhibition of glutathione and thioredoxin metabolism (BSO/AUR). Intracellular levels of total and oxidized glutathione, thioredoxin redu

Oxaloacetate Ameliorates Chemical Liver Injury via Oxidative Stress Reduction and Enhancement of Bioenergetic Fluxes.

Chemical injury is partly due to free radical lipid peroxidation, which can induce oxidative stress and produce a large number of reactive oxygen species (ROS). Oxaloacetic acid is an important intermediary in the tricarboxylic acid cycle (TCA cycle) and participates in metabolism and energy production. In our study, we found that oxaloacetate (OA) effectively alleviated liver injury which was induced by hydrogen peroxide (H₂O₂) in vitro and carbon tetrachloride (CCl₄) in vivo. OA scavenged ROS, prevented oxidative damage and maintained the normal structure of mitochondria. We further confirme

Synergistic effects of bioactive silica and fluoride in enamel protection and repair: an in vitro study.

Erosive tooth wear (ETW) compromises enamel integrity. This study investigated the synergistic effects of bioactive silica and fluoride, as well as the independent performance of bioactive silica, in protecting sound enamel and repairing previously eroded enamel under erosive-abrasive challenges. An in vitro laboratory design was adopted using two models: (1) a protective model with sound enamel and (2) a remineralizing model with pre-eroded enamel. Ninety-six bovine enamel blocks were randomly allocated to four dentifrice groups (n=12/group) within two experimental models: RGS/NaF (bioactive

Structure-activity relationships and rational screening of cyclodextrin polymer-confined nZVI activators for efficient persulfate oxidation of recalcitrant organic pollutants in water.

The preparations of cyclodextrin polymer-confined nZVI activators (CDP-nZVI) with both structural modifiability and pollutant selectivity for water decontamination face the challenge of revealing the structure-activity relationships of CDP-nZVI. Here we developed structure-activity relationship models of both the Fe content and activation potential of CDP-nZVI activators for efficient PS oxidation of recalcitrant pollutants in water. Both models (R2 = 0.927/0.801, n = 28, P < 0.001) were primarily constituted by CD types, Fe3+/2+ dosages, and dosage ratios of the tailoring reagent (citric acid

Mechanism of antimony oxidation and adsorption using immobilized Klebsiella aerogenes HC10 in soil.

Klebsiella aerogenes HC10 is one of the few strains isolated from contaminated soil that efficiently oxidizes Sb. However, the sensitivity of microorganisms to environmental conditions limits Sb-oxidizing bacteria applications in soil remediation. Immobilizing Sb-oxidizing bacteria is a promising strategy to improve colonization rates and microorganism inefficiencies and to strengthen bioremediation in Sb-contaminated soil. This study evaluated the feasibility of an immobilization approach to enhance Sb oxidation and the remediation performance of strain HC10 in soil. The results indicated tha

Mitochondrial pyruvate carrier abundance mediates pathological cardiac hypertrophy.

Cardiomyocytes rely on metabolic substrates, not only to fuel cardiac output, but also for growth and remodelling during stress. Here we show that mitochondrial pyruvate carrier (MPC) abundance mediates pathological cardiac hypertrophy. MPC abundance was reduced in failing hypertrophic human hearts, as well as in the myocardium of mice induced to fail by angiotensin II or through transverse aortic constriction. Constitutive knockout of cardiomyocyte MPC1/2 in mice resulted in cardiac hypertrophy and reduced survival, while tamoxifen-induced cardiomyocyte-specific reduction of MPC1/2 to the att

Glucose is preferentially utilized for biomass synthesis in pressure-overloaded hearts: evidence from fatty acid-binding protein-4 and -5 knockout mice.

AIMS: The metabolism of the failing heart is characterized by an increase in glucose uptake with reduced fatty acid (FA) oxidation. We previously found that the genetic deletion of FA-binding protein-4 and -5 [double knockout (DKO)] induces an increased myocardial reliance on glucose with decreased FA uptake in mice. However, whether this fuel switch confers functional benefit during the hypertrophic response remains open to debate. To address this question, we investigated the contractile function and metabolic profile of DKO hearts subjected to pressure overload. METHODS AND RESULTS: Transv

Reduction of tertiary amine N-oxides by rat liver mitochondria.

Reduction of tertiary amine N-oxides by the mitochondrial fraction of rat liver was investigated. NADPH was required as a cofactor. The rate of reaction was faster with the NADPH-generating system than with NADPH. Isocitrate in the NADPH-generating system revealed the maximum stimulation. A little less activity was observed when NADH was used as a cofactor instead of NADPH. The reductase activity was markedly inhibited under aerobic conditions. The rates of mitochondrial tertiary amine N-oxide reduction expressed in nanomoles per milligram of protein per minute were: N,N-dimethylaniline N-oxid
