A thermostable presentation of the live, attenuated peste des petits ruminants vaccine in use in Africa and Asia.

The research objective was to develop a thermostable vaccine against peste des petits ruminants (PPR), a morbilliviral disease of small ruminants targeted for eradication that is a major constraint on the livelihoods of the rural poor throughout much of Africa and Asia. Although existing PPR vaccines provide life-long immunity, they require continuous refrigeration. This limits their utility in developing countries. Methods for the lyophilization of a related morbillivirus, rinderpest (RP), resulted in vaccine that could be used in the field for up to 30days without refrigeration which was a m

Detection of bacterial pyrogens on the basis of their effects on gamma interferon-mediated formation of neopterin or nitrite in cultured monocyte cell lines.

In a number of mammalian cell types, pteridine biosynthesis from guanosine 5'-triphosphate and formation of nitric oxide from L-arginine are induced by gamma interferon (IFN-gamma) and bacterial lipopolysaccharide (LPS). We assessed the possibility of using such metabolic alterations for the in vitro detection of pyrogens. Products from gram-negative and gram-positive bacteria and related synthetic compounds were tested for their potential to induce either of these pathways. Stimulation of pteridine biosynthesis was monitored as the formation of neopterin in the human myelomonocytic cell line

Studies on the mechanisms of mammalian cell killing by a freeze-thaw cycle: conditions that prevent cell killing using nucleated freezing.

Normally a freeze-thaw cycle is a very efficient method of killing mammalian cells. However, this report describes conditions that prevent killing of cultured mammalian cells by nucleated freezing at -24 degrees C. Optimal protection from cell killing at -24 degrees C was obtained in isotonic solutions containing an organic cryoprotectant such as dimethyl sulfoxide (DMSO; 10%, v/v), a saccharide such as sucrose over a broad concentration range from 50 to 150 mM, and glucose. Glycerol was also an effective cryoprotectant but other organic solvents were ineffective, although in some cases they a

The maltase, glucoamylase and transglucosylase activities of acid -glucosidase from rabbit muscle.

1. The maltase and glucoamylase activities of acid alpha-glucosidase purified from rabbit muscle exhibited marked differences in certain physicochemical properties. These included pH stability, inactivation by thiol-group reagents, inhibition by alphaalpha-trehalose, methyl alpha-d-glucoside, sucrose, turanose, polyols, glucono-delta-lactone and monosaccharides, pH optimum and the kinetics and pH-dependence of cation activation. 2. The results are interpreted in terms of the existence of at least two specific substrate-binding sites or sub-sites. One site is specific for the binding of maltose

From Efficacy to Process: Uric-Acid-Lowering Effect of Limosilactobacillus reuteri W6-12 and Its Production Process Optimization.

Clinical chemical drugs for hyperuricemia (HUA) often cause side effects including liver and kidney injury. Developing safe and effective probiotic preparations for HUA intervention is of considerable application value. However, the large-scale production of probiotics faces two major bottlenecks: low viable cell density during fermentation and poor survival after lyophilization. This study integrated in vivo efficacy evaluation and mechanism exploration with the production process optimization for Limosilactobacillus reuteri W6-12. We first verified its hypouricemic effect in vivo, then adopt

Physiological-Biochemical Characteristics and Omics Landscape of Drought Tolerance in Chlorella sp. from the Taklamakan Desert of China.

Microalgae endemic to desert ecosystems possess exceptional stress-resistance traits; however, the physiological and molecular drought-adaptation mechanisms of desert-dwelling Chlorella remain largely uncharacterized. Herein, we combined physiological-biochemical assays, ultrastructural observation, transcriptomics and metabolomics to dissect drought responses in Taklamakan-desert-origin Chlorella sp. under polyethylene glycol 6000 (PEG-6000)-simulated mild (10%), moderate (20%), and severe (30%) drought stress, with mechanistic focus on moderate-drought conditions. Moderate drought repressed

α-Tocopherol-loaded freeze-dried liposomes and proliposomes: powder properties and thermal performance.

Dry lipid-based formulations can enhance the handling and preservation of lipophilic bioactives, though their efficacy depends heavily on formulation composition and processing conditions. This study compared α-tocopherol-loaded freeze-dried liposomes (FD_LPs) and proliposomes (PLPs) formulated with either mannitol (PLPs_MAN) or trehalose (PLPs_TRE) as carriers. All three formulations were evaluated for reconstitution characteristics, powder properties, and thermal performance after rehydration. Additionally, a four-week storage stability test was conducted at 4, 25, and 40 °C, restricted to F

Neuroprotective Potential of Spermidine in Drosophila sws Neurodegenerative Model.

Neurodegenerative disorders are characterized by progressive neuronal loss and functional decline, yet effective interventions remain limited. The polyamine spermidine was suggested to exert neuroprotective effects, but its concentration-dependent impact on longevity, neuronal integrity, and behavior remains still not well studied. Here, we investigated the effects of spermidine on lifespan, behavioral responses, brain tissue, target gene expression, and antioxidant status in Drosophila melanogaster model of age-dependent neurodegeneration. Wild-type flies and swiss cheese (sws1) mutants were

Evolutionary history and gut microbial genome size in health and disease.

Host-microbe codiversification reflects a shared evolutionary history between hosts and their associated microbial lineages. These patterns indicate stable, multi-generational maintenance by multiple mechanisms, including vertical and familial transmission. While host-microbe codiversification has been observed in mammals, including humans, it remains unclear whether the loss of evolutionarily stable symbionts predicts disease status. In this study, we conducted a meta-analysis of 41 published studies spanning five disease categories (autism, neurodegenerative diseases, diabetes, inflammatory

Cryopreservation of Red-Blood-Cell-Derived Extracellular Vesicles.

Extracellular vesicles (EVs) are crucial in many physiological and pathological processes, and therefore, they are increasingly studied for their potential as diagnostic biomarkers, therapeutic agents, and drug carriers. Red-blood-cell-derived EVs (REVs) have gained particular interest due to their beneficial properties for drug delivery and their unique biophysical and molecular characteristics, which make them a practical model system for EV research. While EV cryopreservation methods have advanced in the past decade, REVs remain relatively understudied, with their hemoglobin-rich compositio

Beyond saponins: An integrated mass spectrometry strategy for profiling, spatial mapping, and rapid authentication of non-saponin constituents in Panax species.

Non-saponin constituents of Panax species, including amino acids, sugars, and nucleosides, have attracted increasing attention due to their nutritional relevance and potential health benefits in food-medicine homologous materials. However, their high polarity and chemical diversity pose significant challenges for comprehensive characterization, limiting their application in food quality evaluation and authenticity assessment. In this study, an integrated mass spectrometry-based workflow was developed to systematically profile water-soluble non-saponins in Panax ginseng, Panax notoginseng, and

Proteome-informed interrogation of a methanogenic archaeon links stress response mechanisms with microbial performance and robustness during biomethanation.

Hydrogenotrophic methanogens are promising biocatalysts for biomethanation and carbon dioxide utilization, yet their robustness under bioprocess perturbations remains insufficiently defined. We quantified robustness in a Methanothermobacter archaeal strain with proven potential for industrial application across temperature shifts, oxidative exposure, nitrogen depletion, and hydrogen starvation, measured five key cellular functions in batch culture, and derived a Fano factor based robustness metric that links data dispersion to functional stability. Thermal and oxidative stress were the primary

Molecular adaptation and clinical progression of Cryptococcus neoformans infection from the lungs to systemic dissemination.

Cryptococcus neoformans is a prime example of accidental virulence, repurposing environmental survival strategies to persist within the hostile milieu of the human host. This review synthesizes recent advances in systems biology and infection modeling to chart the pathogen's dissemination from the lungs to distal organs along the anatomic route of infection. Upon inhalation, mammalian temperature and elevated CO2 act as host-entry signals that engage the thermotolerance and stress-response circuitry required for survival at 37°C. In the lung, capsule expansion, melanization, and trehalose-supp

Non-uniform growth probability of Mycobacterium tuberculosis across inoculum densities partially restored by culture filtrate: Implications for supplemented growth assays.

Differentially culturable (DC) Mycobacterium tuberculosis (Mtb) phenotypes reduce the sensitivity of sputum culture and may be associated with adverse treatment outcomes in tuberculosis patients. Detecting and quantifying DC Mtb in clinical samples remains an important research objective; current approaches rely on culture filtrate (CF) supplemented growth assays that assume CF selectively resuscitates DC Mtb rather than broadly stimulating growth from low density inocula - an untested assumption for Mtb. We performed a half-logarithmic dilution series of Mtb from approximately 10,000-0 total

Evaluation of Indigenous Bacillus Strains from Asian Fermented Foods for Probiotic Properties.

Bacillus species hold particular importance due to their versatile enzymatic repertoire and ability to synthesize diverse bioactive metabolites. In this study, two fermented food-derived strains, Bacillus siamensis BB3 (from douchi) and Bacillus velezensis TMA10 (from tapai) were evaluated for their probiotic, anti-microbial and functional potentials. Both strains exhibited desirable probiotic characteristics, including desirable tolerance to simulated gastric and intestinal conditions, with BB3 showing greater acid tolerance than TMA10. Safety assessments confirmed the absence of hemolytic ac

Fall-like acclimation alters mitochondrial function and substrate use in freeze-tolerant crickets.

Acclimation to low temperatures in insects involves complex metabolic adjustments to ensure survival. Notably, cryoprotectants including glycerol, trehalose and proline can accumulate in insects to promote freeze tolerance. Interestingly, these metabolites can also be converted to metabolic fuels sustaining mitochondrial ATP production. This study investigates the metabolic shifts in the freeze-tolerant cricket Gryllus veletis during fall-like acclimation, focusing on mitochondrial function and its possible link with cryoprotectant accumulation. We hypothesized that fall-like acclimation promo

Exploring chitin metabolite profiles and sensitivity differences in Collembola species exposed to teflubenzuron.

Effective environmental risk assessments of chemical plant protection products, such as benzoylurea pesticides, are crucial for safeguarding ecosystems. These pesticides, including teflubenzuron, target chitin synthesis in arthropods but also pose risks to non-target soil fauna like Collembola, which play essential roles in decomposition and nutrient cycling. This study combines traditional toxicity tests with a metabolomic approach to examine the interspecies specific sensitivity of three Collembola species - Sinella curviseta, Ceratophysella denticulata, and Folsomia candida - to teflubenzur

Elevated atmospheric CO2-induced reprogramming leads to decreased seed protein and nutritional quality in forest trees.

While the global increases in atmospheric CO2 levels have had a beneficial effect on plant growth, the negative impacts of this CO2 fertilization effect on seed quality are often overlooked. Using data from acorns produced by mature oak (Quercus robur) trees in the eighth year of eCO2, we present evidence of negative consequences for seed quality. The acorns produced by the near-200-yr-old oak trees under eCO2 at the free air carbon dioxide enrichment facility at the Birmingham Institute for Forest Research (BIFoR) had higher phytate levels but decreased protein content.
