The role of three calcineurin subunits and a related transcription factor (Crz1) in conidiation, multistress tolerance and virulence in Beauveria bassiana.

The eukaryotic calcineurin (CN) pathway comprising catalytic A (CnA) and regulatory B subunits (CnB) is crucial for many biological processes but functionally unexplored in entomopathogenic fungi. Here, we characterise three CN subunits (CnA1, CnA2 and CnB) and a downstream CN-responsive zinc finger transcription factor (Crz1) in Beauveria bassiana. CN-mediated phosphatase activity decreased by 16-38 % in all deletion mutants compared with wild type. Growth and conidiation were most defective in ΔcnB, which showed a large proportion of abnormally branched germlings but were less defective in Δ

Lack of protective osmolytes limits final cell density and volumetric productivity of ethanologenic Escherichia coli KO11 during xylose fermentation.

Limited cell growth and the resulting low volumetric productivity of ethanologenic Escherichia coli KO11 in mineral salts medium containing xylose have been attributed to inadequate partitioning of carbon skeletons into the synthesis of glutamate and other products derived from the citrate arm of the anaerobic tricarboxylic acid pathway. The results of nuclear magnetic resonance investigations of intracellular osmolytes under different growth conditions coupled with those of studies using genetically modified strains have confirmed and extended this hypothesis. During anaerobic growth in miner

Establishment of a monoclonal antibody directed against Gb3Cer/CD77: a useful immunochemical reagent for a differentiation marker in Burkitt's lymphoma and germinal centre B cells.

A new monoclonal antibody (TU-1) directed against the Gal alpha 1-4Gal beta 1-4Glc residue of the Gb3Cer/CD77 antigen was prepared by the hybridoma technique following immunization of mice with an emulsion composed of monophosphoryl lipid A, trehalose dimycolate, and Gb3Cer isolated from porcine erythrocytes. TU-1 showed reactivity towards Gb3Cer and lyso-Gb3Cer(Gal alpha 1-4Gal beta 1-4Glc beta 1-1'Sph), although the reactivity towards lyso-Gb3Cer was about 10-fold lower than that to Gb3Cer. But it did not react with other structurally-related glycolipids, such as LacCer (Gal beta 1-4Glc beta

Development of a highly sensitive photometric readout platform technology using SPAAC-modified HRP liposomes.

Liposome-based bioassays take advantage of liposomes' immense signaling power and easy surface modification. Photometric, electrochemical, and fluorescent detection methods dominate their application in bioassays. Here, it is studied how covalent modification of the liposome surface with proteins affects the functionality of entrapped enzymes. Horseradish peroxidase (HRP) was selected as a model for entrapment while streptavidin served as a model surface ligand. Several coupling reagents negatively affected the functionality of entrapped HRP. In the case of EDC-sNHS chemistry, membrane-permeab

Functional Characterization of Tret1-like Gene and Its Critical Role in Regulating Growth in Macrobrachium nipponense by RNA Interference.

Macrobrachium nipponense is a commercially significant freshwater prawn species in China, with larger individuals commanding higher market prices than smaller ones. Previous transcriptomic analyses have shown that a facilitated trehalose transporter gene Tret1-like (Tret1) exhibits significantly elevated expression levels in fast-growing individuals compared to their slow-growing counterparts, suggesting a potential involvement of this gene in modulating growth performance. In the present study, we explored the functional roles of Mn-Tret1 in growth regulation using reverse transcription quant

Effects of Sequential Fermentation with Saccharomyces bayanus and Lactobacillus brevis on the Metabolite Composition and Antioxidant Activity of Chinese Yam Juice.

Chinese yam (Dioscorea opposita Thunb.) is a nutrient-rich tuber with recognized health benefits, yet its application in beverage products remains limited due to processing and formulation challenges. In this study, a sequential fermentation strategy was adopted, using Saccharomyces bayanus followed by Lactobacillus brevis to enhance microbial viability and metabolic activity in Chinese yam juice. Samples were collected as an unfermented control (CY), yeast-fermented juice (SP), and sequentially fermented juice (LB). Microbial analysis showed that sequential fermentation supported high LAB via

Integrated physiological and transcriptomic measurements reveal changes in the accumulation and partitioning of storage reserves after prevention of pollination in maize.

BACKGROUND: Reduced sink demand, such as that caused by grain or ear removal, is known to accelerate senescence in maize; nonetheless, how diminished sink strength mechanistically triggers senescence and disrupts grain development remains poorly understood. In particular, the transcriptional and metabolic regulatory networks that link reduced sink demand to the onset of senescence have not been clearly defined. RESULTS: We grew the maize inbred line 08LF, which exhibits early senescence in response to grain removal, under non-pollinated (NONPOL) and normally pollinated (POL) conditions. We th

The role of urinary trehalase levels in the early diagnosis of acute kidney injury in dehydrated children.

BACKGROUND: Trehalase is a brush-border glycoprotein enzyme found in the small intestine and in renal proximal tubules, where it breaks down trehalose into glucose. We examined whether a non-invasive urine measurement of trehalase could help with earlier recognition of acute kidney injury (AKI) in children presenting with acute dehydration. METHODS: We included 80 children who were evaluated in the Pediatric Emergency Department for acute dehydration and 40 healthy controls from the general pediatric outpatient clinic. Baseline laboratory data were extracted from the hospital information syst

Syncephalastrum massiliense sp. nov. and Syncephalastrum timoneanum sp. nov. Isolated from Clinical Samples.

Mucormycosis is known to be a rare opportunistic infection caused by fungal organisms belonging to the Mucorales order, which includes the Syncephalastrum species. These moulds are rarely involved in clinical diseases and are generally seen as contaminants in clinical laboratories. However, in recent years, case reports of human infections due to Syncephalastrum have increased, especially in immunocompromised hosts. In this study, we described two new Syncephalastrum species, which were isolated from human nails and sputum samples from two different patients. We used several methods for genomi

Personalized Clostridioides difficile engraftment risk prediction and probiotic therapy assessment in the human gut.

Clostridioides difficile colonizes up to 30-40% of community-dwelling adults without causing disease. C. difficile infections (CDIs) are the leading cause of antibiotic-associated diarrhea in the U.S. and typically develop in individuals following disruption of the gut microbiota due to antibiotic or chemotherapy treatments. Further treatment of CDI with antibiotics is not always effective and can lead to antibiotic resistance and recurrent infections (rCDI). The most effective treatment for rCDI is the reestablishment of an intact microbiota via fecal microbiota transplants (FMTs). However, t

Multiomics Study of Gut Bacteria and Host Metabolism in Irritable Bowel Syndrome and Depression Patients.

BACKGROUND AND AIMS: Irritable bowel syndrome (IBS) and depression have high tendencies of comorbidity. In particular, diarrhea-predominant IBS (IBS-D) and depression exhibit similar fecal microbiota signatures, yet little is known about their pathogenic mechanism. Here, we propose that the differences in structure and composition of IBS-D and depression gut microbiota give rise to different downstream functions, which lead to distinct clinical phenotypes via host metabolism and further influence the interaction of brain-gut axis. METHODS: We performed multiomics study, including fecal metage

Quercetin Loaded Monolaurate Sugar Esters-Based Niosomes: Sustained Release and Mutual Antioxidant-Hepatoprotective Interplay.

Flavonoids possess different interesting biological properties, including antibacterial, antiviral, anti-inflammatory and antioxidant activities. However, unfortunately, these molecules present different bottlenecks, such as low aqueous solubility, photo and oxidative degradability, high first-pass effect, poor intestinal absorption and, hence, low systemic bioavailability. A variety of delivery systems have been developed to circumvent these drawbacks, and among them, in this work niosomes have been selected to encapsulate the hepatoprotective natural flavonoid quercetin. The aim of this stud

Macrophage-inducible C-type lectin Mincle-expressing dendritic cells contribute to control of splenic Mycobacterium bovis BCG infection in mice.

The macrophage-inducible C-type lectin Mincle has recently been identified to be a pattern recognition receptor sensing mycobacterial infection via recognition of the mycobacterial cell wall component trehalose-6',6-dimycolate (TDM). However, its role in systemic mycobacterial infections has not been examined so far. Mincle-knockout (KO) mice were infected intravenously with Mycobacterium bovis BCG to mimic the systemic spread of mycobacteria under defined experimental conditions. After intravenous infection with M. bovis BCG, Mincle-KO mice responded with significantly higher numbers of mycob

Bio-sensing of cadmium(II) ions using Staphylococcus aureus.

Cadmium, as a hazardous pollutant commonly present in the living environment, represents an important risk to human health due to its undesirable effects (oxidative stress, changes in activities of many enzymes, interactions with biomolecules including DNA and RNA) and consequent potential risk, making its detection very important. New and unique technological and biotechnological approaches for solving this problems are intensely sought. In this study, we used the commonly occurring potential pathogenic microorganism Staphylococcus aureus for the determination of markers which could be used f

Vaccination of BALB/c mice with Escherichia coli-expressed vaccinia virus proteins A27L, B5R, and D8L protects mice from lethal vaccinia virus challenge.

The potential threat of smallpox use in a bioterrorist attack has heightened the need to develop an effective smallpox vaccine for immunization of the general public. Vaccination with the current smallpox vaccine, Dryvax, produces protective immunity but may result in adverse reactions for some vaccinees. A subunit vaccine composed of protective vaccinia virus proteins should avoid the complications arising from live-virus vaccination and thus provide a safer alternative smallpox vaccine. In this study, we assessed the protective efficacy and immunogenicity of a multisubunit vaccine composed o
