alpha(4) integrin-dependent eosinophil recruitment in allergic but not non-allergic inflammation.

1. Although anti-alpha(4) integrin mAbs reduce eosinophil accumulation in several models of allergic inflammation, it is not clear whether this occurs via a direct action to block eosinophil alpha(4) integrins or indirectly on another cell type. The role of alpha(4) integrins on the accumulation of (111)In-labelled eosinophils in allergic and non-allergic inflammation in guinea-pig skin was therefore investigated. 2. Intradermal injection of antigen in sensitized skin sites induced accumulation of (111)In-eosinophils that was reduced up to 70% by two anti-alpha(4) integrin mAbs. In contrast, a

Targeted disruption of the leukotriene B(4) receptor in mice reveals its role in inflammation and platelet-activating factor-induced anaphylaxis.

Leukotrienes are derived from arachidonic acid and serve as mediators of inflammation and immediate hypersensitivity. Leukotriene B(4) (LTB(4)) and leukotriene C(4) (LTC(4)) act through G protein-coupled receptors LTB(4) receptor (BLTR) and Cys-LTR, respectively. To investigate the physiological role of BLTR, we produced mice with a targeted disruption of the BLTR gene. Mice deficient for BLTR (BLTR(-/-)) developed normally and had no apparent hematopoietic abnormalities. Peritoneal neutrophils from BLTR(-/-) mice displayed normal responses to the inflammatory mediators C5a and platelet-activa

Chronic granulomatous disease in an adult male: A proposed X-linked defect.

A 25-year old patient with chronic granulomatous disease of somewhat unusual history is described. The diagnosis of CGD was based on increased susceptibility to infection, granulomatous appearance of tissues, and diminished bactericidal and metabolic response of leukocytes during phagocytosis: the clinical and cellular features considered phenotypic of CGD. A 16-year-old female sibling had bactericidal and metabolic abnormalities of leukocyte function similar to those of the patient's leukocytes. Leukocytes from another sister, 26 years of age, were intermediate in bactericidal capacity. Two p

Metabolism of L-thyroxine by phagocytosing human leukocytes.

Intact normal human leukocytes deiodinated L-thyroxine (T(4)) with the generation of inorganic iodide, chromatographically immobile origin material, and small quantities of L-triiodothyronine (T(3)). When phagocytosis was induced in the leukocytes through the addition of zymosan particles that had been opsonized by coating with plasma, T(4)-deiodination was greatly stimulated. In addition to the stimulation of T(4)-deiodination, the accumulation by the leukocytes of undegraded T(4) was increased. Anoxia, which has previously been shown not to interfere with phagocytosis, did not prevent the in

Interferon gamma applied ex vivo restores function to neutrophils from critically ill patients.

INTRODUCTION: Critically ill patients commonly develop acquired neutrophil dysfunction, which increases susceptibility to intensive care unit-acquired infection (ICU-AI). This study aimed to assess whether interferon gamma (IFN-γ) can restore function in dysfunctional neutrophils from critically ill patients and to uncover potential underlying mechanisms. METHODS: This was an observational cohort study. Neutrophils were isolated from whole blood donated by critically ill patients (n=31) in four separate teaching hospital intensive care units (ICUs). Neutrophils were subsequently treated with

An A549 Cell-Based Approach Using Repeated Fluorescence Readouts for Assessing Reactive Oxygen Species Activity of Atmospheric Particulate Matter.

Exposure to atmospheric particulate matter (PM) is a major public-health concern, in part because PM can perturb cellular redox homeostasis. This study evaluates an in vitro A549/DCFH-DA approach using repeated fluorescence readouts to assess PM2.5-induced oxidative activity. Untreated and assay-specific controls were combined with zymosan and NIST Standard Reference Material® 2584 suspended in PBS, and fluorescence was monitored at multiple readout times over a 15 min-6 h window. Method performance was characterized using the coefficient of variation (CV) and signal-to-noise ratio (SNR). A de

Baicalin ameliorates podocyte injury and renal function impairment in idiopathic membranous nephropathy by inhibiting the AGE/RAGE signaling.

Idiopathic membranous nephropathy (IMN) is an autoimmune glomerular disease. Current immunosuppressive therapies often cause side effects and high recurrence rates, highlighting the need for new treatments. Baicalin (BAI), a flavonoid isolated from Scutellaria baicalensis, has anti-inflammatory and antioxidant properties. This study examined effects of BAI on IMN using in vitro mouse podocyte clone-5 (MPC-5) cells treated with zymosan-activated serum (ZAS) and in vivo mouse models induced with cationic bovine serum albumin (BSA). Treatments included the BAI and advanced glycation end products/

Differential Regulation of Granule Release in Platelets and Immune Cells.

BACKGROUND: Platelets and certain immune cells contain releasable granules that play essential roles in hemostasis and immune regulation. However, the regulatory mechanisms governing degranulation of platelets versus immune cells remain incompletely understood. OBJECTIVES: This study aimed to elucidate the distinct regulatory mechanisms underlying granule release in platelets and immune cells. METHODS: Mouse platelets were freshly isolated from peripheral blood, neutrophils were purified from bone marrow, and mast cells were generated from bone marrow progenitors by in vitro differentiation.

Identification of Arginine-Vasopressin Receptor 1a (Avpr1a/Avpr1a) as a Novel Candidate Gene for Chronic Visceral Pain Sheds Light on the Potential Role of Enteric Neurons in the Development of Visceral Hypersensitivity.

Chronic abdominal pain in the absence of ongoing disease is the hallmark of disorders of gut-brain interaction (DGBIs), including irritable bowel syndrome (IBS). While the etiology of DGBIs remains poorly understood, there is evidence that both genetic and environmental factors play a role. In this study, we report the identification and validation of arginine-vasopressin receptor 1A (Avpr1a) as a novel candidate gene for visceral hypersensitivity (VH), a primary peripheral mechanism underlying abdominal pain in DGBI/IBS. Comparing 2 C57BL/6 (BL/6) substrains (C57BL/6NTac and C57BL/6J) reveale

Shifting the Biosynthesis of Leukotrienes Toward Specialized Pro-Resolving Mediators by the 5-Lipoxygenase-Activating Protein (FLAP) Antagonist BRP-201.

BACKGROUND AND PURPOSE: Lipid mediators (LM) play crucial roles in the complex inflammation process with respect to initiation, maintenance, and resolution. Proinflammatory leukotrienes (LTs), generated by 5-lipoxygenase (LOX) and the 5-LOX-activating protein (FLAP), initiate and maintain inflammation while specialized pro-resolving mediators (SPMs) formed by various LOXs as key enzymes promote inflammation resolution and the return to homeostasis. Since 5-LOX also contributes to SPM biosynthesis, smart pharmacological manipulation of the 5-LOX pathway and accompanied activation of 12-/15-LOXs

Platelets mediate trained immunity against bone and joint infections in a mouse model.

AIMS: Trained immunity confers non-specific protection against various types of infectious diseases, including bone and joint infection. Platelets are active participants in the immune response to pathogens and foreign substances, but their role in trained immunity remains elusive. METHODS: We first trained the innate immune system of C57BL/6 mice via intravenous injection of two toll-like receptor agonists (zymosan and lipopolysaccharide). Two, four, and eight weeks later, we isolated platelets from immunity-trained and control mice, and then assessed whether immunity training altered platel

Immune adherence in renal glomeruli. Complement receptor sites on glomerular capillary epithelial cells.

Several very recent reports have indicated the presence of receptor sites for the third component of complement in human but not other vertebrate renal glomeruli. The present study constitutes a demonstration that the glomerular capillary epithelial cell bears this receptor, detectable with either EAC complexes (EAC1423b) or fluores ceinated zymosan-C3 (ZC3b) complexes, Fresh, unfixed frozen sections of normal or diseased human kidneys, mechanically isolated human glomeruli, dissociated glomerular cells, and glomeruli and golmerular cells maintained in tissue culture were examined with various

Alleviation of experimental arthritis in SKG mice through Nr4a1 agonization.

INTRODUCTION: Rheumatoid arthritis (RA), a chronic autoimmune disease, is characterized by CD4+ T cell-mediated synovial inflammation, with T helper (Th)17 cells being implicated in RA pathogenesis. Nr4a1 is an orphan nuclear receptor functioning as a negative regulator of T cell activation and central tolerance. Cytosporone B (CsnB) is a small-molecule agonist of Nr4a1 and can exert immunomodulatory effects. However, its efficacy in T cell-driven autoimmune arthritis remains unclear. This study aimed to investigate the therapeutic effect of CsnB-mediated Nr4a1 agonization on RA development in

Innate immune recognition of Bartonella bacilliformis: TLR2/TLR4 expression and cytokine modulation.

BackgroundConsider this Carrion's disease (CD) is a biphasic illness-comprising acute and chronic phases-endemic to Peru and caused by Bartonella bacilliformis, a bacterium transmitted by sandflies. Despite its clinical relevance, the mechanisms underlying innate immune activation in response to B. bacilliformis remain poorly understood. Toll-like receptors (TLRs) play a central role in recognizing conserved molecular patterns present in pathogens, thereby initiating innate immune responses. The present study aimed to describe the expression patterns of TLR2 and TLR4, along with cytokine secre

MEK1/2 inhibition decreases pro-inflammatory responses in macrophages from people with cystic fibrosis and mitigates severity of illness in experimental murine methicillin-resistant Staphylococcus aureus infection.

Chronic pulmonary bacterial infections and associated inflammation remain a cause of morbidity and mortality in people with cystic fibrosis (PwCF) despite new modulator therapies. Therapies targeting host factors that dampen detrimental inflammation without suppressing immune responses critical for controlling infections remain limited, while the development of lung infections caused by antimicrobial resistant bacteria is an increasing global problem, and a significant challenge in CF. Pharmacological compounds targeting the mammalian MAPK proteins MEK1 and MEK2, referred to as MEK1/2 inhibito
