Integrative analysis of plasma small-molecule and gut-microbiome markers of sarcopenia in a pilot study within an Indian cohort.

Sarcopenia, the age-associated decline in muscle mass and strength, is influenced by metabolic, inflammatory, and microbiome-related factors. However, integrative analyses combining these dimensions remain limited. This study applies a multi-omics workflow to identify plasma metabolite, lipid, and microbiome signatures linked to sarcopenia in older adults. Forty community-dwelling adults aged 60–87 years were classified as sarcopenic (n = 15) or non-sarcopenic (n = 25) using EWGSOP2 criteria, incorporating dominant hand grip strength (DHGS), chair rise time, psoas muscle cross-sectional area (

Divergent transcriptional programs and constrained metabolic states reveal selective metabolic dependencies in chemoresistant luminal breast cancer.

Aim: Resistance to neoadjuvant chemotherapy remains a major challenge in hormone receptor-positive/human epidermal growth factor receptor 2-negative (HR+/HER2-) breast cancer (BC). Although anthracyclines and taxanes constitute the standard sequential regimen, the metabolic adaptations accompanying resistance to these agents remain poorly defined. We investigated whether chemoresistance is associated with shared or drug-specific metabolic alterations. Methods: Parental MCF-7 and ZR-75-1 cells and their anthracycline-, and taxane-resistant derivatives, were characterized through multi-omics, Se

Native dietary fiber-bound phenolics complex from Lentinula edodes alleviate colitis through gut microbiota and metabolic remodeling.

Dietary fiber-bound phenolics are naturally occurring complexes in foods, yet the functional significance of preserving their native dietary fiber-phenolic assembly remains poorly understood. In this study, using a removal-reconstitution strategy, we compared native soluble dietary fiber-bound phenolics (SDF-BP) from Lentinula edodes with its bound phenolic-removed fraction (SDF-PR), released bound phenolics (RSBP), and their physical mixture (SDF-PR + RSBP). Three isomers of tri-p-coumaroyl spermidine were identified in the bound phenolic for the first time. Bound phenolics were covalently li

Next-Gen Point-of-Care Tool for Ultra-Sensitive Detection of Urinary Spermine for Prostate Cancer Diagnosis.

Prostate cancer (PCa), the second most common cancer in men, demands effective early detection strategies. Elevated spermine levels in the prostate tissue contrast with decreased urinary concentrations in PCa patients. Here, we present a novel sensing platform combining differential pulse voltammetry and an extended gate field-effect transistor (EGFET) with a molecularly imprinted polymer|molecular imprinting (MIP) nanofilm for selective and sensitive spermine detection. Key advancements include successfully constructing and characterizing a pseudoreference electrode and a precisely engineered

Resveratrol alleviates IBD-associated neuropsychiatric comorbidities via microbiota-dependent arginine metabolism reprogramming and microglial M2 polarization through gut-brain axis.

Inflammatory bowel disease (IBD) is intricately linked to neuropsychiatric comorbidities through gut-brain axis dysregulation. This study demonstrates that resveratrol (RSV), a natural polyphenol, alleviates DSS-induced colitis-associated anxiety and depression by reprogramming the microbiota─metabolite-barrier network. RSV (100 mg/kg/day) ameliorated DSS-associated anxiety-like behaviors in open field tests (peripheral zone time ↓12.6%, P< 0.0001) and depression-like phenotypes (TST immobility ↓31.0%, P = 0.0004). It restored colonic barrier integrity via ZO-1 mRNA upregulation (↑80.4%, P < 0

Comparative analysis of bile metabolic profile in patients with biliary obstruction complicated by Clonorchis sinensis infection.

BACKGROUND: Clonorchiasis is an important foodborne parasitic disease. However, eggs of Clonorchis sinensis (C. sinensis) cannot be detected in feces during biliary obstruction. Moreover, many diseases can cause biliary obstruction, such as gallstones, adenocarcinoma, cholangiocarcinoma and Ascaris lumbricoides infection. Therefore, it is of great significance to distinguish between patients of biliary obstruction and biliary obstruction with C. sinensis infection. METHODS: A total of 48 biliary obstruction patients were enrolled, including 23 infected with C. sinensis (C. sinensis) (OB+C.s)

Loss-of-function variants in SAT1 cause X-linked childhood-onset systemic lupus erythematosus.

OBJECTIVES: Families that contain multiple siblings affected with childhood onset of systemic lupus erythematosus (SLE) likely have strong genetic predispositions. We performed whole exome sequencing (WES) to identify familial rare risk variants and to assess their effects in lupus. METHODS: Sanger sequencing validated the two ultra-rare, predicted pathogenic risk variants discovered by WES and identified additional variants in 562 additional patients with SLE. Effects of a splice site variant and a frameshift variant were assessed using a Minigene assay and CRISPR/Cas9-mediated knock-in (KI)

Multiple polyamine transport pathways in cultured pulmonary artery smooth muscle cells: regulation by hypoxia.

The polyamines putrescine (PUT), spermidine (SPD), and spermine (SPM) are a family of low molecular weight organic cations that play essential intracellular regulatory roles in cell growth and differentiation. Consistent with this important function, increases in cellular polyamine contents are necessary for a variety of physiologic and pathologic events in the lung, including development of hypertensive pulmonary vascular disease secondary to chronic alveolar hypoxia. In intact rat lungs, hypoxia depresses ornithine decarboxylase activity, the initial rate-limiting enzyme in de novo polyamine

Production and characterization of monoclonal antibodies against the polyamine, spermine: immunocytochemical localization in rat tissues.

Two monoclonal antibodies of types IgG2b and IgG2a, anti-spermine-(Spm)-1 (ASPM-1) and anti-Spm-2 (ASPM-2) respectively were found among five clones of murine monoclonal antibodies, which were raised against Spm conjugated with bovine serum albumin via the cross-linker N-(gamma-maleimidobutyryloxy) succinimide (GMBS). Antibody specificity was evaluated by a recently developed ELISA binding test, and led to the study of tissue sections by immunocytochemistry (ICC). ASPM-1 showed exclusive immunoreactivity with Spm, with the exception of a negligible cross-reactivity (2.0%) with spermidine (Spd)

Regulatory and antiproliferative effects of N-alkylated polyamine analogues in human and hamster pancreatic adenocarcinoma cell lines.

N-Alkylated polyamine analogues have been shown to exert antiproliferative effects in several tumor models, with the bis-ethyl derivatives exerting the greatest suppression of polyamines by virtue of down-regulation of the polyamine biosynthetic enzymes. Pancreatic adenocarcinoma presents a challenge both clinically and experimentally due to its inherent resistance to conventional therapy, which results in its having the worst 5-year survival rate of all cancers. We have previously shown that N1,N12-bis(ethyl)spermine (BESPM) is much more potent than the polyamine enzyme inhibitor alpha-difluo

Chemoprevention of oral carcinogenesis by DL-alpha-difluoromethylornithine, an ornithine decarboxylase inhibitor: dose-dependent reduction in 4-nitroquinoline 1-oxide-induced tongue neoplasms in rats.

The modifying effect of three doses of DL-alpha-difluoromethylornithine (DFMO) given p.o. during the post-initiation phase of tongue carcinogenesis initiated by 4-nitroquinoline 1-oxide (4-NQO) was studied in male ACI/N rats. Animals were given 4-NQO at 20 ppm for 8 weeks in the drinking water to induce tongue neoplasms. One week after the stop of 4-NQO treatment, rats were transferred to the drinking water containing DFMO at concentrations of 100, 1000, and 2000 ppm for 25 weeks. The other groups consisted of rats given 2000 ppm DFMO alone or untreated rats. Thirty-four weeks after the start

A tryparedoxin-dependent peroxidase protects African trypanosomes from membrane damage.

Hydroperoxide detoxification in African trypanosomes is achieved by 2-Cys-peroxiredoxin (TXNPx)- and non-selenium glutathione peroxidase (Px)-type enzymes which both obtain their reducing equivalents from the unique trypanothione/tryparedoxin system. Previous RNA interference approaches revealed that the cytosolic TXNPx and the Px-type enzymes are essential for Trypanosoma brucei. Because of partially overlapping in vitro substrate specificities and subcellular localisation the physiological function of the individual enzymes was not yet clear. As shown here, TXNPx and Px are expressed at comp

Isolation and characterization of Novosphingobium oxfordense sp. nov. and Novosphingobium mississippiense sp. nov. from soil, with LC-MS/MS and genome-based investigation of their glycosphingolipid production.

Members of the genus Novosphingobium are well known for the biosynthesis of glycosphingolipids. Here, we are reporting two recently discovered species belonging to this genus, which were isolated from a potted tomato plant in Oxford, Mississippi, USA. The cells of both strains grew aerobically, were rod-shaped, and formed yellow colonies. According to the 16S rRNA gene and whole genome-based phylogeny, both strains have the highest similarity to N. clariflavum 164 T . Genomes of both strains had a G + C content of 64 mol %, and assemblies with four circular contigs: two plasmids and two larger

High-dose DFMO alters protein translation in neuroblastoma.

DFMO has been studied as a cancer therapeutic at doses ranging from 500 to 9,000 mg/m2/day. Lower doses are favored for cancer prevention studies while higher doses, often with chemotherapy, are studied in refractory cancers. DFMO inhibits the rate-limiting enzyme in polyamine synthesis, ornithine decarboxylase (ODC), an oncogene transcriptionally regulated by MYC. MYC genes are the principal oncogenic drivers of neuroblastoma, and ODC1 is co-amplified in a subset with dismal outcome, so DFMO is a rational therapeutic candidate. Low-dose DFMO has now been FDA-approved for high-risk patients th

Bleomycin-induced lung fibrosis and dysfunction is exacerbated by G6PD deficiency.

Pulmonary fibrosis (PF) is a major cause of morbidity and mortality. Although increased oxidative stress and altered metabolism are implicated in PF pathobiology, our knowledge regarding the contribution of the glucose metabolism to the synthesis of extracellular matrix (ECM) is still incomplete. Therefore, our objective was to determine altered metabolic pathways that contribute to bleomycin (BLM; 5 mg/kg) sulfate-induced PF in rats. We determined the effects of nebulized BLM on PF in CRISPR-edited rats expressing glucose-6-phosphate dehydrogenase (G6PD) variant (S188F; G6PDS188F) and their w

Changes of Gut Microbiota and Its Correlation With Short Chain Fatty Acids and Bioamine in Piglets at the Early Growth Stage.

The change characteristics of intestinal microbial succession and the correlation with the production of two important types of bacterial metabolites (short chain fatty acids and bioamine) in piglets during the early stage were fully explored in this study. Six piglets from different litters with the same birth time were selected, weighted and euthanized at 1, 7, 14, 21, 28, 35, and 42 days of age. During this stage, the piglets grew quickly with gradual increases in blood levels of growth hormone and insulin, and in the intestinal developmental index and immunity. 16s rRNA analysis indicated

Ozone-induced changes in the serum metabolome: Role of the microbiome.

Ozone is an asthma trigger. In mice, the gut microbiome contributes to ozone-induced airway hyperresponsiveness, a defining feature of asthma, but the mechanistic basis for the role of the gut microbiome has not been established. Gut bacteria can affect the function of distal organs by generating metabolites that enter the blood and circulate systemically. We hypothesized that global metabolomic profiling of serum collected from ozone exposed mice could be used to identify metabolites contributing to the role of the microbiome in ozone-induced airway hyperresponsiveness. Mice were treated for

Anopheles aquasalis transcriptome reveals autophagic responses to Plasmodium vivax midgut invasion.

BACKGROUND: Elimination of malaria depends on mastering transmission and understanding the biological basis of Plasmodium infection in the vector. The first mosquito organ to interact with the parasite is the midgut and its transcriptomic characterization during infection can reveal effective antiplasmodial responses able to limit the survival of the parasite. The vector response to Plasmodium vivax is not fully characterized, and its specificities when compared with other malaria parasites can be of fundamental interest for specific control measures. METHODS: Experimental infections were per
