Effects of graphene oxide and graphene quantum dots on enhancing CPP-ACP anti-caries ability of enamel lesion in a biofilm-challenged environment.

OBJECTIVE: To investigate the anticaries effects of graphene oxide (GO) and graphene quantum dots (GQDs) combined with casein phosphopeptide-amorphous calcium phosphate (CPP-ACP) on enamel in a biofilm-challenged environment. MATERIAL AND METHODS: GO and GQDs were synthesised using citric acid. The antibiofilm and biofilm inhibition effects for Streptococcus mutans were evaluated by scanning electron microscopy (SEM), confocal laser scanning microscopy (CLSM), and colony-forming units (CFU). Remineralisation ability was determined by assessing mineral loss, calcium-to-phosphorus ratio, and su

Immobilized laccase on chitosan-clay beads: an efficient system for synthetic dye removal.

The increasing release of synthetic dyes into aquatic environments has caused severe ecotoxicological impacts. Conventional treatment methods have shown limited efficiency, whereas enzymatic catalysis employing laccase has emerged as a sustainable alternative for dye removal. Here, we report a non-toxic immobilization strategy in which citric acid is used as the sole cross-linking agent to prepare chitosan-montmorillonite beads under mild heating, enabling efficient laccase immobilization without chemical activating agents. CuSO₄-assisted activation was employed to further enhance enzymatic pe

An Optimized and Systematically Integrated Expression, Extraction, and Purification Strategy Enabling Efficient Production of TCR-Like Antibodies in Escherichia coli.

Efficient bacterial production of recombinant antibody fragments is critical for structural characterization and subsequent functional studies. In this study, a systematically optimized workflow was developed for the expression, extraction, and purification of soluble T-cell receptor (TCR)-like antibody fragments in Escherichia coli (E. coli). Two expression hosts, the K-12-derived HB2151 strain and the B-lineage BL21(DE3) pLysS strain, were comparatively evaluated. Although both strains supported recombinant expression, BL21(DE3) pLysS produced higher levels of soluble protein. However, conve

Influence of the relative enamel abrasivity of toothpastes (REA value) on the wear of eroded enamel.

Eroded enamel is more susceptible to abrasion than healthy one. The present study aims to determine the impact of toothpaste abrasiveness (REA-value) and brushing force on enamel wear after erosion. 140 enamel samples from bovine teeth were divided into seven groups (N = 20). Each group was assigned a different toothpaste with varying levels of abrasiveness (groups 1+2: Candida Parodin (REA 2), groups 3+4: Candida Multicare (REA 9), and groups 5+6: Candida White Micro Crystals (REA 19)). The samples were first exposed to a citric acid solution (pH 2.60; 2 min) to simulate an erosive attack and

Sengers Syndrome-Associated Mitochondrial Acylglycerol Kinase Is a Subunit of the Human TIM22 Protein Import Complex.

Acylglycerol kinase (AGK) is a mitochondrial lipid kinase that catalyzes the phosphorylation of monoacylglycerol and diacylglycerol to lysophosphatidic acid and phosphatidic acid, respectively. Mutations in AGK cause Sengers syndrome, which is characterized by congenital cataracts, hypertrophic cardiomyopathy, skeletal myopathy, exercise intolerance, and lactic acidosis. Here we identified AGK as a subunit of the mitochondrial TIM22 protein import complex. We show that AGK functions in a kinase-independent manner to maintain the integrity of the TIM22 complex, where it facilitates the import a

Succination is Increased on Select Proteins in the Brainstem of the NADH dehydrogenase (ubiquinone) Fe-S protein 4 (Ndufs4) Knockout Mouse, a Model of Leigh Syndrome.

Elevated fumarate concentrations as a result of Krebs cycle inhibition lead to increases in protein succination, an irreversible post-translational modification that occurs when fumarate reacts with cysteine residues to generate S-(2-succino)cysteine (2SC). Metabolic events that reduce NADH re-oxidation can block Krebs cycle activity; therefore we hypothesized that oxidative phosphorylation deficiencies, such as those observed in some mitochondrial diseases, would also lead to increased protein succination. Using the Ndufs4 knockout (Ndufs4 KO) mouse, a model of Leigh syndrome, we demonstrate

Biotin deprivation impairs mitochondrial structure and function and has implications for inherited metabolic disorders.

Certain inborn errors of metabolism result from deficiencies in biotin containing enzymes. These disorders are mimicked by dietary absence or insufficiency of biotin, ATP deficit being a major effect, whose responsible mechanisms have not been thoroughly studied. Here we show that in rats and cultured cells it is the result of reduced TCA cycle flow, partly due to deficient anaplerotic biotin-dependent pyruvate carboxylase. This is accompanied by diminished flow through the electron transport chain, augmented by deficient cytochrome c oxidase (complex IV) activity with decreased cytochromes and

A minor catalase/peroxidase from Burkholderia cenocepacia is required for normal aconitase activity.

The opportunistic bacterium Burkholderia cenocepacia C5424 contains two catalase/peroxidase genes, katA and katB. To investigate the functions of these genes, katA and katB mutants were generated by targeted integration of suicide plasmids into the katA and katB genes. The catalase/peroxidase activity of the katA mutant was not affected as compared with that of the parental strain, while no catalase/peroxidase activity was detected in the katB mutant. However, the katA mutant displayed reduced resistance to hydrogen peroxide under iron limitation, while the katB mutant showed hypersensitivity

Glucose production pathways by 2H and 13C NMR in patients with HIV-associated lipoatrophy.

Patients with HIV taking protease inhibitors were selected for the presence (five subjects) or absence (five subjects) of lipoatrophy. Following an overnight fast, subjects were given oral (2)H(2)O in divided doses (5 mL/kg body water), [U-(13)C(3)] propionate (10 mg/kg), and acetaminophen (1000 mg). Glucose (from plasma) or acetaminophen glucuronide (from urine) were converted to monoacetone glucose for (2)H NMR and (13)C NMR analysis. The fraction of plasma glucose derived from gluconeogenesis was not significantly different between groups. However, flux from glycerol into gluconeogenesis re

Chlamydia trachomatis genes whose products are related to energy metabolism are expressed differentially in active vs. persistent infection.

The Chlamydia trachomatis genome encodes glycolysis and pentose phosphate pathway enzymes, two ATP/ADP exchange proteins, and other energy transduction-related components. We asked if and when chlamydial genes specifying products related to energy transduction are expressed during active vs. persistent infection in in vitro models and in synovia from Chlamydia-associated arthritis patients. Hep-2 cells infected with K serovar were harvested from 0-48 h post-infection (active infection). Human monocytes identically infected were harvested at 1, 2, 3, 5 days post-infection (persistent). Renal metabolism of citrate.

Our studies on renal handling of citrate have shown that: (1) citrate enters renal tubular cells from luminal fluid (reabsorption) and peritubular blood; (2) reabsorption becomes maximal, i.e., Tm-limited, at filtered loads 7 to 8 times the normal; (3) administration of malate stimulates net renal production of citrate, leading to release into urine (net secretion) and into peritubular blood; (4) acute metabolic alkalosis, induced while plasma citrate levels are above normal, depressess net citrate reabsorption, stimulates citrate release into peritubular blood and abolishes overall renal upta

The influence of thiamine deficiency on porphyrin synthesis and porphyrin proteins.

1. Studies carried out in cell-free rat-liver extracts showed an inhibitory effect of thiamine deficiency on porphyrin synthesis. 2. Extracts of normal and thiamine-deficient rat livers were incubated with the cofactors and substrates of the thiamine pyrophosphate-dependent enzyme, 2-oxoglutarate glyoxylate carboligase. There was a marked inhibition of porphyrin synthesis when the substances were added either individually or together. 3. Thiamine deficiency resulted in a decrease of haemoglobin concentration and catalase activity whereas a rise in tryptophanpyrrolase activity and cytochrome b5

Elucidation of Sulforaphane-Mediated Effects on the Cellular Human Metabolome Using Metabolic Profiling.

Sulforaphane (SFN) is an isothiocyanate derived from glucoraphanin, which occurs in broccoli. Several human health-promoting effects are attributed to the consumption of cruciferous vegetables or food supplements containing SFN. Its described cancer-preventive, chemoprotective, and antioxidant properties made SFN an increasingly important research topic. The antioxidant properties have previously been connected to stimulation of the Nuclear factor erythroid-2-related factor 2 (Nrf2)/Kelch-like ECH-associated protein 1 (Keap1) signaling pathway. However, the global effects of SFN on the primary

Canagliflozin synergises with serine restriction mediating anti-leukaemic effects in T-cell acute lymphoblastic leukaemia.

T-cell acute lymphoblastic leukaemia (T-ALL) is a haematological malignancy commonly driven by NOTCH1 activating mutations. A concomitant feature associated with NOTCH1 mutations is heightened oxidative metabolism enabling the exponential proliferation of T-ALL blasts. As such, targeting mitochondrial metabolism in T-ALL is an attractive therapeutic avenue. Related to this, canagliflozin (cana), is an FDA-approved sodium glucose co-transporter 2 inhibitor with known off-target effects on complex I and glutamate dehydrogenase, but its potential anti-leukaemic effects remain unexplored. Here, we

Regulation of airway fumarate by host and pathogen promotes Staphylococcus aureus pneumonia.

Staphylococcus aureus is a leading cause of healthcare-associated pneumonia, contributing significantly to morbidity and mortality worldwide. As a ubiquitous colonizer of the upper respiratory tract, S. aureus must undergo substantial metabolic adaptation to achieve persistent infection in the distinctive microenvironment of the lung. We observed that fumC, which encodes the enzyme that converts fumarate to malate, is highly conserved with low mutation rates in S. aureus isolates from chronic lung infections. Fumarate, a pro-inflammatory metabolite produced by macrophages during infection, is

Metabolic rewiring promotes anti-inflammatory effects of glucocorticoids.

Glucocorticoids represent the mainstay of therapy for a broad spectrum of immune-mediated inflammatory diseases. However, the molecular mechanisms underlying their anti-inflammatory mode of action have remained incompletely understood1. Here we show that the anti-inflammatory properties of glucocorticoids involve reprogramming of the mitochondrial metabolism of macrophages, resulting in increased and sustained production of the anti-inflammatory metabolite itaconate and consequent inhibition of the inflammatory response. The glucocorticoid receptor interacts with parts of the pyruvate dehydrog

The ancestral stringent response potentiator, DksA has been adapted throughout Salmonella evolution to orchestrate the expression of metabolic, motility, and virulence pathways.

DksA is a conserved RNA polymerase-binding protein known to play a key role in the stringent response of proteobacteria species, including many gastrointestinal pathogens. Here, we used RNA-sequencing of Escherichia coli, Salmonella bongori and Salmonella enterica serovar Typhimurium, together with phenotypic comparison to study changes in the DksA regulon, during Salmonella evolution. Comparative RNA-sequencing showed that under non-starved conditions, DksA controls the expression of 25%, 15%, and 20% of the E. coli, S. bongori, and S. enterica genes, respectively, indicating that DksA is a p

Switching obese mothers to a healthy diet improves fetal hypoxemia, hepatic metabolites, and lipotoxicity in non-human primates.

OBJECTIVE: Non-alcoholic fatty liver disease (NAFLD) risk begins in utero in offspring of obese mothers. A critical unmet need in this field is to understand the pathways and biomarkers underlying fetal hepatic lipotoxicity and whether maternal dietary intervention during pregnancy is an effective countermeasure. METHODS: We utilized a well-established non-human primate model of chronic, maternal, Western-style diet induced obesity (OB-WSD) compared with mothers on a healthy control diet (CON) or a subset of OB-WSD mothers switched to the CON diet (diet reversal; OB-DR) prior to and for the d
