Quercetin in men with category III chronic prostatitis: a preliminary prospective, double-blind, placebo-controlled trial.

OBJECTIVES: The National Institutes of Health (NIH) category III chronic prostatitis syndromes (nonbacterial chronic prostatitis and prostatodynia) are common disorders with few effective therapies. Bioflavonoids have recently been shown in an open-label study to improve the symptoms of these disorders in a significant proportion of men. The aim of this study was to confirm these findings in a prospective randomized, double-blind, placebo-controlled trial. METHODS: Thirty men with category IIIa and IIIb chronic pelvic pain syndrome were randomized in a double-blind fashion to receive either p

Peptide aldehyde inhibitors of cathepsin K inhibit bone resorption both in vitro and in vivo.

We have shown previously that cathepsin K, a recently identified member of the papain superfamily of cysteine proteases, is expressed selectively in osteoclasts and is the predominant cysteine protease in these cells. Based upon its abundant cell type-selective expression, potent endoprotease activity at low pH and cellular localization at the bone interface, cathepsin K has been proposed to play a specialized role in osteoclast-mediated bone resorption. In this study, we evaluated a series of peptide aldehydes and demonstrated that they are potent cathepsin K inhibitors. These compounds inhib

Contribution of cutinase serine 42 side chain to the stabilization of the oxyanion transition state.

Cutinase from the fungus Fusarium solani pisi is a lipolytic enzyme able to hydrolyze both aggregated and soluble substrates. It therefore provides a powerful tool for probing the mechanisms underlying lipid hydrolysis. Lipolytic enzymes have a catalytic machinery similar to those present in serine proteinases. It is characterized by the triad Ser, His, and Asp (Glu) residues, by an oxyanion binding site that stabilizes the transition state via hydrogen bonds with two main chain amide groups, and possibly by other determinants. It has been suggested on the basis of a covalently bond inhibitor

[Detection and analysis of aldehyde fuchsin positive fibers in the connective tissue of the human oviductal mucosa. I. Preliminary investigations and demonstration of the morphological substratum (author's transl)].

Investigating the biomorphosis of the human uterine tube, it was possible to detect aldehyde fuchsin positive fibers in the lamina propria mucosae. These fibers can be regarded as sulphur containing scleroproteins relating to the histochemical findings of oxidation, thiosulphation, methylation, sensitivity to pepsin and papain, respectively. For the visualization of the morphological substratum it was necessary to treat the fixed or fresh unfixed tissue chemically by oxidation and thiosulfation before the slices were stained with aldehyde fuchsin. Among several oxidantia examined in preliminar

Ionic liquids and lysosomotropic detergents as inhibitors of the SARS-CoV-2 main protease: QSAR modeling, synthesis and biological testing.

SARS-CoV-2 infection is highly contagious, prompting the World Health Organization to classify it as a global public health emergency. The virus has numerous potential hosts, which complicates efforts for effective prevention, diagnosis, and treatment. Consequently, the development of innovative antiviral agents targeting SARS-CoV-2 is essential to mitigate complications and reduce mortality rates associated with COVID-19. The main protease (Mpro), also known as 3CLpro, is a key target for drug development because of its critical role in processing viral polyproteins and its evolutionary conse

A quantitative high-throughput screening pipeline to identify small molecule inhibitors of Chikungunya nsP2 protease.

Chikungunya virus (CHIKV) is a mosquito-borne RNA virus that has emerged as one of the most important global arboviral threats in the last decade. Although the first CHIKV vaccine has recently been FDA approved for use in healthy adults at increased risk, to date, there are no available antiviral drugs for CHIKV infection. CHIKV nsP2 protease plays a crucial role in the processing of the viral polypeptide precursor to release enzymes required for viral replication, thus making it a promising drug target for antiviral discovery. Here, we established a high-throughput pipeline to identify small

Neutrophil elastase activates macrophage calpain as a mechanism for phagocytic failure.

Neutrophil elastase (NE), elevated in the cystic fibrosis (CF) airway, causes macrophage phagocytic failure. We previously reported that NE increases the release of protease calcium ion-dependent papain-like cysteine protease-2 (Calpain-2) in macrophages. We hypothesized that NE mediates macrophage failure through activation of Calpains. We demonstrate that Calpain inhibition rescued NE-induced macrophage phagocytic failure in murine alveolar macrophages in both cftr-null and wild-type genotypes. We then sought to determine how NE regulates Calpain-2. Human monocyte-derived macrophages (hMDMs)

Docking Analysis of Some Bioactive Compounds from Traditional Plants against SARS-CoV-2 Target Proteins.

COVID-19 is still a global pandemic that has not been stopped. Many traditional medicines have been demonstrated to be incredibly helpful for treating COVID-19 patients while fighting the disease worldwide. We introduced 10 bioactive compounds derived from traditional medicinal plants and assessed their potential for inhibiting viral spike protein (S-protein), Papain-like protease (PLpro), and RNA dependent RNA polymerase (RdRp) using molecular docking protocols where we simulate the inhibitors bound to target proteins in various poses and at different known binding sites using Autodock versio

Characteristic features of the SERA multigene family in the malaria parasite.

Serine repeat antigen (SERA) is conserved among species of the genus Plasmodium. Sera genes form a multigene family and are generally tandemly clustered on a single chromosome. Although all Plasmodium species encode multiple sera genes, the number varies between species. Among species, the members share similar sequences and gene organization. SERA possess a central papain-like cysteine protease domain, however, in some members, the active site cysteine residue is substituted with a serine. Recent studies implicate this gene family in a number of aspects in parasite biology and induction of pr

Characterization and differential expression of cathepsin L3 alleles from Fasciola hepatica.

Fasciola hepatica infections cause significant global problems in veterinary and human medicine, including causing huge losses in cattle and sheep production. F. hepatica host infection is a multistage process and flukes express papain-like cysteine proteases, termed cathepsins, which play pivotal roles in virulence through host entry, tissue migration and immune evasion. Expression of these proteases is developmentally regulated. Recent studies indicate that excystment of infective larvae is dependent on cysteine proteases and together FhCL3 and FhCB account for over 80% of total protease act
