Quantitative cytochemistry of glycogen in blood cells. Methods and clinical application.

Quantitative glycogen determinations can be made in single blood and bone marrow cells, using microspectrophotometry or microfluorometry after staining with variants of the periodic acid--Schiff (PAS) reaction. These PAS variant reactions generally do not indicate the presence of non-glycogen PAS-positive substances, known to be prevalent in various hematopoietic cells, possibly due to masking of reactive groups. The specificity of the reaction in blood cells was ascertained by alpha-amylase digestion, which removed more than 95% of the PAS-positive material. Calibration of the PAS reaction wa

Ewing sarcoma breakpoint region 1 (EWSR1) protein binds to nonstructural protein 2 and non-coding regions of chikungunya virus genome to inhibit virus replication.

Chikungunya virus (CHIKV) is an alphavirus with a single-stranded RNA genome. Conserved sequence elements within the 5' and 3' non-coding regions (NCRs) of the CHIKV genome interact with host proteins to regulate viral RNA replication. Electrophoretic mobility shift assays (EMSAs) and mass spectrometry identified several host RNA-binding proteins that associate with these conserved NCR elements, among which EWSR1 uniquely bound the 3' ends of both the plus-sense genomic RNA and the minus-sense replication intermediate RNA. EMSA and biolayer interferometry (BLI) demonstrated that EWSR1 binds st

Genetic determinants of receptor-binding preference and zoonotic potential of H9N2 avian influenza viruses.

Receptor recognition and binding is the first step of viral infection and a key determinant of host specificity. The inability of avian influenza viruses to effectively bind human-like sialylated receptors is a major impediment to their efficient transmission in humans and pandemic capacity. Influenza H9N2 viruses are endemic in poultry across Asia and parts of Africa where they occasionally infect humans and are therefore considered viruses with zoonotic potential. We previously described H9N2 viruses, including several isolated from human zoonotic cases, showing a preference for human-like r

Methylmalonate Overload Despite Glycemic Control Drives Diabetic Heart Damage.

BACKGROUND: Despite optimal glycemic control, the heart failure burden remains substantial in diabetic patients. Metabolic remodeling is involved in this process, yet our current understanding is still in its infancy. Methylmalonic acid (MMA) is conventionally viewed as a marker of cobalamin (Cbl) deficiency. Paradoxically, MMA elevation-related cardiovascular mortality is more pronounced in diabetic patients with normal or high Cbl levels. This study investigated the mechanisms and translational significance of this contradictory MMA accumulation in the diabetic heart. METHODS: We analyzed s

Annexin A1-Targeted d-Peptide-Monomethyl Auristatin E Conjugate Enhanced Antitumor Effect of Monomethyl Auristatin E for Chemotherapy of Prostate Cancer.

BACKGROUND: N-terminus 15 amino acid (MC16) of Annexin A1 (ANXA1) is an effective therapeutic target for tumors and tumor vasculature as it specifically localizes to the cell surface of tumor cells and tumor vascular endothelial cells. We identified the d-type peptide (hpnevrs, dhp7) by mirror-image phage display targeting l-type MC16 and tested the hypothesis that intravenously injected c(vcMMAE)dhp7 eradicates prostate tumor in mice with less adverse side effects than that of mc-Val-Cit-PAB-monomethyl auristatin (vcMMAE) alone. METHODS: The binding affinity of dhp7 to MC16 of ANXA1 was anal

CD24 as a Novel Radiopharmaceutical Target for Hepatocellular Carcinoma.

Hepatocellular carcinoma (HCC) is a leading cause of cancer-related mortality worldwide. After the use of liver-directed therapies, it is challenging to differentiate between nonviable tumor and viable residual or recurrent disease using conventional imaging techniques. Targeted radiopharmaceutical imaging agents with specificity for HCC-selective molecules may address this unmet need. CD24, a glycosylated plasma membrane protein, is overexpressed in HCC. Here, we describe a CD24-targeted antibody-based PET (immuno-PET) tracer for the noninvasive detection of CD24-positive (CD24+) tumors. Meth

Increased Siglec-9/Siglec-9L interactions on NK cells predict poor HCC prognosis and present a targetable checkpoint for immunotherapy.

BACKGROUND & AIMS: Natural killer (NK) cell-based anti-hepatocellular carcinoma (HCC) therapy is an increasingly attractive approach that warrants further study. Siglec-9 interacts with its ligand (Siglec-9L) and restrains NK cell functions, suggesting it is a potential therapeutic target. However, in situ Siglec-9/Siglec-9L interactions in HCC have not been reported, and a relevant interventional strategy is lacking. Herein, we aim to illustrate Siglec-9/Siglec-9L-mediated cell sociology and identify small-molecule inhibitors targeting Siglec-9 that could improve the efficacy of NK cell-based

Emmetropisation under continuous but non-constant light in chicks.

It has been suggested that ambient lighting at night influences eye growth and might play a causal role in human myopia. To test this hypothesis, we reared newly hatched chicks under 12 hr light-dark or light-dim cycles with a light phase intensity of 1500 microW/cm(2) and variable dim phase intensities between 0.01 and 500 microW/cm(2). Other chicks were reared under constant light conditions with intensities between 1 and 1500 microW/cm(2). After three weeks, the chicks were examined by refractometry, ultrasound and caliper measurements of enucleated eyes. To relate ocular parameters with a

Effects of N-Glycosylation on the Structure, Function, and Stability of a Plant-Made Fc-Fusion Anthrax Decoy Protein.

Protein N-glycosylation is an important post-translational modification and has influences on a variety of biological processes at the cellular and molecular level, making glycosylation a major study aspect for glycoprotein-based therapeutics. To achieve a comprehensive understanding on how N-glycosylation impacts protein properties, an Fc-fusion anthrax decoy protein, viz rCMG2-Fc, was expressed in Nicotiana benthamiana plant with three types of N-glycosylation profiles. Three variants were produced by targeting protein to plant apoplast (APO), endoplasmic reticulum (ER) or removing the N-gly

High-affinity chicken scFv antibodies recognize recombinant VP1-CDEF' protein and native virions of feline calicivirus genotype II strain LZ2023.

Feline calicivirus (FCV) is a significant pathogen responsible for upper respiratory tract disease and oral ulceration in felids. Genetic and antigenic variation in circulating FCV strains limits the utility of current diagnostic assays, and no validated genotype II-specific antibodies exist for rapid test development. In this study, we targeted the C, D, E, and partial F regions of the major capsid protein VP1 (termed VP1-CDEF'). The recombinant VP1-CDEF' protein was expressed in a prokaryotic system and used to immunize SPF chickens. A chicken-derived phage display single-chain variable frag

Molecular basis for protection and cross-protection by human antibodies targeting the parainfluenza virus hemagglutinin-neuraminidase protein.

Human parainfluenza viruses (PIVs) are a leading cause of respiratory illness, particularly in vulnerable populations, where infection can lead to severe disease. Despite their clinical impact, there are currently no licensed vaccines or effective antiviral treatments available. PIVs have two large surface proteins, the fusion and hemagglutinin-neuraminidase (HN) proteins, both of which are targets of neutralizing antibodies. In this study, we identified and characterized two human monoclonal antibodies (mAbs), 5217-2 and 5217-9, which bind recombinant PIV3 HN protein, recognize PIV3-infected

IFN-γ PET imaging stratifies and predicts response to CD137 agonism in a syngeneic colorectal tumor model.

BACKGROUND: The secretion of interferon-γ (IFN-γ) by activated immune cells is one of the hallmarks of response to cancer immunotherapy. Hence, detection of this cytokine via imaging can help determine an activated antitumor immune response. We have previously shown that positron emission tomography (PET) imaging of IFN-γ using [89Zr]Zr-Deferoximane (DFO)-anti-IFN-γ measured changes in its intratumoral expression after immune modulation. However, assessment of its potential to predict and identify treated responders (TR) versus treated non-responders (TNR) at the early stages of treatment, bef

Development and Humanization of a Camelid-Derived Neutralizing Nanobody Targeting Botulinum Neurotoxin Type A.

PURPOSE: Botulinum neurotoxin (BoNT), produced by Clostridium botulinum, is one of the most potent neurotoxins, causing botulism, a potentially fatal paralytic disease. Among different serotypes, BoNT/A is the most potent and the primary cause of botulism. The receptor-binding domain (Hc) of BoNT is crucial for targeting host cell receptors, making it a key therapeutic target. This study aims to develop neutralizing antibodies against BoNT/A by isolating camelid-derived nanobodies (Nbs) targeting BoNT/A-Hc (AHc). METHODS: A panel of Nbs specifically targeting AHc was isolated from a camelid i

Quercetin protected the gut barrier in ulcerative colitis by activating aryl hydrocarbon receptor.

BACKGROUND: Ulcerative colitis (UC) is characterized by abdominal pain and bloody diarrhoea and restoring the gut barrier is the core goal of UC treatment. Activation of aryl hydrocarbon receptor (Ahr) was reported to effectively alleviate symptoms and repair the gut barrier damage. Neutrophil extracellular traps (NETs) have been recognized as potential targets in the treatment of UC. Ahr activation has been found to be capable of upregulating Nqo1, thereby reducing the production of reactive oxygen species (ROS), which is important in the formation of NETs.
